Failure to detect Xenotropic murine leukaemia virus-related virus in Chinese patients with chronic fatigue syndrome.

BACKGROUND: Recent controversy has surrounded the question of whether xenotropic murine leukaemia virus-related virus (XMRV) contributes to the pathogenesis of chronic fatigue syndrome (CFS). To investigate the question in a Chinese population, 65 CFS patients and 85 blood donor controls were enrolled and multiplex real-time PCR or reverse transcriptase PCR (RT-PCR) was developed to analyze the XMRV infection status of the study participants. The assay was standardized by constructing plasmid DNAs and armored RNAs as XMRV standards and competitive internal controls (CICs), respectively. RESUL

[Effects of viral infection on transplant recipients].

Viral infection remains an important cause of morbidity and mortality in transplant recipients. The risk of viral infection in these patients depends on several factors, such as the type of organ transplanted, the intensity of immunosuppression, and the recipient's susceptibility. In additional to direct effects, viral infection cause indirect effects, including greater risk of replication of other viruses, graft rejection, opportunistic infections and other specific entities for each type of transplant. These indirect effects result from the immunomodulatory activity of some viruses, such as

Persistence of specific IgM and low avidity specific IgG1 following primary rubella.

Persistence of specific IgM in sera following primary rubella infection was compared with the maturation of the specific IgG1 response. 206 sera, from 171 patients with primary rubella, taken 1 day to 2.5 years after onset of illness, were tested. Rubella-specific IgM was detected by M-antibody capture radioimmunoassay in 100% of sera taken 15-28 days after onset, but in only 9% taken 3-4 months after onset. However, using the diethylamine (DEA) shift value (DSV) method, low avidity specific IgG1 was detected in 91% sera taken at 3-4 months and at 5-7 months 21% of sera remained positive. Usin

Historical perspectives on California encephalitis virus in California.

CEV is primarily associated with Ae. melanimon as its maintenance host and vector in California. A few species of lagomorphs and rodents will develop a transient viremia when infected. Infection of domestic mammals and man is frequent but rarely results in a clinical disease. Extensive field and laboratory studies lead to the conclusion that CEV probably is a mosquito virus that is partially adapted to a narrow range of vertebrate hosts but is not reliant on vertebrates for its continuous existence. JCV has many similarities to CEV in California in that it is dependent on one mosquito, Cu. ino

HPV18 DNA detection and pathological laryngopharyngeal reflux in a young woman with glottic squamous cell carcinoma in situ: a brief report.

Laryngeal cancer predominantly affects elderly males with long-term smoking and alcohol histories, and its occurrence in young females is extremely rare. We report a 26-year-old woman with glottic squamous cell carcinoma in situ who presented with persistent hoarseness. She had no history of smoking, alcohol use, or radiation exposure. Upon evaluation, high-risk HPV18 DNA was detected in FFPE tumor tissue by Lcn PCR, and 24-hour MII-pH monitoring indicated pathological laryngopharyngeal reflux. Targeted sequencing of 535 genes showed no driver mutations, tumor mutation burden elevation, or mic

Suspected fibrosing cholestatic hepatitis in an immunocompetent child with hepatitis C: Case report.

A 12-year-old boy presented to our center with elevated aminotransferases, jaundice, right upper quadrant tenderness, and hepatomegaly. Evaluation revealed a highly elevated blood hepatitis C virus (HCV) ribonucleic acid (RNA) level, and liver biopsy showed canalicular and intrahepatic cholestasis, feathery degeneration of hepatocytes, and periportal/perisinusoidal fibrosis. Though there were atypical biopsy features, including absence of bile ductular reaction, we diagnosed suspected fibrosing cholestatic hepatitis (FCH). FCH secondary to HCV infection has been reported exclusively in immunos

The selected few: Mechanisms of HIV-1 reservoir persistence.

