Phylogeny of whey acidic protein (WAP) four-disulfide core proteins and their role in lower vertebrates and invertebrates.

Proteins containing WAP (whey acidic protein) domains with a characteristic WFDC (WAP four-disulfide core) occur not only in mammals (including marsupials and monotremes) but also in birds, reptiles, amphibians and fish. In addition, they are present in numerous invertebrates, from cnidarians to urochordates. Many of those from non-mammalian groups are poorly understood with respect to function or phylogeny. Those well characterized so far are waprins from snakes, perlwapins from bivalves and crustins from decapod crustaceans. Waprins are venom proteins with a single WAP domain at the C-termin

TPC2 proteins mediate nicotinic acid adenine dinucleotide phosphate (NAADP)- and agonist-evoked contractions of smooth muscle.

Agonists such as those acting at muscarinic receptors are thought to induce contraction of smooth muscle primarily through inositol 1,4,5-trisphosphate production and release of Ca(2+) from sarcoplasmic reticulum. However, the additional Ca(2+)-mobilizing messengers cyclic adenosine diphosphate ribose (cADPR) and nicotinic acid adenine dinucleotide phosphate (NAADP) may also be involved in this process, the former acting on the sarcoplasmic reticulum, the latter acting on lysosome-related organelles. In this study, we provide the first systematic analysis of the capacity of inositol 1,4,5-tris

SmC2P1, a C2 domain protein from Scophthalmus maximus that binds bacterial pathogen-infected lymphocytes and reduces bacterial survival.

C2 domains are protein structural modules found in many eukaryotic proteins involved in signal transduction, membrane trafficking, and immune defense. Most of the studied C2 domain-containing proteins are multi-domained in structure, in which the C2 domain is an independently folded motif and plays an essential role in calcium-dependent membrane-targeting. Although C2 domains isolated from intact proteins have been studied for biological functions, no study on natural proteins containing C2 domain only has been documented. In this study, we identified a Scophthalmus maximus protein SmC2P1 that

Recognition and activation of ryanodine receptors by purines.

Ryanodine receptor (RyR) is a tetrameric, high molecular weight protein that functions as a calcium release channel. It plays a key role in phenomena such as signal transduction, excitation-contraction and excitation-secretion coupling. Hyperthermia maligna, central core disease and myocardial infarction have been related with RyR dysfunction. RyR is present as three isoforms in vertebrates: RyR 1 mainly localized in skeletal muscle, RyR 2 in cardiac muscle, and RyR 3 in nervous system. RyR is regulated by a number of physiological and pharmacological factors. Main physiological modulators: ca

Solution behaviour of myo-inositol hexakisphosphate in the presence of multivalent cations. Prediction of a neutral pentamagnesium species under cytosolic/nuclear conditions.

myo-Inositol hexakisphosphate (InsP6) is an ubiquitous and abundant molecule in the cytosol and nucleus of eukaryotic cells whose biological functions are incompletely known. A major hurdle for studying the biology of InsP6 has been a deficiency of a full understanding of the chemistry of its interaction with divalent and trivalent cations. This deficiency has limited our appreciation of how it remains in solution within cells, and the likely degree to which it might interact in vivo with physiologically important cations such as Ca2+ and Fe3+. We report here the initial part of the descriptio

Analysis of biogenic carbonates by inductively coupled plasma-mass spectrometry (ICP-MS). Flow injection on-line solid-phase preconcentration for trace element determination in fish otoliths.

The aragonite deposits within the ear bones (otoliths) of teleost fish retain a chemical signal reflecting the life history of fish (similar to rings of trees) and the nature of fish habitats. Otoliths dissolved in acid solutions contain high concentrations of calcium and a variety of proteins. Elimination of matrix salts and organic interferences during preconcentration is essential for accurate determination of trace elements in otolith solutions by inductively coupled plasma-quadrupole mass spectrometry. An iminodiacetate-based chelating resin (Toyopearl AF-Chelate 650 M) has been used for

MCP-1 but not CINC synthesis is increased in rat pancreatic acini in response to cerulein hyperstimulation.

Inflammatory mediators including chemokines play a critical role in acute pancreatitis. The precise nature of early inflammatory signals within the pancreas remains, however, unclear. We examined the ability of isolated pancreatic acini to synthesize CC chemokine monocyte chemotactic protein-1 (MCP-1) and CXC chemokine cytokine-induced neutrophil chemoattractant (CINC) and the response to the secretagogue cerulein at physiological and supraphysiological concentrations. Isolated rat pancreatic acini maintained in short-term (< or =48 h) primary culture constitutively synthesized MCP-1 and CINC.

Distinct effects of N-acetylcysteine and nitric oxide on angiotensin II-induced epidermal growth factor receptor phosphorylation and intracellular Ca(2+) levels.

These studies describe inhibitory effects of N-acetylcysteine on several biochemical events associated with the activation of extracellular signal-regulated kinases (ERK) by angiotensin II in the cardiac fibroblast and compare these effects with those of the nitric oxide donor, S-nitroso-N-acetylpenicillamine, an agent we showed previously to inhibit angiotensin II-induced ERK activation and the concomitant phosphorylation of proline-rich tyrosine kinase 2 (Wang, D., Yu, X., and Brecher, P. Regulatory effect of CD9 on calcium-stimulated phosphatidylserine exposure in Jurkat T lymphocytes.

