BACKGROUND: The Holt-Oram syndrome is an autosomal dominant condition characterized by skeletal abnormalities that are frequently accompanied by congenital cardiac defects. The cause of these disparate clinical features is unknown. To identify the chromosomal location of the Holt-Oram syndrome gene, we performed clinical and genetic studies.
METHODS: Two large families with the Holt-Oram syndrome were evaluated by radiography of the hands, electrocardiography, and transthoracic echocardiography. Genetic-linkage analyses were performed with polymorphic DNA loci dispersed throughout the genome

BACKGROUND: Social anxiety disorder (SAD) is among the most common of all psychiatric disorders with lifetime prevalence estimates ranging from 7% to 13%. Although there is evidence that SAD has a strong familial basis, there are few studies of potential candidate genes. In addition to a genetic association, there is also the possibility that temperamental risk factors for the disorder may be genetically transmitted. Against this background, our aims were threefold: i.) to compare patients and controls with respect to personality traits, ii.) to genotype a subgroup of these participants to inv

Glucuronidation, catalysed by UGT enzymes, plays a key role in the metabolism of clozapine (CLZ) and its active metabolite, norclozapine (NCLZ). Genetic polymorphisms in UGT1A4 and UGT2B10 can influence NCLZ plasma levels. Co-administration of valproic acid (VPA), commonly used in Treatment Resistant Schizophrenia, has been linked to decreased NCLZ concentrations and increased neutropenia risk. Additionally, CYP1A2, the main enzyme responsible for CLZ N-demethylation, may affect CLZ and indirectly NCLZ levels, specially considering genetic variability and smoking status. A total of 296 plasma

The parameters for the absorption, distribution, and metabolic clearance of a wide array of chemicals can be calculated and integrated into a physiologically based pharmacokinetic (PBPK) model that simulates plasma drug concentration over time. Typically, pharmacological and toxicological assessments of chemicals involve estimating human clearance through allometric extrapolation from in vivo rat profiles using empirical compartmental and PBPK models. For patients with genetically impaired cytochrome P450 (P450 or CYP) enzymes who are prescribed only certain drugs, moderate plasma exposure sho

The work was performed to assess the relationship of single-nucleotide polymorphisms (SNPs), which determine the concentration of sex hormones (confirmed in previously performed genome-wide studies (GWASs)), with the risk of endometrial hyperplasia (EH). The objects of the study were 1493 women, of which 520 individuals had EH; the control group consisted of 973 women. Nine SNPs that were GWAS-associated with the level of sex hormones were investigated. The correlations of SNPs that determine the level of sex hormones with EH risk were found: minor polymorphic variants rs11031002 (allele A: OR

BACKGROUND: Gene-environment interactions play a critical role in shaping phenotypic heterogeneity in complex psychiatric disorders. Brain-derived neurotrophic factor (BDNF) is a key genetic regulator of stress-sensitive neuroplasticity. Yet, how BDNF polymorphisms are associated with the effect and impact of childhood trauma on clinical phenotypes remains incompletely understood.
METHODS: We conducted a case-control study including 93 patients with first-episode schizophrenia (SZ) and 64 healthy controls. Childhood trauma exposure was assessed using the Childhood Trauma Questionnaire (CTQ),

Toll-like receptors (TLRs) are crucial components of innate immunity. A specific form of genetic variation in TLR genes may increase the chance of developing leukemia. The present investigation conducted a comprehensive meta-analysis to examine the correlation between three TLR polymorphisms, namely TLR2 (rs3804099), TLR4 (rs4986790), and TLR9 (rs187084), within the leukemia risk group. An in-depth literature search was performed using Web of Science, PubMed, and Google Scholar to identify noteworthy research published in these scientific databases from 2012 to 2024. UNLABELLED: Glycated hemoglobin has shown disagreements with other glycemic indices; termed the glycation gap. The glycation gap can be influenced by nonglycemic factors, such as protein deglycation, through the fructosamine-3-kinase (FN3K) enzyme. This cross-sectional study aimed to examine whether single nucleotide polymorphisms (SNPs) in the FN3K gene can explain the glycation gap. Among the 826 participants, 79.8% were female, 22.3% presented with diabetes, and the median age was 53 years. The results suggest that genetic polymorphisms in the FN3K gene may influence the glycation gap in in

Like other plants, wild and domesticated rice species (Oryza nivara, O. rufipogon, and O. sativa) evolve in environments with various biotic and abiotic stresses that fluctuate in intensity through space and time. Microbial pathogens and invertebrate herbivores such as plant-parasitic nematodes and caterpillars show geographical and temporal variation in activity patterns and may respond differently to certain plant-defensive mechanisms. As such, plant interactions with multiple community members may result in conflicting selection pressures on genetic polymorphisms. Here, through assays with

BACKGROUND: Severe aplastic anemia (SAA) is an uncommon life-threatening disorder characterized by hypocellular bone marrow and pancytopenia. It is typically associated with immune-mediated mechanisms, requiring immunosuppressive therapy (IST) or hematopoietic stem cell transplantation (HSCT). Infections, especially invasive fungal infections such as mucormycosis and aspergillosis, constitute principal causes of morbidity and mortality in patients with SAA. Genetic predispositions, including perforin (PRF1) polymorphisms, may further complicate disease outcomes by impairing immune function.
