Antimicrobial activity of rifabutin in combination with two and three other antimicrobial agents against strains of Mycobacterium paratuberculosis.

The inhibitory and bactericidal, synergistic, and antagonistic activities of rifabutin combined with ciprofloxacin, ethambutol, clofazimine, cefazolin, and amikacin in dual and triple combinations against various human and animal isolates of Mycobacterium paratuberculosis were determined. Synergism was observed when rifabutin was combined with either cefazolin or clofazimine in double combinations. The greatest amount of synergy occurred with the rifabutin-cefazolin combination in which bactericidal synergism was present with all strains. Of the triple combinations examined, only rifabutin in …

Extracellular and intracellular activities of clarithromycin used alone and in association with ethambutol and rifampin against Mycobacterium avium complex.

Mycobacterium avium complex bacteria are opportunistic human pathogens, and their chemotherapy remains a challenge since these organisms are resistant to a majority of routine antituberculous drugs. Recently, a wide range of new macrolide antibiotics has been developed, among which the drug clarithromycin appears to have a selective action against M. avium bacteria. In the present study, we have investigated the action of clarithromycin alone (MIC and MBC determinations) and in association with the routine antimycobacterial drugs ethambutol and rifampin at sublethal concentrations (1 microgram…

The role of macrophage activation and of Bcg-encoded macrophage function(s) in the control of Mycobacterium avium infection in mice.

Following the intraperitoneal inoculation of 2.5 x 10(8) colony-forming units of Mycobacterium avium strain ATCC 25291, there was bacillary growth in the liver, spleen and peritoneal cavity of C57BL/6, C57BL/10, DBA/1 and BALB/c mice whereas DBA/2, C3H/He, CBA/Ca and CD-1 mice controlled the infection showing constant or slightly decreasing numbers of viable bacteria in the liver and spleen and effective clearance of the bacilli from the peritoneal cavities. The acquisition of non-specific resistance (NSR) to Listeria monocytogenes during the infection by M. avium was high in C57BL/6, BALB/c a…

Effect of rifabutin on the phagocytosis and intracellular growth of Mycobacterium intracellulare in mouse resident and activated peritoneal and alveolar macrophages.

In order to understand the discrepant results between in vitro and in vivo activity of rifabutin (LM 427) against Mycobacterium intracellulare, we studied its effects upon the phagocytosis of M. intracellulare by resident and activated mouse peritoneal and alveolar macrophages and upon growth of M. intracellulare inside these cells. Rifabutin (ansamycin, LM 427) had no effect on the viability of resident or activated peritoneal or alveolar macrophages or on the phagocytosis of M. intracellulare by resident peritoneal macrophages. In activated peritoneal and alveolar macrophages, it had a dose-…

Experimental disease in infant goats induced by a Mycobacterium isolated from a patient with Crohn's disease. A preliminary report.

Pilot studies were done to assess the pathogenicity of a Mycobacterium which had been recovered from the diseased ileum of a patient with Crohn's disease. In four separate studies, pairs of infant goats served as subjects. One of each pair received an oral inoculum of freshly harvested Mycobacterium species strain Linda suspended in cream. A littermate or stablemate which received only cream served as control. Necropsies were done at three, five, six, and 10 months postinoculation. Each of the four inoculated animals developed segmental granulomatous disease of the ileum or ileum and more prox…

A serological test for leprosy based on competitive inhibition of monoclonal antibody binding to the MY2a determinant of Mycobacterium leprae.

A novel serological assay for leprosy has been devised on the basis of serum inhibition of binding of 125I-labelled ML04 monoclonal antibody to Mycobacterium leprae sonicate-coated microtitre plates. Antibodies were detected in 93% of lepromatous leprosy patients, whereas controls from the endemic area, including leprosy contacts and patients with tuberculosis, were serologically negative. The specificity and efficacy of the test may offer an advantage over previously used techniques.

Serological activity of a characteristic phenolic glycolipid from Mycobacterium leprae in sera from patients with leprosy and tuberculosis.

