Modulation of RASD2 by miRNA-485-5p Drives Thyroid Cancer Progression and Metastasis.

This study investigates the role of RASD2 (Ras Homolog Enriched In Striatum) in thyroid carcinoma progression and its modulation by microRNA-485-5p. Differential RASD2 expression patterns were initially identified through bioinformatic analysis of public databases. Immunohistochemical staining and quantitative reverse transcription PCR (qRT-PCR) validated these findings in clinical specimens and cell lines. Functional characterization of RASD2 was performed through loss-of-function studies, examining cellular proliferation, invasion, and glycolytic parameters. The prognostic significance of RA

Cultural and Molecular Factors Predisposed to Non-Alcoholic Fatty Liver Disease and Type 2 Diabetes Mellitus.

There is an exponential increase in the global prevalence of non-alcoholic fatty liver disease (NAFLD) in all populations. The objective of this review is to examine how different cultures and molecular entities influence the progression of NAFLD. Research databases, including PubMed, Scopus, the American Diabetes Association, the American Liver Foundation, and Diabetes UK, were used to retrieve information. Our data analysis showed that cultural norms shape the perceptions of health, illness, and mortality, thus influencing how individuals view themselves and express their experiences and may

Multi-omics integration identifies ASPH and PTTG1 as potential causal drivers of lung adenocarcinoma progression and immune evasion.

BACKGROUND: Despite advances in therapy, lung adenocarcinoma (LUAD) remains a leading cause of cancer mortality. Angiogenesis and immune evasion critically influence LUAD progression and treatment resistance, yet epithelial-derived regulatory mechanisms and causal genes remain unclear. METHODS: We employed single-cell transcriptomics (scRNA-seq) to identify angiogenesis-related epithelial-specific genes in LUAD. Mendelian randomization (MR) analyses utilizing large-scale genomic databases (eQTLGen, FinnGen) established genetic causality. A prognostic risk model was developed and validated usi

The Role of Viral and Bacterial Infections in the Etiology of Behçet's Disease.

BACKGROUND: Behçet's disease (BD) is a complex systemic vasculitis with a poorly understood etiology that involves genetic, environmental, and immunological factors. Increasing evidence suggests that viral and bacterial infections may trigger or exacerbate BD through immune-mediated pathways. This review aims to clarify how different infectious agents may contribute to BD pathogenesis. METHODS: For this review, articles addressing microbial involvement in BD were collected from established databases such as PubMed, Scopus, and Web of Science. Priority was given to studies evaluating classical

Pregnancy-associated breast cancer: Management of the mother, fetus and tumour.

INTRODUCTION: Pregnancy-associated breast cancer (PABC) is described as breast cancer diagnosed within pregnancy or within 1 year postpartum. PABC is becoming more common due to delayed childbearing, with older maternal age increasing the likelihood of tumorigenesis coinciding with pregnancy. Our review aims to outline the important principles of managing PABC, and discusses future fertility implications, genetic testing and postnatal considera-tions that are not often considered in other existing reviews. METHOD: A literature search was conducted using PubMed, Cochrane and Google Scholar dat

Interplay of Molecular Subtypes Associated with Butyrate Metabolism Elucidates Clinical Characteristics and Tumor Microenvironment in Prostate Cancer.

Emerging evidence underscores the pivotal yet context-dependent role of butyrate metabolism in oncogenic progression; however, the mechanistic landscape of its associated genetic determinants in prostate carcinogenesis remains poorly characterized. This gap presents a critical opportunity to delineate novel metabolic checkpoints that may offer therapeutic vulnerabilities for advanced prostate malignancies. Analyzing prostate cancer (PC) patients from TCGA and GEO databases, we identified 320 BMGs and stratified tumors into two butyrate metabolism-associated clusters. Machine learning and singl

Machine learning-based predictive modeling of foodborne pathogens and antimicrobial resistance in food microbiomes using omics techniques: A systematic review.

The globalization of food systems has heightened the risk of foodborne pathogens such as Salmonella, Listeria monocytogenes, and Campylobacter, exacerbated by rising antimicrobial resistance (AMR). Traditional pathogen identification and AMR risk surveillance methods are often labor-intensive and low-throughput, while single-omics approaches fail to capture microbial complexity. Moreover, reliance on individual machine learning (ML) models limits predictive robustness, posing challenges to food safety and public health. This systematic review evaluates ML-based predictive modeling integrated w

A systematic review of protein post-translational modifications in sepsis.

