Identification of a Candidate Sex Determination Region and Sex-specific Molecular Markers of Spinibarbus caldwelli.

Spinibarbus caldwelli is a commercially significant aquaculture species in China that exhibits pronounced sexual dimorphism in growth-related traits. However, identifying sex based on morphological characteristics remains challenging before sexual maturation, necessitating the development of sex-specific molecular markers for sex-controlled breeding. In this study, whole-genome resequencing was performed on 50 individuals (25 females and 25 males) with an average sequencing depth of 10.05×. A genome-wide association study (GWAS) identified 140 loci significantly associated with sex, all locate…

Multicohort analysis unveils axon guidance pathways linking small for gestational age to spirometric restriction.

Children born small for gestational age (SGA) face elevated risks of metabolic, cardiovascular, respiratory, and neurodevelopmental disorders, as well as premature mortality, yet the underlying mechanisms remain only partly understood. We analyze blood proteomic data from multiple birth cohorts to identify molecular pathways linked to SGA and to later-life lung function. We find that approximately one-third of SGA children exhibit a distinct molecular endotype marked by dysregulation of axon-guidance proteins in cord blood. In peripheral blood collected later in life, these proteins are invers

Shared Genetic Basis Between Systemic Lupus Erythematosus and Inflammatory Bowel Disease in East Asian Ancestry: A Genome-Wide Cross-trait Analysis.

Systemic lupus erythematosus (SLE) and inflammatory bowel disease (IBD) are both categorized as autoimmune disorders and have similar non-specific gastrointestinal symptoms. SLE and IBD have shown shared genetic architecture in European ancestry; however, given the ancestry-specificity of genetic architecture, the shared genetic structure in East Asian ancestry remains unclear. This study reveals significant global and local genetic overlap between SLE and IBD subtypes in East Asian ancestry by using genetic correlation analyses. Cross-trait and colocalization analyses identify 64 shared loci

Proton pump inhibitors use and the risk of osteoporosis and fractures: A two-sample Mendelian randomization study.

The causal relationship between proton pump inhibitor (PPI) use and bone health outcomes remains uncertain. This study employs a Mendelian randomization (MR) approach to investigate the potential causal association between PPI use and the risk of osteoporosis and fractures. We selected 4 representative PPIs, including omeprazole, esomeprazole, lansoprazole, and rabeprazole, for our study. Bone health outcomes were evaluated through femoral neck bone mineral density (BMD), lumbar spine BMD, and the prevalence of osteoporosis and fracture across various anatomical sites, including the upper arm

Genetic analysis of inflammatory proteins following BCG vaccination in Danish children.

The Bacillus Calmette-Guérin (BCG) vaccine, primarily designed for tuberculosis, exerts non-specific immunological effects that vary considerably among children. Limited knowledge exists regarding genetic variants that can impact immune responses post-vaccination. Understanding the genetic factors influencing the immune response in children following BCG vaccination is crucial for optimizing vaccine strategies. Genome-wide pQTL mapping of 65 circulating inflammatory proteins profiled at Month 13 uncovered 11 independent genome-wide significant loci. Following pQTL mapping in BCG-unvaccinated c

Shared genetic architecture between sleep traits and cardiometabolic diseases.

BACKGROUND: Sleep traits, including sleep apnoea (SA), insomnia, daytime sleepiness, and snoring, frequently co-occur with cardiometabolic diseases (CMDs), with shared genetic factors suspected to underlie these associations. However, the contribution of shared genetic determinants to these associations is not fully understood. METHODS: We conducted a genome-wide pleiotropic association study applying sequential genetic methods to identify shared genetic variants, genes, pathways and causal associations between the four sleep traits and seven CMDs, including LDSC, high-definition likelihood a

Causal effects of circulating cathepsins on cholecystitis, cholelithiasis, primary biliary cholangitis, primary sclerosing cholangitis, acute pancreatitis, and chronic pancreatitis: A Mendelian randomization study.

Gallbladder, biliary, and pancreatic disorders are nonneoplastic diseases marked by inflammation, epithelial injury, and fibrosis. Cathepsins are lysosomal proteases involved in inflammatory activation, immune regulation, extracellular matrix turnover, and pancreatic enzyme activation. Observational links between dysregulated cathepsins and hepatopancreatobiliary pathology could reflect confounding or reverse causation. This study used Mendelian randomization (MR) to assess whether circulating cathepsins causally influence gallbladder, biliary, and pancreatic diseases. We conducted a 2-sample

Integrative genetic analysis identifies shared regulation of DNA methylation and gene expression in migraine risk.

BACKGROUND: Migraine affects approximately 14% of adults, but the molecular mechanisms underlying susceptibility remain incompletely understood. Although genome-wide association studies (GWAS) have identified many migraine risk loci, translating these associations into biological insight remains challenging. Environmental and physiological migraine triggers, including hormones, diet and stress, may act partly through epigenetic mechanisms such as DNA methylation. Because many migraine risk variants are non-coding and may influence disease through regulatory effects, integrating genetic associa
