Broad-Spectrum Regulation of Nonreceptor Tyrosine Kinases by the Bacterial ADP-Ribosyltransferase EspJ.

Tyrosine phosphorylation is key for signal transduction from exogenous stimuli, including the defense against pathogens. Conversely, pathogens can subvert protein phosphorylation to control host immune responses and facilitate invasion and dissemination. The bacterial effectors EspJ and SeoC are injected into host cells through a type III secretion system by enteropathogenic and enterohemorrhagic Escherichia coli (EPEC and EHEC, respectively), Citrobacter rodentium, and Salmonella enterica, where they inhibit Src kinase by coupled amidation and ADP-ribosylation. C. rodentium, which is used to

Zika virus: unraveling the complex interplay between viral pathogenesis and host immune defenses.

Zika virus (ZIKV) is a Flavivirus that has spread globally since its initial reporting. Transmitted to humans via multiple mosquito species, its typical symptoms (e.g., Guillain-Barre syndrome [GBS] and neonatal microcephaly) pose severe public health challenges. Based on existing research, the pathogenic characteristics of ZIKV are largely shaped by host molecular pathways, endowing the virus with an enhanced ability toward specific host cell types. On the other hand, ZIKV achieves intracellular survival and increased pathogenicity by evading the human immune system. Host innate immune respon

Kinetic Tracking of Plasmodium falciparum Antigens on Infected Erythrocytes with a Novel Reporter of Protein Insertion and Surface Exposure.

Intracellular malaria parasites export many proteins into their host cell, inserting several into the erythrocyte plasma membrane to enable interactions with their external environment. While static techniques have identified some surface-exposed proteins, other candidates have eluded definitive localization and membrane topology determination. Moreover, both export kinetics and the mechanisms of membrane insertion remain largely unexplored. We introduce Reporter of Insertion and Surface Exposure (RISE), a method for continuous nondestructive tracking of antigen exposure on infected cells. RIS

Illuminating the Dark Host Cell Proteome: A host cell protein coverage method for LC-MS impurity assays.

Host cell protein (HCP) analysis by liquid chromatography-mass spectrometry (LC-MS) enables identification and quantification of individual impurities in biopharmaceutical products, supporting risk-based impurity assessment. However, unlike enzyme-linked immunosorbent assay (ELISA) methods, systematic evaluation of method-specific HCP coverage for LC-MS assays remain poorly defined. In particular, limited attention has been given to proteins that are inherently undetectable under specific analytical conditions, potentially creating blind spots in impurity characterization. Here, we present DAR

Multi-omics profiling reveals associations between gut microbiota and olfactory gene expression in mosquitoes.

INTRODUCTION: The interplay between gut microbiota and host physiological processes has been extensively studied in vertebrates, where it plays a crucial role in regulating appetite, emotion, immunity, and other physiological functions. However, whether a similar regulatory mechanism exists in insects remains unclear, especially regarding the long-distance regulation of olfactory function. This study focused on three Culex subspecies (Culex quinquefasciatus, Culex pipiens pallens, and Culex pipiens molestus) that are closely related but exhibit significant differences in olfaction-dependent ec

Therapeutic depletion of natural killer cells controls persistent infection.

Persistent viral infections are associated with host and viral factors that impair effective antiviral immunity. Natural killer (NK) cells contribute to establishment of persistent lymphocytic choriomeningitis virus (LCMV) infection in mice through suppression of virus-specific T cell responses during the first few days of infection, but NK cell depletion during those early time points can enable severe T cell-mediated immune pathology and death of the host. Here we show that long after their peak in cytolytic activation, NK cells continue to support viral persistence at later times of infecti

Endothelial cells present antigens in vivo.

BACKGROUND: Immune recognition of vascular endothelial cells (EC) has been implicated in allograft rejection, protection against pathogens, and lymphocyte recruitment. However, EC pervade nearly all tissues and predominate in none, complicating any direct test of immune recognition. Here, we examined antigen presentation by EC in vivo by testing immune responses against E. coli beta-galactosidase (beta-gal) in two lines of transgenic mice that express beta-gal exclusively in their EC. TIE2-lacZ mice express beta-gal in all EC and VWF-lacZ mice express beta-gal in heart and brain microvascular

Aromaticity Engineering of Hybrid Cyanine Dyes for Metabolic Labeling and Bioorthogonal NIR-II Imaging of Gut Microbiota.

In the context of diabetes, host immune dysfunction and microcirculation damage significantly increase the susceptibility to bacterial infection. Therefore, the development of high-performance molecular imaging tools to dynamically track the migration and colonization of bacteria in deep living tissues is crucial to distinguish pathology-associated behavioral changes. In this study, we removed heteroatoms from the classical xanthene structure and effectively integrated the more planar and conjugated aminofluorene with the cyanine dye skeleton, enhancing the molecular aromaticity in the excited

Evidence of Japanese Encephalitis Virus Exposure in Feral Pig Populations in North Queensland, Australia.

Several animal species, including horses and pigs, can be infected with Japanese encephalitis virus (JEV). Feral pigs (Sus scrofa) are considered an effective amplifying host for JEV in natural habitats. In this study, 265 dried blood spots were collected from feral pigs in four selected North Queensland local government areas, and antibodies against JEV were detected using a competitive enzyme-linked immunosorbent assay (cELISA). The results showed that 26.4% (70/265) of the feral pigs had antibodies specific to JEV. We found a significant difference in the seroprevalence of JEV in the Wet Tr

Yellow fever beyond the Amazon: surveillance challenges and the role of Callithrix spp. in sylvatic cycles in Brazil.

