Effect of growth hormone on caffeine metabolism in hypophysectomized rats.

Two groups of six male Sprague-Dawley hypophysectomized rats (operated on day 0), 8 weeks old, treated by sc tetracosactid (ACTH, 10 micrograms every 24 hr), thyroxine (5 micrograms/100 g every 24 hr) and desmopressin (240 ng/kg/24 hr continuous infusion) received SC either saline (group I) or human growth hormone (hGH, 120 micrograms/24 hr) (group II) continuous infusion. ACTH and thyroxine were administered on days 7-19 and desmopressin and hGH on days 8-19, after surgery. They received po caffeine 4 mg/kg as citrate salt on day 15. The 0-12, 12-24 and 24-48 hr urine samples were collected a

Role of mesangial cell contraction in adaptation of the glomerular tuft to changes in extracellular volume.

Different chronic states of mesangial cell contraction were induced by variation of extracellular volume in Munich-Wistar rats for 6 days to study the influence of mesangial cells on the geometry of the glomerular tuft. Stereological analysis of superficial glomeruli in volume-expanded rats (VE, treated with enalapril) and volume-reduced rats (VR, treated with indomethacin) revealed a glomerular tuft volume 28.7% smaller, and a capillary luminal volume 32% smaller in VR than in VE rats. The filtration area [defined as glomerular basement membrane (GBM) area facing fenestrated endothelium] was

Diazepam changes risk assessment in an anxiety/defense test battery.

An anxiety/defense test battery was designed to assess defensive reactions of laboratory rats to situations associated with nonpainful threat (exposure to a cat). The battery measured three defense patterns, movement inhibition, risk assessment behaviors, and inhibition of nondefensive behaviors, in two tasks. Diazepam (4.0 mg/kg) altered four of five risk assessment measures, but failed to show an anxiolytic effect on movement inhibition, and had minimal and inconsistent effects on inhibition of nondefensive behaviors. The risk assessment changes with diazepam were all consistent with an anxi

[Modifying effects of neurotropin on post-radiation disorders of neuromediator processes in the autonomic function regulation centers].

In experiments on Wistar rats, it was shown that 6 months after exposure to ionizing radiation (X-radiation 0.5 Gy and intraperitoneal 131I 6.5 mCu/kg) the hypothyroid state was accompanied by changes in inhibitory and excitatory mediation that are characteristic of chronic radiation stress in cerebral structures responsible for regulation of autonomic and animal functions and a relative deficiency of the hypothalamopituitary-adrenocortical axis. Application of therapeutic doses of neurotropin 3 months after radiation levelled the pattern of chronic radiation stress and diencephalic disorders

Endotoxin inhibition of drinking behaviour in the rat.

Intravenous (i.v. 320 and 640 micrograms/kg) and intracerebroventricular (i.c.v.; 1 microgram/rat) injection of Escherichia coli lipopolysaccharide (LPS) powerfully inhibited drinking induced by 24 h water deprivation. Pretreatment with acetylsalicylic acid (ASA) into the preoptic area (POA) completely abolished the effect induced by i.v. LPS, but did not modify that elicited by i.c.v. LPS. Intraperitoneal ASA injections significantly reduced the antidipsogenic effect of i.c.v. LPS. Electrolytic ablation of the subfornical organ (SFO) did not modify the effect induced by either i.v. or i.c.v.

GABAergic projection from nucleus accumbens to ventral pallidum mediates dopamine-induced sensorimotor gating deficits of acoustic startle in rats.

Previous studies have demonstrated that increased mesolimbic dopamine (DA) activity disrupts sensorimotor gating as measured by prepulse inhibition (PPI) of the acoustic startle response (ASR) in rats. Other behavioral changes following mesolimbic DA activation are mediated through GABAergic efferent projections from the nucleus accumbens (NAC) to the ventral pallidum (VP). In this experiment, we examined whether PPI deficits in rats following mesolimbic DA activation are mediated through these same GABAergic substrates. PPI was significantly disrupted in rats following infusion of DA (40 micr

Comparative metabolism of high doses of aspirin in man and rat.

