[Neuropathology of non conventional infectious agents or prions].

The neuropathological diagnosis of infections by non conventional agents relies on four lesions: astrocytic gliosis (cell hypertrophy and proliferation) usually contrasting with absent mononuclear cell infiltrates (lymphocytes, monocytes-macrophages, and/or microglia) revealed by conventional techniques, and neuronal loss in the most affected areas are little specific findings. Amyloid plaques that are inconstantly found, and spongiosis of gray matter, a characteristic and very frequent finding, are most specific. PrP immunohistochemistry brings additional data. The main diagnostic difficultie

Ultrastructural pathology of human T-cell lymphotropic virus type I encephalomyelopathy in a white patient with adult T-cell leukemia/lymphoma.

A white patient with human T-cell lymphotropic virus type I (HTLV-I)-associated adult T-cell leukemia/lymphoma for 10 years died 4 months after the onset of spastic myelopathy. Ultrastructurally, the neuropathologic findings consisted of dystrophic neurites (spheroids) filled with neurofilaments or electron-dense bodies, intense astrocytic reaction with abundant formation of corpora amylacea, and multilamellar bodies. Demyelination and spongiform change were absent. Viruslike particles resembling HTLV-I were detected adjacent to brain endothelial cells.

[Effect of morphine dependence on the hypothalamus of the mother and offspring during postnatal ontogenesis (experimental study)].

Electron microscopy was used to study the time-course of changes in the neuroglia of the hypothalamus. The offspring manifested deceleration of the time of glia maturation with partial destruction of the protein-synthesizing and oxidative-energy structures as well as more frequent occurrence of satellitosis. The mothers and offspring demonstrated membranous bodies in the nucleus and cytoplasm of astrocytes, pronounced osmiophilia of the nucleus of oligodendrocytes accompanied by the impairment of myelination in the function of the latter ones (demyelinization) and axon degeneration by the dark…

Reevaluation of the ultrastructural pathology of experimental Creutzfeldt-Jakob disease. Serial studies of the Fujisaki strain of Creutzfeldt-Jakob disease virus in mice.

We describe the serial ultrastructural pathology of experimental Creutzfeldt-Jakob disease (CJD) in mice. Spongiform vacuoles, widespread myelin and axonal pathology, accompanied by abundant macrophagic reaction and neuroaxonal dystrophy, were consistently found in mice infected with CJD virus. By contrast, intranuclear vacuolation and swelling of astrocytic and neuronal processes were nonspecific changes that also occurred in control animals. We conclude that CJD-related neuropathological phenomena do not accumulate gradually through the incubation period but develop relatively abruptly and i

Malignant gliomas occurring in identical twins.

The occurrence of malignant gliomas in a pair of identical twins is reported. One, in the posterior fossa, presented in the third year of life, the other, in the left cerebral hemisphere, in the eighth. Both ran a rapidly progressive course and died within a few months of the onset of symptoms. The histological structure of the gliomas was similar and both tumours consisted of astrocytic and oligodendroglial parts. Review of the literature revealed only two pairs of histologically proven gliomas occurring in identical twins. The number of cases reported is too small for any valid conclusions o

Neurotropism and Therapeutic Targeting of Brain Metastases in Small Cell Lung Cancer.

Small cell lung cancer (SCLC) is an aggressive malignancy marked by rapid progression, early dissemination, and a pronounced propensity for brain metastases (BM), which develop in up to 80% of patients. SCLC is defined by profound genomic instability, lineage plasticity, and rapid drug resistance. The establishment of BM is promoted by neuronal mimicry, enhanced intercellular adhesion, and dynamic cross-talk with astrocytes and microglia. Emerging therapies targeting delta-like ligand 3 and B7H3 have demonstrated encouraging intracranial activity. Despite these advances, treatment resistance a…

Neuromyelitis Optica: A Deceiving Disorder of Demyelination.

Neuromyelitis optica (NMO), also known as Devic syndrome, is an autoimmune inflammatory and demyelinating disorder that affects the optic nerves and spinal cord. It is believed to be attributed to aquaporin-4 antibodies, a water channel expressed on astrocytes. It commonly presents with isolated or recurrent attacks of myelitis and optic neuritis. Intractable vomiting and hiccups may also be seen as symptoms. Acute treatment is often achieved with high-dose steroids and is imperative to prevent permanent central nervous system damage. Relapse prevention is achieved using long-term immunosuppre…

The Role of Inflammation in Tinnitus: A Systematic Review and Meta-Analysis.

Subjective tinnitus is the perception of sound without the presence of an external source. Increasing evidence suggests that tinnitus is associated with inflammation. In this systematic review, the role of inflammation in subjective tinnitus was studied. Nine animal and twenty human studies reporting inflammatory markers in both humans and animals with tinnitus were included. It was established that TNF-α and IL-1β are increased in tinnitus, and that microglia and astrocytes are activated as well. Moreover, platelet activation may also play a role in tinnitus. In addition, we elaborate on mech…

Immune Mediators of Pathology in Neurobrucellosis: From Blood to Central Nervous System.

