Comparison of interleukin-6 and serum creatinine levels in preeclampsia and normal pregnancy patients: A cross-sectional study in Indonesia.

Preeclampsia (PE) is a complication of pregnancy with high morbidity and mortality, characterized by endothelial dysfunction, systemic inflammation, and impaired renal function. Interleukin-6 (IL-6) and serum creatinine have potential as biomarkers for early detection of PE; however, data from Indonesia are limited. The aim of this study was to compare IL-6 and serum creatinine levels between pregnant women with PE and normotensive controls, as well as their diagnostic performance in identifying PE. A cross-sectional study involving pregnant women with PE and normotensive controls was conducte

Determination of urinary and serum myeloperoxidase and interleukin-6, -8 -10 in dogs with subclinical bacteriuria and urinary tract infections.

Subclinical bacteriuria (SBU) is defined as the presence of significant bacteriuria in dogs, in the absence of clinical signs attributable to lower urinary tract infection (UTI). The local immunological factors potentially associated with the asymptomatic phenotype displayed in dogs with SBU, have not been explored. This study aimed to investigate the local immune response in dogs with SBU through cytokine and myeloperoxidase (MPO) expression in urine. Furthermore, we aimed to investigate the discriminatory potential of urinary interleukin 6 (uIL-6), uIL-8, uIL-10 and uMPO between dogs with SB

Hyperacute Cytokine Kinetics and Early Interleukin-10 Elevation as Predictors of Neurological Outcome in Post-Cardiac Arrest Syndrome.

Background: Post-cardiac arrest syndrome (PCAS) involves a complex interplay between systemic inflammatory and compensatory anti-inflammatory responses. The hyperacute kinetics of these cytokines and their incremental prognostic value beyond established brain injury biomarkers remain inadequately characterized. We aimed to evaluate whether the 6 h post-return-of-spontaneous-circulation (ROSC) cytokine profile predicts neurological outcome, with the Th1/Th2 ratio largely reflecting the IL-10 signal. Methods: This retrospective analysis of prospectively collected data from a single-center cardia

Negative Regulator of Interferon Response Promotes Toll-like Receptor 3-induced Interleukin 6 in Rheumatoid Synoviocyte.

BACKGROUND/AIM: Rheumatoid arthritis (RA) is an autoimmune disease, with synovial inflammation as an important symptom. However, the pathogenesis of RA has not been fully elucidated. Long noncoding RNAs play a role in various biological and pathological situations, and negative regulators of interferon response (NRIR) are long noncoding RNAs that regulate immune reactions. However, the role of NRIR in rheumatoid synovial inflammation remains unclear. MATERIALS AND METHODS: Cultured human rheumatoid fibroblast-like synoviocytes (RFLS) were treated with a synthetic Toll-like receptor 3 (TLR3) l

Interleukin-1β downregulates K+ currents through Kv1.6 channels and promotes apoptosis by enhancing Ca2+ influx in chondrocytes.

Chronic inflammation of the knee joint can induce chondrocyte death, leading to osteoarthritis (OA). Ca2+ influx through Ca2+ channels expressed in the chondrocyte plays a key role in promoting the inflammation and chondrocyte death. Voltage-gated K+ (Kv) channels can modulate this Ca2+ influx by regulating the resting membrane potential (RMP) in chondrocytes. Although it has been reported that an increase in intracellular Ca2+ concentration ([Ca2+]i) is associated with OA progression, the molecular mechanism(s) remain unclear. Therefore, the main goal of this study was to identify the mechani

Genetically modified Scutellaria Radix alleviates rheumatoid arthritis and is associated with suppression of interleukin 1 beta-related signaling.

Scutellaria Radix (SR), the dried root of Scutellaria baicalensis Georgi, has been traditionally used as an antipyretic and analgesic agent. Genetically modified SR (GS), engineered to enhance the active constituents baicalin (BA), baicalein (BE), and wogonin (WG), was hypothesized to exhibit enhanced efficacy at equal or lower doses. Although the pharmacological properties of SR have been reported previously, its therapeutic potential in rheumatoid arthritis (RA) remains unclear. This study aimed to evaluate the therapeutic potential of GS compared with SR in a mouse model of RA. The efficacy

Intraclass switching of interleukin-23 inhibitors in psoriasis: effectiveness, patterns and predictors of response - a retrospective multicentre study in Italy.

BACKGROUND: Interleukin (IL)-23 inhibitors are highly effective therapies for psoriasis, but some patients may need to discontinue the treatment. Intraclass switching is a potential strategy, although data on its effectiveness remain limited. OBJECTIVES: To evaluate the patterns and effectiveness of intraclass switching among IL-23 inhibitors in a real-world setting. METHODS: This retrospective, multicentre study included adult patients with plaque psoriasis who switched between IL-23 inhibitors. Clinical and demographic data were analysed, and univariable and multivariable analyses identifi

Interleukin-12 induces rapid STAT4/DDX5-dependent remodeling of RNA polymerase II occupancy in NK cells.

Natural killer (NK) cells are innate lymphocytes that rapidly respond to inflammatory cytokines during infection, but the mechanisms underlying such swift responses remain incompletely understood. Here, we investigated the RNA polymerase II (Pol II) dynamics during rapid cytokine-induced activation in NK cells. Brief exposure to the proinflammatory cytokine interleukin-12 (IL-12) elicited rapid, genome-wide redistribution of Pol II within minutes, with increased promoter-proximal Pol II pausing at effector loci including Ifng. This IL-12-induced Pol II reorganization was mediated by the transc

Intra-articular interleukin-1β induces early synovitis in an ex vivo autologously perfused equine forelimb model.