Combination antiretroviral therapy durably suppresses HIV-1 replication yet leaves untouched a small, highly specialized population of infected cells that sustains lifelong viral persistence and remains the principal obstacle to cure. In this review, we synthesize emerging insights into viral and host mechanisms that maintain this extremely durable cellular reservoir, with emphasis on proviral transcriptional regulation, interactions between immune effectors and infected cells, cell-intrinsic programs governing survival and proliferation, and microenvironmental cues that confer competitive fit

Pigs with CD163 Mutation Conferred PRRSV Resistance.

Porcine reproductive and respiratory syndrome (PRRS), which is caused by the porcine reproductive and respiratory syndrome virus (PRRSV), results in substantial economic losses for the global pig farming industry. A critical step in the infection process is the binding of PRRSV to the CD163 receptor on the surface of porcine alveolar macrophages. This study successfully generated CD163-/- Landrace pigs using CRISPR/Cas9 gene editing technology. Following an experimental challenge with two distinct Type II PRRSV strains, the edited pigs exhibited complete resistance to infection. Virological an

Successful treatment of vertically acquired Hepatitis C in a child using direct-acting antiviral therapy administered via gastrojejunal tube: A case report.

Hepatitis C virus (HCV) remains a significant global health burden, with vertical transmission being the primary route of infection in children. Direct-acting antivirals (DAAs) have transformed HCV treatment in pediatrics, but limited data exist on their administration in medically complex children who are dependent on enteral tubes. We report the case of a medically complex child with vertically acquired HCV who achieved sustained virologic response at 12 weeks (SVR12) following treatment with crushed sofosbuvir/velpatasvir (SOF/VEL) administered to a patient with a gastrojejunal tube. HIV-specific and resistant CAR T cells promote control of HIV replication in people with HIV.

We conducted a first-in-human evaluation of HIV-specific, HIV-resistant chimeric antigen receptor (CAR) T cells (CARTs) to determine their capacity to modulate viral rebound dynamics following analytic treatment interruption (ATI). Among ten treated individuals who received engineered T cells, one individual did not rebound during the ATI and had a remarkable expansion of their CARTs 43 days after the start of the ATI, suggesting that the CARTs recognized rebounding HIV and prevented its spread. Post-rebound virologic control was observed in 1 participant for 90 weeks and another 5 participant

Case Report: Renal hemangiosarcoma in a free-ranging red fox (Vulpesvulpes).

This report describes the first documented case of primary renal hemangiosarcoma in a male red fox (Vulpes vulpes Linnaeus, 1758) admitted to the Wildlife Rescue Center "Federico II"-Veterinary Hospital of the Local Health Authority (ASL) Napoli 1 Centro in Naples, Italy. The fox, originating from a sparsely urbanized area, presented with progressive debilitation. Diagnostic imaging (ultrasound, radiography, CT) identified a mass in the right kidney. Citology suggested a malignant neoplasm, and histopathology confirmed hemangiosarcoma with multiple metastases to abdominal organs and to the hea

Biomarkers and Risk Factors Associated with Apnea in Hospitalized Infants with Acute Respiratory Infection.

Apnea is a potentially life-threatening complication of severe acute respiratory infection (SARI) in early infancy; however, its determinants remain incompletely understood. Between 2011 and 2013, we conducted a population-based, cross-sectional, multicenter study in Buenos Aires, Argentina. We examined epidemiological, clinical, virological, and immunological factors associated with apnea in a population-based cohort of 2376 infants younger than five months hospitalized due to SARI. Using hierarchical multivariable logistic regression, we identified preterm birth, cesarean section, previous N

A Structural View of Influenza Virus Ribonucleoprotein Complex and Its Functions.

Influenza viruses are a major global health threat because of their recurring seasonal burden and continuing pandemic potential. Central to the viral life cycle is the viral ribonucleoprotein complex (vRNP), the functional unit of the segmented genome, in which each negative-sense RNA segment is encapsidated by oligomeric nucleoprotein (NP) and bound at its termini by the polymerase complex (FluPol). Recent advances in structural biology have revealed high-resolution structures of FluPol in distinct conformations, NP-RNA helical assemblies, and intact vRNP architectures, providing a structural

Refractory and Resistant Herpes Simplex Virus Mucocutaneous Infections in Immunocompromised Patients: Literature Review and Proposed Definitions for Use in Clinical Trials.

