Polycomb targeted loci form long-range chromatin interactions independent of CTCF-cohesion and are demarcated by low DNA methylation(1-3). Polycomb targets form extremely long-range loops (long Polycomb loops) that can separate anchors up to 60 Mb. These loops predominantly occur in cells of self-renewal status, such as human hematopoietic stem cells (HSC) and mouse embryonic stem cells (ESC), but rarely in cell lines. To identify long Polycomb loops in cancer, we initiated a pan-cancer survey of long Polycomb loops in a collection of 264 tumor samples (33 acute myeloid leukemias, 17 T cell ly

BACKGROUND: HIV-1-associated neurocognitive impairment (HIV-1-NCI) is marked by ongoing and chronic neuroinflammation with loss and decline in neuronal function even when antiretroviral drug therapy (ART) successfully suppresses viral replication. Microglia, the primary reservoirs of HIV-1 in the central nervous system (CNS), play a significant role in maintaining this neuroinflammatory state. However, understanding how chronic neuroinflammation is generated and sustained by HIV-1, or impacted by ART, is difficult due to limited access to human CNS tissue.
METHODS: We generated an in vitro mo

Diverse physiological phenotypes in a neuronal population can broaden the range of computational capabilities within a brain region. The avi … urons, we applied dendrotoxin-I (DTX), a potent antagonist of Kv1-type potassium channels, to chick brain stem slices in vitro during whole cell patch-clamp recordings. We found a cell-type specific subset of NA neurons that was sensitive to DTX: single-spiking NA neurons were most profoundly affected, as well as a subset of tonic-firing neurons. Both tonic …

We have shown that vasopressinergic projections to dorsal brain stem are activated during exercise and facilitate exercise tachycardia in both trained (T) and sedentary (S) rats (Dufloth DL, Morris M, and Michelini LC. Am J Physiol Regulatory Integrative Comp Physiol 273: R1271-R1282, 1997). In the present study, we investigated whether oxytocinergic projections to the nucleus of the solitary tract (NTS)-dorsal motor nucleus of the vagus (DMV) complex (NTS/DMV) are involved in the differential heart rate (HR) response to exercise in T and S rats. Arterial pressure (AP) and HR responses to dyna…

OBJECTIVES: This study aimed to develop a MRI-based prognostic nomogram including radiomics and clinical information for brainstem cavernous malformation patients (BSCMs).
METHODS: 114 BSCMs were randomly divided into a training cohort and a validation cohort. Clinical and radiomics nomograms were constructed. Radiomics features were selected using three algorithms: univariate analysis, Pearson correlation, and the elastic net algorithm. A Cox regression model was employed to build the radiomics nomogram. The concordance index (C-index), time-independent receiver operating characteristic (ROC

Intraoperative neurophysiological monitoring (IONM) is widely used during brainstem surgery to reduce the risk of neurological injury, although its prognostic value in adult brainstem glioma (BSG) surgery remains unclear. We retrospectively analyzed 22 consecutive adults who underwent surgery for BSGs with multimodal IONM between 2010 and 2023. Monitoring included somatosensory evoked potentials (SSEPs), motor evoked potentials (MEPs), brainstem auditory evoked potentials (BAEPs), corticobulbar MEPs (CoMEPs), and cranial nerves electromyography. The primary endpoint was the association between

In utero nicotine exposure from maternal smoking is linked to increased risk of auditory processing deficits. This study investigated the impact of developmental nicotine exposure during the critical period on nicotinic acetylcholine receptor (nAChR) functional expression, glutamatergic synaptic transmission and auditory processing in the mouse auditory brainstem. We assessed nAChR function at a central synapse and the consequences of perinatal nicotine exposure (PNE) on synaptic currents and auditory brainstem responses (ABRs) in mice. Our results indicate developmentally regulated changes in

In nonlinear systems, the inclusion of low-level noise can paradoxically improve signal detection, a phenomenon known as stochastic resonance (SR). SR has been observed in human hearing whereby sensory thresholds (e.g., signal detection and discrimination) are enhanced in the presence of noise. Here, we asked whether subcortical auditory processing (neural phase locking) shows evidence of SR. We recorded brainstem frequency-following-responses (FFRs) in young, normal-hearing listeners to near-electrophysiological-threshold (40 dB SPL) complex tones composed of 10 iso-amplitude harmonics of 150

OBJECTIVE: Electrically evoked auditory brainstem responses (EABR) guide placement of the multichannel auditory brainstem implant (ABI) array during surgery. EABRs are also recorded under anesthesia in nontumor pediatric ABI recipients prior to device activation to confirm placement and guide device programming. We examine the influence of anesthesia on evoked response morphology in pediatric ABI users by comparing intraoperative with postoperative EABR recordings.
STUDY DESIGN: Retrospective review.
METHODS: Seven children underwent ABI surgery by way of retrosigmoid craniotomy. General ane

The dichotic frequency following responses (FFR) have been used in studies to infer about dichotic auditory processing. In the present study, we hypothesize that the proximity of the binaural neural generators of the FFR would result in interference of the volume-conducted electrical fields. This might lead to contamination of the scalp-recorded dichotic FFRs due to which it might be difficult to infer about true dichotic processing in the putative neural generators. We investigated this by recording FFRs to binaurally presented 200 Hz pure tone with graded dichotic phase offsets (0°, 90°, 180

The purpose of the present study was to compare auditory brainstem responses (ABRs) using two approaches that allow the use of high stimulation rates, but with different temporal variability in the interstimulus interval: maximum length sequences (MLS) and iterative randomized stimulation and averaging (I-RSA). ABRs were obtained to click stimuli in six bottlenose dolphins (Tursiops truncatus). In experiment 1, click level was held constant and click rate varied from 25 to 1250 Hz. For MLS, interstimulus intervals varied by a factor of 6 at each rate, while for I-RSA the interstimulus interval

Decompressive craniectomy (DC) is used for the management of refractory raised intracranial pressure, but the impact of DC on surgical outcome is still controversial. We report a 21-year-old man admitted to our hospital after a road traffic accident. The brain CT scan revealed a left hemispheric acute subdural hematoma. After DC, he developed a brainstem hemorrhage. Recovery was, however, good.

Auditory brainstem response (ABR) testing is less sensitive in detecting small vestibular schwannomas than medium-size tumors. Magnetic resonance imaging (MRI) is more sensitive than ABR alone for small and large tumors, but it carries with it increased cost and issues of unavailability and patient discomfort. We conducted a prospective pilot study of 7 patients with untreated MRI-proven, unilateral vestibular schwannoma to determine if we could increase the sensitivity of ABR testing in detecting small tumors. Our method involved the use of a new ABR index that is based on threshold differenc
