Cannabidiol improves behavioural and neurochemical deficits in adult female offspring of the maternal immune activation (poly I:C) model of neurodevelopmental disorders.

Cognitive impairment is a major source of disability in schizophrenia and current antipsychotic drugs (APDs) have minimal efficacy for this symptom domain. Cannabidiol (CBD), the major non-intoxicating component of Cannabis sativa L., exhibits antipsychotic and neuroprotective properties. We recently reported the effects of CBD on cognition in male offspring of a maternal immune activation (polyinosinic-polycytidilic acid (poly I:C)) model relevant to the aetiology of schizophrenia; however, the effects of CBD treatment in females are unknown. Sex differences are observed in the onset of schiz

Executive function deficits and glutamatergic protein alterations in a progressive 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine mouse model of Parkinson's disease.

Changes in executive function are at the root of most cognitive problems associated with Parkinson's disease. Because dopaminergic treatment does not necessarily alleviate deficits in executive function, it has been hypothesized that dysfunction of neurotransmitters/systems other than dopamine (DA) may be associated with this decrease in cognitive function. We have reported decreases in motor function and dopaminergic/glutamatergic biomarkers in a progressive 1-methyl-4-phenyl-1,2,3,6-tetrahydropyridine (MPTP) Parkinson's mouse model. Assessment of executive function and dopaminergic/glutamate

Microarray analysis reveals the inhibition of intestinal expression of nutrient transporters in piglets infected with porcine epidemic diarrhea virus.

Porcine epidemic diarrhea virus (PEDV) infection can induce intestinal dysfunction, resulting in severe diarrhea and even death, but the mode of action underlying these viral effects remains unclear. This study determined the effects of PEDV infection on intestinal absorption and the expression of genes for nutrient transporters via biochemical tests and microarray analysis. Sixteen 7-day-old healthy piglets fed a milk replacer were randomly allocated to one of two groups. After 5-day adaption, piglets (n = 8/group) were orally administrated with either sterile saline or PEDV (the strain from

[Septic encephalopathy. Diagnosis und therapy].

23% of all septic patients develop septic encephalopathy which is associated with an increased mortality rate. Symptoms such as agitation, confusion and disorientation ranging from stupor to coma often develop in early sepsis. Severe hypotension is significantly associated with the development of septic encephalopathy. Several other factors which may play a role are also discussed: effects of inflammatory mediators on the brain, inadequate cerebral perfusion pressure, blood-brain barrier derangements, disturbances of the cerebral microcirculation, cerebral ischemia e.g. due to hypocapnia, metab

Glutamatergic modulation of transcranial direct current stimulation over the premotor cortex combined with peripheral nerve stimulation promotes observation-execution-related cortical excitability and motor performance.

Action observation and execution, transcranial direct current stimulation (tDCS) and peripheral nerve stimulation (PNS) enhance motor performance. This study aimed to investigate the effect of concurrent tDCS over the premotor cortex (PMC) and PNS during action observation on motor cortex excitability and motor performance, and its underlying mechanisms. One hundred and twenty-eight healthy adults were randomly assigned to tDCS + PNS combined with presentation of a landscape picture (tDCS + PNSlandscape), and tDCS + PNS, tDCSsham+ PNSsham, tDCSsham + PNS and tDCS + PNSsham combined with action

Targeting glutamate receptors with IEM-1925: A strategy against soman-induced status epilepticus and neurodegeneration.

Exposure to organophosphorus nerve agents (OPNAs) like soman frequently develops status epilepticus (SE), leading to brain damage. Existing antiseizure medications (e.g., diazepam, DZP) often demonstrate insufficient efficacy. To develop more effective treatments for OPNA-induced seizures, this study evaluated the efficacy of glutamate receptor antagonists with distinct mechanisms of action in a soman-induced rat seizure model. After 5 min of subcutaneous exposure to 110 μg/kg soman, which induced SE, rats received intraperitoneal injections (10 mg/kg) of perampanel (PER), fanapanel (FNP), IEM

Short-Term Exposure to Noise and Ethanol Intake Induces Changes in Aminoacidergic Neurotransmisson and Thioredoxin-1 Levels in the Cerebellum and Hippocampus of Adolescent Male Wistar Rats.

