Application of chitin and chitosan derivatives in the pharmaceutical field.

Chitin and chitosan derivatives are used as excipients and drug carriers in the pharmaceutical field. Their derivatization contributed to expansion of application and decrease toxicity. Chitosan is used as an excipient in oral dosage form. Chitosan tablet can exhibit a sustained drug release compared to commercial products. Films prepared using chitin or chitosan have been developed as wound dressings, oral mucoadhesive and water-resisting adhesive by virtue of their release characteristics and adhesion. Intratumoral administration of gadopentetic acid-chitosan complex nanoparticles (approxima

Trypanosoma cruzi carrying a targeted deletion of a Tc52 protein-encoding allele elicits attenuated Chagas' disease in mice.

The intracellular protozoan parasite Trypanosoma cruzi is the etiological agent of Chagas' disease. We have previously characterized a T. cruzi virulence factor named Tc52 sharing structural and functional properties with the thioredoxin and glutaredoxin protein family. Single mutant parasite clones (Tc52(+/-)) exhibiting low virulence in vitro and in vivo were obtained by targeted Tc52 gene replacement. In this report, we have extended our study to analyze the immune response and the disease phenotype in Tc52(+/-)-infected BALB/c mice, during the acute and chronic phases of the disease. Signi

Upregulation of expression from the FRDA genomic locus for the therapy of Friedreich ataxia.

BACKGROUND: Friedreich ataxia is a slowly progressive neurodegenerative disease caused by reduced expression of frataxin as a result of a GAA repeat expansion in the first intron of the FRDA gene. We report here the development of a sensitive cellular assay for frataxin expression from the intact FRDA locus that should facilitate the identification of potentially therapeutic pharmacological agents to treat Friedreich ataxia. METHODS: PAC and BAC clones containing the entire human FRDA functional genomic sequence were identified and shown to express FRDA mRNA. The GET Recombination system was

Na transport in autosomal recessive polycystic kidney disease (ARPKD) cyst lining epithelial cells.

Autosomal dominant (ADPKD) and recessive (ARPKD) polycystic kidney disease are characterized by the progressive growth and expansion of cysts or ectatic collecting ducts, respectively, that ultimately destroy the normal renal parenchyma. Evidence from experimental models of ADPKD suggests that transepithelial Na and fluid secretion contribute to cyst growth, yet little is known about solute transport in ARPKD. This purpose of this study was to begin to characterize the expression and polarity of transport proteins involved in vectorial Na movement in ARPKD epithelium. Immunodetectable alpha1 a

Confocal microscopic observations of the human cornea following overnight contact lens wear.

BACKGROUND: Striae and folds are observed with a slitlamp biomicroscope in the cornea following overnight contact lens wear. These phenomena … ens wear is an optical artefact that can be explained in terms of corneal oedema causing volumetric tissue expansion and a loss of optical clarity, which hampers keratocyte detection. These findings place the onus on researchers postulating a loss of stromal keratocytes following clinical interventions, such as contact lens wear, to account for the effects of oede …

T cell receptor BV gene usage in interstitial cellular infiltrates in active Heymann nephritis.

BACKGROUND: Infiltration of the kidney by mononuclear cells is a prominent feature of active Heymann nephritis (HN). These cells could be present as a part of generalized inflammatory response, or could be proliferating in response to specific antigens. To examine these questions, we have analysed the T cell receptor (TCR) BV repertoire of T cells infiltrating the renal interstitium at regular time intervals throughout the course of the disease. METHODS: HN was induced in Lewis rats by immunization with renal tubular antigen (Fx1A) in complete Freund's adjuvant (CFA). Kidneys were collected 8

Differentiation and expansion of beta cell mass in porcine neonatal pancreatic cell clusters transplanted into nude mice.

Neonatal porcine pancreas has considerable capacity for growth and differentiation, making it an attractive potential source of islet tissue for xenotransplantation. Pancreases from 1-3-day-old newborn pigs were digested with collagenase and cultured for 8 days. The resulting cellular aggregates are called porcine neonatal pancreatic cell clusters (NPCCs). The mean yield of NPCCs from a newborn pig was 28,200 +/- 1700 islet equivalents. Cytokeratin 7 (CK7) was used as a marker for the immunostaining of pancreatic duct cells. In neonatal pancreas, 18% of the insulin-positive cells co-stained fo

Increased tropoelastin and procollagen expression in the lung of nitrofen-induced diaphragmatic hernia in rats.

BACKGROUND/PURPOSE: Collagen and elastin, the predominant components of the lung connective tissue network, have been suggested to have an important influence on lung compliance and maximal expansion. Decrease in lung compliance and distensibility often is seen in human congenital diaphragmatic hernia (CDH) lung as well as in experimentally produced CDH lung. The aim of this study was to investigate mRNA levels of tropoelastin and alpha1 (I) procollagen, the precursors of elastin, and type I collagen, respectively, in CDH lung and to determine whether antenatal dexamethasone treatment has any

The hyaline zone and associated root surface changes in experimental orthodontics in rats: a light and scanning electron microscope study.

