Risk of hepatitis B and tuberculosis reactivation in patients with neuromyelitis optica spectrum disorder undergoing B cell depletion therapy.

INTRODUCTION: B cell-depleting agents are highly effective in preventing relapses in neuromyelitis optica spectrum disorder (NMOSD). Currently, no standardized guidelines exist for preventing hepatitis B reactivation (HBVr) or tuberculosis reactivation (TBr) during the use of B cell-depleting agents in patients with NMOSD who are at risk of HBVr or TBr. Furthermore, data on HBVr and TBr in such patients remain limited. OBJECTIVE: This study aimed to evaluate the risk of HBVr and TBr in patients with NMOSD undergoing B cell depletion therapy and to explore the corresponding prophylactic strate

Shift in the B cell subsets between children with type 1 diabetes and/or celiac disease.

Our purpose was to characterize the pattern of B cell subsets in children with a combined diagnosis of type 1 diabetes (T1D) and celiac disease (C) since children with single or double diagnosis of these autoimmune diseases may differ in peripheral B cell subset phenotype patterns. B cells were analyzed with flow cytometry for the expression of differentiation/maturation markers to identify transitional, naive, and memory B cells. Transitional (CD24hiCD38hiCD19+) and memory Bregs (mBregs; CD24hiCD27+CD19+, CD1d+CD27+CD19+, and CD5+CD1d+CD19+) were classified as B cells with regulatory capacity

Peptide-Loaded HLA Class I Tetramer-Associated Magnetic Bead-Based Enrichment of HBV-Specific CD8+ T Cells.

HBV-specific CD8+ T cells are only present at the low frequency during chronic infection. Thus, they are often undetectable by conventional ex vivo staining methods using peptide-loaded HLA class I tetramers. Detection sensitivity can be increased by magnetic bead-based enrichment strategies following staining with peptide-loaded HLA class I tetramers. Additionally, some downstream applications like e.g., single cell RNA sequencing of virus-specific CD8+ T cells may also require a pre-enrichment step to increase the frequency of the cells of interest. For this, peptide-loaded HLA class I tetra

IMMUNOPHENOTYPE OF LEUKEMIC CELLS IN CHRONIC LYMPHOCYTIC LEUKEMIA PATIENTS WITH NOTCH1 AND SF3B1 GENE MUTATIONS.

BACKGROUND: The typical chronic lymphocytic leukemia (CLL) immunophenotype is vital for diagnosis, but the expression of some antigens varies and has prognostic value. There are data that reduced CD20 expression is associated with NOTCH1 and SF3B1 gene mutations. AIM: To determine a high-risk group of CLL patients for prediction of unfavorable NOTCH1 and SF3B1 gene mutations based on immunophenotyping of leukemic cells. MATERIALS AND METHODS: Flow cytometric and molecular-genetic analysis (mutations of NOTCH1, SF3B1, and TP53 genes using the polymerase chain reaction followed by direct seque

The Role of CTLA-4 in T Cell Exhaustion in Chronic Hepatitis B Virus Infection.

Patients with chronic hepatitis B (CHB) gradually develop T cell exhaustion, and the inhibitory receptor molecule, cytotoxic T-lymphocyte antigen-4 (CTLA-4), may play a role in this phenomenon. This systematic review investigates the role of CTLA-4 in the development of T cell exhaustion in CHB. A systematic literature search was conducted on PubMed and Embase on 31 March 2023 to identify relevant studies. Fifteen studies were included in this review. A majority of the studies investigating CD8+ T cells demonstrated increased expression of CTLA-4 in CHB patients, though one study found this on

Characterization of immune exhaustion and suppression in the tumor microenvironment of splenic marginal zone lymphoma.

The role of the tumor microenvironment (TME) and intratumoral T cells in splenic marginal zone lymphoma (sMZL) is largely unknown. In the present study, we evaluated 36 sMZL spleen specimens by single cell analysis to gain a better understanding of the TME in sMZL. Using mass cytometry (CyTOF), we observed that the TME in sMZL is distinct from that of control non-malignant reactive spleen (rSP). We found that the number of TFH cells varied greatly in sMZL, ICOS+ TFH cells were more abundant in sMZL than rSP, and TFH cells positively correlated with increased numbers of memory B cells. Treg cel

CD5-positive follicular lymphoma: prognostic significance of this aberrant marker?

