Death of a seven-month-old child in a washing machine: a case report.

The authors present a case which brings out a unique modality of child homicide by placing the baby in a washing machine and turning it on. The murder was perpetrated by the baby's mother, who suffered from a serious depressive disorder. A postmortem RX and then a forensic autopsy were performed, followed by histologic examinations and toxicology. On the basis of the results of the autopsy, as well as the histology and the negative toxicological data, the cause of death was identified as acute asphyxia. This diagnosis was rendered in light of the absence of other causes of death, as well as th

Neurological course and correlated computerized tomography findings after severe closed head injury.

This study includes 124 patients with closed head injuries and with Glasgow Coma Scale (GCS) scores of less than or equal to 8, who were admitted over a 7 1/2-month period. The time at which death occurred after injury was bimodal: deaths occurred either within 48 hours or after 7 days or longer after injury. Neurological deterioration, however, occurred with equal frequency on Days 2 to 7 after injury. Patients who survived the first 48 hours and then suffered neurological deterioration did not differ from the total population in age, sex, GCS scores on admission, or pupillary reactivity, but

Ultrasound-Driven Interfacial Electron Modulation Reprograms Mitochondrial Metabolism for Glioblastoma Therapy.

Glioblastoma (GBM) remains difficult to treat because the restrictive blood-brain barrier (BBB), the hypoxic tumor microenvironments, and mitochondrial electron transport chain (ETC)-driven metabolic adaptability jointly limit therapeutic delivery and efficacy. Selective disruption of mitochondrial metabolism in GBM is challenging because ETC function is also essential for normal brain cells, and many therapeutic agents show limited BBB penetration or reduced activity under oxygen-limited conditions. Here, we report Ni/LDH@M, an ultrasound-responsive biomimetic Schottky nanoplatform assembled

CAR-T Cell Therapy for Glioblastoma: Obstacles and Advances.

Glioblastoma (GBM) is the most aggressive primary malignancy of the central nervous system. Chimeric antigen receptor T (CAR-T) cell therapy has shown promising therapeutic potential against GBM, yet its efficacy remains constrained by multiple barriers, including physical barriers imposed by the blood-brain barrier and extracellular matrix, the immunosuppressive tumor microenvironment, spatiotemporal antigen heterogeneity, and safety concerns. In this review, we summarize the major obstacles limiting CAR-T therapy in GBM and discuss emerging strategies to overcome these challenges. Next-gener

A single-cell transcriptomic dataset profiling traumatic brain injury and NeuroD1-based gene therapy in mice.

Traumatic brain injury (TBI), a leading cause of death and disability globally, often results in functional impairments due to complex pathological mechanisms and limited therapeutic options. Recently, astrocytes, the most abundant glia in the central nervous system, have been recognized as therapeutic targets for TBI due to their roles in neuroinflammation and neuroprotection. While NeuroD1-based gene therapy demonstrates therapeutic potential by reversing glial scar, suppressing neuroinflammation, and repairing brain tissue, the underlying mechanisms remain incompletely understood. To elucid

Protein-protein interaction-mediated signaling networks in ischemic stroke: from molecular mechanisms to therapeutic strategies.

The pathological progression of ischemic stroke is driven by a dynamic signaling network mediated by protein-protein interactions (PPI) that involves excitotoxicity, oxidative stress, neuroinflammation, and regulated cell death (RCD), ultimately leading to neurological dysfunctions. Instead of functioning independently, these PPI-driven pathways engage in extensive cross-talk, creating cycles that exacerbate the injury over both space and time. Therapeutic strategies designed to disrupt key nodal PPIs, comprising small molecule inhibitors, peptide mimetics, and chimeras targeting proteolysis (

Advances in small-molecule fluorescent probe development for bioimaging of ischemic stroke-associated biomarkers.

Ischemic stroke initiates with the interruption of cerebral blood flow, and reperfusion following revascularization may subsequently trigger cerebral ischemia-reperfusion injury. This secondary injury is characterized by dynamic molecular changes, including redox imbalance, mitochondrial dysfunction, blood-brain barrier (BBB) disruption, neuroinflammation, regulated cell death, microenvironmental remodeling, and ion homeostasis disturbance. Conventional neuroimaging is indispensable for anatomical and perfusion assessment, but it is less applicable to the real-time visualization of transient m

Mitochondrial DNA heteroplasmy drives cortical neuronal disturbances in human organoids harbouring the common m.3243A>G mutation.

Mitochondrial diseases frequently affect the brain leading to severe and disabling neurological symptoms. The heteroplasmic m.3243 A > G mutation in MT-TL1, encoding mt-tRNALeu, is responsible for ~80% of mitochondrial encephalomyopathy, lactic acidosis, and stroke-like episodes (MELAS), which is one of the most characteristic mitochondrial syndromes, leading to disability and early death. There are no animal models harbouring this mutation to provide precise mechanistic insights informing therapeutic interventions. Here, we generate a human iPSC-derived cerebral organoid slice model that reca

Alcohol-induced structural and cellular brain alterations: molecular and histopathological mechanisms.

Chronic alcohol consumption is a leading cause of acquired neurodegeneration with well-documented structural and ultrastructural brain alterations. This review analyzes the cellular and molecular mechanisms underlying alcohol neurotoxicity, integrating findings from animal models, human post-mortem studies, and neuroimaging investigations. Ethanol crosses the blood-brain barrier and generates toxic metabolites including acetaldehyde and reactive oxygen species, triggering oxidative stress, lipid peroxidation, and mitochondrial dysfunction. Chronic exposure induces glutamatergic and gamma-amino

Anti-herpetic tau preserves neurons via the cGAS-STING-TBK1 pathway in Alzheimer's disease.

