Concise Review: Induced Pluripotent Stem Cell-Based Drug Discovery for Mitochondrial Disease.

High attrition rates and loss of capital plague the drug discovery process. This is particularly evident for mitochondrial disease that typically involves neurological manifestations and is caused by nuclear or mitochondrial DNA defects. This group of heterogeneous disorders is difficult to target because of the variability of the symptoms among individual patients and the lack of viable modeling systems. The use of induced pluripotent stem cells (iPSCs) might significantly improve the search for effective therapies for mitochondrial disease. iPSCs can be used to generate patient-specific neur

A new syndrome of refractory sideroblastic anemia with vacuolization of marrow precursors and exocrine pancreatic dysfunction.

In the past decade, we have studied four unrelated children with what we believe is a previously unreported disorder affecting the bone marrow and exocrine pancreas. During infancy these patients had the onset of severe, transfusion-dependent, macrocytic anemia plus a variable degree of neutropenia and thrombocytopenia. Their bone marrows had normal cellularity but were characterized by remarkable vacuolization of erythroid and myeloid precursors, hemosiderosis, and ringed sideroblasts. The vacuoles probably represented manifestations of cellular degeneration and death. In two patients, in vit

Breast cancer brain metastasis: mechanisms, emerging biomarkers, and therapeutic opportunities.

Breast cancer brain metastasis (BCBM) is a clinically distinct form of metastatic breast cancer that causes substantial neurological morbidity and mortality. This review examines the biological bottlenecks that precede a detectable brain lesion: survival in the circulation, interaction with the cerebral endothelium, passage across the blood-brain barrier (BBB), persistence in neural tissue, and intracranial outgrowth. We emphasize how astrocytes, microglia, neurons, stromal cells, and the nutrient environment of the brain shape these events and influence treatment response. These biological fe

Intestinal epithelial memory: from inflammatory imprinting to metabolite-shaped immune tolerance.

Intestinal epithelial cells are rapidly renewing barrier cells that maintain compartmentalization between the luminal environment and the mucosal immune system through epithelial transport, antimicrobial defense, secretory programs, and cytokine-mediated communication. Emerging evidence suggests that the intestinal epithelium can also retain durable imprints of prior inflammatory, microbial, or metabolic exposures. We refer to this phenomenon as intestinal epithelial memory. This form of memory is likely encoded within stem and progenitor compartments through persistent changes in chromatin ac

Animal Models of Slow-Transit Constipation: Current Progress, Limitations, and Future Perspectives.

PURPOSE: Slow-transit constipation (STC) is a subtype of chronic constipation characterized by delayed colonic transit without obstructive defecation. Because in vivo human experimentation is constrained, animal models remain the primary tool for probing STC pathogenesis and testing therapies, yet no unified framework exists for judging howwell current models reproduce human disease. This review systematically appraises the reported STC animal models against a single validity framework and identifies design criteria for the next generation of models. METHODS: We reviewed pharmacological agent

Repurposing antimalarial drug atovaquone in combination with radiation and PD-L1 checkpoint blockade to enhance antitumour response in head and neck squamous cell carcinoma (HNSCC).

Atovaquone is an approved antimalarial drug which could sensitise tumours to radiation and may modulate tumour immune microenvironment. Here we evaluated the triple combination of radiation with atovaquone plus anti-PD-L1 in HNSCC syngeneic mouse tumour models. We observed significant tumour growth delay induced by the triple combination treatment with differential responses in three different mouse models. There was complete tumour eradication in 6 of 12 mice in the MOCL1 model but not in the other two murine models. The triple combination was associated with increased populations of CD8+-T G

ERX 208 induces endoplasmic reticulum stress and suppresses DNA damage repair to enhance chemotherapy sensitivity.

Ovarian cancer (OCa) remains the most lethal gynecologic malignancy, driven in part by intrinsic and acquired therapeutic resistance to DNA damaging chemotherapies. We previously identified that ERX-208 targets the protein encoded by Lysosomal acid lipase (LIPA) to induce catastrophic endoplasmic reticulum (ER) stress in OCa models. As ERX-class compounds advance to clinical development, rational combination strategies are needed to maximize therapeutic efficacy. To address this question, we conducted an unbiased in vitro screen of the combination of ERX 208 with 147 FDA approved agents. We id

ENO2 drives tumor cell-induced M2 macrophage polarization to promote colorectal cancer liver metastasis.

Liver metastasis is the primary cause of mortality in colorectal cancer (CRC) patients. To decipher the underlying mechanisms, we performed single-cell RNA sequencing (scRNA-seq) on paired primary colorectal tumors, adjacent tissues and liver metastases from three CRC liver metastasis (CRLM) patients, alongside colorectal tumors and adjacent tissues from three non-metastatic CRC patients. Our analysis revealed a significant enrichment of Enolase 2-expressing (ENO2⁺) cancer cells in CRLM patients compared to their non-metastatic counterparts. Functional characterization, supported by bioinforma

CA19-9 induces microenvironment remodeling in pancreatic ductal adenocarcinoma.

