Acute Myocarditis: From Pathophysiology to Risk-Stratified Management-A Clinical Review.

BACKGROUND: Acute myocarditis is an inflammatory disease of the myocardium with an annual incidence of 4-14 per 100,000 individuals, predominantly affecting young adults. Its clinical features are frequently nonspecific, mimicking acute coronary syndrome, which makes early recognition and management challenging. The disease results from infectious (predominantly viral) and noninfectious triggers, including autoimmune disorders, immune checkpoint inhibitors and mRNA vaccines. Pathophysiology involves a dysregulated interplay between innate immunity and adaptive immunity. CLINICAL FEATURES AND

Systematic comparison of temporal hepatotoxicant-induced gene network responses across 3 liver test systems.

Drug-induced liver injury (DILI) arises from dynamic and time-dependent cellular stress responses that remain insufficiently captured by conventional single-timepoint toxicogenomic assessments. We systematically characterized temporal and concentration-dependent transcriptomic responses to the clinically relevant hepatotoxicants ketoconazole, diclofenac, and nitrofurantoin across 3 human liver in vitro models: primary human hepatocytes (PHH), hiPSC-derived hepatocyte-like cells (HLC), and HepG2 cells. Time-resolved RNA sequencing (0 to 48 h) combined with likelihood ratio testing identified ti

SUMOylation in skin homeostasis and disease: Regulation of cutaneous signaling networks.

SUMOylation is a reversible post-translational modification that regulates protein stability, subcellular localization, molecular interactions, and transcriptional activity. The skin is constantly exposed to ultraviolet radiation and other environmental stressors, yet it must maintain barrier function and restore tissue homeostasis. This review examines how SUMOylation contributes to cutaneous stress responses by linking cellular damage, inflammation, and tissue repair. By altering the fate and function of stress-responsive proteins, SUMOylation shapes the strength and duration of damage respo

Construction of biomimetic hydroxyapatite bilayer scaffolds via DLP 3D printing for bio-root regeneration.

Digital light processing (DLP) 3D printing of hydroxyapatite (HA) bioceramics offers a promising strategy for fabricating complex, patient-specific bio-root scaffolds with high resolution and bioactivity. However, achieving adequate mechanical integrity while guiding the formation of a periodontal ligament (PDL)-like enthesis through structural optimisation remains a major challenge. Inspired by interfacial micromorphological cues that regulate both intrinsic mechanical properties and cellular responses, this study proposed a novel bone-root integrated design for a DLP 3D-printed bio-root scaf

The lung-muscle axis in COPD: a conceptual framework integrating evidence, mechanisms, and therapeutic perspectives.

Skeletal muscle dysfunction (SMD) is a clinically important extrapulmonary manifestation of chronic obstructive pulmonary disease (COPD) and is associated with impaired exercise capacity, reduced physical function, increased exacerbation risk, and poorer clinical outcomes. Although traditionally viewed as a consequence of pulmonary disease progression, accumulating evidence suggests that pulmonary impairment and skeletal muscle abnormalities may reflect partially overlapping processes within the systemic pathobiology of COPD. This narrative review synthesizes clinical, imaging, genetic, mechan

Egg Disease in Spodoptera litura (Fabricius) Caused by Aspergillus ochraceus and Its Metabolic Phenotype.

A pathogenic fungus was isolated from unhatched egg masses of Spodoptera litura (S. litura). The pathogen was identified as Aspergillus ochraceus based on morphological characterization and molecular analyses of the internal transcribed spacer (ITS) region and the β-tubulin gene. The isolate exhibited strong pathogenicity toward S. litura eggs, causing 100% mortality. Phenotypic characteristics were further evaluated using the BIOLOG Phenotype MicroArray (PM) system. A total of 950 distinct growth conditions were assessed across PM plates 1-10. The results indicated that A. ochraceus metaboliz

Heterogeneity of Immunological, Coagulation and Complement Clustering to Viral Challenge and Clinical Relevance.

Differences in immune responses affect viral illness outcomes, but the combined contributions of inflammation, coagulation, and complement pathway heterogeneity to disease stratification remain unclear. We analyzed infectious, immunological, coagulation, and complement responses at admission and assessed their associations with cerebrovascular and coagulation comorbidities. Ninety-six PCR-confirmed SARS-CoV-2 patients were sampled at <24 h (T1), 48 h (T2), and 7 days (T3). COVID-19-specific immunity (Spec-IMM domain) was measured by plasma IgG, IgM, and IgA against S and N proteins; nonspecifi

Comparing the molecular correlates of PTSD in first responders and the broader molecular PTSD literature: a hybrid review.

