Child mental health in the Americas: a public health approach.

The systematic, population-wide application of preventive measures based on what is known about the causes and outcomes of psychiatric disorders can markedly reduce morbidity from mental ill health among children in the Americas. The actions proposed here rely partly upon increasing access for all women and their children to thoroughly tested obstetric and pediatric care; in part they depend on improving nutrition and opportunities for cognitive stimulation; and in part they call for enhancing the mental health skills of primary care practitioners by appropriate in-service training. There are

Cognitive-behavioral treatment with and without response prevention for bulimia.

We compared cognitive-behavior therapy (CBT) with and without exposure and response prevention (ERP) in the treatment of eating disorder patients who both binged and purged, and reported abnormal attitudes concerning body weight and shape. Both treatments produced significant and comparable reductions in binge-eating and purging, eating patterns, and attitudes about weight and shape at posttreatment. Treatment effects were generalized to improvements in different measures of general psychopathology, and were maintained over follow-ups of 3 and 12 months. The findings are consistent with prior

The Effect of Psychedelics on Associative Learning: a Systematic Review.

Psychedelics are emerging as potential treatments for neuropsychiatric conditions, with evidence suggesting a single administration can lead to enduring behavioural changes. While the underlying putative mechanism(s) remain unclear, there is evidence supporting altered learning as a key candidate. This systematic review examined studies assessing the effects of psychedelics on associative learning in humans and animals. Electronic databases were searched until 02/02/2026 for studies investigating differences in learning after classical psychedelic administration. 62 studies were included (2 hu

Two Liters a Day Keep the Doctor Away? Considerations on the Pathophysiology of Suboptimal Fluid Intake in the Common Population.

Suboptimal fluid intake may require enhanced release of antidiuretic hormone (ADH) or vasopressin for the maintenance of adequate hydration. Enhanced copeptin levels (reflecting enhanced vasopressin levels) in 25% of the common population are associated with enhanced risk of metabolic syndrome with abdominal obesity, type 2 diabetes, hypertension, coronary artery disease, heart failure, vascular dementia, cognitive impairment, microalbuminuria, chronic kidney disease, inflammatory bowel disease, cancer, and premature mortality. Vasopressin stimulates the release of glucocorticoids which in tur

Executive functions in stuttering across the life span: A scoping review of behavioral and caregiver-reported measures.

Developmental stuttering is a multifactorial speech disorder that emerges during early childhood and can persist into adulthood, affecting communication, social participation, and quality of life. Recent research has increasingly focused on the role of executive functions (EFs)-including working memory, inhibitory control, and cognitive flexibility-in the onset and persistence of stuttering. This scoping review systematically maps the existing literature on EFs in monolingual individuals who stutter across the lifespan, synthesizing findings from 89 studies published between 1993 and 2024. A systematic review of psychological variables associated with juvenile primary fibromyalgia syndrome: Comparison between individuals with and without this condition.

Juvenile primary fibromyalgia syndrome (JPFS) is a pediatric rheumatologic condition characterized by (1) persistent, widespread musculoskeletal pain in multiple locations and (2) other symptoms such as fatigue, headaches, poor sleep or cognitive symptoms, among others. These alterations may be associated with the development of psychological distress, poorer mental health and lower quality of life, although no clear consensus has been reached. Thus, this study aimed to examine the psychological symptoms associated with JPFS, compared with a healthy comparison group of children and adolescents

Virtual Reality as a Platform for Upper-Limb Prosthetic Control Modes Evaluation and Early-Stage Design.

This study investigated the potential of using virtual reality (VR) as a platform for early-stage design of upper-limb prostheses as well as evaluation with a focus on performance, cognitive workload and usability. Three prosthetic device control modes (Direct Control, DC; Pattern Recognition, PR; and Continuous Control, CC) were compared across physical device (PD) and VR settings. Results indicated that task performance was generally lower in VR than in PD for DC and CC modes, likely due to the reduction of haptic cues and stricter spatial-alignment requirements in the VR interaction setting

Pet-induced stress buffering depends on touch: Evidence from brain and cardiac measures.

While there is much evidence for the wellbeing benefits of pet-ownership, the mechanisms through which these benefits emerge have been left uncertain. Here, we pursued the possibility that friendly physical contact with one's pet dampens stress and furthers homeostasis. Sixty-four pet owners visited our lab with their dog. They completed two experimental blocks each comprised of a resting baseline, a stress-inducing cognitive task, and a recovery period. The dog was present only during recovery, with physical contact being possible during one block but not the other. Relative to rest, the cogn

Intranasal administration of neural stem cell-derived extracellular vesicles prevents cognitive decline in both male and female 3×Tg-AD mice by dampening neuroinflammation and epigenetically regulating amyloid β metabolism.