Durable therapeutic efficacy remains a major barrier to improving outcomes for patients with pancreatic ductal adenocarcinoma (PDAC). An immunosuppressive tumor microenvironment (TME) is a hallmark of PDAC and has been demonstrated to be a dominant driver of therapeutic resistance. The aberrant glycan CA19-9 is prevalent in PDAC and drives tumor progression, but the paracrine mechanisms by which it contributes to TME remodeling are unknown. To address this, we mapped TME changes and performed functional analyses using a genetically engineered mouse model (GEMM) harboring KrasG12D mutation and

The GPCR Connection: Linking Alzheimer's Disease and Glioblastoma.

Alzheimer's disease (AD) and glioblastoma multiforme (GBM) are biologically distinct age-related brain disorders with opposing clinical phenotypes. AD is characterised by progressive neurodegeneration and cognitive decline, whereas GBM is characterised by aggressive cellular proliferation and a poor prognosis. Despite these differences, converging evidence indicates that both conditions share molecular pathways and network-level dysfunction that emerge during brain ageing. Central to this convergence are G protein-coupled receptors (GPCRs), which act as integrative signalling hubs that regulat

Patient-specific iPSC-derived ECs identify QKI-7 as a key regulator of endothelial dysfunction and potential therapeutic target in diabetes.

Diabetic vascular complications remain a major cause of morbidity and mortality, yet the molecular mechanisms underlying endothelial dysfunction in diabetes remain incompletely understood. Endothelial dysfunction is a key contributor to vascular pathology, and patient-derived induced pluripotent stem cell-derived endothelial cells (iPSC-ECs) provide a human platform to investigate disease-associated endothelial phenotypes under controlled conditions. Here, we identify the RNA-binding protein Quaking-7 (QKI-7) as a key regulatory factor associated with endothelial dysfunction in patient-derived

--Paneth cell niche function is critical for intestinal stem cells in radiation injury but dispensable in GVHD.

Paneth cells (PCs) produce WNT3 and EGF in the small intestine and are thought to contribute to the intestinal stem cell (ISC) niche, which is a target of graft vs. host disease (GVHD). However, PC deletion models have failed to demonstrate substantial consequences for ISCs or other epithelial pathology. The physiologic role of PC niche function has thus remained opaque, and the significance of this function after bone marrow transplantation (BMT) is poorly understood. Using Lysozyme1-DTR mice to ablate PCs, we found stable WNT3 expression in vivo, and crypts from PC-ablated mice retained orga

Solasonine inhibits growth of gastric cancer by targeting FTL-mediated ferroptosis.

Solasonine (SS), a principal bioactive constituent of Solanum nigrum L., has attracted considerable attention due to its pronounced cytotoxic effects on cancer cells. Nevertheless, the precise mechanism of SS against gastric cancer remains inadequately explored. In this study, the therapeutic effects and potential targets of SS against gastric cancer were investigated. The results showed that SS markedly inhibited cell proliferation and promoted cell death in gastric cancer cells. Pharmacological inhibition studies revealed that the antitumor effects of SS predominantly rely on ferroptosis rat

Metabolic-circadian reprogramming mediates epithelial barrier destabilization during Euphorbia fischeriana toxicity and its mitigation by traditional milk processing.

ETHNOPHARMACOLOGICAL RELEVANCE: EUPHORBIAE EBRACTEOLATAE RADIX (Langdu), the dried root of Euphorbia fischeriana Steud. (E. fischeriana), has been widely used in Mongolian and traditional Chinese medicine for the treatment of inflammatory disorders and malignancies. However, its clinical application is limited by its gastrointestinal toxicity. Milk processing is traditionally used to reduce toxicity, but the underlying mechanisms remain unclear. AIM OF THE STUDY: To investigate the mechanisms underlying E. fischeriana-induced intestinal toxicity and to evaluate the effects of traditional milk

Gut microbial metabolites in inflammatory bowel disease: immunological mechanisms regulating Treg/Th17 balance and therapeutic potential.

Inflammatory bowel disease (IBD) is a chronic inflammatory disorder resulting from a combination of genetic susceptibility, environmental factors, and an abnormal immune response of the intestinal immune system to commensal microorganisms. The gut microbiota and its metabolites play a pivotal role in maintaining intestinal immune homeostasis. Recent advances indicate that dysbiosis of the microbiota is accompanied by alterations in its metabolic functions. Abnormal levels of key metabolites, particularly short-chain fatty acids (SCFAs), tryptophan derivatives, and secondary bile acids, are clo

Micro- and Nanoplastics in Cardiovascular Toxicology: Human Tissue Detection, Clinical Phenotypes, and Adverse Outcome Signals.

Micro- and nanoplastics (MNPs) have emerged from a diffuse environmental concern to a plausible cardiovascular exposure with potential clinical relevance. The current evidence base is heterogeneous: human observational and tissue-detection studies have identified MNPs in blood, thrombi, atherosclerotic plaques, arterial tissue, and cardiac surgical specimens, whereas cell, organoid, and animal studies provide mechanistic plausibility for vascular inflammation, thrombosis, myocardial injury, remodeling, and electrophysiological disturbance. Key clinical signals now include polyethylene detectio
