ASK1/2 signaling promotes inflammation in a mouse model of neutrophilic dermatosis.

Mice homozygous for the Tyr208Asn amino acid substitution in the carboxy terminus of Src homology region 2 (SH2) domain-containing phosphatase 1 (SHP-1) (referred to as Ptpn6spin mice) spontaneously develop a severe inflammatory disease resembling neutrophilic dermatosis in humans. Disease in Ptpn6spin mice is characterized by persistent footpad swelling and suppurative inflammation. Recently, in addition to IL-1α and IL-1R signaling, we demonstrated a pivotal role for several kinases such as SYK, RIPK1, and TAK1 in promoting inflammatory disease in Ptpn6spin mice. In order to identify new kin

Total flavonoids from Ficus virens Aiton leaves alleviate liver injury and protect against lipopolysaccharide-Induced sepsis in mice.

Sepsis is a life-threatening organ dysfunction from dysregulated host response to infection, causing high global mortality. Ficus virens, a traditional Chinese medicinal plant, has anti-inflammatory effects in folk medicine; its main active component is total flavonoids (FLA), with unknown effects on sepsis-induced inflammation. We explored FLA's protective effects and mechanisms in a murine sepsis model induced by intraperitoneal LPS injection. High-dose FLA increased the survival rate from 12.5% to 50%. All FLA doses alleviated LPS-induced weight loss and hepatic injury, reduced serum IL-6,

Potential Roles of Prostaglandins PGD2, PGJ2, and PGE2 in the Pathogenesis of Systemic Lupus Erythematosus.

Prostaglandins (PGs) are eicosanoid compounds with various hormone-like effects within the human body. Prostaglandin D2 (PGD2), prostaglandin J2 (PGJ2, a metabolite of PGD2), and prostaglandin E2 (PGE2) are common prostaglandins. They are produced from arachidonic acid by cyclooxygenase isoenzymes (COX-1 and COX-2) and PG synthases and are involved in many biological metabolisms, including the inflammatory response. Recent studies show PGs are mediators for both proinflammatory and anti-inflammatory processes. In systemic lupus erythematosus (SLE), inflammation caused by autoimmunity damages t

Regulatory T cells in axial spondyloarthritis.

Type 3 immunity underlies both pain and inflammation in axial spondyloarthritis (axSpA), with IL-17A-producing cells such as T-helper 17 cells (TH17) being pathogenic mediators. In contrast, the role regulatory T cells (Tregs) have in maintaining immune homeostasis in axSpA remains poorly defined. Although understudied, emerging research suggests three plausible hypotheses: 1) Tregs in axSpA could be functional but are turned off in response to external signals. 2) Tregs in axSpA have deficiencies that result in reduced suppressive capabilities or stability. 3) Tregs directly contribute to aut

[Research progress on epithelial mucosal barrier impairment contributing to chronic rhinosinusitis].

Chronic rhinosinusitis (CRS) is defined as sinonasal mucosal inflammation persisting for >12 weeks and presents with nasal congestion, rhinorrhea, facial pain/pressure and olfactory impairment. CRS markedly impairs quality of life and imposes a heavy economic burden on health-care systems. Recent research has shifted the pathophysiological paradigm from simple mechanical obstruction to an epithelial-barrier-centred model that emphasizes immune dysregulation and cellular aberrations. Its complex aetiology includes ciliary dysfunction; dysbiosis; cytokine imbalance; epithelial-mesenchymal transi

Saffron as a Retinal Neuroprotectant: A Narrative Review of Preclinical Studies and Clinical Results.

The present narrative review reports the main preclinical and clinical results obtained by using supplementation of saffron or its pure components in neurodegeneration, with special emphasis on age-related macular degeneration. Beyond that, this article will address shared pathways between neurodegenerative diseases of the eye and the brain. It will be shown that saffron treatment might counteract oxidative damage in the retina and brain, as well as inflammation and inflammatory mediators that induce neuronal degeneration and death. The ways of action are multiple, and saffron chemical compone

The Role of Immune Cells and Signaling Pathways in Diabetic Eye Disease: A Comprehensive Review.

Diabetic eye disease (DED) encompasses a range of ocular complications arising from diabetes mellitus, including diabetic retinopathy, diabetic macular edema, diabetic keratopathy, diabetic cataract, and glaucoma. These conditions are leading causes of visual impairments and blindness, especially among working-age adults. Despite advancements in our understanding of DED, its underlying pathophysiological mechanisms remain incompletely understood. Chronic hyperglycemia, oxidative stress, inflammation, and neurodegeneration play central roles in the development and progression of DED, with immun

Aldose reductase is a potential therapeutic target for neurodegeneration.

Neurodegeneration is a complex process involving various inflammatory mediators and cellular responses. Aldose reductase (AR) is a key enzyme in the polyol pathway, which converts glucose to sorbitol. Beyond its metabolic role, AR has also been found to play a significant role in modulating neuroinflammation. This review aims to provide an overview of the current knowledge regarding the involvement of AR inhibition in attenuating neuroinflammation and complications from diabetic neuropathies. Here, we review the literature regarding AR and neuropathy/neurodegeneration. We discuss the mechanism

Role of antimicrobial peptide cathelicidin in thrombosis and thromboinflammation.

Thrombosis is a frequent cause of cardiovascular mortality and hospitalization. Current antithrombotic strategies, however, target both thrombosis and physiological hemostasis and thereby increase bleeding risk. In recent years the pathophysiological understanding of thrombus formation has significantly advanced and inflammation has become a crucial element. Neutrophils as most frequent immune cells in the blood and their released mediators play a key role herein. Neutrophil-derived cathelicidin next to its strong antimicrobial properties has also shown to modulates thrombosis and thus present

Investigation of Platelet Aggregation in Atherosclerosis.

