IRE1α Is Associated with Host Lipid Biosynthesis During Human Coronavirus OC43 Infection.

Research into the replication of human coronaviruses has surged, but much remains unknown regarding the host factors that influence disease. These viruses often activate cellular endoplasmic reticulum (ER) stress responses, benefiting from incompletely understood downstream effects. In this study, we investigated the role of the host ER stress response component, inositol-requiring enzyme 1 α (IRE1α), in regulating lipid metabolism during human coronavirus OC43 (HCoV-OC43) infection. We found IRE1α-dependent induction of lipogenesis-related genes, increased metabolic intermediates, and enhance

Simulated laughter immediately improves lipid layer thickness and tear meniscus height.

PURPOSE: Dry eye disease is a common condition associated with mental health. Daily simulated laughter for eight weeks has been shown to alleviate symptoms and signs of dry eye disease. However, the underlying mechanisms remain unclear. This study aimed to evaluate the immediate effects of laughter on tear film lipid layer thickness and tear meniscus height. METHODS: This was a single-center, self-controlled before-after trial. Participants aged 18 to 45 years with ocular surface disease index scores above 13 and non-invasive tear film break-up time below 8 s were included in the dry eye dise

The ABHD10-KCMF1 Complex Mitigates Cochlear Aging by Regulating Mitochondrial Lipid Metabolism.

Presbycusis is caused by multiple factors, the mechanisms of which are not fully understood. This study investigated mitochondria-related genes in presbycusis, a condition with multifactorial and incompletely understood mechanisms, by combining Mendelian randomization analysis and functional validation. Genetically predicted higher ABHD10 expression was identified as a protective factor against presbycusis and was regulated by methylation sites such as cg15684481, whereas methylation at other sites inhibited ABHD10 expression and alleviated the condition. In contrast, ABHD10 was upregulated in

Lipid abnormalities in chronic viral hepatitis: associations and machine learning-enhanced prediction.

BACKGROUND: Alteration in serum lipids is a well-recognized metabolic accompaniment of chronic hepatitis B (HBV) and hepatitis C (HCV) viral infections, and may reflect a combination of disease severity and metabolic dysregulation. Despite this established association, lipid profile monitoring is not standard practice in these patients, and lipid abnormalities may remain unrecognized until they influence long-term cardiometabolic outcomes or complicate overall risk stratification. This gap in routine practice provided the rationale for the present study. AIMS: This study aimed to evaluate whet…

FABP5 Inhibitor Attenuates HPV11 Pseudovirion Infection by Reducing Lipid Droplet Formation.

Persistent infection with low-risk human papillomavirus (HPV) can result in conditions such as verrucous hyperplasia of the skin and mucous membranes. Among these, anogenital warts (AGW), primarily caused by HPV types 6 and 11, are common sexually transmitted infections with a high recurrence rate. Because early infection with HPV often lacks obvious symptoms and the PsV model primarily reflects the entry phase, timely diagnosis and treatment of full HPV infection are difficult. The incomplete understanding of HPV infection mechanisms highlights the need for further investigation to address ch

Lipid metabolism in γδ T-cell activation: Implications for immunotherapy in ovarian cancer.

Ovarian cancer (OC) remains a leading cause of cancer-related mortality among women worldwide, largely due to asymptomatic progression, late-stage diagnosis, therapeutic resistance, and profound immunosuppression within the tumor microenvironment (TME). This is particularly relevant in advanced disease, where metastatic spread to the omentum creates a lipid-rich niche that promotes tumor growth and weakens anti-tumor immunity. This review examines lipid metabolic reprogramming and γδ T-cell function as a targetable immunometabolic axis in OC, with selected discussion of iNKT and CD8+ T-cells a

Development of Curcumin-Loaded Hybrid Lipid-Polymeric Nanoparticles for Hypoxia-Related Applications.

INTRODUCTION: Hypoxic and ischemia-associated tissues are often characterized by acidic microenvironments, highlighting the need for drug delivery systems that enable enhanced drug release under acidic conditions. Curcumin (CUR) has antioxidant and cytoprotective properties, but its therapeutic application is limited by poor aqueous solubility and low bioavailability. This study aimed to optimize hybrid lipid-polymeric nanoparticles (HLPNs) as a CUR delivery platform and evaluate their physicochemical properties, colloidal stability, CUR release at pH 7.4 and pH 5.5, and cellular effects under

Establishing a chicken embryo model for studying infection control of a novel lipid nanoparticle.

The rise of antibiotic-resistant infections highlights the urgent need for alternative therapeutic strategies. Monoglyceride-based lipid nanoparticles (LNPs) offer both antimicrobial activity and the potential for controlled drug delivery. In this study, LNPs with diameters of 230-350 nm and ζ-potential of approximately -35 mV demonstrated high colloidal stability and strong antibacterial efficacy in vitro, achieving complete growth inhibition of Staphylococcus aureus (S. aureus) at 0.04 mg/mL and Salmonella enterica subsp. enterica serovar Enteritidis (S. Enteritidis) at 0.9 mg/mL. To bridge

Macrophages tune NK cell IFNγ production through direct lipid transfer.

