In Vivo and In Vitro Toxicity Evaluation of Polyprenols Extracted from Ginkgo biloba L. Leaves.

Polyprenols of Ginkgo biloba L. leaves (GBP) are a new type of lipid with 14-24 isoprenyl units, which in humans have strong bioactivity like the dolichols. A large amount of work showed that GBP had good antibacterial activity and powerful protective effects against acute hepatic injury induced by carbon tetrachloride and alcohol, as well as antitumor activity, but the safety of GBP was not considered. The current study was designed to evaluate the toxicity of these polyprenols. Acute toxicity in mice was observed for 14 days after GBP oral dosing with 5, 7.5, 10, 15 and 21.5 g/kg body weight…

Amniotic fluid stem cells prevent development of ascites in a neonatal rat model of necrotizing enterocolitis.

AIM: It has been demonstrated that in a neonatal rat model of necrotizing enterocolitis (NEC), amniotic fluid stem (AFS) cells decrease intestinal damage and improve survival via modulation of stromal cells expressing cyclooxygenase 2 in the lamina propria. Herein, we aimed to evaluate the effect of AFS cells on body weight and fluid retention in this NEC model. Methods AFS cells were obtained from green fluorescent protein (GFP) + pregnant rats at E16 and expanded in culture. A total of 185 neonatal rats had NEC induced by gavage feeding of hypertonic formula + hypoxia + oral lipopolysacchari…

Association of body fat distribution with bone mineral density: evidence from observational and mendelian randomization analyses.

BACKGROUND: Although higher body weight is generally considered protective against low bone mineral density (BMD), the influence of body fat distribution on BMD remains unclear. We aimed to investigate whether specific adipose tissue depots have differential associations with BMD and to assess causality using Mendelian randomization (MR). METHODS: We conducted a cross-sectional analysis using National Health and Nutrition Examination Survey (NHANES) data to evaluate associations between regional fat depots (gynoid, android, visceral, abdominal subcutaneous fat, and total fat percentage) and B…

The relationship between anthropometric measurements and fractures in women.

In a population-based, retrospective study involving postmenopausal women, we investigated the relation between fracture prevalence (risk) and present as well as previous body height and weight. A questionnaire was mailed to 46,353 postmenopausal women aged 50-80 years and there were 29,802 (64.3%) responses. Questions were asked about body height and weight and fractures after the age of 25 and at what year they had occurred. The fractures were grouped according to whether they had occurred within 5 years or 10 years prior to responding to the questionnaire. Main outcome was relative risk (RR

An Autopsy Case of Beare-Stevenson Cutis Gyrata Syndrome Presenting with an Extremely Short Small Intestine.

Beare-Stevenson cutis gyrata syndrome (BSS) is a craniosynostosis caused by an FGFR2 pathogenic variant. Detailed reports of autopsy results are scarce. We herein present an autopsy case of BSS characterized by a disproportionate organ size and severe failure to thrive. Upon autopsy, the weights of the heart, liver, and lungs corresponded to the patient's body weight, whereas the brain, pancreas, and spleen were enlarged. Notably, the small intestine was much shorter in our patient than in infants with a similar body size, which could explain the poor weight gain. This case is the first compre…

Successful Treatment of Varicella Zoster Encephalitis in a Sarcopenic Obese Patient after Acyclovir-induced Acute Kidney Injury: The Necessity of Individualized Dosing.

Acyclovir (ACV) is the first-line agent for varicella zoster virus (VZV) encephalitis. Acute kidney injury (AKI) remains a clinical concern. We report the case of a sarcopenic obese woman with VZV encephalitis. She developed AKI 2 days after receiving intravenous ACV based on her actual body weight. After temporarily discontinuing ACV and confirming recovery of the renal function, ACV was restarted at a dose based on her ideal body weight. The patient recovered without any further complications. This case underscores the importance of dose adjustment of water-soluble medications, such as ACV, …

Distributed amylin receptor signaling and its influence on motivated behavior.

