Cord blood DNA methylation as a potential mediator in the association between prenatal ambient air pollution and early infant allergic symptoms in a Shanghai birth cohort.

BACKGROUND: Prenatal exposure to ambient air pollution has been associated with early-life allergic outcomes; however, underlying biological mechanisms remain incompletely understood. METHODS: A total of 101 mother-infant pairs were ultimately included in this prospective birth cohort study. Maternal exposure to ambient air pollutants was assigned to geocoded residential addresses using spatiotemporal models. Allergic symptoms were defined as cumulative caregiver-reported symptoms occurring from birth to 6 months and assessed at the 6-month follow-up. DNA methylation in naïve CD4+ T cells was…

Gestational exposure to organophosphate ester flame retardants and child growth in weight, height, and body mass index at age 2-10 years: the Environmental influences on Child Health Outcomes Program.

BACKGROUND: Pregnant women are widely exposed to organophosphate esters (OPEs). Few studies have examined how gestational OPE exposures may influence child growth rates, which are important predictors of subsequent cardiometabolic health. METHODS: Among 4566 mother-child dyads from 14 sites within the Environmental influences on Child Health Outcomes (ECHO) Cohort, we evaluated associations of gestational urinary concentrations of nine OPE metabolites with early and mid-childhood rates of change in age- and sex-standardized weight (WAZ), height (HAZ), and body mass index (BMIZ). Using linear

Prenatal exposure to persistent organic pollutants and body mass index trajectories from birth to age 13.

Prenatal exposure to persistent organic pollutants (POPs) has been linked to growth and adiposity development in children. However, only few studies considered the growth dynamic and none have tracked children into adolescence. This study included 450 mother-child pairs from the PELAGIE cohort (France). Polychlorinated biphenyls (PCBs), Organochlorine pesticides (OCs), Polybrominated diphenyl ethers (PBDEs), and Perfluoroalkyl substances (PFAS) were measured in cord blood. A latent class growth model identified four types of BMI z-score (zBMI) trajectories from birth to 13 years. Associations

Prenatal exposure to persistent organic pollutants and sex-specific neonatal outcomes in the ENVIRONAGE birth cohort.

Early-life exposure to environmental contaminants, such as endocrine disrupting persistent organic pollutants (POPs), including polychlorinated biphenyls (PCBs), polybrominated diphenyl ethers (PBDEs) and organochlorine pesticides (OCPs), is linked to adverse neonatal outcomes. However, the sex-specific effects of POP mixtures and the potential mediating roles of biological pathways, such as inflammation, remain insufficiently understood. This study aimed to investigate these aspects within the ENVIRONAGE birth cohort. The study population consisted of newborns (n = 402) from the ENVIRONAGE bi

Prenatal and Postnatal Cigarette Smoke Exposure Is Associated With Increased Risk of Exacerbated Allergic Airway Immune Responses: A Preclinical Mouse Model.

Increased exposure to household air pollution and ambient air pollution has become one of the world's major environmental health threats. In developing and developed countries, environmental cigarette smoke (CS) exposure is one of the main sources of household air pollution (HAP). Moreover, results from different epidemiological and experimental studies indicate that there is a strong association between HAP, specifically CS exposure, and the development of allergic diseases that often persists into later life. Here, we investigated the impact of prenatal and postnatal CS exposure on offspring

Global Perspectives on Immunization During Pregnancy and Priorities for Future Research and Development: An International Consensus Statement.

Immunization during pregnancy has been recommended in an increasing number of countries. The aim of this strategy is to protect pregnant women and infants from severe infectious disease, morbidity and mortality and is currently limited to tetanus, inactivated influenza, and pertussis-containing vaccines. There have been recent advancements in the development of vaccines designed primarily for use in pregnant women (respiratory syncytial virus and group B Streptococcus vaccines). Although there is increasing evidence to support vaccination in pregnancy, important gaps in knowledge still exist a

Associations between PM2.5 and risk of preterm birth among liveborn infants.

PURPOSE: Studies suggest exposure to ambient particulate matter less than 2.5 μg/m3 in aerodynamic diameter (PM2.5) may be associated with preterm birth (PTB), but few have evaluated how this is modified by ambient temperature. We investigated the relationship between PM2.5 exposure during pregnancy and PTB in infants without birth defects (1999-2006) and enrolled in the National Birth Defects Prevention Study and how it is modified by concurrent temperature. METHODS: PTB was defined as spontaneous or iatrogenic delivery before 37 weeks. Exposure was assigned using inverse distance weighting

Risks associated with in utero and lactation exposure to selective serotonin reuptake inhibitors (SSRIs).

BACKGROUND: Prenatal exposure to selective serotonin reuptake inhibitors (SSRIs) may increase risk for congenital malformations and adverse perinatal outcome. OBJECTIVE: This article reviews the published literature on exposure to SSRIs in utero and during lactation. METHODS: Literature search in PubMed. RESULTS: There is no conclusive evidence for increased risk for malformations but paroxetine and possibly fluoxetine use in early pregnancy may be associated with a small increased risk for cardiovascular malformations. Perinatal adverse effects, including respiratory distress and neonatal

Smoking during pregnancy and bulimia nervosa in offspring.