1. Metabolism of aspirin was studied in 10 human volunteers who took a therapeutic dose (600 mg) by mouth and in nine patients who took aspirin in overdose. 2. Salicyluric acid was the major urinary metabolite in volunteers (63.1 +/- 8.4% of dose in 0-8 h). In overdose patients, salicyluric acid in urine was decreased (30.0 +/- 8.2%, 0-24 h, P less than 0.001) and there was increased elimination of salicyclic acid (34.1%, P less than 0.005), salicyl acyl glucuronide (14.4%, P less than 0.05) and gentisuric acid (5.3%). 3. [Low concentration enflurane anesthesia and local administration of epinephrine in rat. Comparison between the lidocaine group and the distilled water group].

In the surgical operation for oral region, local anesthetic agent added with epinephrine is usually given to the site to be operated in order to obtain better hemostatic effect even under general anesthesia. However, there have been relatively few reports, which describe the circulatory changes after the injection into oral region, the fluctuation of plasma catecholamine concentration and the concentration of local anesthetic agent in plasma. In the present study, 1% lidocaine or distilled water added with 10 micrograms/ml epinephrine was locally injected at 0.5 ml/kg into the palatal submucos

Naloxone prevents increased vascular sensitivity in Goldblatt hypertensive rats.

These experiments were designed to determine if the opiate antagonist naloxone affects vascular sensitivity in 2K-1C hypertensive rats. Group 1 was 2K-1C hypertensive rats. Group 2 was 2K-1C rats given a naloxone infusion (100 micrograms/h) for 14 days. Group 3 received naloxone without clipping. Group 4 was untreated rats. At day 14 following clipping, systolic blood pressure was increased significantly in the 2K-1C rats. Those infused with naloxone showed a significant attenuation of the increase in blood pressure. Mortality of metalloporphyrin-treated neonatal rats after light exposure.

Tin protoporphyrin (SnPP) has been used to suppress hyperbilirubinemia in human neonates through inhibition of heme oxygenase. Some of the subjects exhibited mild erythema upon receiving phototherapy. SnPP and three proposed alternatives, tin mesoporphyrin (SnMP), zinc protoporphyrin (ZnPP) and zinc mesoporphyrin (ZnMP) are potential photosensitizers. We therefore studied the phototoxic effects of these compounds in the neonatal rat model. Fed Wistar rats (24-36 h old) were injected intraperitoneally with up to 40 mumol SnPP/kg body weight, 30 mumol SnMP/kg body weight, 60 mumol ZnPP/kg body w

Galanin-induced prolactin release in rats: pharmacological evidence for the involvement of alpha-adrenergic and opioidergic mechanisms.

The mechanism by which galanin (GAL) stimulates prolactin (PRL) secretion was investigated in urethane-anesthetized male rats. The PRL release induced by intracerebroventricular (i.c.v.) injection of porcine GAL (pGAL) was similar to that induced by rat GAL. The PRL release induced by pGAL was partially blocked by alpha-adrenergic antagonists, phentolamine (1 microgram/rat, i.c.v.; 29.1 +/- 4.5 ng/ml vs control 66.9 +/- 10.2 ng/ml, P less than 0.01) and tolazoline (1 microgram/rat, i.c.v.; 25.9 +/- 4.4 ng/ml vs control 59.6 +/- 10.9 ng/ml, P less than 0.05). Possible protective effect of endogenous opioids in traumatic brain injury.

Naloxone (0.1, 1.0, or 20.0 mg/kg), morphine (1.0 or 10.0 mg/kg), or saline was administered systemically intraperitoneally to rats 15 minutes prior to moderate fluid-percussion brain injury. The effects of the drugs were measured on systemic physiological, neurological, and body-weight responses to injury. The animals were trained prior to injury and were assessed for 10 days after injury on body-weight responses and neurological endpoints. Low doses of naloxone (0.1 or 1.0 mg/kg) significantly exacerbated neurological deficits associated with injury. Morphine (10.0 mg/kg) significantly reduc

Antagonistic effect of naloxone on the hypertensive response of intraventricularly administered histamine.

Intracerebroventricular (i.c.v.) injections of histamine (HA) to Vetbutal-anaesthetized rats elicited a biphasic pressure response: a fall of blood pressure (BP) in the first minute, followed by a rise in BP. The heart rate (HR) response was also biphasic: bradycardia in the first minute, followed by tachycardia. The H1-receptor agonist 2-pyridylethylamine (PEA) increased the fall in BP in the first minute in comparison with the HA group, and then elicited a rise in BP that was, however, much lower than that produced by HA. Throughout the experiment PEA produced essentially only a bradycardia.

