Propofol Inhibits the Stem Characteristics and Tumor Growth of NSCLC by Altering the Expression and Localization of HDAC1.

Most treatments for advanced NSCLC are no longer effective due to drug resistance, causing tumor progression. Cancer stem cells drive tumor development and treatment resistance. Moreover, HDAC1 and ARID1A play a role in tumor progression in EGFR-mutated NSCLC. This study aimed to investigate the role of HDAC1 in propofol-mediated antitumor effects in NSCLC using A549 and HCC827 cell lines and NSCLC nude mouse tumor models. Our results demonstrated that propofol effectively inhibited the stemness, activity, proliferation, migration, and invasion of NSCLC cells and tumor growth in NSCLC nude mic

Calcium-Sensing Receptor Promotes Breast Cancer by Stimulating Intracrine Actions of Parathyroid Hormone-Related Protein.

Parathyroid hormone-related protein (PTHrP) contributes to the development and metastatic progression of breast cancer by promoting hypercalcemia, tumor growth, and osteolytic bone metastases, but it is not known how PTHrP is upregulated in breast tumors. Here we report a central role in this process for the calcium-sensing receptor, CaSR, which enables cellular responses to changes in extracellular calcium, through studies of CaSR-PTHrP interactions in the MMTV-PymT transgenic mouse model of breast cancer and in human breast cancer cells. CaSR activation stimulated PTHrP production by breast

Selective lysis of the autologous tumor by delta TCS1+ gamma/delta+ tumor-infiltrating lymphocytes from human lung carcinomas.

Two distinct non-overlapping populations of TcR1+ (gamma/delta) T cells have been described: the first, bearing the disulfide-linked gamma/delta heterodimer, is predominant in the peripheral blood; the second, expressing the non-disulfide-linked form of TcR1, is mostly confined to epithelial tissues (lung, gut, skin). TcR1+ lymphocytes may be cytotoxic and could be involved in anti-tumor immunity, especially against tumors at epithelial sites. Freshly derived tumor-infiltrating lymphocytes (TIL) obtained from two patients with lung cancer were enriched in CD3+WT31- cells. The percentage of thi

Impact of Acidosis-Regulated MicroRNAs on the Expression of Their Target Genes in Experimental Tumors In Vivo.

In comparison to normal tissue, solid tumors show an acidic extracellular pH, which results from hypoxia-induced glycolytic metabolism and the Warburg effect. Since acidosis modulates the expression of different microRNAs (e.g., miR-7, miR-183, miR-203, miR-215), microRNAs and their targets might be mediators between tumor acidosis and malignant behavior. The aim of this study was to investigate how modulation of these microRNAs affects the expression of their targets (Crem, cAMP-responsive element modulator; Gls2, glutaminase 2; Txnip, thioredoxin-interacting protein) in experimental tumors i

Use of Cold Atmospheric Plasma in the Treatment of Squamous Cell Carcinoma: in vitro Effects and Clinical Application in Feline Tumors: A Pilot Study.

Cold atmospheric plasma (CAP) has shown promising results against squamous cell carcinoma (SCC) in both in vivo and in vitro assays, mainly in humans and mice. Its applicability for treatment of feline tumors, however, remains unknown. This study aimed to evaluate the anticancer effects of CAP on a head and neck squamous cell carcinoma (HNSCC) cell lineage and against a clinical case of cutaneous SCC in a cat. Control and treatment groups employing the HNSCC cell line (SCC-25) were used, the latter exposed to CAP for 60 seconds, 90 seconds, or 120 seconds. The cells were subjected to the MTT a

Establishment of a cell line from a Japanese patient useful for generating an in vivo model of malignant pleural mesothelioma.

Malignant pleural mesothelioma is a refractory tumor with increasing incidence. In the present study, we established six mesothelioma cell lines possessing two allele deletions of the p16(INK4A) gene and one allele deletion of the neurofibromatosis type 2 gene, MM16, MM21, MM26, MM35, MM46 and MM56, from pleural effusion fluids or surgically resected tumors of Japanese patients. MM21, MM26 and MM46 cells failed to develop tumors in BALB/c-nude mice following subcutaneous inoculation. MM16 and MM35 cells slowly generated tumors at the site of subcutaneous inoculation in BALB/c-nude mice, but lo

Cucurbitacin B induces immunogenic cell death and activates adaptive immunity in cutaneous melanoma via the ROS-STAT3-eIF2α/IRE1 pathway.

