Aliphatic polycarbonates with acid degradable ketal side groups as multi-pH-responsive immunodrug nanocarriers.

Pharmacokinetics and biodistribution profiles of active substances are crucial aspects for their safe and successful administration. Since many immunogenic compounds do not meet all requirements for safe and effective administration, well-defined drug nanocarrier systems are necessary with a stimuli-responsive drug-release profile. For this purpose, a novel pH-responsive aliphatic cyclic carbonate is introduced with benzyl ketal side chains and polymerized onto a poly(ethylene glycol) macroinitiator. The resulting block copolymers could be formulated via a solvent-evaporation method into well-

Supramolecular Self-Assembly and Metal-Ligand-Enhanced Organic Room-Temperature Phosphorescence for Live Cell Imaging.

A purely organic supramolecular assembly emitting room-temperature phosphorescence (RTP) was constructed from 1-(3-(bis(pyridin-2-ylmethyl)amino)propyl)-4-(4-bromophenyl)pyridin-1-ium bromide (BP-DPA) by complexing with cucurbit[8]uril (Q[8]) and Zn2+ through a supramolecular assembly. Benefiting from the strong affinity and the macrocyclic restriction effect of Q[8], phosphorescence emission was achieved by encapsulating BP-DPA in the Q[8] cavity. Notably, secondary assembly with Zn2+ formed the supramolecular complex BP-DPA@Q[8]⊂Zn2+, which greatly contributed to the enhancement of the phosp

Transdermal sequential delivery of functionalized Nano-Deep eutectic system for enhanced treatment of melanoma.

In the present study, we introduce the concept of "transdermal sequential delivery" as a non-invasive and synergistic approach for the treatment of melanoma. We developed a functionalized Deep Eutectic System (DES) that incorporates both small molecule drugs and nanoparticles. The glycolysis inhibitor 2-deoxy-D-glucose (2-DG) served as the Hydrogen Bond Donor (HBD) to form the DES, while glutathione (GSH)-responsive Mesoporous Organosilicon Nanoparticles (MON) were prepared and encapsulated with chlorin e6 (Ce6). These nanoparticles were incorporated into the DES through surface-modified citri

Intranasal Delivery of Hydrophobic AC5216 Loaded Nanoemulsion into Brain To Alleviate Chronic Unpredictable Stress-Induced Depressive-like Behaviors.

Major depressive disorder (MDD) represents a widespread mental health condition. Efficiently moving therapeutic substances across the blood-brain barrier (BBB) remains a critical obstacle in addressing depressive disorders. AC5216, identified as a translocator protein (TSPO) ligand and considered a potential treatment for major depressive disorder (MDD), faces limitations due to its subpar druggability and oral bioavailability. In this context, an amphiphilic polymer composed of polyethylene glycol, poly-l-lysine, and poly(lactic-co-glycolic acid) (PEG-PLL-PLGA) has been utilized to encapsulat

A smart nanocomposite bioactive ink for controlled siRNA delivery in calvarial mesenchymal stromal cells as a minimally invasive treatment for craniosynostosis.

Craniosynostosis (CS), characterized by the premature fusion of cranial sutures, often results from aberrant activation of Fibroblast growth factor receptor 2 (FGFR2), a major regulator of osteogenic differentiation in cranial mesenchyme. Despite surgical interventions, recurrence and complications remain common, underscoring the need for targeted molecular therapies. In this study, we developed a novel formulation of bioactive nanocomposite hydrogel-based ink designed for localized, sustained delivery of therapeutic small interfering RNAs (siRNAs) targeting FGFR2. The delivery system combines

Hydrogels Targeting Mechanical-biological signaling transitions of interleukin-33 alleviates intervertebral-disc degeneration.

Mechanical-biological signaling transitions (MBSTs) cause intervertebral-disc degeneration (IVDD). The key to MBSTs lies in nucleus pulposus cells (NPCs), but it is not yet fully understood, and relevant therapeutic targets and strategies are lacking. In this study, we found that the key mechanobiological-signaling factor interleukin-33 (IL-33) played an important role in stress-induced IVDD through a piezo-type mechanosensitive ion channel component 1 (PIEZO1)-mediated positive-feedback loop. Specifically, analysis of clinical samples showed that IL-33 was significantly upregulated in NPCs fr

Multifunctional Biomimetic Nanomedicine for Osteoarthritis Alleviation by Mitigating Cartilage Degeneration and Promoting Cartilage Regeneration.

Osteoarthritis (OA) is a long-standing degenerative condition of the joints, defined by the progressive loss of articular cartilage accompanied by ongoing inflammation. For OA to be effectively treated, reducing cartilage breakdown and enhancing its regeneration are essential objectives. However, OA involves chronic sustained inflammation, making it difficult to protect cartilage and promote its regeneration under inflammatory conditions for effective OA treatment. To address these challenges, a kartogenin (KGN)-loaded biomimetic targeting system, designated CM@Lipos-KGN, was developed by inte

"Sandwich" structured nanorods for timely antibiosis, immunoregulation and neuroangiogenesis to accelerate osteogenesis of Zn-based implants in diabetics.

Pathological microenvironment of diabetes predisposes to induce bacterial infection, sustained inflammatory response, and impaired neuroangiogenesis, impeding fracture repair. Zn-based implants have great potential to replace conventional Ti implants as temporary fixations, but their osseointegration ability needs improvement. In this work, "sandwich" structured nanorods were constructed on Zn substrate with ZnO nanorods as core, riboflavin-encapsulated ZIF-8 as intermediate layer and HA as outer shell. Under normal physiological condition, ZIF-8 and HA reduce degradation of Zn substrate, decr

Promoting ROS scavenging and osteogenic differentiation by metal-phenolic network nanoparticle-embedded microenvironment-responsive hydrogel for the repair of senile osteoporotic bone defects.

Senile osteoporosis (SOP) features reduced bone density and degraded trabecular structure, primarily mediated through senescent impairment of osteoblasts that disrupts coupled bone remodeling homeostasis. This pathological process induces systemic bone resorption and localized bone destruction accompanied by architectural deterioration. Existing treatments for senile osteoporosis tend to focus on a single mechanism, making it challenging to simultaneously address bone formation deficits and oxidative stress. Herein, we developed a multifunctional nanoplatform utilizing metal-phenolic networks

Plant-Derived Nanovesicle Enhanced Microribonucleic Acid (MicroRNA) Transfer from Nasal Cavity to the Brain.

The nasal-to-brain route for drug delivery is potentially useful for the treatment of brain disorders. Not only chemically modified molecules but also nanosized carrier vesicles, such as cell culture-derived exosomes, as small molecules, could be delivered via this route. The focus of the present study was to evaluate the potential of vesicular plant-derived nanoparticles (PDNPs), which have been identified as multivesicular bodies, for transporting molecules via the nasal-to-brain route. PDNPs were isolated from the edible parts of onion (Allium cepa), cherry tomato (Lycopersicon esculentum v

Co-encapsulated Ce6 + CpG and biopeptide-modified liposomes for enhanced transdermal photo-immunotherapy of superficial tumors.

Cancer immunotherapy encounters challenges of a low treatment response rate due to the tumor immunosuppressive microenvironment and immune-related adverse events caused by off-target immunotherapy agents delivered through systemic administration in clinical practice. Photodynamic therapy (PDT) offers a viable approach to improve the immunotherapy efficacy through inducing immunogenic tumor cell death and is particularly advantageous in superficial tumor treatment. Therefore, leveraging integrated nanomaterials for photo-immunotherapy appears to be an ideal strategy to improve therapeutic outco
