Abacavir-reactive memory T cells are present in drug naïve individuals.

BACKGROUND: Fifty-five percent of individuals with HLA-B*57:01 exposed to the antiretroviral drug abacavir develop a hypersensitivity reaction (HSR) that has been attributed to naïve T-cell responses to neo-antigen generated by the drug. Immunologically confirmed abacavir HSR can manifest clinically in less than 48 hours following first exposure suggesting that, at least in some cases, abacavir HSR is due to re-stimulation of a pre-existing memory T-cell population rather than priming of a high frequency naïve T-cell population. METHODS: To determine whether a pre-existing abacavir reactive m

A Curious Case of HLH: A Case Report of Hemophagocytic Lymphohistiocytosis Secondary to Atezolizumab.

A 65-year-old man presented to the hospital with multiorgan failure in the setting of recently being started on atezolizumab for his small cell lung cancer. His main clinical findings were white blood cell count of 18.9 K/μL (48% neutrophils, 32% lymphocytes, 15% monocytes, and 0% eosinophils), creatinine of 10.3 mg/dL (increased from a baseline of 1.5), transaminitis with AST of 1365 and ALT of 388, total bilirubin of 2.3, a lactic acid above 11 (the upper detectable limit of our analyzer), and a serum bicarbonate of 2.8. Imaging was only notable for splenomegaly. Upon presentation, he was in

Climate Change: A Major Factor in the Spread of Aedes aegypti (Diptera: Culicidae) and Its Associated Dengue Virus.

Climate change is thought to be responsible for the spread of various vector-borne diseases. The current study was conducted to evaluate the impact of different temperature and relative humidity regimes on the developmental stages of the yellow fever mosquito, Aedes aegypti (Diptera: Culicidae). The study also evaluated the impact of larval density on the survival of Ae. aegypti. In addition, the association between vector larval abundance, dengue incidence, and climatic factors were elucidated during 2016-2019 in three populated districts of Punjab, Pakistan, i.e., Lahore, Rawalpindi, and Mul…

Biased virus transmission following sequential coinfection of Aedes aegypti with dengue and Zika viruses.

BACKGROUND: Mosquito-borne arboviruses are expanding their territory and elevating their infection prevalence due to the rapid climate change, urbanization, and increased international travel and global trade. Various significant arboviruses, including the dengue virus, Zika virus, Chikungunya virus, and yellow fever virus, are all reliant on the same primary vector, Aedes aegypti. Consequently, the occurrence of arbovirus coinfection in mosquitoes is anticipated. Arbovirus coinfection in mosquitoes has two patterns: simultaneous and sequential. Numerous studies have demonstrated that simultan…

Seasonal insights for integrative mosquito management from multi-year baseline entomological data on Aedes aegypti in Lee County, Florida.

The spread of arboviruses like yellow fever, dengue, chikungunya, and Zika, transmitted by the invasive mosquito Aedes aegypti has led to the development of many strategies to suppress mosquito populations. Given the rapid development of resistance to common chemical larvicides and adulticides in some Ae. aegypti populations, as well as the ever-shrinking chemical options for mosquito control, there is a pressing need for new tools and deployment of those innovative tools as a component of integrative mosquito management programs. Prior to the adoption of any mosquito population intervention, …

Using models and maps to inform Target Product Profiles and Preferred Product Characteristics: the example of Wolbachia replacement.

BACKGROUND: The global prevalence of diseases transmitted by Aedes aegypti mosquitoes, such as dengue, Zika and Yellow Fever, is increasing, but development of promising new mosquito control technologies could reverse this trend. Target Product Profiles (TPPs) and Preferred Product Characteristics (PPCs) documents issued by the World Health Organization can guide the research and development pathways of new products and product combinations transitioning from proof of concept to operational use. METHODS: We used high resolution global maps of the case and economic burden of dengue to derive p…

Alanine to glycine substitution in the PyR2 confers sodium channel resistance to Type I pyrethroids.

BACKGROUND: Aedes aegypti is a primary urban vector of dengue, yellow fever, Zika and chikungunya worldwide. Pyrethroid insecticides are the most effective insecticides for controlling Ae. aegypti. However, pyrethroid resistance has developed due to the long-term overuse of the insecticides, and many knockdown resistance (kdr) mutations have been identified in the resistant populations. A1007G, an alanine to glycine substitution, was found in resistant Ae. aegypti from Vietnam and Malaysia, which has always co-existed with F1534C and V1016G. However, the role of A1007G in pyrethroid resistance…

Japanese encephalitis virus inhibits superinfection of Zika virus in cells by the NS2B protein.

Superinfection exclusion (SIE) is a phenomenon in which a preexisting infection prevents a secondary infection. SIE has been described for several flaviviruses, such as West Nile virus vs Nhumirim virus and Dengue virus vs yellow fever virus. Zika virus (ZIKV) is an emerging flavivirus posing threats to human health. The SIE between ZIKV and Japanese encephalitis virus (JEV) is investigated in this study. Our results demonstrate for the first time that JEV inhibits ZIKV infection in both mammalian and mosquito cells, whether co-infects or subsequently infects after ZIKV. The exclusion effect h…

Biological and behavioral features and colonization of the sylvatic mosquito Sabethes identicus (Diptera: Culicidae).

Mosquitoes of the genus Sabethes are exclusively sylvatic species occurring in Central and South Americas, where they play a role in the transmission of arbovirus. Sabethes identicus, a common bamboo-breeder species, has been found naturally infected with yellow fever virus. Our study aimed to describe biological and behavioral features of Sa. identicus in the laboratory, as well as establish and standardize an isolated colony for experimental assays. We tested different larval densities and evaluated larval and pupal development time, oviposition behavior, egg production, and longevity. We al…

[Management and hygiene measures during an outbreak of herpes, influenza, strangles or infections with multidrug resistant bacteria].

The aim of this review is to describe general guidelines of hygiene measures in the horse stable as well as to provide current recommendations for an outbreak of a common infectious disease. General cleanliness, hand hygiene, avoidance of stress, regular deworming, and vaccinations belong to the basic hygiene measures in a horse herd. All new or returning equids should be submitted to a quarantine period as an important prevention measure. Repeated washing and disinfection of hands may prevent spreading of infectious agents to people and horses. The conception of a hygiene plan, including gener

Assessing single-locus CRISPR/Cas9-based gene drive variants in the mosquito Aedes aegypti via single-generation crosses and modeling.

The yellow fever mosquito Aedes aegypti is a major vector of arthropod-borne viruses, including dengue, chikungunya, and Zika viruses. A novel approach to mitigate arboviral infections is to generate mosquitoes refractory to infection by overexpressing antiviral effector molecules. Such an approach requires a mechanism to spread these antiviral effectors through a population, for example, by using CRISPR/Cas9-based gene drive systems. Critical to the design of a single-locus autonomous gene drive is that the selected genomic locus is amenable to both gene drive and appropriate expression of th…

Predicting the Effects of Climate Change on Dengue Vector Densities in Southeast Asia through Process-Based Modeling.

BACKGROUND: Aedes aegypti and Ae. albopictus mosquitoes are major vectors for several human diseases of global importance, such as dengue and yellow fever. Their life cycles and hosted arboviruses are climate sensitive and thus expected to be impacted by climate change. Most studies investigating climate change impacts on Aedes at global or continental scales focused on their future global distribution changes, whereas a single study focused on its effects on Ae. aegypti densities regionally. OBJECTIVES: A process-based approach was used to model densities of Ae. aegypti and Ae. albopictus an…