Follicular lymphomas are the most common of the clinically indolent non-Hodgkin lymphomas. The immunophenotypic pattern of follicular lymphoma classically demonstrates the B-cell markers CD19, CD20, CD22, CD79a, surface immunoglobulin and CD10. The tumour cells are usually negative for CD5. We described two cases of CD5-positive follicular lymphoma. This finding has rarely been described. Our aim was to discuss two cases of CD5-positive follicular lymphoma and review the published literature on the significance of CD5 expression. Bone marrow biopsies showing involvement with follicular lymphom

[Persistent polyclonal B-cell lymphocytosis].

Persistent polyclonal B-cell lymphocytosis (PPBL) is a rare and recently described entity. PPBL is diagnosed predominantly but not exclusively in women, usually smokers and is characterized by a moderate, chronic and absolute lymphocytosis (> 4 x 109/L). Peripheral blood examination show in all cases atypical binucleated lymphocytes. A polyclonal serum IgM is also associated and HLA-DR7 expression is present in most cases. The B cells are polyclonal with kappa and lambda light-chain expression. No clonal rearrangement of immunoglobulin heavy chain genes is observed. Finally, +i(3q) is a recurr

The 'monocytic' cell line I937 displays typical B cell characteristics.

The monocytic cell line I937 was derived from U937 by sorting for cells with high expression of MHC class II molecules. Further analysis of these class II molecules revealed the presence of the HLA-DR3 subtype suggesting that the cell line was a potential candidate for testing antigen presentation to T cells restricted by HLA-DR3. We found that the T cell clones CFTS4:2.80 and CFTS4:2.6 with the required restriction element responded to the house dust mite antigen DPT presented by I937 but not U937, whereas CFTS4:3.1, which is not HLA-DR restricted, did not respond to either cell line. Epstein-Barr virus, infectious mononucleosis, and posttransplant lymphoproliferative disorders.

PTLD may be considered as an "opportunistic cancer" in which the immunodeficiency state of the host plays a key role in fostering the environment necessary for abnormal lymphoproliferation. The following discussion reflects our own current thoughts regarding events which may result in PTLD and its sequelae. Many of the individual steps have not been rigorously proved or disproved at this point in time. Following transplantation and iatrogenic immunosuppression, the host:EBV equilibrium is shifted in favor of the virus. Most seronegative patients will become infected either via the graft or thr

Blood and lymph node T-lymphocyte subsets in non-Hodgkin lymphomas.

Blood and lymph node T-lymphocyte subpopulations have been studied in untreated non-Hodgkin lymphoma (NHL) patients and healthy controls. T- … NHL patients have a reduced pool of T-cells with phenotypic markers of OKT4/LEU3. Monoclonal blood B-lymphocytes were found in 45% of the cases. The presence of such cells in blood was frequently associated with a low OKT4+/OKT8+ ratio.

CD19+ B-Cells, a New Biomarker of Mortality in Hemodialysis Patients.

BACKGROUND AND OBJECTIVES: Mortality of patients on hemodialysis (HD) remains very high despite recent improvements in HD techniques. Cardiovascular (CV) complications and infections are the main causes of death. Some studies suggest that disturbances in the immune system could play a role in this disproportionate mortality, through the links of immunity with inflammation and propensity to infections. However, few studies have addressed the role of lymphocyte populations and the global and CV mortality of HD patients. AIM: To analyze the relationship of peripheral blood lymphocyte populations

Unveiling lymphocyte dynamics: Navigating postoperative immune landscapes in gastric cancer patients undergoing laparoscopic D2 gastrectomy.

INTRODUCTION: Patients with locally advanced gastric cancer often face postoperative complications and insufficient short-term outcomes. Understanding the changes in peripheral lymphocyte subsets following laparoscopic D2 gastrectomy is critical to addressing these challenges and enhancing postoperative recovery. OBJECTIVE: This study investigates the dynamics of peripheral lymphocyte subsets in gastric cancer patients post-laparoscopic D2 gastrectomy. Our goal is to identify factors contributing to postoperative reductions in these immune cells, thereby improving management strategies. METH

Factors affecting lymphocyte subset reconstitution after either related or unrelated cord blood transplantation in children -- a Eurocord analysis.