Yellow fever virus (YFV) remains a major public health concern in tropical and subtropical regions of the Americas and Africa. Since late 2016, sylvatic yellow fever (YF) outbreaks and sporadic cases affecting both humans and non-human primates (NHP) have been recorded in Southern Brazil. These events raised concerns about viral persistence beyond the Amazon Basin, where YFV is endemic, and the potential reemergence of urban transmission. Neotropical NHPs are central to the ecology of sylvatic YF, but many aspects of their infection dynamics and species-specific roles remain poorly understood.

Should I stay or should I go? DNA damage thresholds drive stay-or-go decisions in Acinetobacter baumannii biofilms.

Acinetobacter baumannii is a clinically significant pathogen that is difficult to eliminate from infected individuals; indeed, infections due to this organism cost the health care system tens of millions of dollars per year. This pathogen readily infects immunocompromised individuals and can survive extreme conditions, including the osmotic and immune stresses of the human host and prolonged desiccation on hospital surfaces. The survival success of A. baumannii is in large part due to the formation of multicellular communities, or biofilms, that afford cells protection from external stresses,

Emerging insights into CC and CXC chemokines and their receptors in Mycobacterium tuberculosis infection.

Chemokines and their cognate receptors are central orchestrators of the immune response to Mycobacterium tuberculosis (Mtb) infection. While their overall significance in tuberculosis (TB) is well-established, this review synthesizes recent advances to clarify the distinct roles of CC and CXC chemokines in differentiating active disease, latent infection, and the often overlooked subclinical TB state. We evaluate the potential of specific chemokine signatures as emerging diagnostic biomarkers compared to conventional standards and assess their promise as novel therapeutic targets in personaliz

Functional analysis of Ophraella communa Lesage OcomOBP11 in recognition of Ambrosia artemisiifolia L. volatiles.

Odorant binding proteins (OBPs) play a critical role in insect survival by facilitating the detection of environmental odors. This study identified and characterized OcomOBP11 in the leaf beetle Ophraella communa Lesage. Sequence and phylogenetic analyses demonstrated that OcomOBP11 belongs to the Minus-C OBP family. Tissue-specific expression analysis revealed that OcomOBP11 showed the highest relative expression in the testis. Functional assays demonstrated that OcomOBP11 has strong binding affinity to 24 of the 26 volatiles from Ambrosia artemisiifolia L., a primary host plant. Molecular do

Unlocking data in Klebsiella lysogens to predict capsular type-specificity of phage depolymerases.

Viral entry is a critical step in the infection process. Klebsiella spp. and other clinically relevant bacteria often express complex polysaccharide capsules that act as a barrier to phage entry. In turn, most lytic phages targeting Klebsiella encode depolymerases for capsule removal. This virus-host arms race leads to extensive genetic diversity in both capsules and depolymerases, complicating our ability to understand their interaction. This study exploits the genetic information encoded in Klebsiella prophages to model the interplay between the bacteria, the prophages, and their depolymeras

An Exploration of the Relationship Between Gut Virome and Cardiovascular Disease: A Comprehensive Review.

This study reviews the correlation between the gut virome and cardiovascular diseases (CVDs) and investigates the potential role of the gut virome in CVD progression. The gut virome, which includes bacteriophages and eukaryotic viruses, interacts with the intestinal microbiota and the host immune system, and affects the overall health of the host. Previous studies have demonstrated that alterations in the gut virome are closely associated with various cardiovascular conditions, including hypertension, atherosclerosis, atrial fibrillation, heart failure, and viral myocarditis. Thus, the gut vir

Host-microbiota interactions in genitourinary cancer immunotherapy.

Immunotherapy for genitourinary cancer, including bladder cancer, renal cell carcinoma, and prostate cancer, has made significant progress in recent years; however, it still faces challenges such as low patient response rates, drug resistance and immune-related adverse events. As the "second genome" of the human body, the gut microbiota plays a key factor in modulating the host immune response. Studies have shown that host-microbiome interactions profoundly affect the efficacy and safety of immune checkpoint inhibitors by shaping the tumor immune microenvironment (TIME). This review systematic

N-Glycoproteomics of the Apicomplexan Parasite Toxoplasma gondii.

Protein N-glycosylation influences protein folding, stability, and trafficking, and has prominent functions in cell-cell adhesion and recognition. For the parasite Toxoplasma gondii, N-glycosylation of proteins is crucial for initial adhesion to host cells, parasite motility, and consequently, its ability to invade host cells. However, the glycoproteome of T. gondii remains largely unknown. In this study, we used the α-mannose-specific Burkholderia cenocepacia lectin A (BC2L-A) to enrich glycopeptides from T. gondii tachyzoites and analysed them by tandem mass spectrometry. The data enable the

Microbiological mechanisms of oral mucosal disease: oral-intestinal crosstalk and probiotic therapy.

Balance of the oral-intestinal axis microbiota is essential for maintaining oral mucosal health. The occurrence of oral disease is closely linked to the microbiota, this disorder is closely related to the pathogenesis of oral mucosal diseases, such as oral lichen planus, recurrent aphthous ulcer, oral candidiasis and squamous-cell carcinoma. As a microorganism that is beneficial to host health, probiotics can show multi-dimensional therapeutic effects in oral mucosal diseases by targeting and regulating the immune microenvironment of the oral mucosa, inhibiting the colonization of pathogenic b

Characterizing the microbiome of Dujardinascaris helicina (Nematoda: Ascarididae) isolated from wild American crocodiles in Belize.

Microbiomes are communities of microorganisms that form close associations with metazoan hosts and have important roles in host biological processes. With the advent of Next Generation Sequencing, the microbiomes of myriad animals and plants have been described. However, the microbiomes of parasites have received little attention, which is surprising considering their ecological and medical importance. This study characterizes, for the first time, the microbiome of Dujardinascaris helicina, a gastrointestinal nematode parasite of the American crocodile. Dujardinascaris helicina were isolated f
