Pocket depth reduction as an outcome measure of inflammation and soft tissue changes in periodontitis trials.

Randomized, controlled clinical trials are prospective studies involving human subjects that are designed to assess disease interventions by comparing end-points related to patient well-being. The purpose of this article is to examine the parameter probing (or pocket) depth as a meaningful end-point for evaluating novel periodontitis therapies. Probing depth provides an estimate of the inflamed, ulcerated lesion secondary to periodontitis disease. Probing depth measurements strongly correlate with the concentrations of host inflammatory mediators implicated in the pathogenesis of periodontitis

Marked phenotypic variation in a family with a new myelin protein zero mutation.

Myelin protein zero (MPZ) is a member of the immunoglobulin gene superfamily, which has a role in myelin compaction. MPZ gene mutations cause mostly demyelinating neuropathies of the Charcot-Marie-Tooth 1B type (CMT1B), but axonal CMT have been described as well. There is a broad spectrum of phenotypic manifestation of neuropathies caused by MPZ mutations. Some mutations of MPZ cause severe early-onset neuropathies such as Dejerine-Sottas disease, while others cause the classical CMT phenotype with normal early milestones but development of disability during the first two decades of life. Smoking in adolescence as a predictor of early loss of periodontal attachment.

OBJECTIVES: On the basis of information from studies of older adults, smoking is considered to be an important risk factor for periodontal disease. Examining periodontal loss of attachment among younger adults means a lower contribution from cumulative exposure to other environmental risk factors. The aim of this study was to examine the role of chronic exposure to cigarette smoking as a risk factor for greater prevalence and extent of periodontal loss of attachment among 26-year-old participants in a longstanding prospective cohort study. METHODS: Loss of attachment (LOA) was measured at thr

Bone resorption caused by three periodontal pathogens in vivo in mice is mediated in part by prostaglandin.

Gingival inflammation, bacterial infection, alveolar bone destruction, and subsequent tooth loss are characteristic features of periodontal disease, but the precise mechanisms of bone loss are poorly understood. Most animal models of the disease require injury to gingival tissues or teeth, and the effects of microorganisms are thus complicated by host responses to tissue destruction. To determine whether three putative periodontal pathogens, Porphyromonas gingivalis, Campylobacter rectus, and Fusobacterium nucleatum, could cause localized bone resorption in vivo in the absence of tissue injury

Liver abnormalities associated with chronic mercury accumulation in stranded Atlantic bottlenose dolphins.

Eighteen stranded Atlantic bottlenose dolphins (Tursiops truncatus) examined postmortem were sampled for histologic study. All cases were examined for ferric ion and lipofuscin. Ages were determined from tooth growth layers. Electron microscopic (EM) examination and X-ray spectroscopy (EDAX) were performed. Chemical analysis for mercury was conducted on 12 of the animals by atomic absorption spectrophotometry. Nine animals were found to have excessive lipofuscin in both liver and kidney. Four of these nine animals also exhibited active liver disease (fat globules, central necrosis, lymphocytic

From Axonal Growth to Neurodegeneration: The Dual Role of Neurofilament Dynamics in Health and Disease.

Neurofilaments (NFs) are the predominant type IV intermediate filaments in differentiated neurons, functioning not just as static scaffolds, but as active drivers of radial axonal growth and nerve conduction velocity. While their physical properties are well characterized, a critical gap remains in synthesizing how their dynamic assembly and developmental subunit switching directly dictate neurodegenerative outcomes. This review breaks down the molecular architecture and stepwise kinetic assembly of NFs, detailing their role in polarized transport and the formation of a protective viscoelastic

[Understanding and comments of the series of uncertain viewpoints on the diagnosis and treatment of AAOMS medication-related osteonecrosis of the jaw].

This article focuses on the series of viewpoints on the prevention and treatment of medication-related osteonecrosis of the jaw issued by the American Association of Oral and Maxillofacial Surgery (AAOMS), and puts forward several uncertain issues from a clinical perspective. The points of doubt in diagnosis include the definition of drug categories and the threshold of bone exposure duration, as well as the significance of imaging features of bone lesions for diagnostic and treatment decisions. The concerns in terms of prevention include drug risks and disease incidence, tooth extraction and

Advanced AI-driven detection of interproximal caries in bitewing radiographs using YOLOv8.

Dental caries is a very common chronic disease that may lead to pain, infection, and tooth loss if its diagnosis at an early stage remains undetected. Traditional methods of tactile-visual examination and bitewing radiography, are subject to intrinsic variability due to factors such as examiner experience and image quality. This variability can result in inconsistent diagnoses. Thus, the present study aimed to develop a deep learning-based AI model using the YOLOv8 algorithm for improving interproximal caries detection in bitewing radiographs. In this retrospective study on 552 radiographs, a

Role of regulatory immune cells in pathogenesis and therapy of periodontitis.

Periodontitis disease (PD) is a serious gum infection that progresses from gingivitis. PD is defined by gingival recession and bone loss and can lead to tooth loss. Bacterial infections are the main cause, as they induce inflammation and the development of periodontal pockets. Traditional therapies such as scaling and root planning aim to remove the subgingival biofilm via mechanical debridement but fail to address the fundamental inflammatory imbalance within the periodontium. The immune homeostasis linked to periodontal health necessitates a regulated immuno-inflammatory response, within whi

Agonism of PIEZO1 prevents aggravated periodontitis with traumatic occlusion via MAPK signaling pathway.

