Contrary Effects of Cholesterol on Single-Component and Synaptic Vesicle Mimicking Lipid Bilayers.

Cholesterol is a major determinant of biological lipid membrane properties. Its physiological levels are affected by widely used prescription drugs. The effect of changing the cholesterol content has been extensively studied in model lipid bilayers, but it still remains a matter of debate. Here we compare the effect of cholesterol on synaptic vesicle-mimicking (consisting of POPE, POPC, POPS) and single-component bilayers (containing POPC). We quantify the effects of cholesterol on the mechanical properties of the membrane (e.g., lipid packing, polarity, and the force needed for an AFM tip to

Mechanism of Phosphatidylserine Lipid Scrambling by Human SERINC3, an HIV-1 Restriction Factor.

The HIV-1 restriction factor, hSERINC3, functions as a lipid scramblase, translocating lipids across the bilayer in reconstituted proteoliposomes and the viral envelope. Phosphatidylserine(PS) scrambling and exposure at the outer leaflet are recognized to play important roles in several biological processes. To understand the mechanistic basis for hSERINC3-mediated PS lipid scrambling at atomistic resolution, we implemented the transition-tempered metadynamics (TTMetaD) enhanced sampling method. Our simulations sampled close-to-open hSERINC3 conformational transition during PS scrambling and d…

Immune activation induced by dysregulated lipid metabolism in the pathogenesis of type 2 diabetes.

Type 2 diabetes mellitus (T2DM) is characterized by two core pathological features: Insulin resistance and β-cell dysfunction, with dyslipidemia and immune dysregulation playing critical roles in its pathogenesis. Ectopic lipid deposition and lipotoxicity, resulting from dysregulated lipid metabolism, drive T2DM progression by reshaping immune microenvironments across multiple organs. Over the past two decades, the concept of "immune-metabolic coupling" has gained widespread recognition: Lipotoxicity activates immune cells through pattern recognition receptors, eliciting chronic low-grade infl

Impact of Dapagliflozin on Hepatic Lipid Metabolism and a Dynamic Model of Ketone Body Levels.

The rising prevalence of metabolic-associated steatotic liver disease emphasizes the need to understand its lipid metabolism. Dapagliflozin may improve hepatic steatosis but could also increase the risk of ketoacidosis by elevating β-hydroxybutyrate (KB) levels. This study investigates dapagliflozin's effects on hepatic lipid metabolism and quantifies KB levels in vivo. Male Sprague-Dawley rats were fed either a normal diet or a high-fat diet (HFD) for 12 weeks. The HFD rats were then divided into four subgroups to receive vehicle, 0.5 mg/kg, 1 mg/kg, and 3 mg/kg of dapagliflozin for four week

Efficacious suppression of primary and metastasized liver tumors by polyIC-loaded lipid nanoparticles.

BACKGROUND AND AIMS: So far, there is no effective mechanism-based therapeutic agent tailored for liver tumors. Immune checkpoint inhibitors (ICIs) have demonstrated limited efficacy in liver cancer, often associated with severe adverse effects. Although poly-inosinic:cytidylic acid (polyIC) has shown an adjuvant effect when combined with anti-PD-L1 antibody (αPD-L1) in treating liver tumors in animal models, its systemic toxicity limits its clinical utility. To address this challenge, the original goal of this study was to develop a liver-targeted strategy to minimize toxicity. METHODS AND R

Exploring the role of curcumin in mitigating oxidative stress to alleviate lipid metabolism disorders.

Lipid metabolism plays a crucial role in maintaining homeostasis and overall health, as lipids are essential molecules involved in bioenergetic processes. An increasing body of research indicates that disorders of lipid metabolism can contribute to the development and progression of various diseases, including hyperlipidemia, obesity, non-alcoholic fatty liver disease (NAFLD), diabetes mellitus, atherosclerosis, and cancer, potentially leading to poor prognoses. The activation of the oxidative stress pathway disrupts lipid metabolism and induces cellular stress, significantly contributing to m…

Mechanism of O-GlcNAcylation regulating liver lipid synthesis in mice through FASN.

Nonalcoholic fatty liver disease (NAFLD) has become one of the most common chronic liver diseases. O-Linked attachment of beta-N-acetylglucosamine (O-GlcNAc) are ubiquitous post-translational modifications of proteins as "nutrient sensors" and "stress receptors" in the body that are involved in maintaining normal cellular physiological functions. Increased levels of O-GlcNAcylation have been found in the liver samples of patients with NAFLD and nonalcoholic steatohepatitis. However, the role of O-GlcNAcylation in the development and pathogenesis of NAFLD remains unclear. Here, we sought to det

Influence of Cholesterol on the Insertion and Interaction of SARS-CoV-2 Proteins with Lipid Membranes.

Cholesterol is an essential sterol in cell membranes that regulates organization and fluidity. This biomolecule has been identified as one of the critical factors in the internalization process of various viruses in human cells. Therefore, understanding these mechanisms is crucial for a deeper comprehension of viral pathogenicity in the search for practical therapeutic approaches against viral diseases. The biochemical and biophysical processes related to these diseases are highly complex. For this reason, studying model systems capable of mimicking the interaction of lipid membranes with chol

Targeting fungal lipid synthesis for antifungal drug development and potentiation of contemporary antifungals.

Two of the three most commonly used classes of antifungal drugs target the fungal membrane through perturbation of sterol biosynthesis or function. In addition to these triazole and polyene antifungals, recent research is identifying new antifungal molecules that perturb lipid biosynthesis and function. Here, we review fungal lipid biosynthesis pathways and their potential as targets for antifungal drug development. An emerging goal is discovering new molecules that potentiate contemporary antifungal drugs in part through perturbation of lipid form and function.

Successful endodontic treatment improves glucose and lipid metabolism: a longitudinal metabolomic study.

BACKGROUND: Apical periodontitis (AP) is one of the most prevalent dental diseases. Its presence can increase systemic inflammatory burden and is associated with cardiometabolic disorders including higher cardiovascular risks and impaired glycaemic control. There is currently a paucity of knowledge showing whether successful endodontic treatment is associated with systemic metabolic improvements. This study aims to identify the potential impact of successful endodontic treatment on patients' serum metabolomic profiles, and to evaluate how these impacts are associated with the metabolic syndrom

A Novel Hypothesis for the Protective Rise in Cholesterol Sulfate Against Lipid Metabolic Disorders.

As a cholesterol metabolite, cholesterol sulfate (CS) is widely distributed in the human body, and its role as a regulatory factor has been continuously explored from the 1980s to the present day. However, changes in CS in metabolic disorders have not been systematically investigated. Here, rodent models of insulin resistance, fatty liver, and atherosclerosis were established. The CS content, CS to cholesterol ratio, and CS to total bile acids (TBA) ratio in the serum and liver of these model mice were compared with those of normal mice. Results showed the CS content was increased in fatty liv
