Genomic and clinical landscape of RAS mutations in acute myeloid leukemia.

RAS mutations are recurrent in acute myeloid leukemia (AML); yet, their genomic and clinical landscape remain poorly characterized. We analyzed centralized molecular profiling data in 8,553 patients with newly diagnosed AML. Of these, 1,775 (21%) harbored RAS mutations: NRAS in 1,103 (13%), KRAS in 445 (5%), and both in 227 (3%). RAS mutations primarily clustered in P-loop hotspots (G12/G13, 70%), with NRAS showing higher Q61 mutation prevalence (24% vs. 9%) and KRAS showing additional K117/A146 hotspots. Multiple RAS mutations occurred in 23% of patients. Convergent evolution of RAS signaling…

ELK1 promotes the progress of myeloid leukemia by hindering the differentiation of neutrophils.

BACKGROUND: The genes controlling lineage determination and differentiation tend to be essential for the development of acute myeloid leukemia (AML). Identifying novel target genes capable of promoting the differentiation and maturation of undifferentiated leukemia cells offers a promising therapeutic strategy for the treatment of AML. METHODS: We used conditional Elk1 and KrasG12D expression mice, along with Mx1-Cre, Lyz2-Cre and Elane-Cre drive strains (which enable stage specific control of Elk1 or KrasG12D expression), to investigate the function of Elk1 in the hematopoiesis and leukemoge…

Risk prediction model for thrombosis in leukemia patients: a systematic review.

Thrombosis represents a significant complication in leukemia patients, associated with treatment interruption and reduced survival outcomes. Although multiple risk prediction models have been developed, their methodological quality and applicability remain uncertain. This review aims to evaluate existing risk prediction models for thrombosis in patients with leukemia. We conducted comprehensive literature searches across nine databases from the inception to August 4, 2025. Two reviewers independently performed study selection, data extraction, and quality assessment using the CHARMS checklist …

The role of RAS mutations in leukemia progression, differentiation, and drug resistance.

Mutations in the RAS gene family (NRAS, KRAS) are critical drivers of late-stage acute myeloid leukemia (AML) progression. They are frequently detected in relapsed/refractory AML and AML transformed from myelodysplastic syndrome (MDS). Occurring as late-stage genetic events, RAS mutations synergize with early drivers to promote leukemogenesis. While mutually exclusive with FLT3-ITD mutations, they coexist with KIT, RUNX1, CEBPA mutations and MLL rearrangements. Granulocyte-monocyte progenitors (GMPs) serve as the cellular origin for RAS-mutant leukemia stem cells (LSCs). Ultimately, RAS mutati…

Prevalence and Risk Factors of Acute Pancreatitis in Childhood Acute Leukemia.

BACKGROUND/OBJECTIVES: Acute pancreatitis (AP) is an uncommon but serious complication in children undergoing treatment for acute leukemia. We aimed to determine the prevalence of AP in pediatric patients with acute leukemia, identify its risk factors, and evaluate their impact on treatment outcomes and overall survival. MATERIALS AND METHODS: We retrospectively reviewed the medical records of children with acute leukemia who developed acute abdominal pain suggestive of AP at Songklanagarind Hospital between 2004 and 2024. Demographic data, including leukemia subtypes, treatment protocols, an…

Glycolysis and glycolytic enzymes in acute myeloid leukemia: Warburg and beyond.

The ability to generate and regulate energy, maintain metabolic flux, and preserve redox homeostasis is the basis of all cellular processes … nd their involvement in normal and malignant myelopoiesis, with a particular focus on acute myeloid leukemia (AML). AML cells exhibit high glycolytic activity and frequently overexpress key glycolytic enzymes. Beyond their metabolic functions, these enzymes also exert regulatory roles in signaling, transcriptional control, and redox balance. Here, we discu …

NOTCH1 and SF3B1 mutations in chronic lymphocytic leukemia with their clinical associations.