Periodontitis is a chronic inflammatory disease causing alveolar bone loss and tooth damage. Traumatic occlusion (TO) exacerbates its progression, although the process is mechanistically unclear. This study investigated the role of the mechanosensitive ion channel PIEZO1 in TO-aggravated periodontal destruction. In a rat model combining periodontitis and TO, PIEZO1 expression decreased, with increased inflammation and impaired osteogenesis. Yoda1 treatment alleviated periodontal inflammation and bone loss, demonstrating its therapeutic potential. In vitro, mechanical stress and lipopolysacchar

Novel STAG2 variant expands Mullegama-Klein-Martinez Syndrome phenotype.

We present a novel case of a female patient with a de novo heterozygous splice site pathogenic variant STAG2 (NM_001042749.3):c.1196 + 4_1196+7del. The STAG2 gene encodes "Stromal Antigen 2" (STAG2), a fundamental subunit of the cohesin complex, which regulates the separation of sister chromatids during cell division. Pathogenic STAG2 gene variants are associated with Mullegama-Klein-Martinez Syndrome (MKMS), a rare X-linked cohesinopathy. This patient exhibits symptoms not previously associated with MKMS, thereby expanding the known clinical phenotype of this rare disease. The patient has sev

Periodontitis was associated with mesial concavity of the maxillary first premolar: a cross-sectional study.

The association between the anatomical features of teeth and the pathogenesis of periodontitis is well-documented. This study aimed to evaluate the influence of the mesial concavity of the maxillary first premolar on periodontal clinical indices and alveolar bone resorption rates. Employing a cross-sectional design, in 226 patients with periodontitis, we used cone beam computed tomography(CBCT) to examine the mesial concavity and alveolar bone resorption of 343 maxillary first premolar. Periodontal clinical indicators recorded by periodontal probing in the mesial of the maxillary first premola

Autoantibodies Against Dihydrolipoamide S-Acetyltransferase Are Not Associated With Immune-Mediated Neuropathies.

OBJECTIVES: Dihydrolipoamide S-acetyltransferase (DLAT), the E2 component of the mitochondrial pyruvate dehydrogenase complex (PDC-E2), has recently been suggested to be a biomarker of chronic inflammatory demyelinating polyneuropathy (CIDP). It was particularly associated with sensory variants of CIDP. Antimitochondrial antibodies are important for the diagnosis of primary biliary cholangitis, but insofar, only 2 studies have reported an association with CIDP. Here, we aimed to validate these observations in a cohort of French patients with immune-mediated neuropathy. METHODS: The positivity

CT and MR Appearance of Teeth: Analysis of Anatomy and Embryology and Implications for Disease.

Abnormalities of dental development and anatomy may suggest the presence of congenital or acquired anomalies. The detection of abnormalities, therefore, is an important skill for radiologists to achieve. Knowledge of dental embryology and an understanding of the radiologic appearances of teeth at various stages of maturation are required for the appreciation of abnormal dental development. While many tooth abnormalities are well-depicted on dedicated dental radiographs, the first encounter with a dental anomaly may be by a radiologist on a computed tomographic (CT) or magnetic resonance (MR) e

Skeletal muscle involvement in biallelic SORD mutations: case report and review of the literature.

Biallelic mutations in the sorbitol dehydrogenase (SORD) gene have been identified as a genetic cause of autosomal recessive axonal Charcot-Marie-Tooth disease 2 (CMT2) and distal hereditary motor neuropathy (dHMN). We herein review the main phenotypes associated with SORD mutations and report the case of a 16-year-old man who was referred to our outpatient clinic for a slowly worsening gait disorder with wasting and weakness of distal lower limbs musculature. Since creatine phosphokinase (CPK) values were persistently raised (1.5fold increased) and a Next-Generation Sequencing CMT-associated

The Potential Application of Green-Synthesized Metal Nanoparticles in Dentistry: A Comprehensive Review.

Orodental problems have long been managed using herbal medicine. The development of nanoparticle formulations with herbal medicine has now become a breakthrough in dentistry because the synthesis of biogenic metal nanoparticles (MNPs) using plant extracts can address the drawbacks of herbal treatments. Green production of MNPs such as Ag, Au, and Fe nanoparticles enhanced by plant extracts has been proven to be beneficial in managing numerous orodental disorders, even outperforming traditional materials. Nanostructures are utilized in dental advances and diagnostics. Oral disease prevention me

Genotype-phenotype correlations of KIF5A stalk domain variants.

The kinesin family member 5A (KIF5A) motor domain variants are typically associated with hereditary spastic paraplegia (HSP) or Charcot-Marie-Tooth 2 (CMT2), while KIF5A tail variants predispose to amyotrophic lateral sclerosis (ALS) and neonatal intractable myoclonus. Variants within the stalk domain of KIF5A are relatively rare. We describe a family of three patients with a complex HSP phenotype and a likely pathogenic KIF5A stalk variant. More family members were reported to have walking difficulties. When reviewing the literature on KIF5A stalk variants, we found 22 other cases. The phenot