BACKGROUND: Alzheimer’s disease (AD) is the leading cause of dementia in the elderly and poses a significant socioeconomic burden due to its progressive nature and lack of effective treatments. Recent studies suggest that neural stem cell-derived extracellular vesicles (NSC-EV) hold therapeutic potential against AD by delivering bioactive molecules that counteract neuroinflammation, oxidative stress, and protein dysregulation. METHODS: NSC-EV were intranasally administered to both male and female 3×Tg-AD mice from three to twelve months of age and cognitive function were evaluated at multiple

Wasf1 deletion attenuates tau hyperphosphorylation, microglial state transition, and cognitive deficits in P301S tau mice.

We previously reported that WAVE1, a major activator of Arp2/3 complex-mediated actin polymerization, is downregulated in postmortem brains of Alzheimer's disease (AD) and that WAVE1 regulates amyloid precursor protein trafficking and amyloid-β production. However, its role in tau pathology remains unknown. Here, we demonstrate that WAVE1 activity is suppressed in P301S tau mice through elevated inhibitory phosphorylation. Strikingly, WAVE1 gene ( Wasf1 ) knockout in P301S tau mice significantly reduces tau hyperphosphorylation and improves cognition, suggesting a compensatory role for WAVE1 s

Microglia TFEB activation attenuates Alzheimer's disease pathology by enhancing autophagy-lysosomal function.

Alzheimer's disease (AD) is characterized by amyloid-β (Aβ) accumulation, neuroinflammation, synaptic dysfunction, and cognitive decline. Impairment of microglial autophagy-lysosomal pathway (ALP) is increasingly recognized as a key driver of the disease progression. Transcription factor EB (TFEB), a master regulator of ALP, has emerged as a promising therapeutic target; however, its specific role in microglia remains unclear. Here, we aimed to determine the therapeutic effects of microglial TFEB expression in AD pathogenesis. We established a tamoxifen-inducible, microglia-specific TFEB-overe

Next generation preventive neurology: how artificial intelligence and machine learning are reshaping Alzheimer's disease research.

A neurological condition that worsens over time, Alzheimer’s disease (AD) is typified by memory loss, cognitive decline, and functional degradation. Traditional diagnostic techniques such as neuroimaging, cerebrospinal fluid biomarkers, and neuropsychological testing are often intrusive, costly, or insensitive in the early stages. Recent years have seen the emergence of AI and ML as game-changing technologies for AD risk assessment, early detection, and customized prevention. Using sophisticated models such as deep learning, convolutional neural networks (CNNs), and graph-based algorithms, AI-

Microglial OTUD6A promotes neuroinflammation and Alzheimer's disease pathogenesis by deubiquitinating C/EBPβ.

Alzheimer's disease (AD) is a progressive neurodegenerative disorder characterized by β-amyloid (Aβ) deposition, neuroinflammation, and cognitive decline. Microglia, the brain's primary immune cells, play a central role in AD pathogenesis by driving neuroinflammatory responses. Deubiquitinating enzymes (DUBs) regulate microglial activation, but the role of the ovarian tumor domain-containing DUB OTUD6A in AD remains unclear. In this study, we demonstrate that OTUD6A is upregulated in microglia across multiple AD models, including Aβ-infused mice, 3 ×Tg mice, and APP/PS1 mice. Otud6a KO or micr

AMPK Activator MK-8722 Improves Spatial Memory Deficit and Alters Metabolome in APP/PS1 AD Mouse Model.

Alzheimer's disease (AD) involves early synaptic vulnerability and systemic metabolic dysfunction, yet the impact of peripheral metabolic state on brain function remains unclear. Previous studies indicate a link between AMP-activated protein kinase (AMPK) signaling dysregulation and AD pathophysiology. Here we tested whether systemic treatment of a potent pan-AMPK activator MK-8722 influences cognitive function, synaptic plasticity, and circulating metabolic profiles in APP/PS1 AD model mice at the age of 6-8 months. MK-8722 was well tolerated and activated AMPK in the liver but not in the hip

Cross-region neuron co-firing mediated by ripple oscillations supports distributed working memory representations.

High-frequency (~90-Hz) ripple oscillations may promote integrative processing in mammalian brains. Co-occurrence of ripple oscillations has been associated with enhanced temporal binding of neural activity between nearby human cortical neurons, but whether co-ripple facilitation of neuronal coupling supports cognitive processing or occurs at greater distances remains unclear. Here we analyze intracranial recordings from patients implanted with microwire electrodes in the hippocampus, amygdala, ventromedial prefrontal cortex, anterior cingulate cortex and pre-supplementary motor area, bilatera

Social physical activity and sedentary behaviour as key determinants of humour expression and loneliness in older adults: a cross-sectional study using bayesian variable selection approach.

OBJECTIVE: Social isolation among older adults is a growing concern, partially owing to diminished social cognitive function. Physical activity can potentially enhance social cognitive function. This study focused on humour as an important facilitator of social interaction and examined how physical activity was associated with humour expression and loneliness. METHODS: This study included 5,212 Japanese older adults. We examined the associations between physical activity patterns (past and present exercise experiences, physical activity levels, sedentary behaviour), humour-related outcomes (hu
