An unexpected role for cyclin E-CDK2 in regulating responses to cancer immunotherapy.

In this issue of Molecular Cell, Chu et al.1 describe that cancers with increased cyclin E-CDK2 activity suppress responses to immunotherapy through phosphorylation of BRD4, resulting in downregulation of interferon-stimulated genes. Their findings suggest approaches to enhance immunotherapy responses.

Decoding the functional landscape of long non-coding RNAs in hepatocellular carcinoma: molecular mechanisms, clinical implications, and therapeutic prospects.

Hepatocellular carcinoma (HCC) remains the third leading cause of cancer-related mortality worldwide, with limited treatment efficacy and poor prognosis, particularly in advanced-stage disease. Despite progress in diagnostic imaging and systemic therapies, early detection and effective targeted interventions remain major clinical challenges. Long non-coding RNAs (lncRNAs), a class of regulatory RNA molecules exceeding 200 nucleotides in length, have emerged as essential regulators of oncogenic signaling, metabolism, and tumor microenvironment in HCC. This review provides a comprehensive overvi

REST and RASSF1A Tumor Suppressor Genes in Peripheral Blood: Potential Molecular Markers in Breast Cancer.

Tumor suppressor genes, such as RASSF1A and REST, play a central role in regulating cell proliferation. RASSF1A is frequently inactivated in various cancers, being associated with poor prognosis and metastasis. REST loss promotes the activation of genes related to invasion and estrogen sensitivity. We aimed to evaluate the expression of REST and RASSF1A in peripheral blood of breast cancer patients at different treatment stages and to associate the results with clinical and laboratory variables. Peripheral blood samples from breast cancer patients were collected at diagnosis and at 3 and 6 mon

A polysaccharide from Arthrospira platensis alleviates pancreatic cancerassociated with negatively regulating galectin-3 and glypican-6 expression.

Pancreatic cancer, specifically pancreatic ductal adenocarcinoma (PDAC), ranks among the most prevalent malignancies and is a leading cause of cancer-related mortality worldwide. Therefore, there is an urgent need to identify novel anti-pancreatic cancer agents. This study reports a newly identified homogeneous polysaccharide, designated ESPPW, isolated from Arthrospira platensis (A. platensis). The molecular weight of ESPPW is estimated at 356 kDa, and it consists predominantly of glucose and rhamnose, with minor amounts of mannose, glucuronic acid, galacturonic acid, galactose, xylose, arabi

Metabolic targeting of pantothenate by medium chain fats in glioblastoma cells.

Ketogenic diets are commonly used for epilepsy therapy and are increasingly being considered in cancer treatment, where metabolic reprogramming triggered by glucose restriction and ketosis alters energy provision as the therapeutic mechanism. However, a recently developed medium chain triglyceride product, with a high decanoic acid (DA) to octanoic acid (OA) ratio (DA: OA) has enabled a much less restrictive diet and is clinically effective as an epilepsy treatment, yet no studies have explored the product in cancer models. Here we investigate metabolic mechanisms of DA, OA and DA: OA using a

Amoeboid-mesenchymal transition and the proteolytic control of cancer invasion plasticity.

Invasion plasticity allows malignant cells to toggle between collective, mesenchymal, and amoeboid phenotypes while traversing extracellular matrix (ECM) barriers. Current dogma holds that collective and mesenchymal invasion programs trigger the mobilization of proteinases that digest structural barriers dominated by type I collagen, while amoeboid activity allows cancer cells to marshal mechanical forces to traverse tissues independently of ECM proteolysis. Here, we use cancer spheroid-3-dimensional matrix models, single-cell RNA sequencing, and human tissue explants to identify the mechanism

The compartment-specific interactome of VDAC1 reveals its emerging role beyond mitochondria in breast cancer.

Voltage-dependent anion channel 1 (VDAC1) is a multifunctional protein that is primarily localized in the outer mitochondrial membrane, where it regulates metabolite flux and apoptosis. Aberrant expression of VDAC1 has been implicated in tumorigenesis. Moreover, recent reports suggested that VDAC1 is localized at the plasma membrane, but its role in this compartment remains unclear. This study aimed to evaluate the relationships between VDAC1 expression and clinicopathological parameters in breast cancer and to map its interaction networks in the mitochondrial and plasma membrane compartments.

A non-complement role for C1R rewires integrin and death-receptor signaling to drive renal cancer metastasis.

BACKGROUND: Metastasis is the leading cause of death in clear cell renal cell carcinoma (ccRCC) patients. Anoikis, a form of programmed cell death induced by the loss of cell-extracellular matrix interactions, is a critical factor in hindering metastasis. Nevertheless, the regulatory mechanisms underlying anoikis resistance in ccRCC remain poorly characterized and warrant further investigation. METHODS: We created a single-cell transcriptomic atlas of ccRCC metastasis and used multi-omics data to identify the key role of complement C1R during metastasis. Anoikis-related cell experiments and m

Zebrafish xenografts reveal line-specific patterns of growth, survival, and metastases in canine mammary cell lines: CMT-U27, CMT-U131, and CMT-U229.

Canine mammary tumours (CMTs) are the most common malignancies in female dogs and represent a valuable comparative model for human breast cancer. This study aimed to characterise the molecular and functional profiles of three CMT cell lines: CMT-U27, CMT-U131 and CMT-U229, using a zebrafish xenograft model. Gene ex-pression analysis targeted proliferation markers (MKI67, CCNB1, MYBL2, BIRC5, AURKA), receptor signalling (ESR1, PGR, AR, ERBB2, GRB7, EGFR), apoptosis and survival (BCL2, BAG1), and angiogenesis (VEGFA). CEP72 promotes malignant progression and shapes the tumor immune microenvironment in hepatocellular carcinoma: integrated multi-cohort bioinformatics and in vitro validation.

