C1QBP Promotes Prostate Cancer Progression and Lipid Accumulation by Negatively Regulating ALDH9A1.

Prostate cancer (PCa) relies heavily on lipid metabolism for energy acquisition, and lipid metabolic reprogramming plays a crucial role in its progression. Here, we utilized publicly available PCa databases and immunohistochemistry to evaluate C1QBP expression in PCa. We found that C1QBP is highly expressed in PCa, potentially due to promoter hypomethylation. Functional assays showed that C1QBP promotes cell proliferation, migration, and lipid accumulation in PCa cells. We identified differentially expressed proteins associated with C1QBP by using liquid chromatography-tandem mass spectrometry

Hydrophilicity and topology interplay determines positioning of guest molecules in lipid cubic phases.

Lipid nanostructures with inverse bicontinuous cubic symmetries are of paramount importance as delivery structures of active compounds in the pharmaceutical, cosmetic and food science fields. By atomistic molecular dynamics, here we study the internalization of three molecules of varying hydrophilicity, fructose, caffeine and vitamin D, within a cubic phase with primitive symmetry, allowing us to assess how the incorporation of the guest molecule is affected by the interplay between its hydrophilicity and the topology of the host membrane. For lipophilic molecules our results reveal the detail

Hypoxia induced lipid droplet accumulation promotes resistance to ferroptosis in prostate cancer.

Ferroptosis is a mode of cell death that relies on iron metabolism and lipid peroxidation. Preclinical and clinical studies indicate that fe … ly reduced the expression of genes related to incorporating polyunsaturated fatty acids into phospholipids (ACSL4, LPCAT3), and parallel lipidomic analysis demonstrated that hypoxia significantly decreased the levels of the ferroptosis-prone lipid class, phosphatidylethanolamine (PE) and increased production of neutral lipid species, cholesteryl ester (C …

Lipid mesophases-nanoconfined water rules crystal symmetry during in-meso crystallization.

Polymorphism plays a critical role in the pharmaceutical industry, as it directly influences the solubility, stability, and bioavailability of drug compounds. In this work, nanoconfinement via cubic lipid mesophases (Ia3¯d and Pn3¯m geometries) has been used to study the polymorphism of the model drugs theophylline, paracetamol and caffeine. By exploiting the interactions of the drug with nanoconfined water and lipid head groups, this approach drives the nucleation towards polymorphs that are otherwise not stable at ambient conditions. In the lipid mesophases, theophylline crystallizes as meta

Targeting Neuroinflammation in Central Nervous System Diseases by Oral Delivery of Lipid Nanoparticles.

Neuroinflammation within the central nervous system (CNS) is a primary characteristic of CNS diseases, such as Parkinson's disease, Alzheimer's disease (AD), amyotrophic lateral sclerosis, and mental disorders. The excessive activation of immune cells results in the massive release of pro-inflammatory cytokines, which subsequently induce neuronal death and accelerate the progression of neurodegeneration. Therefore, mitigating excessive neuroinflammation has emerged as a promising strategy for the treatment of CNS diseases. Despite advancements in drug discovery and the development of novel the

PBAE-PEG-based lipid nanoparticles for lung cell-specific gene delivery.

Exemplified by successful use in COVID-19 vaccination, delivery of modified mRNA encapsulated in lipid nanoparticles (LNPs) provides a framework for treating various genetic and acquired disorders. However, LNPs that can deliver mRNA into specific lung cell types have not yet been established. Here, we sought to determine whether poly(β-amino ester)s (PBAE) or PEGylated PBAE (PBAE-PEG) in combination with 4A3-SC8/DOPE/cholesterol/DOTAP LNPs could deliver mRNA into different types of lung cells in vivo. PBAE-PEG/LNP was similar to Lipofectamine MessengerMAX followed by PBAE/LNP for mRNA transfe

Phagophore Ceramide Promotes LC3 Lipidation and Autophagy in T-Lymphoid Cell Death.

The bioactive sphingolipid ceramide induces various types of cell death, including autophagy-dependent cell death (ADCD). However, its role in autophagy and ADCD is not fully understood. Here, we demonstrated that amino acid deprivation (AA[-]) in murine lymphoid WR19L cells promoted the accumulation of ceramide at phagophores and autophagy, resulting in ADCD. Control of ceramide production by sphingomyelin synthase 1 and neutral sphingomyelinase 2 altered cell fate by regulating AA(-)-induced autophagy and ADCD. The Regulatory Effects of Pea Protein Hydrolysate on Lipid Metabolism in Gestational Diabetic Mice.

Gestational diabetes mellitus (GDM) poses significant risks to maternal health and adverse metabolic outcomes in offspring. Pea protein hydrolysate (PPH), a kind of bioactive peptide with anti-diabetic properties, was evaluated for its therapeutic potential in the GDM mouse model. While PPH exhibited mild glucose-lowering effects, high-dose PPH (1000 mg/kg/day) significantly ameliorated insulin resistance and systemic dyslipidemia, including reductions in serum triglycerides, total cholesterol, low-density lipoprotein cholesterol, and hepatic lipid accumulation. In addition, the PPH treatment

Coenzyme Q10 deficiency disrupts lipid metabolism by altering cholesterol homeostasis in neurons.

Coenzyme Q10 (CoQ10) is a critical component of the mitochondrial respiratory chain. CoQ10 deficiencies cause a variety of clinical syndrome … that CoQ10 deficiency causes alterations in cholesterol homeostasis, fatty acids oxidation, phospholipids and sphingolipids synthesis in neurons. These alterations depend on the molecular defect, and on the residual CoQ10 levels. Our results imply that CoQ10 deficiencies can induce pathology by altering lipid homeostasis and the composition of cellular …

Sperm activation for fertilization requires robust activity of the TAT-5 lipid flippase.

During fertilization, sperm and egg membranes signal and fuse to form a zygote and begin embryonic development. Here, we investigated the role of lipid asymmetry in gametogenesis, fertilization, and embryogenesis. We find that phosphatidylethanolamine asymmetry is lost during meiosis prior to phosphatidylserine exposure. We show that TAT-5, the P4-ATPase that maintains phosphatidylethanolamine asymmetry, is required for both oocyte formation and sperm activation, albeit at different levels of flippase activity. Loss of TAT-5 significantly decreases fertility in both males and hermaphrodites an
