Compulsive singing: another aspect of punding in Parkinson's disease.

We report on two patients with advanced Parkinson's disease who were exhibiting a peculiar and stereotyped behavior characterized by an irrepressible need to sing compulsively when under high-dose dopamine replacement therapy. Sharing many features with punding, this singing behavior is proposed as a distinct manifestation of the dopamine dysregulation syndrome in Parkinson's disease.

Neurotoxicity mechanism of polychlorinated biphenyl mediated by dopaminergic system.

Polychlorinated Biphenyls (PCBs) are persistent organic pollutants in the environment. With high lipophilicity, PCBs are easily accumulated … ms are likely to involve multi-targets. Here, we focused on two highly correlated components of the dopaminergic system-the dopamine transporter (DAT) and dopamine receptor (DR), to investigate whether there is a synergetic effect between them. By combining multiple computational methods and experimental assays, this study elucidated that DAT was a potenti …

Spinal dopaminergic D1 and D5 receptors contribute to reserpine-induced fibromyalgia-like pain in rats.

Fibromyalgia is a complex pain syndrome without a precise etiology. Reduced monoamines levels in serum and cerebrospinal fluid in fibromyalgia patients has been reported and could lead to a dysfunction of descending pain modulatory system producing the painful syndrome. This study evaluated the role of D1-like dopamine receptors in the reserpine-induced fibromyalgia-like pain model in female Wistar rats. Reserpine-treated animals were intrathecally injected with different dopamine receptors agonists and antagonists, and small interfering RNAs (siRNAs) against D1 and D5 receptor subtypes. Withd…

[Neurochemistry of Parkinson's disease and parkinsonism plus].

The neurochemistry of Parkinson's disease and other degenerative parkinsonisms is reviewed, emphasizing the changes described in the dopaminergic system. Presynaptic dopaminergic markers are reduced over the striatum in all these degenerative parkinsonisms, dopamine receptor changes being more heterogeneous. While in Parkinson's disease D1 and D2 receptors remain preserved as compared to controls, in progressive supranuclear palsy there is a loss of nigral D1 receptors and of striatal D2 receptors. This finding has also been described in striatonigral degeneration. There are no clear data abou…

The extrapyramidal effects of phenothiazines on patients.

The phenothiazines have been used for many years in the treatment of psychoses. They act by blocking dopamine receptors. Unfortunately, they can also produce extrapyramidal side-effects (EPS). The pharmacological origins of these side-effects are explored in this paper and their presentation, prevalence and reversibility are considered. Non-motor adverse effects are also described, and implications for patient monitoring and education are also noted.

Role of tyrosinase in ocular growth and myopia development in a guinea pig model.

Tyrosinase participates in melanin synthesis and may also contribute to L-3,4-dihydroxyphenylalanine (L-DOPA)-related dopamine metabolism during development. Because dopamine (DA)-related signaling has been implicated in visually guided ocular growth, this study investigated whether tyrosinase-related pathways are associated with refractive development and susceptibility to lens-induced myopia (LIM) in guinea pigs. A total of 48 pigmented and 32 albino guinea pigs at 3 weeks of age were included. Animals were divided into five groups: pigmented guinea pigs with LIM and kojic acid treatment (PL…

The impact of a competitive and a non-competitive NMDA receptor antagonist on dopaminergic neurotransmission in the rat ventral tegmental area and substantia nigra.

The study compares effects of the competitive and non-competitive NMDA receptor antagonists, CGP 40116 and MK-801 respectively, on the metabolism of dopamine and on the density of D-1 and D-2 dopaminergic receptors in the rat ventral tegmental area and substantia nigra. The effects of CGP 40116 were tested in a range of doses which either were devoid of or had locomotor- or stereotypy-stimulating effects. It was found that (1) CGP 40116 given in a dose of 5 mg/kg enhanced the locomotor activity of rats and evoked a stereotypy-like activity; doses of 1.25 and 2.5 mg/kg were devoid of such effec…

[A mouse model of N-methyl-4-phenyl-1,2,3,6-tetrahydropyridine induced parkinsonism: effect of norepinephrine terminal destruction].

N-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) has been reported to cause chronic Parkinsonism in humans, primates, and long lasting striatal dopamine depletion in mice. Acute animal models thus produced closely resemble Parkinson's disease. There are, however, two major differences. The one is a lack of Lewy bodies and the other is that norepinephrine system is relatively well preserved in the model. So the acute animal model is better considered a nigrostriatal dopamine deficiency model. We have produced another model by adding N-2-chloroethyl-N-ethyl-2-bromobenzyl-amine (DSP4) to MPTP.…

Simultaneous perfusion and dopaminergic imaging using dual-isotope CZT SPECT/CT in dementia with Lewy bodies.

