Identification of Two Distinct Working Memory-Related Brain Networks in Healthy Young Adults.

Working memory (WM) is an important cognitive domain for everyday life functioning and is often disturbed in neuropsychiatric disorders. Functional magnetic resonance imaging (fMRI) studies in humans show that distributed brain areas typically described as fronto-parietal regions are implicated in WM tasks. Based on data from a large sample of healthy young adults (N = 1369), we applied independent component analysis (ICA) to the WM-fMRI signal and identified two distinct networks that were relevant for differences in individual WM task performance. A parietally-centered network was particular

Frontoparietal processing of stress-relevant information differs in individuals with a negative cognitive style.

Prior research indicates that cognitive vulnerabilities can render individuals more susceptible to psychopathology in the wake of stressful events. However, little work has directly targeted the neural mechanisms involved. In this study, we examined functional magnetic resonance imaging (fMRI) activity as a function of negative cognitive style, a well-studied cognitive vulnerability for depression. We adapted a robust paradigm in which undergraduate students completed fMRI testing after a known ecologically valid stressor (a midterm exam). Negative cognitive style correlated with brain activit

Cerebellar ataxia, neuropathy, vestibular areflexia syndrome (CANVAS) with chronic cough and preserved muscle stretch reflexes: evidence for selective sparing of afferent Ia fibres.

The aim of this study was to describe five patients with cerebellar ataxia, neuropathy and vestibular areflexia syndrome (CANVAS) with chronic cough and preserved limb muscle stretch reflexes. All five patients were in the seventh decade of age, their gait imbalance having been initiated in the fifth decade. In four patients cough antedated gait imbalance between 15 and 29 years; cough was spasmodic and triggered by variable factors. Established clinical picture included severe hypopallesthesia predominating in the lower limbs with postural imbalance, and variable degree of cerebellar axial an

Awareness of perception and sensory-motor integration: ERPs from the anterior insula.

The present work follows recent evidences of studies showing that visual stimuli evoke two early prefrontal event-related potentials (ERP) concomitant to the canonical occipital activities, but originating within the anterior insula (the pN1 and the pP1 components). To clarify the exogenous/endogenous nature of these components, we performed two experiments in which stimulus physical features (Experiment 1) and motor demands of the task (Experiment 2) were considered. In a simple response task (SRT), low-visibility stimuli evoked larger pN1 over the prefrontal areas (Experiment 1) with respect

Learning Processes and Brain Connectivity in A Cognitive-Motor Task in Neurodegeneration: Evidence from EEG Network Analysis.

Electroencephalographic (EEG) rhythms are linked to any kind of learning and cognitive performance including motor tasks. The brain is a complex network consisting of spatially distributed networks dedicated to different functions including cognitive domains where dynamic interactions of several brain areas play a pivotal role. Brain connectome could be a useful approach not only to mechanisms underlying brain cognitive functions, but also to those supporting different mental states. This goal was approached via a learning task providing the possibility to predict performance and learning alon

Circuit-based frameworks of depressive behaviors: The role of reward circuitry and beyond.

Major depressive disorder (MDD) is a common but serious neuropsychiatric affliction that comprises a diverse set of symptoms such as the inability to feel pleasure, lack of motivation, changes in appetite, and cognitive difficulties. Given the patient to patient symptomatic variability in MDD and differing severities of individual symptoms, it is likely that maladaptive changes in distinct brain areas may mediate discrete symptoms in MDD. The advent and recent surge of studies using viral-genetic approaches have allowed for circuit-specific dissection of networks underlying motivational behavi

Decreased functional connectivity within the default-mode network in acute brainstem ischemic stroke.

PURPOSE: Ischemic stroke within the brainstem is associated with an increased risk of cognitive dysfunction. This study aimed to explore the integrity of a default-mode network (DMN) and its relationship with clinical variables in patients with acute ischemic brainstem stroke using an independent component analysis (ICA) approach. MATERIALS AND METHODS: Twenty-one patients with acute ischemic brainstem stroke and 25 well-matched healthy subjects were enrolled in this study and underwent resting-state functional magnetic resonance imaging. The ICA was adopted to extract the DMN, including its

Spinal Cord Injury Disrupts Resting-State Networks in the Human Brain.

Despite 253,000 spinal cord injury (SCI) patients in the United States, little is known about how SCI affects brain networks. Spinal MRI provides only structural information with no insight into functional connectivity. Resting-state functional MRI (RS-fMRI) quantifies network connectivity through the identification of resting-state networks (RSNs) and allows detection of functionally relevant changes during disease. Given the robust network of spinal cord afferents to the brain, we hypothesized that SCI produces meaningful changes in brain RSNs. RS-fMRIs and functional assessments were perfor

Functional hierarchy of oculomotor and visual motion subnetworks within the human cortical optokinetic system.

Optokinetic look nystagmus (look OKN) is known to engage cortical visual motion and oculomotor hubs. Their functional network hierarchy, however, and the role of the cingulate eye field (CEF) and the dorsolateral prefrontal cortex (DLPFC) in particular have not been investigated. We used look OKN in fMRI to identify all cortical visual motion and oculomotor hubs involved. Using these activations as seed regions, we employed hierarchical clustering in two differing resting state conditions from a separate public data set. Robust activations in the CEF highlight its functional role in OKN and in

Resting state fMRI reveals the altered synchronization of BOLD signals in essential tremor.

