The effects of changing intracellular Ca2+ buffering on the excitability of cultured dorsal root ganglion neurones.

The whole cell patch clamp technique was used to study the effects of changes in [Ca2+]i on the excitability of cultured dorsal root ganglion (DRG) neurones. Increases in [Ca2+]i caused membrane depolarization, altered the characteristics of evoked action potentials and activated potassium, chloride and non-selective cation conductances. Mobilization of Ca2+ from intracellular stores with caffeine (1 mM) or ryanodine (10 microM) or photorelease of Ca2+ from DM-nitrophen gave rise to depolarizations suggesting a dominant inward current under our recording conditions of low [Ca2+]i buffering cap

Startle-induced epilepsy in a patient with epidermal nevus syndrome.

Startle-induced epilepsy was observed in a 5-year-old boy with epidermal nevus syndrome. He manifested linear nevus on the face and neck, mild mental retardation, and right hemiparesis. Massive myoclonus, followed by tonic seizures, had been triggered by unexpected auditory stimuli since 3 years of age. The startle-induced seizures were the only epileptic manifestation. Interictal EEG occasionally depicted spontaneous focal spikes and waves in the left frontotemporal area, and ictal EEG depicted vertex spikes and then diffuse slow spike-and-wave complex bursts. Left frontal and perisylvian cor

Sclerosteosis: report of a case in a black African man.

Sclerosteosis is a rare genetic disorder of bone modelling, similar to, but distinct from, van Buchem disease; it has been described almost … rophy, facial palsy and trigeminal neuralgia. Radiologic examination, visual and brainstem auditory evoked potentials, computerized tomography and magnetic resonance imaging of the skull were performed. This seems to be the first case of the disease in a black African individual, with no known relationship with Dutch ancestry.

Action of babycurus-toxin 1 from the east African scorpion Babycurus centrurimorphus on the isolated cockroach giant axon.

A toxin named babycurus-toxin 1 (mol. wt 8191), from telson extracts of the scorpion Babycurus centrurimorphus, was found to depolarize the cockroach giant axon. It progressively blocked the evoked action potentials after a short period of limited repetitive activity and after 30 min of toxin action it became impossible to evoke responses to current stimulations. Voltage-clamp experiments on the sodium current indicated that the toxin in micromolar concentrations progressively decreased the transient inward peak sodium current, but also slowed the activation phase of this sodium current and ma

Desensitization of AMPA receptors limits the amplitude of EPSPs and the excitability of motoneurons of the rat isolated spinal cord.

Intracellular recording was used to study the effect of cyclothiazide, a selective blocker of alpha-amino-3-hydroxy-5-methyl-4-isoxazolepropionate (AMPA) receptor desensitization, on lumbar motoneurons of the rat isolated spinal cord. Cyclothiazide (25 microM) enhanced the responses to AMPA in a tetrodotoxin-insensitive fashion, without affecting those produced by N-methyl-D-aspartate or gamma-aminobutyric acid. Excitatory postsynaptic potentials (EPSPs) evoked by dorsal root stimulation were strongly potentiated in amplitude while paired-pulse depression (produced by applying pairs of pulses

Neural conduction velocity of the human auditory nerve: bipolar recordings from the exposed intracranial portion of the eighth nerve during vestibular nerve section.

We measured the conduction velocity of the intracranial portion of the auditory nerve in 3 patients undergoing vestibular nerve section to treat Ménière's disease. The conduction velocity varied from patient to patient, with an average value of 15.1 m/sec. The latency of peak III of the brain-stem auditory evoked potentials (BAEPs) increased by an average of 0.5 msec as a result of exposure of the eighth nerve, and if that increase is assumed to affect the entire length of the auditory nerve (2.6 cm) evenly, then the corrected estimate of conduction velocity would be 22.0 m/sec. Estimates of c…

[Retrocochlear hearing disorders].

Sensorineural hearing loss is a common disorder that results from damage to the inner ear in over 95% of all cases; therefore, retrocochlear hearing disorders are rare and cannot be differentiated from sensory losses by clinical symptoms alone. Associated vestibular and other neurotological symptoms must lead to appropriate diagnostic procedures. Besides audiological and neurotological tests, e.g. auditory-evoked potentials, otoacoustic emissions or electronystagmography, modern imaging methods are the most valuable tools to detect tumorous, vascular and inflammatory processes. Acoustic neurom…

[Optic neuropathy in type-1 diabetes and acetylsalicylic acid-refractory thrombocyte activation].

A 26-year-old female with severe complications from type I diabetes mellitus of 17 years' duration (proliferative retinopathy, nephropathy with renal failure and nephrotic syndrome) developed rapid deterioration of vision in the right eye to 6/60 over a period of several weeks. There were no other neurological signs. Ophthalmological examination showed no worsening of the diabetic retinopathy, but the presence of bilateral optic atrophy, confirmed by visual evoked potentials. CT scan did not reveal any retrobulbar process, and MR scans of both the optic nerves and the visual pathways were unre…

Ageing and problems in differential diagnosis of psychoorganic syndromes.