Comparison of residual glomerular function in experimental papillary necrosis and renal infarction.

The purpose of this study was to examine the course of residual glomerular function in two models of renal ablation both with equivalent degrees of nephron reduction (1 1/3 nephrectomy). The remnant group underwent right nephrectomy and infarction of one third of the left kidney, yielding constant proportions of remnant deep and superficial nephrons, while the bromoethylamine (BEA) group underwent right nephrectomy and chemical ablation by BEA (50 mg intravenously [IV]) of the deep nephrons of the left kidney, leaving superficial nephrons. Ten weeks following ablation, greater whole kidney per

Effect of nifedipine on morphine-induced analgesia.

Experimental data show that opiates interfere with calcium influx in the cell and that some calcium-channel blockers are analgesic. We therefore studied the effect of the calcium-receptor blocker nifedipine on the analgesic effect of morphine in the rat, using tail-flick responses, and in humans, using measurements of the intensity of postoperative pain. In both the experimental animals and humans nifedipine significantly (P less than 0.001) increased the analgesic effect of morphine independently of any effect on the metabolism of morphine. Respiratory and cardiovascular functions were not si

Plasma beta-endorphin and cortisol levels in morphine-tolerant rats and in naloxone-induced withdrawal.

The relationship between morphine tolerance and pituitary-adrenocortical activity was examined. In rats made tolerant to morphine by implantation of morphine-containing pellets, there was a significant reduction in plasma levels of beta-endorphin-like immunoreactivity (beta-END-LI), whereas no significant changes in cortisol levels were seen. Naloxone treatment induced an increase in plasma beta-END-LI and cortisol levels in morphine-tolerant animals. Additionally, acute morphine administration induced an increase in plasma levels of beta-END-LI and cortisol, an effect which was prevented by n

Effects of naloxone on renin and pressor responses to acute renal hypotension in rats.

Reduction of renal perfusion is followed by increases in plasma renin activity (PRA) and arterial pressure. The present experiments were designed to determine if an opiate antagonist would alter pressor or renin responses to acute reduction of renal arterial pressure (RAP) in anesthetized rats. Male Sprague-Dawley rats were anesthetized with Inactin, and an adjustable constrictor device was placed around the abdominal aorta proximal to the renal arteries. One-half of the animals were pretreated with the opiate antagonist naloxone (2 mg/kg iv), and the other one-half were pretreated with saline

Regulation of nuclear scaffold protease activity by calcium.

Recent investigations have disclosed protease activity which is intimately associated with the nuclear scaffold. This protease activity shows a marked selectivity for lamins A/C, and from them may produce the 46-kDa NTPase thought to participate in nucleocytoplasmic RNA transport. Preliminary characterizations suggested that the protease activity might require Ca2+. In this report, we present evidence showing that nuclear scaffold protease activity is stringently regulated by Ca2+ within the physiologic range. Further, calmodulin antagonists such as calmidazolium and chlorpromazine produce mar

Blocking the calcium cascade in experimental acute renal failure.

Calcium is believed to be responsible for initiating a deleterious cascade of events that leads to irreversible cell injury during prolonged ischemia. Theoretically, the calcium-dependent cascade of events can be interrupted at three distinct points: a) by reducing calcium inflow into the cytosol using a calcium channel blocker such as verapamil, b) by increasing the mitochondrial capacity to sequester calcium using ethane-1-hydroxy-1:1-diphosphonic acid (EHDP), and c) by inhibiting the activation of the calcium-calmodulin complex using trifluoperazine (TFP). To evaluate the protective role of

Calcium-independent GABA release from striatal slices: the role of calcium channels.

We have investigated the role of Ca2+ and Ca2+ channels in the modulation of GABA release. Brain slices prepared from rat striatum were preincubated with [3H]GABA, superfused with Krebs bicarbonate buffer, and exposed to electrical field stimulation (2 Hz for 3 min). Tritium efflux was measured as an index of GABA release. Both resting and evoked efflux were greatly accelerated by deleting Ca2+ from the medium and adding EGTA (1 mM). However, when the concentration of Mg2+ in the buffer was elevated to 10 mM, no effect of the Ca2(+)-deficiency was observed on resting release and its impact on