Neurobrucellosis, which is the most morbid form of brucellosis disease, presents with inflammatory signs and symptoms. Recent experimental evidence clearly indicates that deregulation of astrocytes and microglia caused by Brucella infection creates a microenvironment in the central nervous system (CNS) in which secretion of pro-inflammatory mediators lead to destabilization of the glial structure, the damage of the blood brain barrier (BBB) and neuronal demise. This review of Brucella interactions with cells of the CNS and the BBB is intended to present recent immunological findings that can e…

MeCP2 in central nervous system glial cells: current updates.

avMethyl‑CpG binding protein 2 (MeCP2) is an epigenetic regulator, which preferentially binds to methylated CpG dinucleotides in DNA. MeCP2 mutations have been linked to Rett syndrome, a neurodevelopmental disorder characterized by severe intellectual disability in females. Earlier studies indicated that loss of MeCP2 function in neuronal cells was the sole cause of Rett syndrome. Subsequent studies have linked MeCP2 expression in CNS glial cells to Rett syndrome pathogenesis. In this review, we have discussed the role of MeCP2 in glial subtypes, astrocytes, oligodendrocytes and microglia, and…

Lactulose enhances neuroplasticity to improve cognitive function in early hepatic encephalopathy.

Lactulose is known to improve cognitive function in patients with early hepatic encephalopathy; however, the underlying mechanism remains poorly understood. In the present study, we investigated the behavioral and neurochemical effects of lactulose in a rat model of early hepatic encephalopathy induced by carbon tetrachloride. Immunohistochemistry showed that lactulose treatment promoted neurogenesis and increased the number of neurons and astrocytes in the hippocampus. Moreover, lactulose-treated rats showed shorter escape latencies than model rats in the Morris water maze, indicating that la…

Role of Wnt1 and Fzd1 in the spinal cord pathogenesis of amyotrophic lateral sclerosis-transgenic mice.

Amyotrophic lateral sclerosis (ALS) is a fatal neurodegenerative disease characterized by chronic progressive degeneration of motor neurons resulting in muscular atrophy, paralysis, and ultimately death. We have investigated the expression of Wnt1 and Fzd1 in the spinal cords of SOD1G93A ALS transgenic mice, SOD1G93A-transfected N2a cells, and primary cultured astrocytes from SOD1G93A transgenic mice. In addition, we provided further insight into the role of Wnt1 and Fzd1 in the pathogenesis of ALS transgenic mice and discuss the mechanisms underlying the Wnt signal pathway which may be useful…

Endoscopic treatment of intranasal glioma in an infant presenting with dyspnea.

Nasal gliomas are uncommon congenital lesions arising from abnormal embryonic development. Clinically, these masses are firm and uncompressible. Histologically, they are made up of astrocytes and neuroglial cells, embedded in fibrous and vascular connective tissue. Proper management of a nasal glioma requires a multidisciplinary approach including an otorhinolaryngologist, radiologist, and neurosurgeon. Radiological investigations such as computed tomography or magnetic resonance imaging should be performed to exclude intracranial extension. The mainstay of treatment is complete surgical extir…

Production and characterization of immortal human neural stem cell line with multipotent differentiation property.

We document the protocols and methods for the production of immortalized cell lines of human neural stem cells from the human fetal central nervous system (CNS) cells by using a retroviral vector encoding v-myc oncogene. One of the human neural stem cell lines (HB1.F3) was found to express nestin and other specific markers for human neural stem cells, giving rise to three fundamental cell types of the CNS: neurons, astrocytes, and oligodendrocytes. After transplantation into the brain of mouse model of stroke, implanted human neural stem cells were observed to migrate extensively from the site…

Neuronal injury regulates fractalkine: relevance for HIV-1 associated dementia.

Fractalkine (FKN), a chemokine highly expressed in the central nervous system, participates in inflammatory responses operative in many brain disorders including HIV-1 associated dementia (HAD). In this report, HIV-1 progeny virions and pro-inflammatory products led to FKN production associated with neuronal injury and apoptosis. FKN was produced by neurons and astrocytes; but differentially produced by the two cell types. Laboratory tests paralleled those in infected people where cerebrospinal fluid FKN levels in HIV-1 infected cognitively impaired (n=16) patients were found to be increased w…

[Subacute sclerosing panencephalitis (SSPE)].

Subacute sclerosing panencephalitis is a rare progressive neurological disorder of childhood and early adolescence caused by persistent measles virus. The diagnosis is based upon characteristic clinical manifestations, periodic EEG discharges, raised antibody titre against measles/SSPE in the plasma and cerebrospinal fluid and increase of gamma-globulins in the cerebrospinal fluid. Histopathological examination shows neuronal loss, astrogliosis, demyelination, infiltration of inflammatory cells, and intranuclear inclusions in neurons, oligodendrocytes and astrocytes. In most cases nucleocapsid…

Genetics and brain injury: apolipoprotein E.

Apolipoprotein E (apo E) is a lipoprotein produced by astrocytes and microglia and has a proposed role in transporting lipids to injured neurons. There are three known isoforms of apo E, coded for by the APOE epsilon2, APOE epsilon3, and APOE epsilon4 genes. The APOE epsilon4 genotype has been implicated as a risk factor for Alzheimer's disease. Recent studies have suggested that APOE epsilon4 may influence the central nervous system's response to injury. This article presents an overview of the relationship between apo E, Alzheimer's disease, and head injury and reviews recent studies implica…
