BACKGROUND: Chronic hepatitis B virus (HBV) infection remains a major global health challenge and a leading cause of liver cirrhosis and hepatocellular carcinoma. Interleukin-6 (IL-6), a key pro-inflammatory cytokine, plays an important role in immune regulation and hepatic inflammation. However, its relationship with HBV viral load and disease severity remains incompletely understood.
METHODS: A hospital-based cross-sectional study was conducted among 293 HBsAg-positive patients. Serum IL-6 levels were measured using ELISA, and HBV DNA was quantified using real-time quantitative PCR. Patient

INTRODUCTION: Hepatocellular carcinoma (HCC) remains a major cause of cancer-related mortality worldwide, and postoperative recurrence continues to limit long-term survival. Liver-resident natural killer (NK) cells are key effectors of antitumor immunity in the hepatic microenvironment. Interleukin-37 (IL-37), an anti-inflammatory cytokine, has recently been shown to modulate immune responses and enhance NK-cell activity.
METHODS: Liver mononuclear cells were isolated from 24 deceased donors. For functional experiments, liver NK cells from six donors were cultured with interleukin-2 (IL-2) in

Despite the recent scientific advancements, pancreatic cancer remains the seventh leading cause of cancer-related mortality. Pancreatic cancer progression is closely associated with inflammation, and we previously showed that short hairpin RNA-mediated knockdown of sST2 expression, a soluble decoy receptor for the proinflammatory molecule interleukin-33 (IL-33), in mouse Panc02 pancreatic cancer cells reduced malignant growth following pancreatic (orthotopic) implantation. Furthermore, this growth suppression was accompanied by decreased tumor angiogenesis, reduced expression of the neutrophil

Severe fibrosis, predominantly driven by the activation of pancreatic stellate cells (PSCs), plays a crucial role in the poor prognosis associated with pancreatic ductal adenocarcinoma (PDAC). Understanding the intricate interplay between tumor cells and their microenvironment is essential for deciphering the regulatory mechanisms underlying PSC activation. This study sheds light on the critical role of tumor-derived interleukin-35 (IL-35) in modulating PSC activation, thereby unveiling a promising therapeutic target for mitigating PDAC progression. This study demonstrates that IL-35, secreted

T cells contribute to immunotherapy and autoimmune pathogenesis and Langerhans cells (LCs) have a substantial ability to activate T cells. In vitro-generated monocyte-derived LCs (Mo-LCs) are useful models to study LC function in autoimmune diseases and to test future LC-based immunotherapies. Although dendritic cells (DCs) expressing high levels of Delta-like 4 (DLL4+ DCs), which is a member of the Notch ligand family, have greater ability than DLL4- DCs to activate T cells, the induction method of human DLL4+ DCs has yet to be determined. The aim of this study is to establish whether Mo-LCs

Cancer-associated fibroblasts (CAFs) are a crucial component in the tumor microenvironment (TME) of peritoneal metastasis (PM), where they contribute to tumor progression and metastasis via secretion of interleukin-6 (IL-6). Here, we investigated the role of IL-6 in PM of gastric cancer (GC) and assessed whether anti-IL-6 receptor antibody (anti-IL-6R Ab) could inhibit PM of GC. We conducted immunohistochemical analysis of IL-6 and α-smooth muscle (α-SMA) expressions in clinical samples of GC and PM, and investigated the interactions between CAFs and GC cells in vitro. Anti-tumor effects of an

Type I interferons (IFNs-I), a group of pleiotropic cytokines, critically modulate host response in various inflammatory diseases. However, the role of the IFN-I pathway in periodontitis remains largely unknown. In this report, we describe that the IFN-β levels in the gingival crevicular fluid of human subjects were negatively associated with periodontitis and clinical gingival inflammation. Disruption of IFN-I signaling worsened alveolar bone resorption in a ligature-induced periodontitis murine model. Deficiency of the IFN-I pathway resulted in an exaggerated inflammatory response in myeloid

