Biomaterials Combined With Physical Modulation Strategies for Spinal Cord Injury Repair: Material Design, Mechanism, and Challenges.

Spinal cord injury (SCI) causes persistent neurological deficits because primary damage initiates dynamic secondary cascades that restrict endogenous repair. Functional biomaterials can bridge lesions, provide structural and biochemical support, guide axonal growth, and remodel the inhibitory microenvironment. However, their cues are generally static or slowly evolving and cannot match the changing pathological and electrophysiological states of injured tissue. Physical modulation complements these functions by delivering electrical, magnetic, ultrasonic, optical/photothermal, or mechanical cu

Porcine Small Intestinal Submucosa Mitral Valve Material Responses Support Acute Somatic Growth.

Background: Conceptually, a tissue engineered heart valve would be especially appealing in the pediatric setting since small size and somatic growth constraints would be alleviated. In this study, we utilized porcine small intestinal submucosa (PSIS) for valve replacement. Of note, we evaluated the material responses of PSIS and subsequently its acute function and somatic growth potential in the mitral position. Methods and Results: Material and mechanical assessment demonstrated that both fatigued 2ply (∼65 μm) and 4ply (∼110 μm) PSIS specimens exhibited similar failure mechanisms, but at an

Classic and Current Opinions in Human Organ and Tissue Transplantation.

Graft tolerance is a pathophysiological condition heavily reliant on the dynamic interaction of the innate and adaptive immune systems. Genetic polymorphism determines immune responses to tissue/organ transplantation, and intricate humoral and cell-mediated mechanisms control these responses. In transplantation, the clinician's goal is to achieve a delicate equilibrium between the allogeneic immune response, undesired effects of the immunosuppressive drugs, and the existing morbidities that are potentially life-threatening. Transplant immunopathology involves sensitization, effector, and apopt

Human pluripotent stem cell-derived cardiac stromal cells and their applications in regenerative medicine.

Coronary heart disease is one of the leading causes of death in the United States. Recent advances in stem cell biology have led to the development and engineering of human pluripotent stem cell (hPSC)-derived cardiac cells and tissues for application in cellular therapy and cardiotoxicity studies. Initial studies in this area have largely focused on improving differentiation efficiency and maturation states of cardiomyocytes. However, other cell types in the heart, including endothelial and stromal cells, play crucial roles in cardiac development, injury response, and cardiomyocyte function.

Animal models of soft-tissue sarcoma.

Soft-tissue sarcomas (STSs) are rare mesenchymal tumors that arise from muscle, fat and connective tissue. Currently, over 75 subtypes of STS are recognized. The rarity and heterogeneity of patient samples complicate clinical investigations into sarcoma biology. Model organisms might provide traction to our understanding and treatment of the disease. Over the past 10 years, many successful animal models of STS have been developed, primarily genetically engineered mice and zebrafish. These models are useful for studying the relevant oncogenes, signaling pathways and other cell changes involved

bFGF-chitosan scaffolds effectively repair 20 mm sciatic nerve defects in adult rats.

The repair of peripheral nerve injury is still a great challenge in clinic. Autologous nerve transplantation is the gold standard for the treatment of long-distance peripheral nerve defects, but this method remains associated with high morbidity of the donor site and lack of matching donor. In this study, a novel chitosan scaffold (CS) loaded with control-released basic fibroblast growth factor (bFGF) was used to repair 20 mm sciatic nerve defects in adult rat. The ultrastructure of bFGF-CS was observed by scanning electron microscope. The tensile tester and nano-indentation were used to evalu

Long-term evaluation of elongating columella using conchal composite graft in bilateral secondary cleft lip and nose deformity.

BACKGROUND: A short columella is one of the most severe problems in bilateral cleft lip and nose deformity. In particular, with tissue insufficiency or scar contracture on the upper lip, it is very hard to achieve a good nasal profile with conventional methods. METHODS: One hundred thirty-seven patients with a short columella were operated on from February of 1987 to December of 2008. The average age at operation was 7.3 years. The conchal cartilage was harvested. The preauricular or postauricular skin-subcutaneous fat composite tissue was harvested. Through a transcolumellar incision, the co

Peripheral nerve defects repaired with autogenous vein grafts filled with platelet-rich plasma and active nerve microtissues and evaluated by novel multimodal ultrasound techniques.

BACKGROUND: Developing biocompatible nerve conduits that accelerate peripheral nerve regeneration, lengthening and functional recovery remains a challenge. The combined application of nerve microtissues and platelet-rich plasma (PRP) provides abundant Schwann cells (SCs) and various natural growth factors and can compensate for the deficiency of SCs in the nerve bridge, as well as the limitations of applying a single type of growth factor. Multimodal ultrasound evaluation can provide additional information on the stiffness and microvascular flow perfusion of the tissue. This study was designed

Cytoskeletal and functional changes in bioreactor assembled thyroid tissue organoids exposed to gamma radiation.

Fischer rat thyroid cells were grown under low-shear stress in a bioreactor to a stage of organization composed of integrated follicles resembling small thyroid glands prior to exposure to 3 Gray-gamma radiation. Bioreactor tissues and controls (both irradiated and non-irradiated) were harvested at 24, 48, 96 and 144 hours post-exposure. Tissue samples were fixed and fluorescently labeled for actin and microtubules. Tissues were assessed for changes in cytoskeletal components induced by radiation and quantified by laser scanning cytometry. ELISA's were used to quantify transforming growth fact

Novel Adult-Onset Systolic Cardiomyopathy Due to MYH7 E848G Mutation in Patient-Derived Induced Pluripotent Stem Cells.