Chronic lymphocytic leukemia (CLL) exhibits heterogeneous clinical outcomes influenced by chromosomal aberrations and genetic mutations. NOTCH1 and SF3B1 mutations are critical prognostic markers linked to disease progression and therapy resistance. This study analyzed 60 CLL patients from Hiwa Hospital (Sulaymaniyah, Iraq). Hematological parameters were assessed, and genomic DNA was sequenced for NOTCH1 exon 34 and SF3B1 exons 15-16. In silico pathogenicity was predicted using I-Mutant and PolyPhen-2. Mutations were found in 23.3% of patients (14/60), including the recurrent NOTCH1 c.7541_754…

Mitochondrial heteroplasmy is a risk factor for the development of chronic lymphocytic leukemia.

Chronic lymphocytic leukemia (CLL) can arise from lymphoid clonal hematopoiesis of indeterminate potential (L-CHIP), but many individuals who develop CLL lack detectable L-CHIP prior to diagnosis. To identify additional predictors of CLL risk, we analyze mitochondrial heteroplasmy in 419,154 individuals from the UK Biobank (UKB). Heteroplasmy is associated with a 1.5-fold increased risk of developing CLL, and this risk rises to 4-fold when accounting for deleterious heteroplasmic variants. These findings are confirmed in an independent cohort, the All of Us Research Program (AoU). Notably, the…

The role of chemokine receptors in leukemia: implications for prognosis and therapeutic strategies.

In leukemia, chemokines and their receptors are central regulators of immune cell signaling and migration, profoundly influencing disease progression, cellular proliferation, differentiation, and treatment response. Receptors from the CXC and CC families play key roles in leukemia pathogenesis. In acute myeloid leukemia, CXCR1, CXCR2, and CXCR4 are associated with poor prognosis, whereas elevated levels of ligands such as CXCL8, CXCL9, and CXCL10 generally indicate better outcomes. Reduced levels of CCL5 and CCL2 are also associated with a favorable prognosis. Conversely, decreased CXCR3 expre…

Epigenetics and In Silico Transcriptome Analysis of Pediatric Acute Myeloid Leukemia.

Pediatric acute myeloid leukemia (AML) is a heterogeneous hematologic malignancy that accounts for about 15%-20% of childhood leukemias. Despite therapeutic advances, relapses remain common, and survival for high-risk patients is below 60%. Unlike adult AML, pediatric AML displays distinct genetic mutations, including FLT3-ITD, NPM1, KMT2A rearrangements, and core-binding factors (CBF) fusions, as well as extensive epigenetic dysregulation. Aberrant DNA methylation, histone modifications, and altered non-coding RNA expressions disrupt hematopoietic differentiation and activate oncogenic transc…

EHA Guidelines on management of chronic lymphocytic leukemia and Richter transformation.

Previous editions of the European guidelines for the management of chronic lymphocytic leukemia (CLL) were developed by experts in CLL under the auspices of the European Society for Medical Oncology (ESMO). These previous editions have served as a reference text for many physicians caring for patients with CLL. The current, 2026 edition, represents the new, updated guidelines that, for the first time (and in agreement with ESMO), were written on behalf of the European Hematology Association (EHA), which will be solely responsible for subsequent editions, published annually to keep pace of the …

Co-option of lineage plasticity as a hallmark of multipotent acute leukemias.

Leukemias are classified by hematopoietic lineage and genetic alterations. Mixed-lineage and biphenotypic leukemias have long challenged diagnostic classification because of the coexpression of markers of distinct lineages. Recent studies have revealed previously unappreciated multilineage potential in a subset of B-cell acute lymphoblastic leukemias (B-ALL) as well as leukemias with markers of myeloid and T-cell differentiation with shared genetic features variably classified as acute myeloid leukemia, early T-cell progenitor ALL, or T/myeloid mixed-phenotype acute leukemias. We propose that …
