Presence of double-stranded RNAs in human and calf isolates of Cryptosporidium parvum.

We examined the occurrence of 2 virus-like double-stranded (ds)RNAs in human and calf isolates of Cryptosporidium parvum senso latu and othe … widespread distribution of the dsRNAs, the similarity of these molecules between isolates, and high host specificity, these nucleic acids may prove to represent species-specific molecular markers for C. parvum. Evidence also suggests that the dsRNA can be utilized for molecular genotyping of C. parvum.

Phylogenetic analysis of Cryptosporidium parasites based on the small-subunit rRNA gene locus.

Biological data support the hypothesis that there are multiple species in the genus Cryptosporidium, but a recent analysis of the available … C. muris, C. baileyi, and C. serpentis, which is consistent with the biological characteristics and host specificity data. The Cryptosporidium species formed two clades, with C. parvum and C. baileyi belonging to one clade and C. muris and C. serpentis belonging to the other clade. Within C. parvum, human genotype isolates and guinea pig isolates (known as Cryptos …

HLA-target antigens and T-cell receptor diversity of activated T cells invading the skin during acute graft-versus-host disease.

To study the repertoire and specificity of T lymphocytes infiltrating skin lesions during graft-versus-host disease (GVHD), we performed an exhaustive molecular and functional analysis of 146 T-cell clones derived from the skin of three patients undergoing an acute GVHD after allogeneic bone marrow transplantation (BMT) from HLA-mismatched related donors. Analysis of T-cell receptor (TCR) rearrangement and TCR chain junctional sequences demonstrated the presence of 11 distinct clones among the 64 derived from patient UPN1, six among the 58 derived from patient UPN2, and seven among the 24 deri

Trophozoite-to-cyst ratio increases during recovery from Pneumocystis carinii pneumonia in rats.

Pneumocystis carinii has two developmental stages: the trophozoite and the cyst. The differential specificity of monoclonal antibodies SMoAb (cysts only) and CMoAb (cysts and trophozoites) was used to analyse the trophozoite-to-cyst (T: C) ratio during recovery from P. carinii pneumonia (PCP) in a dexamethasone model in Wistar rats. In tissue homogenates the cysts decreased from week 0 to week 4 (4.10 +/- 1.57 10(6)/g vs 0.069 +/- 0.021 10(6)/g, p < 0.001), whereas the trophozoites decreased much less, leading to a change in T: C ratio from week 0 and 1 (72 +/- 33 and 49 +/- 14, respectively)

Heat shock proteins and molecular chaperones: implications for adaptive responses in the skin.

Recent advances in the biology of heat-shock proteins (hsps) are reviewed. These abundant and evolutionarily highly conserved proteins (also called stress proteins) act as molecular escorts. Hsps bind to other cellular proteins, help them to fold into their correct secondary structures, and prevent misfolding and aggregation during stress. Cytoplasmic hsp70 and hsp60 participate in complicated protein-folding pathways during the synthesis of new polypeptides. Close relatives of hsp70 and hsp60 assist in the transport and assembly of proteins inside intracellular organelles. Hsp90 may have a un

The role of neutrophil elastase in chronic inflammation.

Passively released or actively secreted elastase from neutrophils has been linked to the pathologic processes of a variety of inflammatory diseases, including idiopathic pulmonary fibrosis, rheumatoid arthritis, adult respiratory distress syndrome, and cystic fibrosis. The serine proteinase has a broad substrate specificity and may attack a number of host proteins outside of the neutrophil, including lung elastin and fibronectin. Such a proteolysis may change the normal surrounding tissue and the protein pattern of an inflammatory focus. Additionally, it acts as a potent secretagogue in minute

The detection of Aspergillus spp. by the polymerase chain reaction and its evaluation in bronchoalveolar lavage fluid.

Invasive pulmonary aspergillosis (IPA) is an important cause of mortality and morbidity in the immunocompromised host. However, the diagnosis of this condition may be difficult, and it is sometimes missed because of the lack of sensitivity of available tests. Therefore, we used polymerase chain reaction (PCR)-based amplification of fragments of genes-encoding alkaline proteases from Aspergillus fumigatus and A. flavus to detect these organisms in bronchoalveolar lavage fluid specimens. The predicted size of the product (747 base pairs) after amplification of A. fumigatus was larger than that f

Parasite enzymes as a tool to investigate immune responses.

Previous evidences reported by us and by other authors revealed the presence of IgG in sera of Schistosoma mansoni-infected patients to immunodominant antigens which are enzymes. Besides their immunological interest as possible inductors of protection, several of these enzyme antigens might be also interesting markers of infection in antibody-detecting immunocapture assays which use the intrinsic catalytic property of these antigens. It was thus thought important to define some enzymatic and immunological characteristics of these molecules to better exploit their use as antigens. Four differen

Identification of macrophage cell-surface binding sites for cationized bovine serum albumin.

Autoimmune diseases are characterized by the presence of autoantibodies often restricted to host proteins exhibiting charge rich domains. Charged polypeptides elicit strong immune responses, and cationized bovine serum albumin and other cationic proteins are significantly more immunogenic than their less charged counterparts. These phenomena may involve enhanced protein uptake by macrophages, resulting in greater processing and presentation of antigenic peptide-MHC complexes to T-cells. We compared macrophage cell-surface binding and uptake of native and cationized bovine serum albumin. Specif

Histochemical localization of gene expression in Onchocerca volvulus: in situ DNA histohybridization and immunocytochemistry.