Platelet activation and aggregation is implicated in all stages of inflammation-related atherosclerosis from the initial steps of endothelial dysfunction and plaque formation, to plaque rupture and atherothrombotic events, such as acute coronary syndrome, myocardial infarction, and ischemic incidences. Platelet aggregometry assays are the mainstream for evaluating and monitoring platelet reactivity in such conditions and for the investigation of prophylactic and therapeutic approaches. The most established methodology is light transmittance aggregometry (LTA). Here we describe the appropriate

Controlling leukocyte trafficking in IBD.

Inflammatory bowel disease (IBD) is characterized by the accumulation of immune cells, myeloid cells and lymphocytes in the inflamed intestine. The presence and persistence of these cells, together with the production of pro-inflammatory mediators, perpetuate intestinal inflammation in both ulcerative colitis and Crohn's disease. Thus, blockade of leukocyte migration to the intestine is a main strategy used to control the disease and alleviate symptoms. Vedolizumab is the only anti-integrin drug approved for the treatment of IBD but several other drugs also targeting integrins, chemokines or r

Inflammation-induced foam cell formation in chronic inflammatory disease.

Atherosclerosis is the leading cause of cardiovascular disease and is both a metabolic and inflammatory disease. Two models describe early events initiating atherosclerotic plaque formation, whereby foam cells form in response to hyperlipidaemia or inflammation-associated stimuli. Although these models are inextricably linked and not mutually exclusive, identifying the unique contribution of each in different disease settings remains an important question. Circulating monocytes are key mediators of atherogenesis in both models as precursors to lipid-laden foam cells formed in response to eithe

Hypertrophy of infected Peyer's patches arises from global, interferon-receptor, and CD69-independent shutdown of lymphocyte egress.

Lymphoid organ hypertrophy is a hallmark of localized infection. During the inflammatory response, massive changes in lymphocyte recirculation and turnover boost lymphoid organ cellularity. Intriguingly, the exact nature of these changes remains undefined to date. Here, we report that hypertrophy of Salmonella-infected Peyer's patches (PPs) ensues from a global "shutdown" of lymphocyte egress, which traps recirculating lymphocytes in PPs. Surprisingly, infection-induced lymphocyte sequestration did not require previously proposed mediators of lymphoid organ shutdown including type I interferon

Autoantibodies and resident renal cells in the pathogenesis of lupus nephritis: getting to know the unknown.

Systemic lupus erythematosus is characterized by a breakdown of self-tolerance and production of autoantibodies. Kidney involvement (i.e., lupus nephritis) is both common and severe and can result in permanent damage within the glomerular, vascular, and tubulo-interstitial compartments of the kidney, leading to acute or chronic renal failure. Accumulating evidence shows that anti-dsDNA antibodies play a critical role in the pathogenesis of lupus nephritis through their binding to cell surface proteins of resident kidney cells, thereby triggering the downstream activation of signaling pathways

Therapeutic down-modulators of staphylococcal superantigen-induced inflammation and toxic shock.

Staphylococcal enterotoxin B (SEB) and related superantigenic toxins are potent stimulators of the immune system and cause a variety of diseases in humans, ranging from food poisoning to toxic shock. These toxins bind directly to major histocompatibility complex (MHC) class II molecules on antigen-presenting cells and specific Vβ regions of T-cell receptors (TCR), resulting in hyperactivation of both monocytes/macrophages and T lymphocytes. Activated host cells produce massive amounts of proinflammatory cytokines and chemokines, activating inflammation and coagulation, causing clinical symptom

Low-dose colchicine: a potential augmenting agent in the treatment of refractory mood disorders.

Treatment-resistant mood disorders, including major depressive disorder (MDD) and bipolar disorder (BD), remain a significant clinical challenge. A confluence of emerging evidence implicates chronic low-grade inflammation in the pathophysiology of at least a subset of mood disorders, potentially mediating their poor response to traditional therapies. Colchicine, a well-established anti-inflammatory agent used in gout and more recently (at low dose) cardiovascular disease, inhibits microtubule polymerisation and suppresses NLRP3 inflammasome activation. Recent advances in immunopsychiatry sugge

Adipose tissue-derived extracellular vesicles as drivers of endothelial dysfunction in obesity.

Obesity and (white) adipose tissue (AT) inflammation are closely linked to endothelial dysfunction (ED) and neurological conditions associated with an impaired blood-brain barrier (BBB). AT-derived extracellular vesicles (AT-EVs), cell-derived particles involved in intercellular and interorgan communication, have emerged as active mediators of this process, yet the underlying mechanisms remain largely elusive. Molecular and functional profiling of AT-EVs' cargo has revealed obesity-related changes in miRNAs, proteins, and lipids that drive ED, primarily by activating proinflammatory signaling

A Comparative Study on the Anti-Inflammatory Activity of Kaempferol and Its O-Glycosides in LPS-Induced RAW264.7 Cells.

Kaempferol and its O-glycosides are prevalent dietary flavonoids, yet their comparative anti-inflammatory activities and underlying mechanisms remain poorly defined. This study evaluated the effects of kaempferol and three O-glycosides (K3G, K3R, K3S) on LPS-induced inflammation in RAW264.7 macrophages. Compared with its O-glycosides, kaempferol exhibited markedly stronger anti-inflammatory activity, shown by greater suppression of inflammatory mediators (NO, PGE2, IL-6, TNF-α) and ROS production. Mechanistically, kaempferol inhibited G0/G1 to S phase progression and attenuated MAPK/NF-κB/IκBα
