Selective insolubility of alpha-synuclein in human Lewy body diseases is recapitulated in a transgenic mouse model.

alpha-Synuclein (alpha-SYN) is deposited in intraneuronal cytoplasmic inclusions (Lewy bodies, LBs) characteristic for Parkinson's disease (PD) and LB dementias. alpha-SYN forms LB-like fibrils in vitro, in contrast to its homologue beta-SYN. Here we have investigated the solubility of SYNs in human LB diseases and in transgenic mice expressing human wild-type and PD-associated mutant [A30P]alpha-SYN driven by the brain neuron-specific promoter, Thy1. Distinct alpha-SYN species were detected in the detergent-insoluble fractions from brains of patients with PD, dementia with LBs, and neurodegen

The development of the vermiculate pattern in the brow region of crania from Indian Knoll, Kentucky.

A convoluted brow surface consisting of fine ridges, grooves, and depressions, first identified in ancient fossil hominids and termed the vermiculate pattern (VP), is often low in frequency and of moderate rugosity in crania of anatomically modern human populations. Burials at Indian Knoll, Kentucky, constitute an excellent series for study of the development of the VP, since cranial surfaces are usually well preserved, there are hundreds of immature and mature individuals previously assessed for age, and the VP is high in frequency and rugosity. While a surface pattern resembling a miniature

Mechanisms of homology-facilitated illegitimate recombination for foreign DNA acquisition in transformable Pseudomonas stutzeri.

Sound for the human voice is produced via flow-induced vocal fold vibration. The vocal folds consist of several layers of tissue, each with differing material properties. Normal voice production relies on healthy tissue and vocal folds, and occurs as a result of complex coupling between aerodynamic, structural dynamic, and acoustic physical phenomena. Voice disorders affect up to 7.5 million annually in the United States alone and often result in significant financial, social, and other quality-of-life difficulties. Understanding the physics of voice production has the potential to significant

Physical, chemical, and structural properties of human gastric organoid-derived mucus.

The gastric mucus layer protects the epithelium from gastric acid and ingested pathogens. However, studies of human gastric mucus have been limited due to poor accessibility of native human mucus and the abundance of contaminants in these samples. Here, we explored the potential of human gastric organoids as models for mucus production. Immunofluorescence staining confirmed that the organoids produced mucus containing MUC5AC and MUC6. The luminal mucus had viscoelastic properties similar to those of native human gastric mucus, as determined by particle tracking microrheology. To collect organo

Distinct Host Tropism Protein Signatures to Identify Possible Zoonotic Influenza A Viruses.

Zoonotic influenza A viruses constantly pose a health threat to humans as novel strains occasionally emerge from the avian population to cause human infections. Many past epidemic as well as pandemic strains have originated from avian species. While most viruses are restricted to their primary hosts, zoonotic strains can sometimes arise from mutations or reassortment, leading them to acquire the capability to escape host species barrier and successfully infect a new host. Phylogenetic analyses and genetic markers are useful in tracing the origins of zoonotic infections, but there are still no

Cyclic mechanical stretching and interleukin-1alpha synergistically up-regulate prostacyclin secretion in cultured human uterine myometrial cells.

Prostacyclin (PGI2), a potent uterine smooth muscle relaxant, is postulated to be a major prostaglandin (PG) secreted from the human myometrium. PGI2 metabolite concentrations in the maternal plasma were reported to be elevated during pregnancy, especially during labor. Recently, we developed cultured human myometrial cells from pregnant women and reported that cyclic mechanical stretching mimicking labor increased PGI2 secretion from these cells by up-regulating PGI2 synthase promoter activities. Since elevation of cervical/vaginal interleukin-1alpha (IL-1alpha) concentrations is also a chara

Hydroxyl-Terminated Polyethylene Glycol Evades Human Pre-existing Anti-polyethylene Glycol Antibodies.

Polyethylene glycol (PEG) has been extensively utilized in food, cosmetics, and pharmaceutical fields, especially in the realm of nanomedicines, where it serves as a pivotal excipient to prolong the nanoparticles' circulation half-life. Contrary to its historical perception as being nonimmunogenic, pre-existing anti-PEG antibodies have been widely detected in human even in individuals without prior exposure to PEGylated therapeutics and are associated with clinically relevant adverse effects of PEGylated nanomedicines including infusion reactions and hypersensitive reactions. Herein, we elucid

Single-cell transcriptome delineating asymmetric dynamics of CD4+ T and CD8+ T cell lineage commitment in human prenatal thymus.