BACKGROUND: Sepsis originates from the host's dysregulated response to pathogens, and its pathophysiological mechanisms are extremely complex. Recent research has found that post-translational modifications (PTMs) can regulate gene transcription without altering the genetic sequence, thereby playing a key role in the occurrence and development of sepsis. OBJECTIVE: This review aims to systematically categorize the main types of PTMs and elucidate their roles in the pathogenesis of sepsis, thereby providing new perspectives for a deeper understanding of the complex pathophysiological processes

Cohesins: Crossroad Between Cornelia de Lange Spectrum and Cancer Predisposition.

The cohesin complex plays crucial roles in DNA repair, chromatid separation, and gene transcription regulation. Pathogenic variants in cohesins or dysfunctional transcriptional regulators lead to cohesinopathies, a broader group of disorders including Cornelia de Lange Spectrum (CdLSp), for which the prevalence of cancer cases remains unclear. Here, we aimed to assess the prevalence of oncological events in CdLSp and elucidate the role of cohesin variants in cancer predisposition. We developed a custom next-generation sequencing (NGS) panel targeting predisposition and pathogenic genes, which

Adverse Childhood Experiences and Antidepressant Therapy: A Systematic Review.

Adverse childhood experiences (ACEs) increase depression risk and may affect antidepressant (ADM) treatment outcomes; however, cumulative research on each ACE in conjunction with ADM outcomes is lacking. This review aims to examine the relationship between ACEs and ADM. We examined PsychInfo and PubMed/MEDLINE databases from 2010-2025 and identified 26 publications that met review criteria (i.e., peer-reviewed and concurrently examined ACEs and ADM). ACEs were associated with an increased propensity to use ADMs, an interaction between DNA methylation and ADM efficacy, an increased risk of suic

Biallelic variants in GTF3C3 encoding a subunit of the TFIIIC2 complex are associated with neurodevelopmental phenotypes in humans and zebrafish.

RNA polymerase III transcribes essential non-coding RNAs, a process regulated by transcription factors TFIIIB and TFIIIC. Although germline variants in TFIIIC subunit genes have been described in a few patients with neurodevelopmental disorders, the associated pathogenesis and clinical spectrum are not yet well defined. Herein, we describe the identification of biallelic variants in GTF3C3, which encodes a key component of the TFIIIC subunit, in four patients from three unrelated families of different ethnicities collected through GeneMatcher. The patients exhibited microcephaly, developmental

Abnormal Splicing of GALC Transcripts Underlies Unusual Cases of Krabbe Disease.

Background/Objectives: Krabbe disease (KD) is a hereditary lysosomal disorder whose hallmark is progressive demyelination, with variable involvement of the central nervous system. It is caused by pathogenic variants in the GALC gene that disrupt the function of its gene product, the lysosomal enzyme galactosylceramidase. We analyzed two unrelated cases (one early infantile and one adult) with a clinical suspicion of KD. Methods: We used a combination of biochemical techniques (high-performance liquid chromatography-tandem mass spectrometry), NGS (resequencing gene panels), splicing assays, and

Value of Optical Genome Mapping (OGM) for Diagnostics of Rare Diseases: A Family Case Report.

Optical genome mapping (OGM) is a novel method enabling the detection of structural genomic variants. The method is based on the laser image acquisition of single, labeled, high-molecular-weight DNA molecules and can detect structural genomic variants such as translocations, inversions, insertions, deletions, duplications, and complex structural rearrangements. We aim to present our experience with OGM at the Clinical Institute of Genomic Medicine, University Medical Centre Ljubljana, Slovenia. Since its introduction in 2021, we have used OGM for the testing of facioscapulohumeral muscular dys

The fetal exposome and preterm birth: a systematic synthesis of environmental exposures and multi-omics evidence.

OBJECTIVES: Preterm birth (PTB), defined as delivery before 37 weeks of gestation, is a leading cause of neonatal mortality and long-term developmental impairment. Its complex etiology, spanning environmental, genetic, psychosocial, and socio-economic domains, limits effective prediction and prevention. We systematically synthesized evidence on how environmental exposures influence PTB risk through multi-omic disruptions within a fetal exposome framework. METHODS: A comprehensive literature search was conducted in major biomedical databases, following PRISMA guidelines. Ninety-five human stud
