The effect of anticonvulsant drugs on daytime and nighttime N-acetyltransferase (NAT) and hydroxyindole-O-methyltransferase (HIOMT) levels was investigated. Some of the drugs caused a significant reduction in daytime NAT and HIOMT levels, possibly owing to beta-receptor blockade although other mechanisms cannot be excluded. Sulthiame caused an increase in daytime NAT levels, and this may represent a compensatory mechanism by the pineal gland to allow normal melatonin production. None of the drugs affected nocturnal HIOMT levels, and only phenobarbitone affected NAT levels, apparently causing a…

Sleep disorders are highly prevalent and significantly impair physical, cognitive, and psychological functioning. Conventional management strategies such as pharmacotherapy and cognitive behavioral therapy for insomnia are effective for many individuals, but may be limited by side effects, accessibility issues, adherence challenges, and patient preference for non-pharmacological options. Consequently, complementary and alternative medicine approaches have gained increasing attention as potential adjuncts or alternatives for managing sleep disturbances. This review examines the role of alternat

Tryptophan is an essential amino acid for the human body and is crucial for protein synthesis. It is primarily metabolized through the kynurenine, 5-hydroxytryptamine, and indole pathways. The various bioactive compounds produced through tryptophan metabolism can regulate a range of physiological functions, including digestive, metabolic, immune responses, and neurological functions. New evidence supports a close relationship between tryptophan metabolism and embryo implantation; the levels or ratios of tryptophan metabolites, such as kynurenine, serotonin, and melatonin, significantly correla…

PURPOSE OF THE REVIEW: This narrative review examines the newest findings from clinical trials of chronotherapeutics for psychiatric disorders. We reviewed the potential effects of the most commonly-studied chronotherapeutics such as bright light therapy, dark therapy, melatonin, and chronotherapy on the psychiatric disorders of depression, bipolar disorder, and anxiety disorders.
RECENT FINDINGS: The preponderance of recent clinical trials in chronotherapeutics has focused on bright light therapy in depression. However, there is an emerging body of preliminary studies testing chronotherapeut…

Management of rapid eye movement sleep behavior disorder (RBD) includes reducing injurious dream-enactment behaviors, risk of injury to self and bedpartner, and vivid or disruptive dreams and improving sleep quality and bedpartner sleep disruption. Safety precautions should be reviewed at each visit. Medications to reduce RBD symptoms such as melatonin, clonazepam, pramipexole, and rivastigmine should be considered for most patients. Isolated RBD confers a high lifetime risk of neurodegenerative diseases with a latency often spanning many years. A patient-centered shared decision-making approa…

Experimental and interventional studies show that light can regulate sleep timing and sleepiness while awake by setting the phase of circadian rhythms and supporting alertness. The extent to which differences in light exposure explain variations in sleep and sleepiness within and between individuals in everyday life remains less clear. Here, we establish a method to address this deficit, incorporating an open-source wearable wrist-worn light logger (SpectraWear) and smartphone-based online data collection. We use it to simultaneously record longitudinal light exposure (in melanopic equivalent

The use of light-emitting diodes (LEDs) has increased considerably in the 21st century with humans living in a modern photoperiod with brighter nights and dimmer days. Prolonged exposure to LEDs, especially at night, is considered a new source of pollution because it may affect the synthesis and secretion of retinal melatonin and dopamine, resulting in negative impacts on retinal circadian clocks and potentially disrupting retinal circadian rhythms. The control of ocular refraction is believed to be related to retinal circadian rhythms. Moreover, the global prevalence of myopia has increased a…

BACKGROUND: The internal clock is driven by circadian genes [e.g., Clock, Bmal1, Per1-3, Cry1-2], hormones [e.g., melatonin, cortisol], as well as zeitgeber ['synchronisers']. Chronic disturbances in the circadian rhythm in Objectives: The aim of this review is to summarise the current knowledge and literature regarding circadian rhythms in the context of mood disorders, focussing on the role of circadian genes, hormones, and neurotransmitters.
METHODS: The review presents the current knowledge and literature regarding circadian rhythms in mood disorders using the Pubmed database. Articles wi…

2-Hydroxymelatonin (2-OHMT) is an important metabolite produced through melatonin interaction with oxygenated compounds. 2-OHMT pretreated seeds (50 µM, 100 µM, and 150 µM) were grown in soil contaminated with 50 mg kg-1 cadmium. Cadmium imposed stress reduced seed germination, growth, biomass production, and chlorophyll (Chl) content in Cucumis sativus seedlings. 2-OHMT application emphatically revamped germination, shoot length, root length, and plant biomass production. The 2-OHMT pretreatment modulated expression levels of plasma membrane H+-ATPase genes of C. sativus including CsHA2, CsHA…

Pineal gland phylogenic antiquity, its involvement in the stress reaction and many-sided activity of its compounds permit to consider the gland to be the organ taking an active part in the body protection against injury, which is essential for everyone since its appearance on the planet. The available data prove that almost all the reactions developing in the organism from the moment of trauma up to the complete wound reparation are under the pineal control. Being always an urgent one the problem of traumatism increased nowadays because of pineal dysfunction. It makes the premises to apply pin

In the aging male, endocrine changes and decline in endocrine function involve tissue responsiveness as well as reduced secretory output from peripheral glands and alterations in the central mechanism controlling the temporal organization of hormonal release. The latter are likely to be responsible for the dampened circadian hormonal and non-hormonal rhythms. These are in part responsible of the age dependent decrease of the peripheral levels of T, DHEA, the thyroid hormones, GH, IGF-I, and melatonin. These hormonal changes which develop in most men at about the age of 50 are in part responsib…

The ultrastructure of the pineal gland of 32 sheep embryos was studied from day 54 of development through birth. Embryos were arranged in four age-groups, defined in terms of the most relevant histological features: group 1 (54 to 67 days of prenatal development), group 2 (71 to 92 days), group 3 (98 to 113 days), and group 4 (118 to 150 days). A primary cell type, designated the pinealoblast, was observed from 54 days until birth; ultrastructurally, this cell was found to contain all the organelles required for hormone synthesis. A second cell population, classified as interstitial cells by v

In recent years some people expressed concern about possible health effects of electric and magnetic fields produced by exposure to extremely low frequency bands (ELF: less than 300 Hz). This was first mentioned by epidemiological studies. However, the epidemiological method has several limitations that are difficult to overcome. One problem is the detection of relationships when the risk ratios are relatively low. Another obstacle may be how to define 'strength' of magnetic or electric fields. In animal studies, there have been published several papers that melatonin was depressed by exposure…
