A Self-Assembling Cross-Protective Antigen Against Multiple Gram-Positive Nosocomial Pathogens.

ESKAPE pathogens are responsible for complicated nosocomial infections worldwide and are usually resistant to commonly used antibiotics in clinical settings. Among these bacteria, vancomycin-resistant Enterococcus faecium and methicillin-resistant Staphylococcus aureus are the two most important Gram-positive pathogens for which alternative treatments and preventions are urgently needed. We previously designed a multipresenting antigen, embedding the main epitope displayed by the AdcA protein of E. faecium, that conferred protection against different Gram-positive pathogens both in passive and

Genome Sequencing of the Antibiotic-Resistant Leucobacter sp. HNU-1 and Its Developmental Toxicity in Caenorhabditis elegans.

To date, Leucobacter species have been identified from diverse sources with various ecological and functional roles. However, the genomic features and pathogenic potential of antibiotic-resistant Leucobacter strains remain understudied. Here, we isolated the Leucobacter sp. HNU-1 from tropical Hainan Province, China, and found it can induce diapause in Caenorhabditis elegans following ingestion, while exhibiting no significant effects on the nematode's lifespan, survival rate, locomotion, and intestinal epithelial cells. This bacterium demonstrates resistance to multiple antibiotics, including

From Isolation to Application: Utilising Phage-Antibiotic Synergy in Murine Bacteremia Model to Combat Multidrug-Resistant Enterococcus faecalis.

Enterococcus species, natural inhabitants of the human gut, have become major causes of life-threatening bloodstream infections (BSIs) and the third most frequent cause of hospital-acquired bacteremia. The rise of high-level gentamicin resistance (HLGR) in enterococcal isolates complicates treatment and revives bacteriophage therapy. This study isolated and identified forty E. faecalis clinical isolates, with 30% exhibiting HLGR. The HLGR5 isolate, resistant to fosfomycin, vancomycin, and linezolid, was used to isolate the vB_EfaS_SZ1 phage from effluent water. This phage specifically lysed 42

Bosutinib Stimulates Macrophage Survival, Phagocytosis, and Intracellular Killing of Bacteria.

Host-acting compounds are emerging as potential alternatives to combating antibiotic resistance. Here, we show that bosutinib, an FDA-approved chemotherapeutic for treating chronic myelogenous leukemia, does not possess any antibiotic activity but enhances macrophage responses to bacterial infection. In vitro, bosutinib stimulates murine and human macrophages to kill bacteria more effectively. In a murine wound infection with vancomycin-resistant Enterococcus faecalis, a single intraperitoneal bosutinib injection or multiple topical applications on the wound reduce the bacterial load by approx

Elucidating the potential of isorhapontigenin in targeting the MgrA regulatory network: a paradigm shift for attenuating MRSA virulence.

As methicillin-resistant Staphylococcus aureus (MRSA) exhibits formidable resistance to many drugs, the imperative for alternative therapeutic strategies becomes increasingly evident. At the heart of our study is the identification of a novel inhibitor through fluorescence anisotropy assays, specifically targeting the crucial multiple gene regulator A (MgrA) regulatory network in S. aureus. Isorhapontigenin (Iso), a natural compound, exhibits outstanding inhibitory efficacy, modulating bacterial virulence pathways without exerting direct bactericidal activity. This suggests a paradigm shift to

Enterococcus casseliflavus Infection: A Review of Clinical Features and Treatment.

Some Enterococcus species, including Enterococcus faecalis and E. faecium, are increasingly becoming a common cause of nosocomial infections, accounting for the majority of human enterococcal infections, while other species, such as E. casseliflavus, have also been shown to be pathogenic to humans due to the increase in immunocompromised patients. These infections vary widely in their mode of transmission, symptoms, and other characteristics. Treatment is difficult in some cases because enterococci are resistant to numerous antimicrobial agents. Enterococcus faecalis and E. faecium are the bes

Bacteriophage endolysin Ply113 as a potent antibacterial agent against polymicrobial biofilms formed by enterococci and Staphylococcus aureus.

Antibiotic resistance in Enterococcus faecium, Enterococcus faecalis, and Staphylococcus aureus remains a major public health concern worldwide. Furthermore, these microbes frequently co-exist in biofilm-associated infections, largely nullifying antibiotic-based therapy. Therefore, it is imperative to develop an efficient therapeutic strategy for combating infections caused by polymicrobial biofilms. In this study, we investigated the antibacterial and antibiofilm activity of the bacteriophage endolysin Ply113 in vitro. Ply113 exhibited high and rapid lytic activity against E. faecium, E. faec

Prophylactic Antibiofilm Activity of Antibiotic-Loaded Bone Cements against Gram-Negative Bacteria.

Gram-negative bacilli can be responsible for prosthetic joint infection (PJI) even if staphylococci are the main involved pathogens. Gram-negative PJIs (GN-PJI) are considered difficult-to-treat infections due to the increase in antimicrobial resistance and biofilm formation. To minimize the risk of infection in cases of arthroplasties with cemented prosthesis, bone cement can be loaded with antibiotics, especially gentamicin. In this study, we aimed to compare the prophylactic antibiofilm activity of ready-to-use antibiotic-loaded bone cements (ALBC), already commercialized or new prototypes.

Size-dependent photothermal antibacterial activity of Ti3C2Tx MXene nanosheets against methicillin-resistant Staphylococcus aureus.

Developing antibiotics-independent antibacterial materials is of great importance for combating drug-resistant bacteria, such as methicillin-resistant Staphylococcus aureus (MRSA). MXene (transition metal carbides and nitrides), a class of novel 2D nanomaterials, has shown great potentials in biomedical areas. However, the effect of MXene size on its properties and bioactivity is still unknown. Herein, we report for the first time that the antibacterial photothermal therapy efficacy of Ti3C2Tx MXene nanosheets is size-dependent. Three MXene suspensions with small size of 196 nm (MX-s), medium
