[Analysis of the frequencies of genotype combinations of 4 polymorphisms of genes acting on the folate cycle in the Spanish population].

BACKGROUND AND OBJECTIVE: Studies on different populations have shown a great variability of the frequencies of different polymorphisms in genes acting in the folate cycle. The present study was aimed to analyze the frequency in the Spanish population of each genotype combination of four polymorphisms, one of them -1561C-T of the glutamate carboxypeptidase II (GCPII) gene- being the first time that is studied in Spain. The study included a meta-analysis of the published data. SUBJECTS AND METHOD: Using the Spanish Collaborative Study of Congenital Malformations (ECEMC) Network, blood samples

Design, synthesis, and evaluation of gamma-phosphono diester analogues of glutamate as highly potent inhibitors and active site probes of gamma-glutamyl transpeptidase.

Gamma-glutamyl transpeptidase (GGT, EC 2.3.2.2) catalyzes the transfer of the gamma-glutamyl group of glutathione and related gamma-glutamyl amides to water (hydrolysis) or to amino acids and peptides (transpeptidation) and plays a central role in glutathione metabolism. GGT is involved in a number of biological events, such as drug resistance and metastasis of cancer cells by detoxification of xenobiotics and reactive oxygen species through glutathione metabolism, and is also implicated in physiological disorders, such as Parkinson's disease, neurodegerative disease, diabetes, and cardiovascu

Flash photolysis using a light emitting diode: an efficient, compact, and affordable solution.

Flash photolysis has become an essential technique for dynamic investigations of living cells and tissues. This approach offers several advantages for instantly changing the concentration of bioactive compounds outside and inside living cells with high spatial resolution. Light sources for photolysis need to deliver pulses of high intensity light in the near UV range (300-380 nm), to photoactivate a sufficient amount of molecules in a short time. UV lasers are often required as the light source, making flash photolysis a costly approach. Here we describe the use of a high power 365 nm light em

Targeting ionotropic receptors with polyamine-containing toxins.

This review summarises current knowledge of polyamine-containing spider toxins and their interactions with ionotropic receptors of invertebrate and vertebrate excitable cells. Their diverse actions on ionotropic glutamate and acetylcholine receptors, which include potentiation, closed channel block and open channel block, are discussed in the context of toxin and target structures. Factors that complicate attempts to identify and pharmacologically characterise the binding sites for these toxins include their ability to permeate channels of some ionotropic receptors and their apparent accumulat

Glutamate receptors in RVLM modulate sympathetic baroreflex in conscious rabbits.

In this study, we examined the effect of excitatory amino acid (EAA) receptor blockade in the rostral ventrolateral medulla (RVLM) on the renal sympathetic baroreflex in conscious rabbits. Rabbits were implanted with guide cannulas for bilateral microinjections into the RVLM (+2 to +3 mm from the obex, n = 8) or into the intermediate ventrolateral medulla (IVLM; 0 to +1 mm from the obex, n = 5) and with an electrode for measuring renal sympathetic nerve activity (RSNA). After 7 days of recovery, microinjection of the EAA receptor antagonist kynurenate (10 nmol) into the RVLM did not affect res

Decreased plasma and cerebrospinal fluid glutamine concentrations in a patient with bialaphos poisoning.

A 47-year-old Japanese woman undergoing maintenance hemodialysis (HD) was admitted to our hospital because of poisoning with the herbicide bialaphos. Respiratory arrest and loss of consciousness ensued rapidly, accompanied by convulsions and nystagmus. Treatment with HD and direct hemoperfusion, followed by HD alone, effectively removed bialaphos and its chief toxic metabolite (L-AMPB) from the circulation (bialaphos decreased from 0.33 to < 0.05 microg/ml and L-AMPB from 14 to 0.86 microg/ml). The glutamate concentration improved gradually after the removal of bialaphos and L-AMPB from plasma

[A study of lipid peroxidation in patients with chronic gastritis during administration of a food additive monosodium glutamate].

The levels of diene conjugates (DC) and TBA-reactive substances (TBA-RS) were studied in serum blood of patients with gastritis before and after including in diet of monosodium glutamate (MSG). For investigation 15 patients were selected who had unacid or subacid signs determined by histamine test. It was shown that levels of DC and TBA-RS were increased in serum of patients. MSG increased the gastric secretion in patients but had no effect of levels of DC and TBA-RS in serum.

Effect of ferric nitrilotriacetate on rostral mesencephalic cells.

After murine fetal cells from the rostral mesencephalic tegmentum were isolated, prepared, and cultured; neuronal and glial cells in primary mixed cell cultures were exposed to ferric nitrilotriacetate (Fe-NTA) at varying concentrations. Studies were performed at 23 days in culture after 14 day exposure to Fe-NTA. In addition to morphologic studies, biochemical assays including specific [3H]flunitrazepam (FLU) binding, clonazepam (CLO)-displaceable [3H]-FLU binding, Ro5-4864-displaceable [3H]-FLU binding, [3H]-FLU binding, [3H]dopamine (DA) uptake, [3H]haloperidol (HAL) binding, [3H]spiperone

Amino acid requirements of a Chlamydia trachomatis genital strain in McCoy cell cultures.

Amino acid requirements of a Chlamydia trachomatis were defined in McCoy cell monolayer cultures, depleted of their nutrient reserves by maintenance in Earle balanced salt solution for at least 2 days, and then treated at infection with 1 microgram of cycloheximide per ml, an inhibitor of host cell protein and deoxyribonucleic acid synthesis. Serum-free Waymouth MB 752/1 medium lacking one amino acid or a group of related amino acids was added to depleted cultures before infection. Eight of the amino acids normally present in MB 752/1 were essential for chlamydial growth, six were advantageous

The carotid chemoreceptor input to the respiratory neurones of the nucleus of tractus solitarus.

1. An investigation has been made into the connexions between the carotid body chemoreceptors and the dorsal respiratory neurones of the cat's medulla.2. In confirmation of previous work these neurones were found to be all inspiratory in firing pattern and to fall into two categories, Ralpha (forty-four units) which fire only with the central inspiratory rhythm and Rbeta (thirty-two neurones) that are also excited by lung inflation. Both categories were shown to be excited by stimuli delivered to the carotid bodies during inspiration but, with a single exception, not during expiration.3. When

Effects of ethanol on neurotransmitter release by rat brain cortical.

Using a double label technique to preload rat brain cortex slices with different radioactive neurotransmitters (or precursor choline), we have studied the effects of ethanol on the electrically stimulated release of these transmitters. Ethanol inhibited the release of these transmitters, acetylcholine being the most sensitive and occurring at concentrations compatible with moderate to severe intoxication in the rat (IC50 equals 0.17 M). The order of sensitivity to ethanol was acetylcholine greater than serotonin greater than dopamine greater than norepinephrine greater than glutamate greater g

A radiometric technique for measuring L-asparagine in picomole quantities.

A radiometric technique for measuring l-asparagine in small samples of biological origin is presented. After removal of l-glutamate and l-aspartate by enzymic decarboxylation, l-asparagine in the sample is hydrolysed to l-aspartate with l-asparaginase from Escherichia coli. The l-aspartate so generated is then enzymically transaminated with 2-oxo[1-(14)C]glutarate yielding l-[1-(14)C]glutamate. After specific chemical removal of unchanged 2-oxo[1-(14)C]glutarate, the l-[1-(14)C]glutamate generated is determined by scintillation counting of the radioactivity. This technique can measure the conc