Because smoking during pregnancy is implicated in influencing appetite and impulse control in offspring, the aim of this study was to establish if it is associated with bulimia nervosa in offspring. Bulimia was identified at age 30 years among 4046 females, born 5-11 April, 1970. After adjustment for potential confounding factors including body mass index (BMI) and maternal psychiatric morbidity, smoking during pregnancy was associated with bulimia in offspring by age 30 years. Compared with non-smoking mothers, the adjusted odds ratios (95% confidence intervals) for bulimia in offspring were

A neuroprotective peptide antagonizes fetal alcohol exposure-compromised brain growth.

We evaluated a 9-amino-acid peptide, SALLRSIPA (SAL), an agonist of activity-dependent neurotrophic factor (ADNF), for its protective properties against fetal alcohol-related brain growth retardation, using an established liquid diet model of alcohol-related neurodevelopmental disorder (ARND) in C57BL/6 mice. Alcohol exposure during neurulation reduced body weight, head size, and specifically brain weight and volume. Major gross brain deficits include underdevelopment of brain areas, cortical thinning, ventricle enlargement, and restricted midline neural tissue growth leading to openings at th

Immunologic effects of background prenatal and postnatal exposure to dioxins and polychlorinated biphenyls in Dutch infants.

Immunologic effects of pre- and postnatal polychlorinated biphenyl (PCB)/dioxin exposure in Dutch infants from birth to 18 mo of age are explored. The total study group consisted of 207 healthy mother-infant pairs, of which 105 infants were breast-fed and 102 children were bottle-fed. Prenatal PCB exposure was estimated by the PCB sum (PCB congeners 118, 138, 153, and 180) in maternal blood and the total toxic equivalent (TEQ) level in human milk (17 dioxin and 8 dioxin-like PCB congeners). Postnatal PCB/dioxin exposure was calculated as a product of the total TEQ level in human milk multiplie

Abdominopelvic computed tomography during pregnancy and the risk of congenital malformations: protocol for a nationwide population-based cohort study in South Korea.

INTRODUCTION: The use of CT has increased markedly over recent decades, and this trend is also observed among pregnant women. According to current clinical guidelines, fetal radiation exposure below a threshold of 100 to 200 mGy does not appear to increase the risk of congenital malformations. Although the estimated fetal dose from a maternal abdominopelvic CT examination is generally lower than this range, evidence regarding the risks associated with such relatively low-dose diagnostic exposures during pregnancy remains scarce. This study aims to evaluate the association between abdominopelvi

The association between in-utero exposure to stressful life events during pregnancy and male reproductive function in a cohort of 20-year-old offspring: The Raine Study.

STUDY QUESTION: Is exposure to gestational stress in the critical time window for the normal differentiation and growth of male reproductive tissue associated with male reproductive function in offspring in later life? SUMMARY ANSWER: Exposure to stressful life events (SLEs) in early, but not late gestation, are associated with reduced adult male reproductive function, consistent with the hypothesis that events during early prenatal life programme adult male reproductive function. WHAT IS ALREADY KNOWN: Animal studies suggest that gestational stress may impact on the reproductive function of

Early Life Origins of Neurodevelopmental Disabilities in the Boston Birth Cohort: Research findings and future directions.

Neurodevelopmental disabilities (NDD) are a group of conditions with onset in early development period and is characterized by limitations in several functional domains. Autism spectrum disorder (ASD) and Attention-Deficit Hyperactivity Disorders (ADHD), the most common NDDs, have complex etiologies and possibly multiple pathways leading up to the manifestation of these disorders. Boston Birth Cohort (BBC) is a preterm enriched birth cohort, and over the years, researchers have used the BBC dataset to study a broad spectrum of early life protective and risk factors in the context of NDDs. Broa

Fetally-encoded GDF15 and maternal GDF15 sensitivity are major determinants of nausea and vomiting in human pregnancy.

Human pregnancy is frequently accompanied by nausea and vomiting that may become severe and life-threatening, as in hyperemesis gravidarum (HG), the cause of which is unknown. Growth Differentiation Factor-15 (GDF15), a hormone known to act on the hindbrain to cause emesis, is highly expressed in the placenta and its levels in maternal blood rise rapidly in pregnancy. Variants in the maternal GDF15 gene are associated with HG. Here we report that fetal production of GDF15, and maternal sensitivity to it, both contribute substantially to the risk of HG. We found that the great majority of GDF15

Using a two-sample Mendelian randomization design to investigate a possible causal effect of maternal lipid concentrations on offspring birth weight.

BACKGROUND: The intrauterine environment is critical for fetal growth and development. However, observational associations between maternal gestational lipid concentrations and offspring birth weight (BW) have been inconsistent and ascertaining causality is challenging. METHODS: We used a novel two-sample Mendelian randomization (MR) approach to estimate the causal effect of maternal gestational high-density lipoprotein cholesterol (HDL-C), low-density lipoprotein cholesterol (LDL-C) and triglyceride concentrations on offspring BW. Single nucleotide polymorphisms (SNPs) associated with serum

Maternal infections during pregnancy and cerebral palsy: a population-based cohort study.

BACKGROUND: Cerebral palsy (CP) is a common motor disability in childhood. We examined the association between maternal infections during pregnancy and the risk of congenital CP in the child. METHODS: Liveborn singletons in Denmark between 1997 and 2003 were identified from the Danish National Birth Registry and followed from 1 year of life until 2008. Redemption of antibiotics from the National Register of Medicinal Product Statistics and maternal infections reported by the National Hospital Register were used as markers of maternal infection during pregnancy. CP diagnoses were obtained from