First responders are trained civilian professionals, including firefighters, law enforcement, emergency medical services, and frontline medical workers, who have a vocational duty and are expected to respond in person to emergency situations. The chronic exposure to adverse traumatic events while working with public health and safety can lead to the clinical presentation of post-traumatic stress disorder (PTSD), which also occurs in other trauma-exposed populations such as military personnel, veterans, refugees, and individuals who experience interpersonal violence. Examining epigenetic and tr

Chronic toxicity of triazine-based and non-triazine ultraviolet filters toward Chlorella vulgaris: Low ecological risk of triazine-based compounds.

Low-dose chronic toxicity can induce sublethal biological effects, which is of great significance for assessing the ecological toxicity of UV filters. In this study, we investigated the chronic toxicity of three triazine-based UV filters including bis-ethylhexyloxyphenol methoxyphenyl triazine (BEMT), diethylhexyl butamido triazone (DBT), and ethylhexyl triazone (EHT) and three non-triazine UV filters: diethylamino hydroxybenzoyl hexyl benzoate (DHHB), avobenzone (AVB), and octocrylene (OCR) on Chlorella vulgaris. Results indicated that the triazine-based UV filters exerted no statistically si

Infection, inflammation and immune dysregulation: evolving perspectives on the host response.

Host inflammatory responses contribute substantially to infection-related morbidity, and in severe cases such as sepsis, acute respiratory distress syndrome (ARDS) and haemophagocytic lymphohistiocytosis, immune-mediated tissue injury may exceed the direct effects of pathogens themselves. Although traditional wisdom has considered immunosuppression risky when antimicrobial therapy is required, modern insights into immune biology reveal a more nuanced landscape in which targeted immunomodulators can attenuate harmful inflammation without broadly compromising host defence. This evolution is part

Management of a Severe Traumatic Intrusion of a Permanent Incisor With Spontaneous Re-Eruption After Maxillary Expansion: A Surgical-Orthodontic Case Report.

BACKGROUND Traumatic intrusion of permanent incisors is one of the most severe forms of dental injury and is frequently associated with complications such as pulp necrosis, root resorption, and ankylosis. Although IADT guidelines provide structured recommendations based on the stage of root development and degree of intrusion, some clinical situations remain borderline, particularly cases presenting an open apex in an otherwise nearly mature root, making classification and treatment planning challenging. CASE REPORT A 7-year-old girl presented with severe intrusive luxation (>10 mm) of the per

Investigation of the Effect of TiO2 as a Dietary Marker on Broiler Intestinal Fermentation: Combination of Ex Vivo Simulation and In Vivo Approach.

The impact of dietary inert digestibility markers on gut microbiota and intestinal fermentation remains poorly understood. This study investigated the effects of dietary titanium dioxide (TiO2) supplementation at 4 kg/t feed, representing a typical dose used in animal nutrition studies, on fermentation dynamics and microbial composition in broiler chickens using combined ex vivo and in vivo approaches. Ex vivo fermentations were conducted using ileal and caecal microbiota and substrates collected from 32-day-old broiler chickens. Titanium dioxide (TiO2) was supplemented directly to the ferment

Predisposition to ALL and Solid Tumors Rather Than Bone Marrow Failure in FANCM-Associated Fanconi Anemia.

PURPOSE: Fanconi anemia (FA) is a genetic disorder typically characterized by progressive bone marrow failure (BMF) during childhood, leading to diagnosis at that stage. In adolescence or adulthood, patients are predisposed to myelodysplastic syndrome (MDS), acute myeloid leukemia, and solid tumors. However, some individuals present atypically, delaying FA recognition and resulting in life-threatening complications. This study describes the distinctive phenotype associated with biallelic FANCM pathogenic variants. PATIENTS AND METHODS: Clinical and biologic data were analyzed from eight patie

A comprehensive mechanistic multicellular model of the human immune system spanning 11 diseases.

INTRODUCTION: The immune system is a dynamic, complex network of interacting cells, molecules, and signals central to health. Immune digital twins - virtual representations of the immune system that evolve in tandem with their biological counterparts - offer a path toward predictive, patient-specific simulations, but their realization requires mechanistic frameworks that generalize across multiple cell types and disease contexts. METHODS: We constructed a logic-based mechanistic model of the human immune system built exclusively on human experimental data extracted from 449 publications. The

Tissue-resident memory T cells in atopic dermatitis: mechanisms of residual inflammation, relapse, and therapeutic persistence.

BACKGROUND: Atopic dermatitis (AD) is a chronic, relapsing inflammatory skin disease in which disease flares frequently recur at previously affected anatomical sites. This distinctive clinical pattern suggests the presence of residual inflammation and long-lasting local immune memory that persists beyond apparent clinical remission. OBJECTIVES: To synthesize current evidence on the role of tissue-resident memory T (TRM) cells in the pathogenesis, chronicity, and relapse of AD, and to discuss how TRM biology may explain disease recurrence after treatment withdrawal and inform future therapeuti
