Congenital non-parasitic hepatic cyst complicated by hepatoduodenal fistula: a rare case report.

INTRODUCTION AND IMPORTANCE: Simple hepatic cysts are benign and usually asymptomatic; however, they can rarely develop serious complications, such as hepatoduodenal fistula, due to infection, cyst size, or mechanical pressure. CASE PRESENTATION: A 68-year-old male presented with fever, hematochezia, and severe right upper quadrant pain. Imaging revealed a 10 × 12 cm segment V-VII hepatic cyst with an air-fluid level, and endoscopy showed a fistulous opening in the second duodenum adjacent to a necrotic diverticulum. After stabilization, laparotomy confirmed a hepatoduodenal fistula; a 2 cm o

Association of Parasite Biomass with Disease Severity in Plasmodium vivax and Plasmodium falciparum Malaria Infections.

The clinical and parasitological factors underlying severe malaria pathogenesis remain incompletely understood, particularly in Plasmodium vivax (P. vivax). To address this issue, clinical and parasitological metrics, including circulating peripheral parasitemia and the parasite biomass (the total number of parasites in the body consisting of both circulating peripheral and noncirculating parasites), were measured in patients with severe and uncomplicated monoinfections of P. vivax and Plasmodium falciparum (P. falciparum) malaria who presented at a hospital in southwest India. Peripheral para…

Evolutionary Rate Covariation Across Malaria Parasite Species Enables Inference of Protein Interactions.

Despite publication of the Plasmodium falciparum reference genome over 20 years ago, one-third of its genes remain functionally unannotated, and information is limited for many others. Proteins that act in the same pathway or complex tend to experience similar shifts in evolutionary pressure, such that correlated constraints across species can indicate co-functional proteins. To investigate this connection, we calculated relative evolutionary rates across the genome on a phylogeny of 22 Plasmodium species and assigned each protein-protein pair a score representing the strength of evolutionary

Temperature and Rainfall Associate With Patterns of Parasitism in Free-Ranging Capuchin Monkeys (Cebus imitator).

Ecological variables such as climate and habitat composition can impact the infection patterns of gastrointestinal parasites in animal host populations by affecting host behavior and parasite survivability and distribution in the external environment. Data from wild primate populations provide insights into these processes. To contribute new insights from a well-characterized population, we collected 200 fecal samples from five groups of Costa Rican capuchin monkeys from August 2020 to August 2021 and screened them for gastrointestinal parasites. We estimated annual home range sizes for the gr…

Effect of Cestodes Parasitizing the Small Intestine on Host Protease Activities in Herring Gull Chicks.

Total protease activity and activities of protease subclasses in the small intestine were assayed in herring gull (Larus argentatus (Pontoppidan, 1763)) chicks infested by the cestodes Tetrabothrius erostris (Loennberg, 1889) (Cestoda: Tetrabothriidae), Microsomacanthus ductilus (Linton, 1927) (Cestoda: Hymenolepididae), Wardium cirrosa (Krabbe, 1869) (Cestoda: Aploparaksidae), Arctotaenia tetrabothrioides (Loennberg, 1890), and Alcataenia larina (Krabbe, 1869) (Cestoda: Dilepididae). Serine protease activity was found to decrease in small intestine sites with the highest cestode abundance (M.

Genomic and Transcriptomic Insights into the Evolution and Parasitic Strategy of the Woody-Plant Nematode Pratylenchus vulnus.

The root-lesion nematode Pratylenchus vulnus parasitizes a wide range of hosts, including woody perennials such as walnut (Juglans regia) and grapevine (Vitis vinifera), significantly damaging roots and reducing yields. Here, we present a high-quality, chromosome-level genome assembly of P. vulnus (61.7 Mb across six chromosomes). Comparative genomic analysis revealed high collinearity in protein-coding genes between P. vulnus and the root-knot nematode included in this analysis, Meloidogyne graminicola, indicating a close evolutionary relationship between these genera of plant parasites. Larg…

Artificial Intelligence-Based Tools for Automated Genus-Level Identification of Plant-Parasitic Nematodes.