Depressive syndrome was investigated in 132 psychiatric and neurologic patients by Zung's self-rating depression scale. The dexamethasone suppression test (DST) was carried out in 62 patients. Visual evoked potentials (VEP) (stimulation with flash (F-VEP) and pattern (P-VEP)), serum levels of LH, FSH, prolactin, ACTH, estradiol and cortisol were measured in 15 female patients between 85 and 94 years of age. Abnormal DST results indicated depression rather than dementia. P-VEP latencies correlated with cognitive symptoms, while F-VEP latencies correlated with affective symptoms. Gonadotropin le…

[Interaction between continuous and impulse noise: anatomic and functional evaluation in relation to the intensity of the exposure].

Interaction between continuous and impulse noise was studied. One group of chinchillas was exposed to octave band of noise of 0.5 kHz 95 dB SPL intensity. Three groups were exposed to impulse noises of 113, 119 and 125 prak SPL emitted at 1 imp/sec, 1 imp/4 sec and 1 imp/16 sec respectively. Three groups were exposed to noise created by a combination of above continuous and impulse noises. All exposures lasted for 5 days. Hearing thresholds were measured in 35 animals using auditory evoked potentials. The combined continuous and impulse noises showed equal energy contents, but the groups expos…

[Intraoperative monitoring auditory function during surgery of internal auditory canal and C-P angle].

Intraoperative measurement of electrocochleography, auditory brainstem responses and eighth nerve action potentials are utilized to assess the functional integrity of the cochlea and cochlear nerve. All such assessments have only one purpose: for warning the surgeon the possibility of damage the auditory system or its surrounding structure. Eighteen patients with various otoneurological diseases undergoing surgery had intraoperative monitoring of such evoked electrophysiological responses. Hearing was preserved in sixteen patients. However, the CM/N1 in ECochG still keep for a few minutes even

[Somatosensory disorders in patients with extrapyramidal pathology: the effect of a stereotaxic intervention].

The authors compared somatosensory evoked potentials (SSEP) recorded in 16 patients with infantile cerebral paralysis (ICP), 16 patients with parkinsonism (P), 18 patients with torticollis (T), and 15 healthy individuals. Structural changes of SSEP were found in all patients with ICP and in most patients with P and T. The structural changes of SSEP in these patients were mainly associated with the absence or suppression of one or two components of SSEP of a healthy individual. It is assumed that one and the same pathological process underlies these three diseases, but it differs in the degree …

[Benign encephalitis of the brain stem].

Seven cases of an encephalitic brainstem syndrome are reported. All had an initial flu-like phase, with a febrile state. Then, all had eye movements disturbances and ataxia. Six had a variable dysfunction of other cranial nerves, five had dysmetria of limbs and three had pyramidal signs. In all, the topographic diagnosis was based on clinical, neurophysiological (electromyography, evoked potentials) and neuroradiological (swelled brainstem on CT scan) findings. Analysis of cerebro-spinal fluid revealed pleocytosis (mean; 165 cells/ml), without profile indicative of multiple sclerosis. Serologi…

[Neurophysiologic analysis of disorders of the functional activity of projection systems of the brain in patients with neuroses].

To study the neurophysiological mechanisms of neurasthenia, hysterical neurosis and obsession neurosis with a view to the practical employment of the results, the authors examined 100 patients and 40 normal subjects using the method of evoked potentials (EP). EP changes indicated an neurasthenia-related decrease in the functional activity of the cortex, weakening of its inhibitive functions and an increase in the reactivity of the system of negative-emotional support. Patients with hysterical neurosis displayed the greatest excitability of the structures of the projective pathways and insuffic…

Treatment of adult and obstetrical brachial plexus injuries.

Twenty-five percent of patients with injuries to the plexus require surgical intervention for optimal results. Patients with loss of serratus and rhomboids, Horner's syndrome, positive myelogram, or negative evoked potentials have a high incidence of lower root avulsions and a probability of upper root ruptures. These should be explored early. Results depend on the amount of remaining axonal input available for grafting. Using intraoperative electrical studies, elements which will recover spontaneously may be saved and improved function obtained by grafting only damaged elements. Obstetrical p…

Changes of neurochemical and electrophysiological indices of rat brain under ethanol intoxication.

It was found that acute ethanol intoxication caused an imbalance of the neurotransmitters in the CNS: accumulation of GABA and serotonin and depletion of catecholamines. Alcohol depression was characterized by suppression of the evoked potentials of the various rat brain structures. Under chronic ethanol intoxication of animals, relative stabilization of the electrophysiological indices of the rat brain activity was observed. This reflects the CNS adaptation to the constant ethanol presence in the blood. This state was also characterized by the relative stabilization of the serotonin system an…

Chronic cerebellar stimulation in the monkey. Preliminary observations.

In a single monkey, the surface of the cerebellum was stimulated electrically for 205 hours with electrodes and parameter values similar to those currently used in humans for treatment of epilepsy. Im pedance of stimulating and nonstimulating control electrodes remained unchanged throughout an observation period of six months. Potentials evoked by cerebellar stimulation could be recorded from the cranium, providing a noninvasive technique of determining the level of current delivered to cerebellum. Examination of the implantation site showed marked meningeal thickening surrounding the stimulat