A novel myosin heavy chain 7 mutation (E848G) identified in a familial cardiomyopathy was studied in patient-specific induced pluripotent stem cell-derived cardiomyocytes. The cardiomyopathic human induced pluripotent stem cell-derived cardiomyocytes exhibited reduced contractile function as single cells and engineered heart tissues, and genome-edited isogenic cells confirmed the pathogenic nature of the E848G mutation. Reduced contractility may result from impaired interaction between myosin heavy chain 7 and cardiac myosin binding protein C.

Cell viability mapping within long-term heart valve organ cultures.

BACKGROUND AND AIM OF THE STUDY: Organ cultures maintain cells within their native microstructural environment, and thus offer greater potential for studying tissue disease and remodeling than do monolayer cell cultures or pathological examinations of diseased tissue. To validate an in-vitro heart valve organ culture model, cell viability was examined within valve tissues over sustained culture periods. METHODS: Following culture of blocks of valve tissue for 1 to 49 days, cross-sections were cut with a vibratome, stained with a LIVE/DEAD kit, and imaged with confocal microscopy to quantify t

Bioinspired smart dentin ECM-chitosan hydrogels for dentin-pulp complex regeneration.

OBJECTIVES: Approximately two-thirds of the world's population suffers from dental caries, and clinical treatment of the resulting lesions varies from dental fillings to devitalization. Despite recent advances in regenerative endodontic therapy, scaffolds intended for the regeneration of the dentin-pulp complex with innate odontogenic potential and microarchitecture similar to these dental tissues are still scarce. Taking advantage of the physio-biological properties of demineralized dentin extracellular matrix (ECM), we developed a functional hydrogel that combines insoluble molecules of dent

A sustainable tissue-specific hydrogel from decellularized corneal ECM for 3D In vitro ocular models.

In vitro models are increasingly replacing animal testing due to their enhanced ability to mimic human physiology. In this study, we developed and characterized a hydrogel derived from decellularized extracellular matrix (C-dECM) of bovine corneas-a tissue-specific and sustainable biomaterial obtained from food industry waste. The hydrogel was produced through decellularization, enzymatic solubilization, and gelation, preserving key ECM components such as glycosaminoglycans and collagen. C-dECM hydrogels demonstrated stable gelation behavior (T50 = 51.6 ± 7.5 min), biocompatibility, and struct

Polydopamine-coated electroconductive nanofibrous scaffold with sustained NGF release for peripheral nerve regeneration.

Peripheral nerve injuries (PNIs) remain a major clinical challenge due to permanent functional impairment and limited effective therapeutic options. To address this, we developed a novel tissue-engineered nerve scaffold that integrates multiple regenerative elements to mimic a favorable microenvironment for nerve regeneration. The scaffold consists of electrospun aligned polycaprolactone (PCL) nanofibers for structural guidance of axonal growth, reduced graphene oxide (rGO) to provide electrical conductivity for enhanced neural signaling, and polydopamine (PDA) surface modification to improve

Collagen silver-doped hydroxyapatite scaffolds reinforced with 3D printed frameworks for infection prevention and enhanced repair of load-bearing bone defects.

Osteomyelitis, a severe bone infection, is an extremely challenging complication in the repair of traumatic bone defects. Furthermore, the use of long-term high-dose antibiotics in standard treatment increases the risks of antibiotic resistance. Herein, an antibiotic-free, collagen silver-doped hydroxyapatite (coll-AgHA) scaffold reinforced with a 3D printed polycaprolactone (PCL) framework was developed with enhanced mechanical properties to be used in the repair of load-bearing defects with antimicrobial properties as a preventative measure against osteomyelitis. The AgHA particles were fabr

From the sea to the bee: Gellan gum-honey-diatom composite to deliver resveratrol for cartilage regeneration under oxidative stress conditions.

Carbohydrate-based porous scaffolds are promising biomaterials to support cartilage regeneration. In this respect, their composition could be designed to face clinical challenges, i.e., articular load bearing, infections and oxidative stress. Herein, an innovative scaffold has been developed, combining raw materials belonging to different kingdoms of life. Indeed, gellan gum, a bacterial-derived carbohydrate, was blended with a beehive product (Manuka honey) with prominent antibacterial features. Moreover, resveratrol, a phytoalexin with powerful antioxidant activity, was loaded into the silic

Encapsulation of Mesenchymal Stem Cells in 3D Ovarian Cell Constructs Promotes Stable and Long-Term Hormone Secretion with Improved Physiological Outcomes in a Syngeneic Rat Model.

Loss of ovarian function (e.g., due to menopause) leads to profound physiological effects in women including changes in sexual function and osteoporosis. Hormone therapies are a known solution, but their use has significantly decreased due to concerns over cardiovascular disease and certain cancers. We recently reported a tissue-engineering strategy for cell hormone therapy (cHT) in which granulosa cells and theca cells are encapsulated to mimic native ovarian follicles. cHT improved physiological outcomes and safety compared to pharmacological hormone therapies in a rat ovariectomy model.
