We describe the genetic analysis of an Alzheimer's disease (AD) sample derived from a genetically isolated population. Genetic assessment included the analysis of genes involved in AD, such as the genes for amyloid precursor protein (APP), presenilin 1 (PSEN1) and presenilin 2 (PSEN2). We also assessed genes for some proteins that constitute the gamma-secretase complex: nicastrin (NCSTN), presenilin enhancer-2 (PEN2), in addition to the AD risk factor apolipoprotein E (APOE). Using polymerase chain reaction and single strand conformational polymorphism method, screens for APP, PSEN1 and PSEN2

In mixed alcohol-water solvents, bovine beta-lactoglobulin undergoes a cooperative transition from beta-sheet to a high alpha-helix content conformer. We report here the characterization of beta-lactoglobulin by compressibility and spectroscopy measurements during this transconformation. Both the volume and compressibility increase as a function of alcohol concentration, up to maximal values which depend on the chemical nature of the three alcohols used: hexafluoroisopropanol, trifluoroethanol, and isopropanol. The order of effectiveness of alcohols in inducing the compressibility transition i

PrP106-126, a peptide corresponding to residues 107-127 of the human prion protein, induces neuronal cell death by apoptosis and causes proliferation and hypertrophy of glia, reproducing the main neuropathological features of prion-related transmissible spongiform encephalopathies, such as bovine spongiform encephalopathy and Creutzfeldt-Jakob disease. Although PrP106-126 has been shown to form amyloid-like fibrils in vitro, their structural properties have not been elucidated. Here, we investigate the conformational characteristics of a fibril-forming fragment of the mouse prion protein, MoPr

BACKGROUND/AIMS: We developed a single-chain antibody fragment (scFv) to the non-structural 3 protein (NS3) of hepatitis C virus (HCV) and tested its ability to interfere with the HCV replication cycle in infected hepatocytes.
METHODS: The variable regions of the human monoclonal antibody CM3.B6 that recognizes a conformational epitope within the helicase domain of NS3 were introduced into adenoviral vectors for expression in mammalian hepatocytes. Expression and binding properties of the scFv were analyzed by immunological assays. Effects of intracellular expression of the scFv on HCV replic

Escape mutants of tick-borne encephalitis (TBE) virus were selected using neutralizing monoclonal antibodies (MAbs) that react with three different and previously unrecognized epitopes in the envelope protein E of TBE virus. Two of these variants (V-IC3 and V-IE3) exhibited a significantly reduced reactivity with their selecting MAbs, as determined by ELISA, whereas with one variant (V-IO3), reactivity was completely unchanged. Comparative sequence analyses demonstrated that each of the variants differed from the wild-type virus by a single amino acid substitution located at exposed positions

Prion diseases, also known as transmissible subacute spongiform encephalopathies (TSSE), are rare neurodegenerative disorders of both humans and animals. Their biochemical hallmark is an accumulation in the brain of an abnormal form of the host-encoded prion protein (PrP). This pathological accumulation could result from a protein conformational change under the influence of unknown factors. The normal function of PrP is unknown. The abnormal form is thought to induce neurodegeneration when experimentally or accidentally introduced in recipient hosts. Such a possibility would explain the trans…

Histological lesions in man, presumably indicating kidney damage, may occur during therapy with lithium carbonate. We studied the cellular effects of lithium carbonate applied in vitro to slices of kidney from adult female Sprague-Dawley rats. Alternate slices were immersed in saline solution containing 1.1 mEq/L of lithium carbonate; the other slices were immersed in ordinary saline solution as controls. Compared with control specimens, a two-hour incubation in lithium carbonate solution showed (1) with toluidine blue 0, in the cells of the collecting ducts a fibrous metachromatic network tha

Goat milk, distinguished by its high β-casein content and superior digestibility compared with bovine milk, represents a promising source of bioactive peptides. Dipeptidyl peptidase-IV (DPP-4) inhibitors are vital for managing type 2 diabetes mellitus by enhancing incretin-mediated glycemic control. Naturally derived DPP-4 inhibitory peptides are of interest as food-compatible candidates for functional ingredient development. This study aimed to identify and characterize DPP-4 inhibitory peptides from goat milk proteins through an integrated computational and experimental approach. Eleven majo

BACKGROUND: Although carcinoma of the tongue usually occurs in patients older than 60 years, up to 4% of these tumors may occur in patients younger than 40 years. Many of the younger patients with this tumor have had no exposure or brief exposure to tobacco smoke or alcohol consumption, to which oral carcinoma is usually attributed. The molecular mechanism responsible for carcinogenesis in this group of patients is not known.
OBJECTIVE: To assess the role of p53 gene mutation in oral carcinogenesis in a group of patients younger than 40 years with squamous carcinoma of the tongue.