Early and accurate identification and quantification of plant-parasitic nematodes (PPNs) is crucial for their effective control. Although valuable, the current techniques for identifying PPNs, such as morphology and molecular marker-based methods, can be time- and resource-intensive. This study aimed to develop and validate cutting-edge computer vision tools for automated, accurate, and reproducible PPN detection. To achieve this goal, we captured microscopic images of the three economically important PPN genera associated with potato crop: root-lesion nematode (RLN; Pratylenchus spp.), root-k

mRNA vaccination overcomes haemozoin-mediated impairment of whole-parasite malaria vaccines in mice.

Immunization with radiation-attenuated sporozoites (RAS) drives effective sterilizing immunity against liver-stage Plasmodium infection. However, protection is compromised in individuals living in malaria endemic regions and the mechanisms of vaccine failure are unclear. Here we show that previous blood-stage exposure in a mouse model of Plasmodium yoelii infection compromises Plasmodium berghei RAS-induced essential CD8+ T cell responses and subsequent protection. The persisting malarial pigment haemozoin mediates impaired CD8+ T cell responses owing to impaired antigen uptake by dendritic ce

An Unexpected Intraoperative Discovery: A Parasitic Leiomyoma in a Perimenopausal Patient With Complex Abnormal Uterine Bleeding.

Parasitic leiomyomas are an uncommon subtype of extra-uterine smooth muscle tumors characterized by complete detachment from the uterus and subsequent development of an autonomous vascular supply. Their atypical anatomical locations, independent blood flow, and frequent coexistence with large fibroid uteri make preoperative diagnosis difficult. Abnormal uterine bleeding (AUB) is one of the leading indications for hysterectomy worldwide, and leiomyomas remain the most common structural cause within the International Federation of Gynecology and Obstetrics (FIGO) PALM-COEIN classification. Accuracy of Plasmodium falciparum genetic data for estimating parasite prevalence and malaria incidence in Uganda.

BACKGROUND: Genetic metrics derived from Plasmodium falciparum infections offer a potential complement to conventional malaria surveillance by utilizing features of parasite diversity and relatedness to estimate transmission intensity. However, the performance of genetic metrics to predict epidemiologic metrics across a wide range of transmission settings remains understudied. METHODS: Dried blood spots from 3563 symptomatic malaria cases were collected from 26 sentinel health facilities across Uganda during two collections in 2023. Amplicon deep sequencing of 165 polyallelic microhaplotypes

Host iron deficiency protects against Plasmodium infection and drives parasite molecular reprofiling.

Iron deficiency, anemia, and Plasmodium infection are global health challenges with overlapping geographical distributions, particularly aff … chelation substantially changed the transcriptomic and proteomic profile of cultured P. falciparum parasites. Intravenous iron supplementation did not increase parasitemia when coupled with malaria prevention. These findings demonstrate that iron deficiency protects against Plasmodium infection and support World Health Organization recommendations for iron …

The Trichinella Super-Pangenome Reveals the Evolution of Encapsulation and Predicted Host-Parasite Protein Interactions.

The muscle capsule of Trichinella is a critical structure that impedes immune attacks and drug penetration, yet the molecular mechanisms underlying its formation remain poorly understood. Using a high-quality super-pangenome comprising 12 Trichinella species, we compared extensive genomic variations between encapsulating and non-encapsulating lineages. Pangenomic analysis revealed an open-genome architecture dominated by dispensable genes, with nonencapsulated lineages exhibiting a high load of structural variation (SV). These findings suggest that the capsule represents an evolutionary innova

Oxidative Stress and Redox Imbalance in Acanthamoeba Keratitis: Host-Parasite Crosstalk and Therapeutic Perspectives.

Acanthamoeba keratitis is a severe, sight-threatening corneal infection caused by free-living amoebae widely distributed in soil, natural waters, and domestic or recreational water systems. Following entry into the corneal microenvironment, disease develops within a redox conflict in which host-derived reactive oxygen species (ROS) and reactive nitrogen species (RNS) may contribute to parasite control while simultaneously promoting inflammation and tissue injury. Acanthamoeba responds through an adaptable defense network involving superoxide dismutases, catalase, thioredoxin- and glutathione-d