Immune checkpoint inhibitors (ICIs) benefit only a subset of patients with hepatocellular carcinoma (HCC), and biomarkers linking ICI response with tumor biology remain limited. We investigated CEP72 as an ICI-response–associated gene and evaluated its prognostic, immune, and functional relevance in HCC. Anti–PD-1 response–associated differentially expressed genes (DEGs) were identified in GSE35640 and screened for prognostic relevance in TCGA-LIHC using univariate Cox models for overall survival (OS), disease-specific survival (DSS), and progression-free interval (PFI). Mediator subunit MED4 enforces metastatic dormancy in breast cancer.

Long-term survival in breast cancer is often limited by metastatic recurrence arising from disseminated cancer cells that persist in a dormant state. The mechanisms that enable these dormant cells to survive and subsequently reawaken remain incompletely understood. Here an unbiased genome-scale genetic screen identified Med4 as a cancer cell-intrinsic gatekeeper in metastatic reactivation. Correspondingly, MED4 haploinsufficiency was found to be prevalent in metastatic breast cancer and associated with poorer clinical outcomes. Syngeneic mouse metastasis models revealed that MED4 enforces meta

Molecular insights into DDX3X-androgen receptor mRNA regulation via non-canonical G-quadruplex in castration-resistant prostate cancer.

Prostate cancer (PC) is one of the most common malignancies in men, and the emergence of androgen receptor-low/negative castration-resistant PC (ARL/- CRPC) following androgen receptor signaling inhibitor (ARSI) therapy remains a critical clinical challenge. The RNA-binding protein DEAD-box helicase 3 X-linked (DDX3X) has been implicated in the translational regulation of androgen receptor (AR) mRNA; however, the underlying binding mechanisms are not well defined. Here, we show that DDX3X colocalizes with AR mRNA in ARL/- CRPC cells and selectively recognizes non-canonical RNA G-quadruplex (rG

VprBP drives prostate tumorigenesis by its kinase activity targeting histone H2A.

VprBP has been recently identified as an oncogenic kinase and a promising drug target in human malignant tumors. Although VprBP can phosphorylate histone H2A and some non-histone proteins, it seems to selectively target specific substrates in a cancer type-dependent manner by an unknown mechanism. Here we report that VprBP is highly expressed in prostate cancer cells and inactivates a group of genes encoding critical regulators of cell growth and proliferation in a manner dependent on its kinase activity toward H2AT120. As an extension of our previous finding of VprBP inhibitor B32B3, we also

LD-associated signatures identified by perilipin-based proximity labeling proteomics reveal GNA14 as a therapeutic and prognostic target in renal cell carcinoma.

Renal cell carcinoma (RCC), especially clear cell RCC (ccRCC), is characterized by metabolic reprogramming, notably disordered lipid metabolism and prominent intracellular lipid droplet accumulation. In addition to abnormal triglyceride and cholesterol ester storage, lipid droplets promote tumor proliferation, survival, and drug resistance by supplying energy, membrane components, and signaling platforms, thereby representing potential therapeutic targets. We integrated TMU-RNAseq and TCGA-KIRC datasets and performed proximity-labeling mass spectrometry targeting PLIN2/3 to identify perilipin-

Double-negative prostate cancer: Evolutionary mechanisms, microenvironmental remodeling, and clinical translation.

The management of metastatic castration-resistant prostate cancer (mCRPC) is continuously challenged by the emergence of therapeutic resistance. Under the selective pressure of profound androgen inhibition, a significant proportion of tumors undergo lineage plasticity, evolving into double-negative prostate cancer (DNPC). Defined by the concomitant loss of androgen receptor (AR) dependence and the absence of classic neuroendocrine markers, DNPC represents a highly aggressive, independent evolutionary steady state. The fundamental drivers of this phenotypic transition include the concurrent fun

CD47 as a prognostic biomarker and potential immunotherapy target in penile squamous cell carcinoma.

Penile squamous cell carcinoma (PSCC) remains a global healthcare concern, characterized by poor outcomes and limited therapeutic options. CD47, a “don’t-eat-me” signal that enables tumor immune evasion, has not been extensively evaluated in PSCC. This study aims to explore the expression and significance of CD47 in PSCC. RNA-sequencing was performed on eight paired PSCC and adjacent normal tissues. Protein expression was assessed by immunohistochemistry (IHC) in an independent of 131 PSCC cases, and associations with clinicopathological factors and prognosis was evaluated. In addition, the in

[MiR-21 targets PDCD4 to regulate proliferation, apoptosis, and invasion of osteosarcoma MG63 cells].

Objective MicroRNAs (miRNAs) are small endogenous single-stranded non-coding RNAs, approximately 20-25 nucleotides in length, that regulate the expression of potential target mRNAs at the post-transcriptional level. miRNAs are abnormally expressed in many malignant tumors, and are closely associated with tumorigenesis and development, functioning either as proto-oncogenes or oncogenes. Studies have revealed that microRNA-21 (miR-21) is one of the most common oncogenes across various cancer types. However, the function of miR-21 in osteosarcoma has not been fully elucidated. This study aims to