During adolescence, the Central Nervous System (CNS) undergoes key developmental processes, making it vulnerable to exogenous agents. Ethanol (EtOH) consumption and exposure to noise, common in human adolescent environments, have been linked to behavioral alterations. The hippocampus (HC) and the cerebellum (CE), key CNS regions involved in behavior regulation, are affected by these stimuli, though the mechanisms remain unclear. This study evaluated the effects of short-term voluntary EtOH consumption, alone or combined with noise exposure, on aminoacidergic neurotransmission and thioredoxin-1

Identification of a Novel Thermal Promoter and Its Application in Glutamate Decarboxylase Protein Expression in Bacillus licheniformis.

As a thermotolerant bacterium, Bacillus licheniformis is an attractive chassis for high-temperature biomanufacturing. Here, we identified a novel temperature-responsive promoter, PycgM, which maintained strong transcriptional activity at 37-52 °C. In a promoter-mCherry reporter system, PycgM exhibited 2287.3-fold higher activity than P2 at 52 °C, demonstrating excellent compatibility with a thermotolerant host. Truncation analysis identified a 150-bp core functional region responsible for optimal activity under induction and heat stress. When applied to drive glutamate decarboxylase expression

Fabrication of Co-Doped Covalent Organic Framework Nanosheets with Mild Interlayer Stress for Quantitative Detection of Alzheimer's Disease Biomarkers.

Alzheimer's disease (AD) seriously affects human health worldwide. Nicotinamide adenine dinucleotide (NADH) and glutamate are important biomarkers of AD, which play an indispensable role in the pathogenesis of AD. Herein, two ligands were used to synthesize a layered covalent organic framework (TPCOF) via amide bond formation, which was loaded with cobalt to obtain Co-TPCOF. TPCOF has a twisted 2D layered structure, along with large interlayer spacing and porosity, enabling precise Co coordination and stable loading of metal nanoparticles/enzymes to support electrocatalysis. A Co-TPCOF was imm

Role of GPR55 in glutamatergic and dopaminergic signaling: A review of preclinical evidence in neuropsychiatric disorders.

G protein-coupled receptor 55 (GPR55) has gained increasing attention for its involvement in various neuropsychiatric disorders. Emerging evidence highlights the critical role of this receptor in modulating brain functions through complex interactions with glutamate and dopamine neurotransmitters. Recent studies suggest that GPR55 is involved in key processes, including synaptic plasticity and reward signaling. Moreover, central GPR55 participated in the pathophysiology of neuropsychiatric conditions such as schizophrenia, depression, anxiety disorders, and addiction. This review aims to provi

Audiogenic kindling activates glutamatergic system in the hippocampus of rats with genetic predisposition to audiogenic seizures.

Temporal lobe epilepsy (TLE) development is associated with dysregulation of glutamatergic transmission in the hippocampus; however, detailed molecular mechanisms of pathological changes are still poorly understood. In the present study, we performed the complex analysis of glutamatergic system in the hippocampus of Krushinsky-Molodkina (KM) rats genetically prone to audiogenic seizures (AGS). Daily AGS stimulations (audiogenic kindling) were used to reproduce the dynamics of TLE development. Naïve KM rats were used as a control. After 14 AGS, at the stage of developing TLE, KM rats demonstrat

Dysregulation of kappa opioid receptor neuromodulation of lateral habenula synaptic function following a repetitive mild traumatic brain injury.

Mild traumatic brain injury (mTBI) increases the risk of affective disorders, anxiety and substance use disorder. The lateral habenula (LHb) plays an important role in pathophysiology of psychiatric disorders. Recently, we demonstrated a causal link between mTBI-induced LHb hyperactivity due to excitation/inhibition (E/I) imbalance and motivational deficits in male mice using a repetitive closed head injury mTBI model. A major neuromodulatory system that is responsive to traumatic brain injuries, influences affective states and also modulates LHb activity is the dynorphin/kappa opioid receptor

Synaptic modulation of glutamate in striatum of the YAC128 mouse model of Huntington disease.

BACKGROUND: Altered balance between striatal direct and indirect pathways contributes to early motor, cognitive and psychiatric symptoms in Huntington disease (HD). While degeneration of striatal D2-type dopamine receptor (D2)-expressing indirect pathway medium spiny neurons (iMSNs) occurs prior to that of D1-type dopamine receptor (D1)-expressing direct pathway neurons, altered corticostriatal synaptic function precedes degeneration. D2-mediated signaling on iMSNs reduces their excitability and promotes endocannabinoid (eCB) synthesis, suppressing glutamate release from cortical afferents. D2
