Snake venom: From fieldwork to the clinic: Recent insights into snake biology, together with new technology allowing high-throughput screening of venom, bring new hope for drug discovery.

Snake venoms are recognized here as a grossly under-explored resource in pharmacological prospecting. Discoveries in snake systematics demon … is threatening the very snake populations on which hopes for new venom-derived medications depend. Biomedical researchers, pharmacologists, clinicians, herpetologists, and conservation biologists must combine their efforts if the full potential of snake venom-derived medications is to be realized.

[Efficacy of electroconvulsive therapy: a systematic review of scientific evidences].

We carried out a systematic study of bibliographical review of scientific evidence provided by clinical trials that assessed the short, medium and long-term efficacy of electroconvulsive therapy (ECT) from 1965 until June 2003. The studies with the following features have been excluded: a) those in which ECT is not the aim of the research; b) those that do not compare ECT with another different treatment; c) those in which the aim of the research is not to evaluate the efficacy of ECT, and d) those in which the studies are not randomized clinical trials. We have used the biomedical databases M

Implementing broad consent for research with routinely collected clinical data and residual biosamples in a cancer hospital: using mixed methods approach to evaluate consent rates and patients' perspectives.

BACKGROUND: Patients are generally willing to contribute to research with routinely collected health data and residual biosamples, but transparency and being able to (to some extent) have control over data are important conditions. A broad consent procedure ensures that patients are informed, without overloading the patients with too many or repeated study-specific consents. In the context of implementation of broad consent, we investigated five aspects: response rates, whether patients felt informed and were able to reach a decision, whether the registered consent was in line with their desir

Establishment of Uveal Melanoma Patient-Derived Organoids Using an Air-Liquid Interface System for Translational Research.

PURPOSE: Uveal melanoma (UM) is the most common primary intraocular malignancy in adults, with a unique genetic and epigenetic profile and a high metastatic risk. The lack of experimental models to replicate tumor biology, including its epigenetic landscape, limits progress in translational research. Recently, patient-derived organoid (PDO) models have emerged as a promising tool for the modeling of various tumors, including UM. This study aims to establish an air-liquid interface (ALI) PDO culture system for UM, evaluate its similarity to the primary tumors, and assess its utility for drug sc

Targeting Immunologic Pathways in Eosinophilic Granulomatosis With Polyangiitis: Translating Emerging Evidence Into Clinical Practice.

Eosinophilic granulomatosis with polyangiitis (EGPA) is a rare and potentially life-threatening systemic, inflammatory disease with multi-organ manifestations, variable presentation and complex pathology. Multiple interconnected immunological pathways are implicated in EGPA pathology, including a type-2 immune response driving predominantly eosinophilic inflammation, B-cell mediated autoimmunity, neutrophil activation, and the generation of pathogenic anti-neutrophil cytoplasmic antibodies, all of which can contribute to tissue/organ damage. High-dose glucocorticoids are the mainstay treatment

Translational framework combining machine learning and in vivo screening for aminoglycoside ototoxicity prevention.

Aminoglycosides are potent antibiotics, but their clinical use is limited by ototoxicity. Traditional drug screening is labor-intensive and inefficient in identifying safer alternatives. To address this, we developed a rapid screening framework combining a graph attention-based transformer model with zebrafish-based validation. Due to the limited availability of ototoxicity-specific datasets, the model was trained using handcrafted labels derived from extensive literature review, along with transfer learning techniques. It was then used to screen FDA-approved drug libraries, prioritizing candi

The NLRP3 inflammasome in chronic kidney disease progression: systematic review of preclinical models reveals a critical gap in pyroptosis research and therapeutic translation.

Chronic kidney disease (CKD) affects ~ 850 million people globally. The NOD-like receptor protein 3 (NLRP3) inflammasome has emerged as a central regulator of inflammatory, fibrotic, and mitochondrial injury during CKD pathogenesis. Nevertheless, its consistency across disease models and translational status has not been systematically evaluated to date. We conducted a PubMed and Scopus search for research articles examining NLRP3 inflammasome activation across various CKD models, following PRISMA guidelines, and selected 17 animal studies for further analysis. We found that across all model t

Silent Myocardial Infarction Revisited: Immuno-metabolic Mechanisms, Multimodal Biomarkers, and Translational Diagnostics.

Silent Myocardial Infarction (SMI) is a clinically underrecognized phenotype along the myocardial infarction continuum that progresses without anginal symptoms. Its prevalence in diabetes, chronic kidney disease, and the elderly reflects contributions from neuropathy, autonomic dysfunction, and neurogenic silencing. Emerging evidence indicates that SMI reflects a biologically biased phenotype within the myocardial infarction continuum shaped by immune-metabolic and neurogenic modulation rather than representing a distinct entity. Biomarkers such as sCD36, galectin-3, sST2, and GDF-15 capture f

From bench to bedside: the untold story behind the clinical development of enoxaparin.