Alteration in membrane-phosphatidylserine (PS) asymmetry occurs during lymphocyte apoptosis, but the mechanism and regulation of this process are not well understood. We investigated the possible involvement of CD9, a member of the tetraspan family of membrane proteins, in the PS-exposure process in Jurkat T cells; flow cytometry with FITC-annexin V was used to detect the PS-positive cells. We found that antibody to CD9 inhibited the prompt (within 5 min) PS exposure stimulated by calcium ionophore A23187, whereas it had no effect on resting cells. Antibodies against other control antigens (CD

Impaired potassium-induced insulin secretion in chronic renal failure.

Extrarenal disposal of potassium load is impaired in chronic renal failure (CRF). This has been attributed to excess PTH since extrarenal disposition of potassium is normal in CRF-PTX animals. Insulin augments potassium entry into cells and hyperkalemia stimulates insulin secretion. Since glucose-induced insulin secretion is impaired in CRF and normal in CRF-PTX, it is possible that K(+)-induced insulin secretion is also impaired in CRF due to excess PTH. Such a defect would contribute to the abnormality in extrarenal disposal of potassium in CRF. We examined K(+)-induced insulin secretion, cy

Activation of amino acid uptake at fertilization in the sea urchin egg. Requirement for proton compartmentalization during cytosolic alkalosis.

The comparative importance of the release of intracellular ionic calcium, Na+/H+ exchange and cytosolic alkalosis as activator signals was studied on the development of amino acid uptake at fertilization in sea urchin eggs. We show that, once stimulated, the rate of valine uptake is greatly dependent upon intracellular pH. Suppression of the Na+/H+ exchange at the time of activation, by applying ionophore (A23187) in sodium-free artificial sea water (ONaASW), inhibits the development of valine influx. This cannot be restored by a further (30 min later) alkalosis by transferring eggs into sea w

Spinal Dural Ossifications Are Hyperintense on T1-Weighted Sequence and May Mimic Dural Lesions.

Spinal dural ossifications (SDOs) are bone plaques on the dura mater surface. They are usually incidental findings in humans and dogs, and rarely can cause clinical signs. The authors noticed that SDOs on magnetic resonance imaging (MRI) may appear hyperintense on a pre-contrast T1-weighted (T1W) sequence. This study aimed to assess the MRI signal of SDOs in dogs. For this retrospective descriptive study, the hospital archive was searched for dogs that underwent both a pre-contrast computed tomography (CT) study and at least one pre-contrast T1W MRI sequence, from January 2020 to April 2024. The Role of Neuritin 1 in Synaptic Plasticity and Sensory Nerve Function: Integrator of Neurotrophic, Metabolic and Injury Signals.

Neuritin 1 (NRN1) has emerged as a multifaceted regulator of synaptic plasticity, neuronal excitability and structural remodelling. This review synthesises knowledge of NRN1 function across the central and peripheral nervous systems, with a focus on its roles in sensory neurones and neuronal repair following injury. We discuss evidence that NRN1 interacts with classical neurotrophic pathways, including brain-derived neurotrophic factor (BDNF) and nerve growth factor (NGF), while engaging distinct cellular mechanisms that span activity-dependent trafficking, modulation of calcium and potassium

Irisin modulates the transcriptomic profile of porcine anterior pituitary cells isolated from gilts on days 15-16 of pregnancy.

Irisin, an adipomyokine primarily known for its role in metabolic regulation, has recently emerged as a potential regulator of reproductive function. While its ovarian actions have been partially characterised, its influence on the anterior pituitary remains largely unexplored. This study aimed to investigate the in vitro effects of irisin on the transcriptome of porcine anterior pituitary cells (APc) isolated from gilts on days 15-16 of pregnancy. Primary APc were isolated from pigs on days 15-16 of pregnancy and treated with 300 ng/mL of irisin. RNA sequencing (NovaSeq 6000, Illumina) was pe

Butyrylcholinesterase alleviates hepatic stellate cell activation via paracrine muscarinic signalling.

BACKGROUND: Butyrylcholinesterase (BuChE) is a metabolic enzyme implicated in Alzheimer's disease, detoxification and depression. Although reduced BuChE expression is recognised in cirrhosis, its functional role in fibrogenesis remains undefined. OBJECTIVE: We investigated whether BuChE regulates fibrosis by modulating acetylcholine (ACh) bioavailability and identified downstream cholinergic signalling. DESIGN: BuChE expression was assessed via qPCR and immunostaining in patient biopsies. Its contribution was studied using Buche ΔHep, Buche KI/KI-Hep mice, pharmacological inhibition, mutant

Lack of vitamin D signalling per se does not aggravate cardiac functional impairment induced by myocardial infarction in mice.

Epidemiological studies have linked vitamin D deficiency to an increased incidence of myocardial infarction and support a role for vitamin D signalling in the pathophysiology of myocardial infarction. Vitamin D deficiency results in the development of secondary hyperparathyroidism, however, the role of secondary hyperparathyroidism in the pathophysiology of myocardial infarction is not known. Here, we aimed to explore further the secondary hyperparathyroidism independent role of vitamin D signalling in the pathophysiology of myocardial infarction by inducing experimental myocardial infarction
