Use of a job-exposure matrix to assess occupational exposures in relation to birth defects.

Accurate exposure assessment remains a challenge in occupational epidemiology. We evaluated one approach, use of a job-exposure matrix (JEM), by applying the National Institute for Occupational Safety and Health (NIOSH) JEM to a large case-control birth defects study that included parental occupation information. We investigated the JEM exposure predictions in several ways and found that for a substantial proportion of the parents in the birth defects study, the JEM yielded either no exposure data or nonsense predictions. Among exposure predictions that were plausible, most were of low probabi

Fathers of children on welfare: their impact on child well-being.

Although it is generally assumed that fathers are absent from the lives of children being raised on AFDC, evidence from a case-control study in Worcester, Massachusetts, suggests that there is considerable father-child contact. In addition, multivariate modeling indicates that contact with fathers had a modest beneficial effect on children's behavior. Based on these study findings, negative traits of fathers (e.g., substance abuse, physical violence) appear to be associated with increased child behavior problems.

Parental exposure to 2,2',4,4'5 - pentain polybrominated diphenyl ethers (BDE-99) causes thyroid disruption and developmental toxicity in zebrafish.

Although polybrominated diphenyl ethers (PBDEs) are known to disturb thyroid hormone signaling, the mechanisms underlying the effects of 2,2',4,4'5 - pentain polybrominated diphenyl ethers (BDE-99) in fish remain unclear. In order to reveal these mechanisms, adult zebrafish (Danio rerio) were exposed to different concentrations of BDE-99 (0, 0.5, 5, or 50 μg/L) for 28 days and spawned by mating naturally in clean water (without BDE-99). Females exposed to BDE-99 showed significantly lowered thyroxine (T4) levels. Expression of transthyretin (ttr) and uridine diphosphate glucuronosyl transferas

Pregnancy in Patients With Chronic Myeloid Leukemia.

Estimates suggest that nearly 30% of patients diagnosed with chronic myeloid leukemia (CML) are aged <49 years, with approximately half being women. For many of these women, childbearing concerns are a major factor as they initiate treatment with tyrosine kinase inhibitors, which are known to be teratogenic. During her presentation at the NCCN 23rd Annual Conference, Dr. Berman identified the challenges in helping women undergoing treatment for CML who want to have children, and emphasized the importance of an individualized and multidisciplinary approach to management. In addition, she encour

Preconceptional and perinatal exposure to traffic-related air pollution and eczema in preschool children.

BACKGROUND: Evidence linking prenatal exposure to outdoor air pollution with eczema in early childhood is scare, and the role of components of air pollution and exposure timing remains unclear. OBJECTIVES: We investigated the association between exposure to air pollution during preconceptional and perinatal period and the risk of eczema in preschool children. METHODS: We conducted a prospective cohort study of 2598 children aged 3-6 years in Changsha, China. The prevalence of eczema was assessed by a standardized health questionnaire administered by the parents. Individual exposures to nitro

Father absence and adolescent development: a review of the literature.

Rapid social change has seen increasing numbers of woman-headed single-parent families, meaning that more and more children are growing up without a father resident in the home. Father absence is a term that is not well defined and much of the literature does not discriminate between father absence due to death, parental relationship discord or other causes. This article presents a critical review of the extant literature on father absence, particularly as it relates to adolescent well-being and development. Findings from the literature point to the importance of father presence in children's

Including the fathers' perspective in holistic care. Part 2: Findings on the fathers' hospital experience including restraining the child patient for treatment.

Part 2 of this discussion provides a detailed examination of specific issues associated with the fathers' personal involvement in their child's hospital treatment for paediatric acute lymphoblastic leukaemia (ALL). In particular, fathers testify to the difficulties they encounter when involved with the restraining of their child-patient for treatment. Issues raised by parents during this study have been addressed by recommendations detailing factors which need to be considered when providing holistic care for the family and the child with leukaemia.

Stepwise pairing and fast transition regulated by recombination drive meiotic chromosome interactions.

Maternal and paternal homologous chromosomes recognize each other and pair during meiosis to ensure accurate chromosome segregation. However, the mechanisms that drive chromosome pairing and their regulation remain poorly understood. To investigate this, we develop a fluorescence reporter-operator system based on the lac operator-lac repressor system, enabling real-time observation of specific genomic loci in live mouse seminiferous tubule tissues. Using 3D time-lapse imaging, we visualize and quantify telomere-led rapid chromosome movements and homologous chromosome pairing. Our results revea

The Intergenerational Clock: How Advanced Parental Age Shapes Offspring Biological Aging, Longevity and Neurodegenerative Risk.

BACKGROUND: As delayed parenthood becomes a global demographic norm, understanding the biological consequences of advanced parental age or APrA (≥35 years in mothers; ≥40 years in fathers) is a critical geroscience priority. This shift represents a significant public health challenge, as parental senescence dictates implications far beyond immediate fertility. APrA is a key determinant of offspring healthspan, linked to increased risks of congenital anomalies, premature mortality, neurodevelopmental and metabolic dysfunction, as well as neurodegenerative diseases. This inherited biological leg

Biallelic CYB5A disruptions in 46, XY DSD: Novel missense and deep intronic variants.