The path to the development of enoxaparin (Lovenox/Clexane), one of the most widely used low-molecular-weight heparins worldwide, was far from smooth. This narrative review, presented from the personal perspective of the senior author who lived this journey, describes the pivotal research conducted at McMaster University in Canada that transformed enoxaparin from a promising laboratory compound into a cornerstone of prevention and treatment of thrombosis. The story begins in the late 1970s with the observation that low-molecular-weight heparin, produced through controlled depolymerization, exh

Strengthening microbial genomics capacity in Africa for epidemic preparedness: key lessons from the AFROSCREEN programme.

The AFROSCREEN initiative, implemented in 13 countries, has strengthened regional genomic surveillance through investments in sequencing infrastructure, workforce training, and cross-country coordination, evolving from a SARS-CoV-2-focused effort into a multi pathogen platform. By building local capacity, AFROSCREEN reduced dependence on external laboratories, helped close critical surveillance gaps, and generated genomic data that could inform mitigation of national health risks. However, persistent structural challenges related to inequity, including procurement delays, limited bioinformatic

A Narrative Review of the Regional Prospective Observational Research in Tuberculosis International Consortium: Sharing Diverse Data and Specimens in the Fight Against Tuberculosis.

BACKGROUND: Clinical tuberculosis (TB) research has been hampered by a lack of reliable predictors of Mycobacterium tuberculosis infection progression and active TB treatment's cure, relapse, and failure. Given host and bacterial variability, large sample sizes from harmonized datasets are needed to study infrequent endpoints. Therefore, global observational research consortia, such as Regional Prospective Observational Research in Tuberculosis (RePORT) International (RI), can help make meaningful inroads into successful TB prevention, diagnosis, and treatment. METHODS: RePORT International w

SGLT2 inhibitors in translational medicine: A paradigm shift for diabetes and heart health.

Diabetes mellitus (DM) is a major contributor to a number of catastrophic health disorders, including heart attacks, strokes, kidney failure, blindness, and lower limb amputations. In 2023, DM was recognized by the World Health Organization (WHO) as a major cause of death. Diabetes is becoming more and more common, and any family member could be impacted. Due to the limited availability of current antidiabetic drugs, the urgent need for new active pharmaceutical ingredients becomes clear. In recent decades, sodium-glucose cotransporter-2 (SGLT2) has drawn interest as a key target for type 2 di

Challenges in clinical translation of artificial intelligence and real-time imaging navigation in radical gastrectomy.

Radical gastrectomy for gastric cancer demands meticulous pre-operative staging and real-time intra-operative guidance to optimise oncologic margins and minimize complications. Recent advances in artificial-intelligence algorithms reliably integrate multimodal clinical, imaging and pathological data, producing highly reproducible tumour-staging and risk-stratification models that inform personalised operative strategies. Concurrently, navigation platforms that fuse computed-tomography, magnetic-resonance, ultrasound and fluorescence datasets generate patient-specific three-dimensional reconstr

Transient receptor potential channels in hair follicle biology: physiological significance and translational relevance.

Transient receptor potential (TRP) channels are a family of versatile ion channels that act as sensor for a wide range of chemical and physical stimuli. These cation-permeable channels are widely expressed in various tissues and play pivotal roles in numerous biologic processes. Studies have demonstrated that six specific TRP channels -TRPV1, TRPV3, TRPV4, TRPM5, TRPML1 and TRPML3 - are actively involved in hair follicle (HF) cell proliferation, modulation of hair growth-related signalling pathways, regulation of hair cycle progression, and control of hair growth and pigmentation in humans and

Suzetrigine, a Non-Opioid Small-Molecule Analgesic: Mechanism of Action, Clinical, and Translational Science.

The discovery and approval of Suzetrigine (VX-548, Journavx) marks a significant breakthrough in pain management. It is the first non-opioid analgesic approved since celecoxib in 1998. Suzetrigine selectively blocks voltage-gated sodium channel Nav1.8 and acts exclusively on peripheral nociceptors without crossing the blood-brain barrier, providing analgesia while sparing central nervous system side effects such as dependence, addiction, sedation, and respiratory depression. In vitro experiments have demonstrated that Suzetrigine is a state-dependent inhibitor with nanomolar potency against hu

Translating animal models of SARS-CoV-2 infection to vascular, neurological and gastrointestinal manifestations of COVID-19.

Since the emergence of severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) initiated a global pandemic resulting in an estimated 775 million infections with over 7 million deaths, it has become evident that COVID-19 is not solely a pulmonary disease. Emerging evidence has shown that, in a subset of patients, certain symptoms - including chest pain, stroke, anosmia, dysgeusia, diarrhea and abdominal pain - all indicate a role of vascular, neurological and gastrointestinal (GI) pathology in the disease process. Many of these disease processes persist long after the acute disease has bee