OBJECTIVE: To evaluate an ex vivo model of synovitis induced by IA lipopolysaccharide (LPS) and IL-1β in isolated, perfused equine forelimbs. METHODS: Forelimbs and blood were collected from 12 clinically unremarkable horses following slaughter at a licensed abattoir. In 8 limbs, 2 of 3 phalangeal joints were injected with LPS (0.5 to 5 ng, 4 limbs) or IL-1β (100 ng, 4 limbs) diluted in 1 mL of distilled water. All limbs were perfused with autologous perfusate for 8 hours. Synovial fluid cytology and synovium sample histology scores for synovitis (0 = no changes and 3 = marked changes) were a

β-Glucan alleviates colitis by remodeling the colonic microenvironment and promoting interleukin-33-driven active eosinophil differentiation.

Eosinophils play an essential role in intestinal homeostasis, yet the mechanisms governing their functions in intestine remain poorly defined. β-Glucan, an immunomodulator, has been shown to alleviate colitis, but whether it acts through eosinophils and the underlying mechanisms remains unclear. Here we show that β-Glucan pretreatment significantly attenuated dextran sulfate sodium (DSS)‑induced colitis in wild-type mice but not in eosinophil-deficient mice, indicating an eosinophil-dependent protective effect. β‑Glucan increased the frequency and absolute number of colonic active eosinophils

Massive idiopathic pericardial effusion in a pediatric patient responsive to interleukin-1 receptor antagonist: Unveiling an undercover hyperinflammation?

We report a child with massive, chronic idiopathic pericardial effusion (PE), initially detected incidentally and asymptomatically. Despite multiple treatment attempts, including Nonsteroidal anti-inflammatory drugs, corticosteroids, colchicine, and repeated pericardiocentesis, the effusion persisted and recurred. Advanced imaging revealed a stable but severe PE over the years, with no hemodynamic compromise. Cytokine analysis of the pericardial fluid showed markedly elevated interleukin (IL)-6 levels, suggesting an underlying inflammatory process despite the absence of classic clinical signs.

Independent prognostic value of semaphorin-4D, interleukin-1β and complement activation in newly diagnosed multiple myeloma patients.

Multiple myeloma represents a systemic disease of the bone marrow (BM) niche, in which immune and skeletal pathways are tightly interconnected. However, the independent prognostic significance of bone and immune-related markers in newly diagnosed multiple myeloma (NDMM) remains incompletely understood. Semaphorin (Sema) 4D, activin-A, and periostin ELISA, LEGENDplex™ Human Bone Metabolism Panel and proteomic analysis for novel biomarker identification were conducted in 71 consecutive samples from NDMM patients. In 25 patients, genomic analysis was performed on sorted clonal plasma cells. NDMM

The Interleukin-17-T helper 17 axis in primary sclerosing cholangitis: A narrative review of an emerging pathogenic frontier.

Interleukin-17 (IL-17) and IL-17+ secretory cells, including T helper 17 (Th17) cells play pivotal roles in autoimmune diseases such as psoriasis. Emerging evidence implicates the IL-17 pathway in primary sclerosing cholangitis (PSC), a chronic cholestatic liver disease lacking approved pharmacological therapies, for which liver transplantation remains the only life-extending option. The IL-17 pathway and Th17 biology is increasingly implicated in PSC pathogenesis through its interactions with hepatic parenchymal and non-parenchymal cells, including cholangiocytes, neutrophils and hepatic stel

Interleukin-1α Mediates Pancreatic Fibroblast Activation, Regulates Immune Cell Recruitment and Fibrosis in Acute and Chronic Pancreatitis.

Pancreatitis is a life-threatening inflammatory disease of the pancreas. The cytokine interleukin-1α has been demonstrated to act as an alarmin released by necrotic cells. In the present study, we investigated the influence of IL-1α on the immune response during acute and chronic pancreatitis. Following tissue injury, pancreatic acinar cells released IL-1α, which activates tissue-resident fibroblasts to differentiate toward a pro-inflammatory phenotype. By secreting chemokines and cytokines such as CXCL5, CCL2, and IL-6, these fibroblasts recruit immune cells to the pancreas. The absence of IL

Interleukin-38 attenuates Obesity-induced cardiomyopathy through AMPK/autophagy-mediated suppression of endoplasmic reticulum stress.

Obesity-induced cardiomyopathy involves cardiac hypertrophy, apoptosis, and fibrosis, contributing to cardiac dysfunction. Autophagy activation alleviates endoplasmic reticulum stress (ERS). Interleukin (IL)-38, an IL-1 family cytokine with anti-inflammatory and metabolic regulatory properties, protects against cardiovascular diseases. This study investigated whether IL-38 attenuates obesity-induced cardiomyopathy by activating the AMP-activated protein kinase (AMPK)/autophagy pathway to suppress ERS. Mice fed a high-fat diet (HFD) for 20 weeks received IL-38 for the final 8 weeks, with or wit

Molecular characterization of the type 2 diabetes-associated interleukin-6 receptor single nucleotide polymorphism p.V385I (rs2228146).

The cytokine Interleukin-6 (IL-6) activates its target cells through the membrane-bound IL-6 receptor (IL-6R). IL-6 crucially controls important metabolic functions of the human body. Low-grade inflammation, including increased IL-6 serum levels, contribute to the development of autoimmune diseases like type 2 diabetes (T2D). In this study, we have analyzed the single nucleotide polymorphism of the IL-6R rs2228146 that is associated with T2D and results in the exchange of a single amino-acid residue within the IL-6R protein (p.V385I). We show that the affected amino-acid residue is located wit