Serological activity against a purified phenolic glycolipid from Mycobacterium leprae, which may be obtained in large amounts from M. leprae infected armadillo liver, was investigated using immunodiffusion and an enzyme linked immunosorbent assay (ELISA). Generally a good correlation was obtained between these techniques, but the ELISA was more sensitive and convenient. Relatively high IgG and IgM anti-glycolipid antibody levels were found in lepromatous leprosy patients. The antibody titres to the glycolipid were, however, low when compared with antibody titres to crude sonicates. Since the g…

Multiple skin testing of tuberculosis patients with a range of new tuberculins, and a comparison with leprosy and Mycobacterium ulcerans infection.

Four hundred and seventy tuberculosis patients were each skin tested with four of a range of 17 mycobacterial reagents in four countries in … a. It was also noted, in Burma particularly, that sensitization to mycobacterial species other than Mycobacterium tuberculosis, especially to the slow growers, plays a role in determining responses to different mycobacterial species. In tuberculosis patients enhanced skin test responses were also seen but only in those countries, e.g. Libya, where the prevalence …

Use of cetylpyridinium chloride and sodium chloride for the decontamination of sputum specimens that are transported to the laboratory for the isolation of Mycobacterium tuberculosis.

A method is presented for the decontamination, liquefaction, and concentration of sputum specimens that are in transport more than 24 h. The method is inexpensive, and culture results compare well with those obtained with the accepted N-acetyl-L-cysteine and sodium hydroxide method for the isolation of tubercle bacilli. The working solution, 1% cetylpyridinium chloride and 2% sodium chloride, is mixed in equal volumes with sputum before the specimens are shipped. Tubercle bacilli remained viable after 8 days of exposure to this solution. Only Lowenstein-Jensen medium was used because the cetyl

Effect of pretreatment with Mycobacterium bovis (strain BCG) and immune syphilitic serum on rabbit resistance to Treponema pallidum.

Stimulation of the rabbit reticuloendothelial system with viable Mycobacterium bovis (strain BCG), and other agents, had no effect on the development of syphilitic lesions after intradermal or intravenous inoculation with graded doses of Treponema pallidum (virulent Nichol's strain; mean infective doses less than 10). The simultaneous administration of immune syphilitic rabbit serum retarded the development of lesions, but this appeared to be due solely to the immune serum, suggesting no synergism between the activated reticuloendothelial system and the anti-T. pallidum antibodies. The adminis…

Characterization of the cellular immune defect in lepromatous leprosy: a specific lack of circulating Mycobacterium leprae-reactive lymphocytes.

The blastogenic response of leucocyte cultures from patients with tuberculoid and lepromatous leprosy has been studied. The leucocytes from the two groups were studied simultaneously and cultivated in the same pool of normal human serum. While the leucocytes from twenty-eight tuberculoid patients responded quite strongly to Mycobacterium leprae after 7 days of culture (average lymphocyte transformation 11·1%), there was a complete lack of response in similar cultures from twenty-seven lepromatous patients (average 0·1% transformed cells). These results were confirmed by studies on cellular inc…

PstS1-loaded exosomes promote Mycobacterium tuberculosis infection via miR-122-mediated PI3K/AKT/mTOR activation and autophagy suppression.

Tuberculosis (TB), caused by Mycobacterium tuberculosis (MTB), is a chronic respiratory infectious disease. With the continuous increase in new cases, TB has become the second leading infectious disease worldwide, posing a significant challenge to public health. Previous studies have shown that exosomes play a crucial role in MTB infection, and microRNA-122 (miR-122) is highly expressed in MTB-infected cells. Exosomes secreted under the induction of phosphate-specific transport system substrate-binding protein 1 (PstS1) were characterized using transmission electron microscopy (TEM) and Wester…

Structure-based and machine learning-assisted identification of natural phytochemicals targeting Mycobacterium leprae InhA for anti-mycobacterial therapeutics.