We report here the development of in situ hybridization and immunohistochemistry protocols which permit the histological identification of gene expression of a cloned antigen of Onchocerca volvulus, OI5, in the parasite. Skin nodules containing female adult worms were fixed in a modified Carnoy's fixative and embedded in paraffin. Histological staining of tissue sections revealed uniformly excellent morphology and RNA preservation. To localize mRNA by in situ hybridization, tissue sections were incubated with biotin-labeled pOI5, the plasmid containing the genomic sequence of the antigen, and

Pathogenesis of experimental lupus nephritis: a role for anti-basement membrane and anti-tubular brush border antibodies in murine chronic graft-versus-host disease.

The pathogenesis of renal involvement was studied in murine chronic graft-versus-host disease (GVHD), which is a model for human systemic lupus erythematosus. GVHD was induced by four i.v. injections of lymphocytes from DBA/2 donor mice into (C57BL/10 x DBA/2)F1 hybrids at 3-4-day intervals. Two weeks after the first injection, antibodies were found to have been deposited in the mesangium and along the glomerular basement membrane (GBM) in a linear arrangement, which changed to a granular pattern after 6-8 weeks. In this stage, large electron-dense complexes were present both subepithelially a

Cardiology across continents, case series session #15: complicated post-op course in an infant with Tetralogy of Fallot.

Clinical experience during fellowship training and post-graduation depends on training length, resources, and time at specific institutions, contributing to knowledge deficits. Institutional collaboration overcomes such barriers wherein case discussions share knowledge and management strategy, enhancing patient outcomes. Our discussion of the management of Tetralogy of Fallot highlights key insights between programmes differing in geography and resources, hosted by Heart University.

Pathogen-defending deubiquitinase possesses distinct specificity towards K6-polyubiquitination.

The bacterial pathogen Legionella pneumophila encodes numerous effectors to manipulate host ubiquitin signaling. Recently, Warren et al. revealed the structural basis of K6-polyubiquitination recognition by Legionella deubiquitinase LotA, while validating its potential as an enzymatic tool to study linkage-specific ubiquitination. During Legionella infection, LotA counteracts valosin-containing protein (VCP) recruitment to the Legionella-containing vacuole.

Subcutaneous dematiaceous fungal infection.

Subcutaneous dematiaceous fungal infections, which include chromoblastomycosis and phaeohyphomycosis, are a heterogeneous group of clinical entities that are caused by dematiaceous or pigmented fungi found in soil. These infections have a wide spectrum of clinical presentations that depend largely on the specific causative organism and on the integrity of the host's immune response. Treatment is challenging and involves a highly individualized plan that often combines both surgical and long-term medical treatment.

Type IV secretion and Brucella virulence.

The type IV secretion system, encoded by the virB region, is a key virulence factor for Brucella. The 12 genes of the region form an operon that is specifically induced by phagosome acidification in cells after phagocytosis. We speculate that the system serves to secrete unknown effector molecules, which allow Brucella to pervert the host cell endosomal pathways and to create a novel intracellular compartment in which it can replicate.

[Fulminant erysipelas].

An apparent resurgence of invasive and life-threatening group A streptococcal infections in young and previously healthy hosts is reported in several countries. We report a case of fatal necrotizing fasciitis and review the spectrum of these clinical syndromes, including the invasive soft tissue infections. This changing pattern of infection could be explained by an increased incidence of some serotypes of group A Streptococcus carrying specific virulence factors, such as pyrogenic exotoxins.

Histopathology in hosts parasitized by Sarcoptes scabiei.

Histopathologic evaluation of nondermal tissue in rabbits infested with Sarcoptes scabiei var. canis was investigated. Severe infestation resulted in deviant serological and serum biochemical values. Histological study revealed structural changes in the tissues of specific organs. The most prominent histological finding was amyloidosis in the liver, glomerulus of the kidney, red pulp of the spleen, intestines, and tongue. Hosts treated for infestation exhibited no abnormal organ histology.

[Impact of stress on the mononuclear phagocytic system and hepatic parenchymal structure in vaccine-induced granulomatous inflammation].

Stress substantially modulates the mononuclear phagocytic system (its center and periphery), the time of changes in granuloma formation in animals infected with BCG vaccine. This determines the specific features of development of vaccine-induced granulomatous inflammation as a reflection of changes in the host's resistance system and possibly mycobacteria. An additional influence (to Mycobacteria tuberculosis) of stresses during 30 days enhances destructive processes in the hepatic parenchyma.

Recent advances in cryptosporidiosis: the immune response.

An increased understanding of host immune responses to Cryptosporidium parvum which are responsible for clearance of primary infection and resistance to reinfection, and characterization of the parasite molecules to which they are directed, are essential for discovery of effective active and passive immunization strategies against cryptosporidiosis. In this article, recent advances in knowledge of humoral and cellular immune responses to C. parvum, their antigen specificities, and mechanisms of protection are briefly reviewed.

Ineffective airway clearance related to airway infection.

Clearance of secretions is a common problem in patients who experience acute respiratory tract infections. Although the offending agent is a major factor, the degree of severity of ineffective airway clearance depends on many other factors: pulmonary host defenses, presence of underlying pulmonary disease, age, lifestyle habits, environments, airway clearance self-management skills, and time of specific treatment.
