Personality disorders and cardiometabolic diseases: A Mendelian randomization study.

Observational studies have suggested the associations between personality disorders (PDs) and cardiometabolic diseases (CMDs), though these may be biased by unmeasured confounding, including lifestyle factors or reverse causality. Mendelian randomization (MR) provides a novel approach to address these limitations by using genetic variants as instrumental variables, thereby minimizing environmental confounding and enabling causal inference. This study leverages MR to systematically evaluate causal relationships across 9 clinically relevant PDs and 8 major CMDs, offering genetic evidence to clar

Causal Relationship Between Chronic Obstructive Pulmonary Disease and Respiratory Tuberculosis Susceptibility: A Two-Sample Bayesian Weighted Mendelian Randomization Study.

The causal relationship between chronic obstructive pulmonary disease (COPD) and respiratory tuberculosis (TB) remains unclear due to limited evidence from prospective studies. Therefore, this study aimed to assess the causal relationship between COPD and respiratory TB using a two-sample Bayesian weighted Mendelian randomization (BWMR) study. The genetic instrumental variants (IVs) for COPD and respiratory TB were obtained from the IEU Open GWAS project in 2021. The inverse variance weighted (IVW) method was used as the main statistical analysis method, and the results supplemented with weigh

A Mendelian randomization study: Genetically predicted dopamine 4-sulfate to dopamine 3-O-sulfate ratio mediates the association between chemokine C-C motif ligand 11 and polycystic ovary syndrome.

This Mendelian randomization (MR) study aims to elucidate the causal pathways between circulating chemokine C-C motif ligand 11 (CCL11) levels and the pathogenesis of polycystic ovary syndrome (PCOS), with a specific focus on the mediating role of dopamine sulfate isomers. Using genome-wide association data from 14,824 European participants (for CCL11 quantitative trait loci) and 2,01,005 individuals (including 1424 PCOS cases), we performed 2-sample MR analyses, with inverse-variance weighting as the primary estimator. Sensitivity analyses included MR-Egger regression and weighted median appr

Multiple association studies identify 3 novel candidate genes for teat number trait in Danish Landrace and Large White pigs: BRINP3, LIN52, and UBE3B.

Milk is an essential source of nutrition for preweaning piglets. Therefore, in the breeding process, sows were expected to have sufficient teats to suckle their piglets. However, in Danish Landrace and Large White pigs, the number of piglets born currently exceeds the number of teats, making it urgent to select and breed for an increased teat number. In this study, the samples of 491 Danish Landrace pigs and 1,047 Danish Large White pigs with teat number phenotype were used to perform genome-wide association studies to identify SNPs associated with total teat number (TTN) based on SNP-chip dat

The causality between chronic rhinosinusitis and inflammatory bowel disease: A bidirectional 2-sample Mendelian randomization analysis.

Prior observational studies suggested an association between chronic rhinosinusitis (CRS) and inflammatory bowel disease (IBD), but causality remains unclear. This study employed a bidirectional 2-sample Mendelian randomization (MR) analysis to investigate potential causal relationships. Genetic variants associated with CRS and IBD (including Crohn's disease and ulcerative colitis) were sourced from the IEU Open genome-wide association study project. Primary analysis used inverse-variance weighted, supplemented by weighted median and MR-Egger methods. Sensitivity analyses included Cochran's Q …

Early Vascular Aging Determined by 3-Dimensional Aortic Geometry: Genetic Determinants and Clinical Consequences.

BACKGROUND: Vascular aging is an important phenotype characterized by structural and geometric remodeling. Some individuals exhibit supernormal vascular aging, associated with improved cardiovascular outcomes; others experience early vascular aging, linked to adverse cardiovascular outcomes. The aorta is the artery that exhibits the most prominent age-related changes; however, the biological mechanisms underlying aortic aging, its genetic architecture, and its relationship with cardiovascular structure, function, and disease states remain poorly understood. METHODS: We developed sex-specific

Genetic causal effects of multi-site chronic pain on post-traumatic stress disorder: Evidence from a two-sample, two-step Mendelian randomization study.

BACKGROUND: Existing evidence supports a correlation between multi-site chronic pain and post-traumatic stress disorder (PTSD), but it is yet to be determined if this correlation is causal and in what direction the causation works. METHODS: Applying two-sample Mendelian randomization (MR) analysis to data from available genome-wide association studies in populations of European ancestry, we estimated the causal association between multi-site chronic pain and no pain versus PTSD. Moreover, we used multivariable and mediation MR analysis to assess the mediating effects of 13 lifestyle factors o

HHIP protein interactions in lung cells provide insight into COPD pathogenesis.

Chronic obstructive pulmonary disease (COPD) is the third leading cause of death worldwide. The primary causes of COPD are environmental, including cigarette smoking; however, genetic susceptibility also contributes to COPD risk. Genome-Wide Association Studies (GWASes) have revealed more than 80 genetic loci associated with COPD, leading to the identification of multiple COPD GWAS genes. However, the biological relationships between the identified COPD susceptibility genes are largely unknown. Genes associated with a complex disease are often in close network proximity, i.e. their protein pro