The intake- and body weight-suppressive hormone amylin exerts effects on energy balance control at a variety of nuclei within the brain, including sites that have been referred to in the literature as mediating homeostatic versus hedonic aspects of feeding. Here, we review key central nervous system sites of action for amylin signaling in the neural control of feeding and body weight, and discuss how these sites may interact to mediate the effects of amylin within the brain. Additionally, we review recent findings suggesting that amylin influences alcohol intake, suggesting broader effects of …

Usefulness of sugammadex in a patient with Becker muscular dystrophy and dilated cardiomyopathy.

A 54-year-old patient with Becker muscular dystrophy and dilated cardiomyopathy underwent laparoscopic cholecystectomy under total intravenous anesthesia. Muscle relaxation was induced by rocuronium (0.4 mg/kg body weight) under train-of-four (TOF) ratio monitoring. The TOF ratio was 0 at intubation, and 0.2 at the end of surgery. Residual muscle relaxant activity was successfully reversed by sugammadex (2 mg/kg body weight) without any hemodynamic adverse effects (TOF ratio 1.0 at extubation). The clinical and hemodynamic findings suggest that sugammadex can be safely used in patients with Be…

Anesthetic Pharmacology and the Morbidly Obese Patient.

Anesthesiologists are increasingly being faced with treating obese patients. Physiologic and anthropometric associated with obesity-most notably increases in cardiac output, changes in tissue perfusion and increases in total body weight (TBW), lean body weight (LBW), and fat mass affect the pharmacokinetics (PK) of anesthetic agents. In addition, redundancy of airway tissue, obstructive and central sleep apnea and CO2 retention affect the pharmacodynamics (PD) of anesthetics and narrow the therapeutic window of numerous anesthetic drugs. Safe and effective pharmacologic management of the obese…

Efficacy of Cymelarsan in the treatment of natural chronic Trypanosoma evansi infection in camels in the Sudan.

The efficacy of Cymelarsan, administered by intramuscular (i/m) injection in the treatment of chronic cases of camel trypanosomosis due to Trypanosoma evansi, was tested under controlled laboratory conditions. It was confirmed that Cymelarsan is a safe drug for use in dromedary camels when administered i/m at dose rates of 0.25 or 0.50 mg/kg body weight. During the 90 days post-treatment no relapses occurred at either dose rate. Hence, the drug was found to be fully effective against the chronic form of the natural disease. A dose of 0.25 mg/kg body weight of Cymelarsan given i/m is recommende…

Ventral tegmental area amylin / calcitonin receptor signaling suppresses feeding and weight gain in female rats.

The pancreatic peptide amylin promotes negative energy balance in part through activation of amylin receptors (AmyRs) expressed in the ventral tegmental area (VTA), but studies have been limited to male rodents. We evaluated whether VTA amylin signaling governs feeding and body weight in female rats. Indeed, pharmacological VTA AmyR activation suppressed chow intake and body weight in females. Viral-mediated knockdown of VTA calcitonin receptor (GPCR of AmyR) supports the physiological relevance of VTA amylin signaling for energy balance control in females. Collectively, these data support the…

Hypothermia in a patient with anorexia nervosa.

A young woman with anorexia nervosa had severe recurrent hypothermia. Initial evaluation revealed a thiamine deficiency. Administration of pharmacologic doses of thiamine improved voluntary food intake, and her temperature normalized. Following the restoration of body weight, withdrawal of supplemental thiamine produced a clinical relapse. After reinstitution of supplemental thiamine, the patient again became asymptomatic until she chose to consume a calorically-restricted diet. With progressive weight loss, hypothermia recurred and was unresponsive to thiamine therapy. Fortuitously, the patie…

Maternal Betaine and Methylation Potential during Pregnancy Are Associated with Child Growth at Birth and Early Childhood: Results of the Chilean Maternal & Infant Cohort Study-II.

BACKGROUND: One-carbon metabolism regulates growth through methyl group transfer required for DNA synthesis and epigenetic processes. In folate-replete populations, the contribution of betaine-dependent remethylation and methylation potential to postnatal growth remains unclear. OBJECTIVES: This study examined if maternal plasma concentrations of betaine, methionine, S-adenosylmethionine (SAM), S-adenosylhomocysteine (SAH), cystathionine, and the SAM:SAH ratio are associated with offspring growth and body composition at birth and during early childhood. METHODS: This study included 150 mothe