Herpes simplex virus (HSV) infection is one of the most prevalent viral infections worldwide. In general, host immunity is sufficient to clear viral shedding and recurrences, although it is insufficient to prevent subsequent virologic reactivations. In immunocompromised patients, prolonged and difficult-to-treat HSV infections may develop. The diagnosis of refractory HSV infection is based on the lack of clinical response to nucleoside analogs. Antiviral resistance is confirmed via genotypic and/or phenotypic testing. To provide consensus definitions of refractory and/or resistant (R/R) HSV mu

A Guide to the Evolving Clinical Landscape of Low-Level Viremia Among Persons with HIV in the Modern Treatment Era.

PURPOSE OF REVIEW: Low-level viremia (LLV) remains common in the modern HIV treatment era. We sought to clarify a unified definition of LLV, examine current LLV risk factors, assess outcomes for people with LLV, explore sex and gender differences, and evaluate management updates for people who develop LLV. RECENT FINDINGS: There continue to be varied definitions of LLV without standardization, making comparison of clinical research findings challenging. Potential LLV risk factors have expanded and early data suggest that poor antiretroviral therapy adherence is unlikely to be the cause of mos

A Promising Anti-dengue Virus Carboxylated-Sesquiterpene Lactone from Stevia entreriensis (Asteraceae).

In the ongoing search for effective antiviral agents against dengue virus (DENV), we have isolated a promising compound from a relatively unexplored natural source. This study describes the isolation, structural elucidation, and biological evaluation of a carboxylated sesquiterpene lactone, desacyl grazielia acid tiglate, from Stevia entreriensis (Asteraceae). Using an integrated approach that combines phytochemical and virological techniques, we reveal the potent anti-DENV activity of this compound, characterized by low cytotoxicity and high selectivity against DENV serotype 2 (EC50 = 2.36 μM

Transcriptional impacts of substance use disorder and HIV on human ventral midbrain neurons and microglia.

For people with HIV (PWH), substance use disorders (SUDs) are a prominent neurological risk factor, and the impacts of both on dopaminergic pathways are a potential point of deleterious convergence. Here, we profile, at single nucleus resolution, the substantia nigra (SN) transcriptomes of 90 postmortem donors in the context of chronic HIV and opioid/cocaine SUD, including 67 prospectively characterized PWH. We report altered microglial expression for hundreds of pro- and anti-inflammatory regulators attributable to HIV, and separately, to SUD. Stepwise, progressive microglial dysregulation, c

How HIV-1 Uses the Metabolite Inositol Hexakisphosphate to Build Its Capsid.

The HIV-1 capsid is one of virology's most iconic structures, yet how it assembles has long remained elusive. Remarkably, the capsid is made from just a single protein, CA, which forms a lattice of ~250 hexamers and exactly 12 pentamers. Conical capsids form inside budded virions during maturation, but early efforts to reproduce this in vitro resulted instead in open-ended tubes with a purely hexameric lattice. The missing component in capsid assembly was finally identified as the metabolite inositol hexakisphosphate (IP6). Simply mixing soluble CA protein with IP6 is sufficient to drive the s…

New perspectives in hepatocellular carcinoma surveillance after hepatitis C virus eradication.

Achieving a sustained virologic response (SVR) through direct-acting antivirals for hepatitis C virus (HCV) infection significantly reduces the long-term risk of hepatocellular carcinoma (HCC), particularly in patients with advanced fibrosis (F3) or cirrhosis (F4). However, despite this improvement, the risks associated with HCC and the optimal surveillance strategies for patients who have achieved SVR remain topics of debate. This controversy is compounded by challenges in reliably staging liver fibrosis non-invasively, especially at advanced fibrosis (F3), and the unclear cost-effectiveness,