INTRODUCTION: The thymus provides a specialized microenvironment for T cell development and selection, yet the cellular heterogeneity and molecular dynamics that govern human prenatal thymopoiesis remain incompletely characterized. METHODS: We constructed an integrative single-cell atlas of human prenatal thymocytes from 7 to 23 post-conception weeks by combining five published datasets. Selection intermediates were classified based on coreceptor expression patterns. Pseudotime analysis, regulon profiling, metabolic analysis, and cell-cell communication modeling were applied to characterize d

Translational Pharmacokinetic-Pharmacodynamic Modeling and Efficacious Human Dose Prediction of DNDI-6148 for the Treatment of Cutaneous Leishmaniasis.

Cutaneous leishmaniasis is a neglected tropical disease with only one oral treatment option. DNDI-6148 is an orally bioavailable compound with potent antiparasitic activity in preclinical studies. Establishing skin target-site pharmacokinetic/pharmacodynamic (PK/PD) relationships is essential to enable its clinical development. The objective was to characterize the target-site PK and PD of DNDI-6148 in a murine L. major model and to predict a human efficacious dose. A nonlinear mixed-effects PK/PD model was developed using data from L. major-infected BALB/c mice orally administered DNDI-6148 (

Distinct cytotoxicity of water disinfection byproducts in human uroepithelium and Chinese hamster ovary cells.

Water disinfection unintentionally produces hundreds of disinfection byproducts (DBPs). Although the cytotoxicity of over 100 DBPs has been extensively studied in Chinese hamster ovary (CHO) cells, toxicity data in human cell models remain limited. Given the epidemiological association of DBPs with bladder cancer risk, it is necessary to conduct studies using cell lines that exhibit biological characteristics resembling human bladder cells, e.g., the immortalized human uroepithelium SV-HUC-1 cells. In this study, the cytotoxicity of 14 regulated and unregulated DBPs in SV-HUC-1 and CHO-K1 cell

Epithelial and stromal remodeling in murine Crohn's disease-like ileitis.

BACKGROUND AND AIMS: Inflammatory bowel disease is characterized by progressive epithelial, immune, and stromal dysfunction. Human datasets yield important insights yet are inherently descriptive, requiring animal models for in vivo mechanistic interrogation. The SAMP1/YitFc (SAMP) mouse model develops spontaneous Crohn's disease (CD)-like ileitis while allowing controlled manipulation of pathophysiologic pathways. To understand cell-specific disease-associated transcriptional changes, we defined the single-nucleus transcriptomes of inflamed SAMP ilea using single-nucleus RNA-sequencing (snRNA

Multifaceted Role of Specialized Neuropeptide-Intensive Neurons on the Selective Vulnerability to Alzheimer's Disease in the Human Brain.

Regarding Alzheimer's disease (AD), specific neuronal populations and brain regions exhibit selective vulnerability. Understanding the basis of this selective neuronal and regional vulnerability is essential to elucidate the molecular mechanisms underlying AD pathology. However, progress in this area is currently hindered by the incomplete understanding of the intricate functional and spatial diversity of neuronal subtypes in the human brain. Previous studies have demonstrated that neuronal subpopulations with high neuropeptide (NP) co-expression are disproportionately absent in the entorhinal

PFASs in Soil: How They Threaten Human Health through Multiple Pathways and Whether They Are Receiving Adequate Concern.

Per- and polyfluoroalkyl substances (PFASs) have been mass-produced and widely applied in consumer and industrial products, resulting in their widespread presence in the environment. Features such as environmental persistence, bioaccumulation, and high toxicity even at low doses have made PFASs an increasing concern. This brief review focuses on soil PFASs, especially the effect of soil PFASs on other environmental media and their potential threats to human health through daily diet. Specifically, soil PFASs contamination caused by different pathways was first investigated. Soil pollution from

Up-Regulation of microRNA-424 Causes an Imbalance in AKT Phosphorylation and Impairs Enteric Neural Crest Cell Migration in Hirschsprung Disease.

Insights into the role of microRNAs (miRNAs) in disease pathogenesis have made them attractive therapeutic targets, and numerous miRNAs have been functionally linked to Hirschsprung disease (HSCR), a life-threatening genetic disorder due to defective migration, proliferation, and colonization of enteric neural crest cells (ENCCs) in the gut. Recent studies have demonstrated that miR-424 strongly inhibits migration in a variety of cell types and its potential target RICTOR is essential for neural crest cell development. We therefore sought to interrogate how miR-424 and RICTOR contribute to the