Alzheimer's disease (AD) diagnosis relies on the presence of extracellular β-amyloid (Aβ) and intracellular hyperphosphorylated tau (p-tau). Emerging evidence suggests a potential link between AD pathologies and infectious agents, with herpes simplex virus 1 (HSV-1) being a leading candidate. Our investigation, using metagenomics, mass spectrometry, western blotting, and decrowding expansion pathology, detects HSV-1-associated proteins in human brain samples. Expression of the herpesvirus protein ICP27 increases with AD severity and strongly colocalizes with p-tau but not with Aβ. Modeling in

ML-Driven Alzheimer's disease prediction: A deep ensemble modeling approach.

Alzheimer's disease (AD) is a progressive neurological disorder characterized by cognitive decline due to brain cell death, typically manifesting later in life.Early and accurate detection is critical for effective disease management and treatment. This study proposes an ensemble learning framework that combines five deep learning architectures (VGG16, VGG19, ResNet50, InceptionV3, and EfficientNetB7) to improve the accuracy of AD diagnosis. We use a comprehensive dataset of 3,714 MRI brain scans collected from specialized clinics in Iraq, categorized into three classes: NonDemented (834 image

Pre-infection cerebral cortex structure predicts murine sepsis outcome.

Sepsis is a life-threatening condition caused by an inappropriate host response to an infection that can lead to multi-organ failure and death. Predictive markers that can inform the trajectory and outcome for a septic patient are necessary to inform treatment and increase the likelihood for patient survival. Here, by leveraging the concept of Lethal Dose 50 (LD50), which is the dose of a pathogen that will kill 50% of a genetically identical host population, we tested the hypothesis that variations in brain structure can be readily used to predict trajectory and outcome in a murine model of s

O-GlcNAcylation Suppressed Apoptosis and Ferroptosis in Traumatic Brain Injury by Enhancing Mitophagy.

O-linked-N-acetylglucosaminylation (O-GlcNAcylation), a distinctive post-translational modification (PTM), is ubiquitously present in numerous nuclear and mitochondrial proteins. The emerging role of O-GlcNAcylation is increasingly recognized for its involvement in various diseases. However, its role in traumatic brain injury (TBI) has not been explored. This study was aimed to explore the neuroprotection of O-GlcNAcylation in both in vivo and in vitro TBI models. Our results revealed that the levels of O-GlcNAcylation were increased after TBI. Up-regulation of O-GlcNAcylation by Thiamet G (TM

Quercetin attenuated ischemic stroke induced neurodegeneration by modulating glutamatergic and synaptic signaling pathways.

Источник пометил публикацию как отозванную

Ischemic strokes originate whenever the circulation to the brain is interrupted, either temporarily or permanently, resulting in a lack of oxygen and other nutrients. This deprivation primarily impacts the cerebral cortex and striatum, resulting in neurodegeneration. Several experimental stroke models have demonstrated that the potent antioxidant quercetin offers protection against stroke-related damage. Multiple pathways have been associated with quercetin's ability to safeguard the brain from ischemic injury. This study examines whether the administration of quercetin alters glutamate NMDA a

3D Neurovascular Unit Tissue Model to Assess Responses to Traumatic Brain Injury.

The neurovascular unit (NVU) is a critical interface in the central nervous system that links vascular interactions with glial and neural tissue. Disruption of the NVU has been linked to the onset and progression of neurodegenerative diseases. Despite its significance the NVU remains challenging to study in a physiologically relevant manner. Here, a 3D cell triculture model of the NVU is developed that incorporates human primary brain microvascular endothelial cells, astrocytes, and pericytes into a tissue system that can be sustained in vitro for several weeks. This tissue model helps recapit

Therapeutic Potentials of MicroRNA-126 in Cerebral Ischemia.

Stroke is a leading cause of death and disability worldwide. It is among the most common neurological disorders with an 8-10% lifetime risk. Ischemic stroke accounts for about 85% of all strokes and damages the brain tissue via various damaging mechanisms. Following cerebral ischemia, the disrupted blood-brain barrier (BBB) leads to cerebral edema formation caused by activation of oxidative stress, inflammation, and apoptosis, targeting primarily endothelial cells. Activation of the protective mechanisms might favor fewer damages to the neural tissue. MicroRNA (miR)-126 is an endothelial cell-

Endocrine Dysfunction After Traumatic Brain Injury: An Ignored Clinical Syndrome?

Traumatic brain injury (TBI) incurs substantial health and economic burden, as it is the leading reason for death and disability globally. Endocrine abnormalities are no longer considered a rare complication of TBI. The reported prevalence is variable across studies, depending on the time frame of injury, time and type of testing, and variability in hormonal values considered normal across different studies. The present review reports evidence on the endocrine dysfunction that can occur after TBI. Several aspects, including the pathophysiological mechanisms, clinical consequences/challenges (i

Cannabidiol's neuroprotective properties and potential treatment of traumatic brain injuries.

Cannabidiol (CBD) has numerous pharmacological targets that initiate anti-inflammatory, antioxidative, and antiepileptic properties. These neuroprotective benefits have generated interest in CBD's therapeutic potential against the secondary injury cascade from traumatic brain injury (TBI). There are currently no effective broad treatment strategies for combating the damaging mechanisms that follow the primary injury and lead to lasting neurological consequences or death. However, CBD's effects on different neurotransmitter systems, the blood brain barrier, oxidative stress mechanisms, and the