BACKGROUND: Current diagnostic criteria for dementia with Lewy bodies (DLB) include indicative and supportive biomarkers, some of which are typically assessed separately using dopamine transporter or perfusion imaging. Cadmium-zinc-telluride (CZT) Single Photon Emission Computed Tomography (SPECT) cameras improve simultaneous dual isotope acquisition using [99mTc]-hexamethylpropyleneamine oxime ([99mTc]TcHMPAO) (perfusion) and N-(3-Fluoropropyl)-2β-carbomethoxy-3β-(4-[123I]iodophenyl)nortropane ([123I]FP-CIT; dopamine transporter), allowing the assessment of one indicative and two supportive b…

Sex differences in the pituitary transforming growth factor-β1 system: studies in a model of resistant prolactinomas.

Dopamine and estradiol interact in the regulation of lactotroph cell proliferation and prolactin secretion. Ablation of the dopamine D2 receptor gene (Drd2(-/-)) in mice leads to a sexually dimorphic phenotype of hyperprolactinemia and pituitary hyperplasia, which is stronger in females. TGF-β1 is a known inhibitor of lactotroph proliferation. TGF-β1 is regulated by dopamine and estradiol, and it is usually down-regulated in prolactinoma experimental models. To understand the role of TGF-β1 in the gender-specific development of prolactinomas in Drd2(-/-) mice, we compared the expression of dif…

[Apomorphine in the treatment of Parkinson's Disease].

Apomorphine has long been used in many medical specialties. It is a highly potent D2-, D3- and D4-dopamine receptor agonist with a particularly high D1-dopamine receptor affinity. Due to its almost complete inactivation during liver passage it is usually applied subcutaneously. After rapid onset its effect is waning after about one hour. Apomorphine's action is not dependent on enteral dopamine resorption and on praesynaptic dopamine storage and dopamine secretion mechanisms. When applied subcutaneously its most common side effect is cutaneous nodules at the injection sites. Peripheral dopamin…

The dopaminergic D1 receptor modulates the hyperactivity of Bapa mutant mice.

The mutant bate-palmas ("claps"; symbol - bapa) mice induced by the mutagenic chemical ENU present motor incoordination and postural alterations. A previous study showed that bapa mice present increased motor/exploratory behaviors during the prepubertal period due to increased striatal tyrosine hydroxylase expression, suggesting striatal dopaminergic system hyperactivity. This study aimed to evaluate the involvement of striatal dopaminergic receptors in the hyperactivity of bapa mice. Male bapa mice and their wild strain (WT) were used. Spontaneous motor behavior was observed in the open-field…

Dopaminergic inhibition of human neutrophils is exerted through D1-like receptors and affected by bacterial infection.

Dopamine (DA) affects immune functions in healthy subjects (HS) and during disease by acting on D1-like (D1 and D5) and D2-like (D2, D3 and D4) dopaminergic receptors (DR); however, its effects on human polymorphonuclear leukocytes (PMN) are still poorly defined. We investigated DR expression in human PMN and the ability of DA to affect cell migration and reactive oxygen species (ROS) production. Experiments were performed on cells from HS and from patients (Pts) with bacterial infections as well, during the acute phase and after recovery. Some experiments were also performed in mice knockout …

Putamen dopaminergic dysfunction is associated with sleep disturbance in drug-naïve patients with Parkinson's disease.

BACKGROUND: Sleep disturbance (SD) is common in Parkinson's disease (PD) and adversely affect the quality of life (QOL). Although dopamine dysfunction has been implicated, the specific role of dopaminergic activity in SD among patients with PD remains unclear. Given that dopamine-related medications can affect sleep, it is essential to assess SD in drug-naïve patients. This study investigated the association between SD and uptake of striatal dopamine transporters using Dopamine Transporter Single-Photon Emission Computed Tomography with 123I-Ioflupane (DAT-SPECT). METHODS: We retrospectively …

Mechanisms Underlying Dopaminergic Regulation of Nicotine-Induced Kinetic Tremor.

Nicotine induces kinetic tremor, which resembles pharmacological features of essential tremors, via activating the inferior olive (IO) neurons. Since nicotine is known to enhance dopamine release by stimulating α4β2 and/or α6 nACh receptors, we examined the effects of various dopamine receptor ligands on nicotine-induced tremor to clarify the role of the dopaminergic system in modulating nicotine tremor. A tremorgenic dose of nicotine increased the dopamine level in the pons and medulla oblongata (P/MO), and the levels of dopamine metabolites in the hippocampus, P/MO, and striatum. Treatment o…

Subthalamic nucleus stimulation and compulsive use of dopaminergic medication in Parkinson's disease.

BACKGROUND: Behavioural disorders associated with compulsive use of dopaminergic drugs for Parkinson's disease (PD) such as dopamine dysregulation syndrome (DDS) and impulse control disorders (ICDs) may have devastating consequences and are challenging to manage. Whether or not such patients should undergo subthalamic nucleus (STN) deep brain stimulation (DBS) is controversial. A few case reports and small series have reported contrasting effects of STN DBS on dopamine misuse and ICDs, while a recent prospective study found clear beneficial effects of STN DBS on these disorders. METHODS: We c…