BACKGROUND & AIMS: Alcohol-associated liver fibrosis (ALF) is a key, potentially reversible stage leading to alcohol-associated liver cirrhosis, but effective treatments are lacking. This study explored whether interleukin (IL)-22, a hepatocyte survival factor, plays an anti-fibrotic role in ALF by modulating the Nrf2-ARE antioxidant pathway.
METHODS: IL-22 and Nrf2-ARE inhibitor, ML385, were administered to rat hepatic stellate cells (HSCs) exposed to acetaldehyde. Cell proliferation, cell cycle distribution, and Nrf2-ARE activation were investigated. An ALF mouse model was used to evaluate

Surgical myocardial revascularization, regardless of the technique used, causes ischemia-reperfusion injury (IRI) in the myocardium mediated by inflammation and degradation of the endothelial glycocalyx (EG). We investigated the difference between on-pump and off-pump techniques in terms of the concentration of proinflammatory interleukin (IL)-18 and the EG degradation products syndecan-1 and hyaluronic acid measured by ELISA in the peripheral and cardiac circulation during open heart surgery and in the early postoperative period. The concentration of IL-18, C-reactive protein (CRP), and cardi

Osteoporosis is an inflammatory disease characterised by low bone mass and quality, resulting in weaker bone strength and fragility fractures. Periostin is a matricellular protein expressed in the periosteum of bone by osteoblasts. It regulates cell recruitment and differentiation in response to fracture and contributes to extracellular matrix (ECM) formation. The aim of the following study was to determine the splice variants of Periostin expressed in human osteoblasts and Periostin's function in the pathophysiology of osteoporosis. Osteoblasts isolated from femoral heads from 29 patients wit

Iron is one of the indispensable trace elements in living organisms. However, excessive iron deposition in organisms is prone to induce dysfunction of the liver and other vital organs. The present study aimed to investigate the mechanism how aquatic high iron affects iron transport and induces hepatic injury in zebrafish. Our results showed that the iron levels in zebrafish liver and serum were significantly increased after the fish treated with aquatic high iron (200 mg/L ferric ammonium citrate, FAC) for 21 days. Meanwhile, hepatic fibrosis was observed in zebrafish with high iron treatment.

Propofol is widely used in anesthesia, but its role in breast cancer progression remains controversial. This study investigated the molecular mechanisms of propofol in breast cancer, focusing on IL-6 and tumor microenvironment modulation. Bioinformatics analysis identified IL-6 as a potential target of propofol. MCF-7 cells were treated with varying propofol concentrations (0-100 μg/mL), and cell viability was assessed via CCK-8 assay. Propofol at 50 μg/mL significantly reduced viability, while 25 μg/mL (a non-cytotoxic dose) was selected for further experiments. Western blot confirmed propofo

BACKGROUND: Interleukin-6 plays an essential role in cytokine storm and cytokine release syndrome, which occur in response to pathogen infection or tissue injury and are associated with severe symptoms. Neutralizing IL-6 can help reduce symptom severity. Chicken eggs serve as an excellent alternative antibody source compared to mammalian serum. The immunoglobulin Y (IgY) in the chicken's blood is transferred to and deposited within the egg yolk in large amounts. Several IgY products have been developed for therapeutic applications in various diseases. This study focuses on producing anti-human

Negative energy balance (NEB) during early lactation links spontaneous lipolysis (SL) with inflammatory signaling, yet the molecular response in dual-purpose breeds remains insufficiently characterized. This study investigated how NEB regulates circulating concentrations of interleukin-6 (IL-6) and plasminogen (PL) in Simmental cows, contextualizing these changes within concurrent metabolic adaptation. Forty-two cows were monitored from approximately two weeks prepartum to 150 days in milk across six defined stages. Energy balance (EB) was calculated from feed intake and energy-corrected milk