Immune recovery after cord blood transplantation (CBT) is of concern owing to the low number of lymphocytes transferred with the graft and their immaturity. Risk factors influencing lymphocyte subset reconstitution related to disease, patient, donor and transplant were studied in 63 children (< 16 years), given either related (n = 14) or unrelated (n = 49) CBT for malignant (n = 33) or non-malignant diseases (n = 30). Only children with sustained myeloid engraftment were analysed. Absolute numbers of T (CD3(+), CD4(+), CD8(+)), B and natural killer (NK) cells were reported 2--3, 6, 9, 12 and 1

[Isolation of FDC-lymphocyte clusters from human tonsillar tissues].

Follicular B cells and follicular dendritic cells (FDCs) form FDC-lymphocyte clusters and play a central role in events related to humoral immunity in the lymphoid follicle (LF). The secondary LF is divided into five zones, each of which exhibits functional differences. However, the distribution of the clusters across the five follicular zones remains unclear. We here report a procedure for isolating FDC-lymphocyte clusters from fixed tonsillar tissues and compare of the structure of clusters isolated from three follicular zones: the mantle, light and dark zones. First, the germinal centers (G

Idelalisib for the treatment of chronic lymphocytic leukemia/small lymphocytic lymphoma.

Idelalisib is a first-in-class selective oral PI3Kδ inhibitor for the treatment of patients with relapsed chronic lymphocytic leukemia (CLL) and small lymphocytic lymphoma, a predominantly elderly population with high comorbidity. The drug promotes apoptosis in primary CLL cells ex vivo, independent of common prognostic markers and inhibits CLL cell homing, migration and adhesion to cells in the microenvironment. Idelalisib has shown efficacy with acceptable safety as monotherapy and combination therapy in relapsed/refractory CLL. Idelalisib has clinical activity in patients with CLL with del(

Composite lymphoma: a unique case with two immunologically distinct B-cell neoplasms.

The authors recently have encountered a unique case of composite lymphoma arising in a 69-year-old man with severe long-standing rheumatoid arthritis. The neoplasm was composed of two B-cell lymphomas occurring within a single axillary lymph node: nodules of IgG-kappa nodular poorly differentiated lymphocytic lymphoma were surrounded by a diffuse growth of IgM-lambda well-differentiated lymphocytic lymphoma. It was the immunohistologic studies performed in this case that demonstrated the simultaneous occurrence of two separate monoclonal lymphocyte proliferations. Without the immunohistologic

The effectiveness of the high-LET radiations from the boron neutron capture [10B(n,α) 7Li] reaction determined for induction of chromosome aberrations and apoptosis in lymphocytes of human blood samples.

Provided that a selective accumulation of (10)B-containing compounds is introduced in tumor cells, following irradiation by thermal neutrons produces high-LET alpha-particles ((4)He) and recoiling lithium-7 ((7)Li) nuclei emitted during the capture of thermalized neutrons (0.025 eV) from (10)B. To estimate the biological effectiveness of this boron neutron capture [(10)B(n,α)(7)Li] reaction, the chromosome aberration assay and the flow cytometry apoptosis assay were applied. At the presence of the clinically used compounds BSH (sodium borocaptate) and BPA (p-boronophenylalanine), human lymphoc

Homing receptors and the control of lymphocyte migration.

The traffic of lymphocytes is controlled in part by the selective interaction of circulating lymphocytes with specialized high endothelial venule (HEV) cells at sites of lymphocyte exit from the blood. At least three independent receptor systems are responsible for controlling lymphocyte traffic to different lymphoid organs or to sites of inflammation: one mediates lymphocyte interaction with HEV in peripheral lymph nodes, another in mucosa-associated lymphoid tissues, and a third in inflamed synovium. The receptors mediating lymphocyte recognition of HEV in different organs appear to be struc