Mycobacterium leprae (M. leprae) is the bacterium that causes leprosy. It is a public health problem in many regions, especially in developing countries. The situation is getting worse and worse as drug resistance spreads. InhA is one of the most important proteins for M. leprae's survival. It helps make mycolic acid, an important part of the bacterial cell envelope; hence, InhA is a good target for developing new anti-leprosy drugs. In this study, we focused on identifying natural plant-derived compounds (phytochemicals) capable of inhibiting InhA function using sophisticated computer-based t…

Linking lipid profile alterations to antibiotic tolerance and natural product synergy in drug-resistant Mycobacterium tuberculosis clinical isolates.

Despite global control efforts, tuberculosis remains the leading infectious cause of death, with rising incidence, pediatric cases, and drug-resistant strains posing major public health challenges. Mycobacteria, including Mycobacterium tuberculosis, possess a lipid-rich, dual-membrane cell envelope that contributes to their impermeability, drug resistance, and unique pathogenic mechanisms. Some lipids play key roles in modulating host immune responses, enabling survival within macrophages, and promoting granuloma formation. Since it is known that lipid remodeling of the cell envelope is correl…

Molecular Identification of Mycobacterium bovis in Human Pulmonary Tuberculosis: Insights from a Tertiary Care Hospital in Gujarat, India.

BACKGROUND: Mycobacterium bovis, the causative agent of bovine tuberculosis, is a zoonotic pathogen capable of infecting cattle and humans. Human contraction of bovine tuberculosis, particularly pulmonary infection, remains a significant public health concern. The differentiation between Mycobacterium bovis and Mycobacterium tuberculosis is challenging due to limitations in conventional diagnostic methods, leading to an underestimated burden of M. bovis in human population. This study focuses on the prevalence of M. bovis in cases of pulmonary tuberculosis in a tertiary care teaching hospital …

Can Mycobacterium avium subsp. paratuberculosis Survive During Pecorino Cheesemaking Process? An Experimental Evaluation Through a Microbiological Challenge Test.

Paratuberculosis is an infectious disease mainly affecting domestic ruminants, caused by Mycobacterium avium subsp. paratuberculosis (MAP). Affected animals shed the pathogen through feces, also during the subclinical stages of the disease. Although there is still no conclusive evidence of MAP involvement in human diseases, particularly Crohn's disease and autoimmune disorders, exposure to MAP through the consumption of contaminated dairy products has become a public health concern. Therefore, in this study, the survival of MAP during the production of Pecorino, an Italian cheese made from raw…

Bovine tuberculosis resulting from infection with Mycobacterium orygis in a closed herd of Indian water buffalo (Bubalus bubalis).

Mycobacterium orygis (M. orygis), a member of the Mycobacterium tuberculosis complex, has emerged as an important cause of animal and human tuberculosis in South Asia. Despite the widespread prevalence of bovine tuberculosis (bTB) in India and the major role of over 100 million Indian water buffaloes in dairy production, the pathological and epidemiological features of M. orygis infection in buffaloes are largely unexplored. A closed herd of 279 female water buffaloes in Haryana, India, was screened for bTB by single intradermal cervical tuberculin skin (SCT) test, the comparative cervical tub…

IL-36γ enhanced bactericidal effects of macrophages to Mycobacterium tuberculosis via the IFN-γ/HIF-1ɑ/glycolysis pathway.

BACKGROUND: IL-36γ coordinates macrophage activation and is essential for defense against Mycobacterium tuberculosis (Mtb), but the mechanisms remains poorly understood. Aerobic glycolysis plays a critical role in macrophages intrinsic control of Mtb infection. This study aimed to investigate the potential effects of IL-36γ on macrophages energy metabolism transformation from mitochondrial oxidative phosphorylation to aerobic glycolysis in response to Mtb infection. METHODS: The expression of IL-36γ in lung tissues, PBMCs and serum was analyzed using Immunohistochemistry, ELISA and RT-qPCR, w…