BACKGROUND: Immunotherapy is the first-line treatment for advanced cases in cutaneous melanoma (CM). However, nearly half of patients develop resistance or recurrence, mainly attributed to an immunosuppressive tumor immune microenvironment (TIME). Cucurbitacin B (CuB), a tetracyclic triterpenoid from Cucurbitaceae plants, has broad-spectrum anticancer activity, yet its role and mechanism in regulating CM-associated immunogenic cell death (ICD) and TIME remain unclear. PURPOSE: To investigate the antitumor effects of CuB on CM, and explore its mechanisms in inducing ICD. STUDY DESIGN AND METH

Modulating STAT3 pathway in breast cancer: Improved activity of T40214 aptamer by peptide-based nanofibers delivering.

G-quadruplex (G4)-forming aptamers are highly selective oligonucleotides with significant potential in cancer therapy, acting as inhibitors of oncogenic proteins such as Signal Transducer and Activator of Transcription 3 (STAT3). Among them, the G4-forming aptamer T40214 (STAT) effectively modulates STAT3 signaling; however, its clinical translation is currently hindered by poor cellular internalization. In this study, we developed a multifunctional peptide amphiphile (PA)-based nanofiber (NF) to enhance the intracellular delivery and tumor selectivity of the G4 STAT aptamer in breast cancer c

Decorated gold nanoparticles on baicalein/chitosan-modified zinc oxide nanoparticles for one-pot preparation of pyrano[2,3-d]pyrimidines and inhibiting the gastrointestinal stromal tumor progression by controlling the gene expression of the cell cycle.

A composite of gold nanoparticles decorated on baicalein/chitosan-modified zinc oxide nanoparticles (ZnO@CS-baicalein/Au NPs) was synthesized and tested in a gastrointestinal stromal tumor cell line. The prepared nanocomposite was characterized using SEM and TEM, FT-IR, EDX, XRD, ICP-OES, and elemental mapping. The heterogeneous catalyst demonstrated excellent catalytic performance in synthesizing pyrano[2,3-d]pyrimidines via a three-component reaction involving malononitrile, aryl aldehydes, and barbituric acid. Additionally, ZnO@CS-baicalein/Au NPs showed good recyclability, maintaining cata

T-type voltage-activated calcium channel Cav3.1, but not Cav3.2, is involved in the inhibition of proliferation and apoptosis in MCF-7 human breast cancer cells.

T-type voltage-gated Ca2+ channels have unique electrophysiological properties, suitable for generating Ca2+ oscillations and waves and thus controlling the proliferation of various tumor cells. In the present study, we investigated the role of Cav3.1, a candidate tumor suppressor gene, in neoplastic processes, and compared the differences between Cav3.1 with Cav3.2 channels. While the overexpression of a full-length Cav3.1 clone suppressed cell proliferation, the knockdown of the Cav3.1 gene by siRNA, or treatment with ProTx-I, a relatively selective inhibitor for Cav3.1, promoted the cell pr

A chemoresistance-associated proliferation gene signature identified by single-cell and machine learning analysis predicts prognosis in high-grade serous ovarian carcinoma.

BACKGROUND: High-grade serous ovarian carcinoma (HGSOC) features extensive intratumoral heterogeneity, frequent chemoresistance and poor prognosis. Tumor proliferation kinetics reflected by tumor doubling time (TDT) are linked to therapeutic response, yet molecular drivers of chemoresistance-associated proliferation remain incompletely defined. METHODS: We integrated scRNA-seq (GSE154600) with bulk data (TCGA-OV, GTEx). Differential analyses of refractory/resistant versus sensitive tumors and tumor versus normal tissues, intersected with TDT-associated genes, identified 105 chemoresistance-as

GPR39 Suppresses Ferroptosis via the Nrf2/SLC7A11 Axis and Reduces the Sensitivity of Colorectal Cancer to Anti-PD-1 Immunotherapy.