Essential tremor (ET) is one of the most common movement disorders in humans. Nevertheless, there remain several controversies surrounding ET, such as whether it is a disorder of abnormal neuronal oscillations within the tremor network. In this work, the resting-state fMRI data were collected from 17 ET patients and 17 age- and gender-matched healthy controls. First, using FOur-dimensional (spatiotemporal) Consistency of local neural Activities (FOCA) the abnormal synchronization of fMRI signals in ET patients were investigated. Then, global functional connectivity intensity (gFCI) and density

Comparison of tractography-assisted to atlas-based targeting for deep brain stimulation in essential tremor.

BACKGROUND: Direct targeting of the dentato-rubro-thalamic tract is efficacious in DBS for tremor suppression. OBJECTIVES: We sought to compare outcomes and optimal stimulation parameters for tremor control using the technique of directly targeting the dentato-rubro-thalamic tract to those who underwent indirect targeting of the ventral intermediate nucleus thalamus. METHODS: Twenty consecutive essential tremor patients obtained preoperative diffusion MRIs, where the dentato-rubro-thalamic tract was individually drawn and used to directly target the ventral intermediate nucleus of the thalam

Altered Functional Connectivity of the Executive Functions Network During a Stroop Task in Children with Reading Difficulties.

Children with reading difficulties (RDs) often receive related accommodations in schools, such as additional time for examinations and reading aloud written material. Existing data suggest that these readers share challenges in executive functions (EFs). Our study was designed to determine whether children with RDs have specific challenges in EFs and define neurobiological signatures for such difficulties using magnetic resonance imaging (MRI) data. Reading and EFs abilities were assessed in 8-12-year-old children with RDs and age-matched typical readers. Functional MRI data were acquired duri

Functional Connectivity Alterations in Children with Spastic and Dyskinetic Cerebral Palsy.

Cerebral palsy (CP) has long been investigated to be associated with a range of motor and cognitive dysfunction. As the two most common CP subtypes, spastic cerebral palsy (SCP) and dyskinetic cerebral palsy (DCP) may share common and distinct elements in their pathophysiology. However, the common and distinct dysfunctional characteristics between SCP and DCP on the brain network level are less known. This study aims to detect the alteration of brain functional connectivity in children with SCP and DCP based on resting-state functional MRI (fMRI). Resting-state networks (RSNs) were established

Functional brain stem circuits for control of nose motion.

Rodents shift their nose from side to side when they actively explore and lateralize odors in the space. This motor action is driven by a pair of muscles, the deflector nasi. We studied the premotor control of this motion. We used replication-competent rabies virus to transsynaptically label inputs to the deflector nasi muscle and find putative premotor labeling throughout the parvocellular, intermediate, and gigantocellular reticular formations, as well as the trigeminal nuclei, pontine reticular formation, midbrain reticular formation, red nucleus, and superior colliculus. Two areas with ext

Role of Anterior Intralaminar Nuclei of Thalamus Projections to Dorsomedial Striatum in Incubation of Methamphetamine Craving.

Relapse to methamphetamine (Meth) seeking progressively increases after withdrawal from drug self-administration (incubation of Meth craving). We previously demonstrated a role of dorsomedial striatum (DMS) dopamine D1 receptors (D1Rs) in this incubation. Here, we studied the role of afferent glutamatergic projections into the DMS and local D1R-glutamate interaction in this incubation in male rats. We first measured projection-specific activation on day 30 relapse test by using cholera toxin b (retrograde tracer) + Fos (activity marker) double-labeling in projection areas. Next, we determined

Characterization of McDonald's intermediate part of the Central nucleus of the amygdala in the rat.

The actual organization of the central nucleus of the amygdala (CEA) in the rat is mostly based on cytoarchitecture and the distribution of several cell types, as described by McDonald in 1982. Four divisions were identified by this author. However, since this original work, one of these divisions, the intermediate part, has not been consistently recognized based on Nissl-stained material. In the present study, we observed that a compact condensation of retrogradely labeled cells is found in the CEA after fluorogold injection in the anterior region of the tuberal lateral hypothalamic area (LHA

Abnormal Functional Connectivity of Frontopolar Subregions in Treatment-Nonresponsive Major Depressive Disorder.

BACKGROUND: Approximately 30% of patients with major depressive disorder develop treatment-nonresponsive depression (TNRD); novel interventions targeting the substrates of this illness population are desperately needed. Convergent evidence from lesion, stimulation, connectivity, and functional neuroimaging studies implicates the frontopolar cortex (FPC) as a particularly important region in TNRD pathophysiology; regions functionally connected to the FPC, once identified, could present favorable targets for novel brain stimulation treatments. METHODS: We recently published a parcellation of th

Frontal-Brainstem Pathways Mediating Placebo Effects on Social Rejection.

Placebo treatments can strongly affect clinical outcomes, but research on how they shape other life experiences and emotional well-being is in its infancy. We used fMRI in humans to examine placebo effects on a particularly impactful life experience, social pain elicited by a recent romantic rejection. We compared these effects with placebo effects on physical (heat) pain, which are thought to depend on pathways connecting prefrontal cortex and periaqueductal gray (PAG). Placebo treatment, compared with control, reduced both social and physical pain, and increased activity in the dorsolateral

Disruption of the Right Temporoparietal Junction Impairs Probabilistic Belief Updating.

Generating and updating probabilistic models of the environment is a fundamental modus operandi of the human brain. Although crucial for various cognitive functions, the neural mechanisms of these inference processes remain to be elucidated. Here, we show the causal involvement of the right temporoparietal junction (rTPJ) in updating probabilistic beliefs and we provide new insights into the chronometry of the process by combining online transcranial magnetic stimulation (TMS) with computational modeling of behavioral responses. Female and male participants performed a modified location-cueing