Light and scanning electron microscopic (SEM) examinations were used to study the hyaline zone and associated root resorption after orthodontic treatment of the upper first molars of rats. The orthodontic treatment consisted of a fixed buccal expansion appliance with an initial force of 250 mN. The animals were divided into nine experimental groups with orthodontic treatment and one control group without orthodontic treatment. Three groups were sacrificed immediately after 1, 3, and 7 days of treatment, and six groups after 7 days of treatment followed by 1, 2, 3, 4, 5, and 6 weeks without tre

The pressure/volume relationship of the calf: a measurement of vein compliance?

OBJECTIVES: The role of compliance changes in the patho-physiology of venous disease is not well known mainly because of difficulty to measure compliance of veins in situ. This study suggests a method to determine the calf pressure/volume relationship by utilizing venous occlusion plethysmography combined with dorsal vein pressure. DESIGN: Comparison between two techniques of measuring calf pressure/volume relationship using air plethysmography with validation against popliteal vein diameter changes detected by duplex ultrasound. SETTING Vascular laboratory. MATERIALS: In 6 normal and 6 radi

Intraluminal vapor bubble induced by excimer laser pulse causes microsecond arterial dilation and invagination leading to extensive wall damage in the rabbit.

BACKGROUND: Previous in vitro studies demonstrated that during excimer laser ablation of aortic tissue in saline, a fast-expanding and imploding vapor bubble is formed. The present in vivo study was designed to demonstrate the formation of a fast-expanding intraluminal bubble in flowing blood and to assess any damage to the adjacent arterial wall. METHODS AND RESULTS: Excimer laser pulses (one to 10, at 55 mJ/mm2 per pulse) were delivered coaxially in the femoral and iliac arteries of nine normal rabbits. Time-resolved flash photography of dissected arteries in situ demonstrated a 50% diamete

The Impact of GATA2 Expression and Methylation on Patients with Hirschsprung's Disease.

INTRODUCTION: Hirschsprung's (HIRSH-sproongz) disease is a multifactorial disorder characterized by the failure of enteric nervous system (ENS) development and is associated with loss-of-function variants, primarily in the RET gene. Rearranged during transfection (RET) expression is tightly regulated within a complex gene regulatory network (GRN) involving many transcription factors, such as GATA2. Aberrant DNA methylation in GATA2 may indicate a reduced enhancer activity, resulting in the silencing of RET and contributing to the failure of ENS development in HIRSH. However, the epigenetic mec

A Case of Massive Air Leakage After Lung Cancer Surgery Successfully Treated With the "Tachosuture" Technique and Intensive Management Under One-Lung Ventilation.

Massive air leakage after lung resection can be difficult to control, especially in patients with vulnerable lung tissues due to underlying diseases. TachoSil® (Corza Medical, Linz, Austria) patch is applied to the site of air leakage during surgery to control air leakage; however, in cases of massive air leakage, it may peel off before the tissue has healed. This report describes a method for securely positioning the TachoSil® patch at the site of air leakage to control massive air leakage after lung resection. The patient was a 50-year-old man with a history of heavy smoking and alcoholic li

NKG2A-HLA-E axis contributes to an immunosuppressive checkpoint network in hepatocellular carcinoma.

BACKGROUND: The immunosuppressive tumor microenvironment in hepatocellular carcinoma (HCC) limits the efficacy of current immunotherapies. The interplay between the emerging inhibitory receptor NKG2A and its activating counterpart NKG2C, both binding HLA-E, and its interaction with the PD-1-PD-L1 pathway in cancer remains unclear. This study aims to delineate the functional network formed by these pathways in shaping the immunosuppressive landscape of HCC. METHODS: We analyzed public databases and performed immunohistochemistry on HCC tissues to assess HLA-E expression and prognostic relevanc

The morphology of the subacromial and related shoulder bursae. An anatomical and histological study.

Shoulder bursae are essential for normal movement and are also implicated in the pathogenesis of shoulder pain and dysfunction. The subacromial bursa (SAB), within the subacromial space, is considered a primary source of shoulder pain. Several other bursae related to the subcoracoid space, including the coracobrachial (CBB), subcoracoid (SCB) and subtendinous bursa of subscapularis (SSB), are also clinically relevant. The detailed morphology and histological characteristics of these bursae are not well described. Sixteen embalmed cadaveric shoulders from eight individuals (five females, three

Scid mouse model of psoriasis: a unique tool for drug development of autoreactive T-cell and th-17 cell-mediated autoimmune diseases.

In both skin and synovial tissues of psoriatic arthritis (PsA) patients, there are prominent lymphocytic infiltrates localized to the dermal papillae in the skin and the sublining layer stroma in the joint. T-cells, with a predominance of CD4+ lymphocytes, are the most significant lymphocytes in the tissues; in contrast, this ratio is reversed in the epidermis, synovial fluid compartment, and at the enthesis, where CD8+ T-cells are more common. This differential tropism of CD8+ T-cell suggests that the CD8+ T-cells may be driving the immune response in the joint and skin. This is supported by