BACKGROUND: PD-1 blockade has yet to achieve broad clinical success in colorectal cancer (CRC), with microsatellite-stable tumors proving especially resistant. At the same time, ferroptosis has emerged as a mechanistic link between redox control in tumor cells and the antitumor immune response. GPR39 is overexpressed in CRC, but its functional role in ferroptosis and immunotherapy resistance is currently unclear. METHODS: GPR39 expression was analyzed in human and mouse CRC cell lines. GPR39 knockdown, with or without the ferroptosis inhibitor liproxstatin-1, was performed to determine whethe

HCG11 up-regulation induced by ELK4 suppressed proliferation in vestibular schwannoma by targeting miR-620/ELK4.

BACKGROUND: Vestibular schwannoma (VS) is a kind of benign tumor deriving from the acoustic nerve sheath. Substantial long non-coding RNAs (lncRNAs) were illustrated to have crucial roles in multiple cancers. However, few lncRNAs were elucidated in VS. METHODS: HCG11, miR-620 and ELK4 expression were tested by RT-qPCR. Gain-of-function experiments were conducted to confirm the effect of HCG11 on VS. RESULTS: HCG11 possessed a low expression in VS cell lines. Overexpression of HCG11 repressed cell proliferation but accelerated apoptosis of VS cells. Moreover, we identified ELK4 stimulated the

Combination of the MEK inhibitor pimasertib with BTK or PI3K-delta inhibitors is active in preclinical models of aggressive lymphomas.

BACKGROUND: Lymphomas are among the most common human cancers and represent the cause of death for still too many patients. The B-cell receptor with its downstream signaling pathways represents an important therapeutic target for B-cell lymphomas. Here, we evaluated the activity of the MEK1/2 inhibitor pimasertib as single agent and in combination with other targeted drugs in lymphoma preclinical models. MATERIALS AND METHODS: Cell lines derived mature B-cell lymphomas were exposed to increasing doses of pimasertib alone. Immunoblotting and gene expression profiling were performed. Combinatio

Variable effects of retinoids on two pigmenting human melanoma cell lines.

Two melanoma cell lines, each derived from a different patient with metastatic disease, were very similar in their appearance, their growth characteristics, and their tendency to differentiate and to pigment in culture as they become confluent. These lines, UCT-Mel 1 and UCT-Mel 2, were used to study the effects of retinoic acid and other derivatives of vitamin A. When added to UCT-Mel 1 cells, retinoids had only a modest effect on plasminogen activator release and were without measurable effect on morphology, growth, or tyrosinase synthesis. In contrast, when added to UCT-Mel 2 cells, retinoi

Effect of GEN1 interference on the chemosensitivity of the breast cancer MCF-7 and SKBR3 cell lines.

Chemotherapy is a notable method for the treatment of breast cancer. Numerous genes associated with the sensitivity of cancer to chemotherapy have been found. In recent years, evidence has suggested that a particular structure termed Holliday junction (HJ) plays a crucial role in cancer chemosensitivity. Targeting HJ resolvases, such as structure-specific endonuclease subunit SLX4 (Slx4) and MUS81 structure-specific endonuclease subunit (Mus81), significantly increases the chemosensitivity of tumor cells. Flap endonuclease GEN homolog 1 (GEN1) is a HJ resolvase that belongs to the Rad2/xeroder

Aberrant melanogenesis and melanocytic tumour development in transgenic mice that carry a metallothionein/ret fusion gene.

We generated four independent transgenic mouse lines that showed severe melanosis of the whole body by introducing the ret oncogene fused to the mouse metallothionein (MT)-I promoter-enhancer (MT/ret). Whereas melanogenesis was accelerated without distinct proliferative disorders in one line, melanocytic tumours frequently developed in the other three lines. Northern hybridization and in situ hybridization analyses showed that tumour cells and non-tumorous melanin-producing cells expressed the transgene at high levels. The aberrant melanogenesis and tumour development were influenced by geneti

Targeting thymidylate synthase enhances CD8 + T-cell infiltration and inhibits tumor growth in cervical cancer.

Cervical cancer (CESC) presents a significant clinical challenge, primarily due to an incomplete understanding of the immunometabolic crosstalk within the tumor microenvironment (TME) and the lack of reliable biomarkers for immunotherapy stratification. Thymidylate synthase (TYMS), a pivotal enzyme in nucleotide synthesis, has been implicated in tumor progression, but its role as an immunometabolic regulator in CESC remains unexplored. Through an integrative approach combining single-cell RNA sequencing (scRNA-seq) of 47,589 cells, bulk transcriptomics, functional assays, and in vivo modeling,