Natural killer (NK) cells are critical effectors of innate immunity, but their activity is strongly influenced by metabolic state. While intrinsic NK metabolism has been studied extensively, less is known about how surrounding immune cells shape NK cell function. Here, we identify a direct metabolic communication axis between macrophages and NK cells. Using co-culture and in vivo models, we show that lipopolysaccharide-stimulated macrophages induce lipid accumulation in NK cells that suppresses mTORC1 activity and the production of IFNγ. This lipid accumulation is visualised as increased lipid

Myeloid deficiency of RORα exacerbates lipid dysregulation and laser-induced choroidal neovascularization.

Dysregulated lipid and cholesterol metabolism is implicated in the pathogenesis of age-related macular degeneration (AMD), a leading cause of vision loss. Retinoic acid receptor-related orphan receptor alpha (RORα) is a lipid-sensing nuclear receptor genetically associated with neovascular AMD. We investigated the role of RORα in regulating AMD-like pathologies and laser-induced choroidal neovascularization (CNV) through mediating myeloid cell lipid homeostasis and function. Both systemic (Rorasg/sg) and myeloid-specific (Rorafl/fl;LysMCre) RORα deficient mice exhibited fundus lesions with agi

TALDO1 promotes lipid metabolic reprogramming and immunosuppressive microenvironment remodelling in hepatocellular carcinoma.

BACKGROUND: Hepatocellular carcinoma (HCC) is characterized by pronounced metabolic reprogramming and is frequently accompanied by the development of an immunosuppressive microenvironment. However, the key molecular mediators linking tumour metabolic dysregulation to immune microenvironment remodelling remain insufficiently defined. This study aimed to identify critical metabolic genes in HCC and to investigate their roles in lipid metabolic reprogramming and immunosuppression. METHODS: A deep autoencoder was used to extract latent metabolic features from HCC and identify TALDO1 as a key cand

Elevated lipid peroxidation biomarkers in autoimmune diseases: A systematic review and meta-analysis.

BACKGROUND: Ferroptosis, an iron-dependent cell death pathway driven by lipid peroxidation (LPO), is implicated in the pathogenesis of autoimmune diseases (AIDs). However, comprehensive clinical evidence establishing the association between specific LPO biomarkers and AIDs is lacking. OBJECTIVE: To systematically evaluate the clinical evidence for elevated LPO in major AIDs through a meta-analysis, focusing on key biomarkers including malondialdehyde (MDA) and 8-iso-prostaglandin F2α (8-iso-PGF2α). METHODS: We searched four databases for studies reporting serum, plasma, or urinary LPO levels

AARS1-mediated lactylation reprograms macrophage lipid metabolism to restrict antitumor immunity.

BACKGROUND: Tumor-derived lactate has long been regarded as a metabolic waste product. However, accumulating evidence indicates that lactate also functions as a signaling molecule that actively remodels the tumor immune microenvironment. How lactate-driven post-translational modifications in immune cells contribute to immune evasion in hepatocellular carcinoma (HCC) remains incompletely understood. This study aimed to identify the immune cell population responsible for lactylation-driven immunosuppression in HCC and to elucidate the molecular mechanism by which lactylation rewires macrophage m

Interplay between lipid droplets and alpha-synuclein: implication in Parkinson's disease pathogenesis.

Lipid droplets (LDs), once considered inert lipid stores, are now recognized as active regulators of lipid metabolism, stress responses, and protein quality control in the brain. Their dysregulation is increasingly linked to neurodegenerative diseases, notably Parkinson's disease (PD). This review explores the emerging bidirectional relationship between LDs and α-synuclein (α-Syn), a key pathological hallmark of PD. α-Syn can promote LD accumulation by modulating lipid metabolism and inhibiting lipolysis, while LDs can facilitate α-Syn aggregation through specific lipid-protein and membrane in

Lipid packing and cholesterol content regulate membrane wetting and remodeling by biomolecular condensates.

Biomolecular condensates play a central role in cellular processes by interacting with membranes driving wetting transitions and inducing mutual remodeling. While condensates are known to locally alter membrane properties such as lipid packing and hydration, it remains unclear how membrane composition and phase state in turn affect condensate affinity. Here, we show that it is not only the membrane phase itself, but rather the degree of lipid packing that determines the condensate affinity for membranes. Increasing lipid chain length, saturation, or cholesterol content, enhances lipid packing,

Diagnosis and Management of Lipid Transfer Protein Allergy-A BSACI Clinical Practice Statement.

Although the most prevalent plant food allergy in the United Kingdom (UK) is pollen food syndrome (PFS), there is increasing evidence that reactions to plant foods could also be due to sensitisation to Lipid Transfer Proteins (LTP). These proteins, highly resistant to heat and processing, are present in raw, cooked and processed plant foods and often provoke moderate to severe symptoms. LTP allergy is common in Mediterranean countries, but there is a lack of epidemiological data from Northern Europe, although small case series have been published characterising LTP allergy in both England and