alpha 2-Macroglobulin, a 720 kDa tetramer, and Pregnancy Zone Protein, a 360 kDa dimer, are strongly homologous glycoprotein proteinase inhibitors of human plasma. alpha 2M and PZP monomers contain two regions of decisive functional importance i.e. the bait- and the thiol ester regions. The bait region serves as a substrate to a variety of proteinases, and cleavage in this domain causes a conformational change of the molecule. In addition, PZP forms tetramers when reacted with proteinase. As a result of the conformational change the thiol ester, which is hidden in the native structure, becomes

Water-soluble B-complex vitamins have been proven to undergo significant interactions with proteins both in vivo and in vitro. Understanding the impact of major vitamin B-interacting residues (VBIRs), like Gly, His, Asn, Ser, Thr, Trp and Tyr, is relevant to designing experiments related to the beneficial effects of protein-vitamin interactions. The maintenance of native folds and the monomeric form is necessary for the functioning of therapeutic proteins like insulin. Some fatal cases of amyloid generation before (during storage and in the delivery pump) and/or after insulin application (at t

The strong, polar covalent nature of C-F bonds contributes to the permanent nature of per- and polyfluoroalkyl substances (PFAS). PFAS are toxic to humans. Here, we have examined the ability of the small, globular milk protein β-lactoglobulin to bind PFAS. This protein transports hydrophobic and amphiphilic compounds, including retinol and fatty acids, for vision and brain development; therefore, understanding its interactions with PFAS is significant. The crystal structures of β-lactoglobulin complexed with PFOA (perfluorooctanoic acid) at 2.0 Å, PFOS (perfluorooctanesulfonic acid) at 2.5 Å,

Noonan syndrome (NS) is the most common RASopathy, a developmental disorder that derives from dysregulation of the mitogen-activated protein kinase (MAPK) pathway. NS results from modestly activating mutations in proteins throughout the pathway. Trametinib, a MEK inhibitor, has shown promising results for certain NS complications, but NS-specific therapeutic options are lacking. CRAF activity, which is governed by the adaptor protein 14-3-3, represents a key NS regulatory node that has not been exploited. When phosphorylated (p) at CRAF S259, the 14-3-3/CRAF-pS259 complex adopts an inactive co

The performance of soybean protein isolate-dextran (SPI-Dex) Maillard conjugates as O/W emulsion stabilizers is fundamentally limited by their structure and modification degree. However, cold plasma (CP) technology offers a compelling approach to overcome these limitations. Therefore, this study investigates the impact of CP treatment time on the structural and emulsifying properties of SPI-Dex conjugates to develop a high-performance, protein-based emulsion delivery system. Results indicated that compared with the control group (0 min), a CP treatment for 5 min (SPI-Dex5) reduced the disulfid

Apolipoprotein E (apoE), a major protein for lipid transport in circulation and the brain, has three common isoforms, apoE2, apoE3 and apoE4. APOE4 is the strongest genetic risk factor for late-onset Alzheimer's disease (AD). Recently identified rare apoE variants, the apoE3(R136S)-Christchurch, apoE3(V236E)-Jacksonville and apoE4(R251G), appear to exert protective functions against AD and reduce the disease risk, but the molecular basis behind these effects is unknown. ApoE is a structurally dynamic protein, undergoing significant rearrangements that are important for its biological function.

Dynamic attenuated total reflectance-Fourier transform infrared (ATR-FTIR) spectroscopy at both solutions and coatings of a semicrystalline silk material derived from Bombyx mori was applied to monitor the β-sheet conformation, which is known to correlate with silk protein crystallinity. The secondary structure-sensitive Amide I band was analyzed. Two silk protein samples were studied: native-based silk buffer fibroin (NSF) was extracted from silk glands and regenerated silk fibroin (RSF) was extracted from degummed cocoons. Solutions of both NSF and RSF at 2 mg/mL featured low initial β-sheet

The spontaneous aggregation of infectious or misfolded forms of prion protein is known to be responsible for neurotoxicity in brain cells, which ultimately leads to the progression of prion disorders. Bovine spongiform encephalopathy (BSE) in animals and Creutzfeldt-Jakob disease (CJD) in humans are glaring examples in this regard. Square-planar complexes with labile ligands and indole-based compounds are found to be efficiently inhibitory against protein aggregation. Herein, we report the synthesis of an indole-based cyclometallated palladium complex. The ligand and complex were characterized

Invasion of human red blood cells (RBCs) by Plasmodium falciparum (Pf) merozoites relies on the interaction between two parasite proteins, apical membrane antigen 1 (AMA1) and rhoptry neck protein 2 (RON2) 1,2 . Antibodies to AMA1 confer limited protection against P. falciparum in non-human primate malaria models 3,4 . However, clinical trials with recombinant AMA1 alone (apoAMA1) saw no protection, likely due to inadequate levels of functional antibodies 5-8 . Notably, immunization with AMA1 in its ligand bound conformation using RON2L, a 49 amino acid peptide from RON2, confers superior prot