BACKGROUND: Cytochrome b5, encoded by CYB5A, is an essential co-factor for CYP17A1-mediated 17,20-lyase activity. CYB5A deficiency is a rare cause of 46,XY DSD. We report two siblings with 46,XY DSD from a Vietnamese family, presenting with phenotypic female. METHODS: Clinical evaluation and hormone profiling were conducted. Whole genome sequencing (WGS) was performed on extracted DNA from both affected siblings and identified variants were classified according to the ACMG/AMP 2015 guidelines. A minigene RNA splicing assay in HEK293T cells was used to evaluate the functional impact of the CYB

Noncoding Variants in Intron 2 of HK1 Associated With Hyperinsulinism With Variable Clinical Phenotype.

CONTEXT: Noncoding variants in hexokinase 1 (HK1) were first associated with congenital hyperinsulinism (HI) in a large family with diazoxide-responsive HI in 2008. Since then, additional cases have been reported in the literature with noncoding variants in HK1 associated with variable HI phenotypes. METHODS: We sequenced a 350 bp region in intron 2 of HK1 in 281 individuals with genetics-negative HI to identify additional cases related to non-coding HK1 variants and to characterize the clinical features of these cases. RESULTS: We identified 16 unique non-coding variants in intron 2 of HK1

Parental Stress and Caregiver Role Modulate Child-Caregiver Prosodic Synchrony in Autism: A Computational Analysis.

Parental stress influences parent-child interactions in typical development and is a prognostic factor of autism outcome. However, we still do not know to what extent parental stress affects parent-child interactions and whether caregiver role matters. This study explored the relationship between parental stress and prosodic synchrony in parent-child vocal interactions, drawing on complex dynamic systems and affective computing frameworks. We assessed 62 dyads (31 autistic preschoolers, interacting separately with their mother and father) during structured play interactions at two time points

Factors Influencing Breastfeeding Knowledge and Attitudes Among Parents of Moderate-To-Late Preterm Infants: A Cross-Sectional Study.

BACKGROUND: Prematurity constitutes a major threat to perinatal health, with moderate-to-late preterm infants accounting for over 80% of all preterm births in China. This places significant challenges on breastfeeding for the affected families. As the optimal enteral nutrition for preterm infants, human milk reduces the incidence of necrotizing enterocolitis and fulfills their growth needs. PURPOSE: The aim of this study was to explore factors associated with breastfeeding knowledge and attitudes among parents of moderate-to-late preterm infants in the context of mother-infant separation. Predicting sub-clinical risk of complex PTSD in adolescents using multi-informant ecological data: A machine learning approach.

BACKGROUND: Complex Post-Traumatic Stress Disorder (CPTSD) is a significant mental health concern in adolescents, however traditional diagnostic tools often fail to capture early risk patterns. Adolescence is a unique developmental window marked by neurobiological remodeling and heightened interpersonal sensitivity. This study aims to move beyond binary clinical diagnosis toward a subclinical prevention framework by predicting continuous CPTSD symptom severity using multidimensional ecological data. METHODS: The data were collected from a triad of 4177 Chinese adolescents and their parents. A

Association of parental prenatal mental health with offspring neurodevelopmental disorders: a systematic review and meta-analysis.

BACKGROUND: Parental prenatal mood and anxiety disorders (PMADs) are linked to child neurodevelopmental disorders (NDDs), but evaluations of the magnitude and mechanisms of this association are limited. This study estimates the strength of the association and whether it is impacted by genetic and environmental factors. METHODS: A systematic search of PubMed, CENTRAL, PsycINFO, OVID, and Google Scholar was performed for articles published from January 1988 to September 2025. Of 2,420 articles screened, 74 met the inclusion criteria. Meta-analyses were conducted on 21 studies, and 53 were inclu

Pulmonary exacerbations in cystic fibrosis after fatherhood with and without CFTR modulators.

BACKGROUND: Advances in cystic fibrosis (CF) management, quality of life, and fertility treatments have encouraged more men with CF (mwCF) to pursue parenthood. However, data on the impact of fatherhood on CF disease course, particularly in the era of CFTR modulators (CFTRm), remain limited. METHODS: We conducted a multicenter retrospective cohort study in six CF centers (2007-2023). Demographic and clinical parameters, including forced expiratory volume in 1 s (FEV1), body mass index (BMI), bacterial colonization, and pulmonary exacerbation (PEx) rates were compared for the year before and t

Investigating the impact of early adversity on perceived support from parents and friends in preadolescence: Do genetic predispositions and timing of exposure matter?

BACKGROUND: Perceived social support contributes to youth resilience, but perceptions of support differ substantially between youth. Genetic predispositions and early life adversity are potential sources for this variation, but research has yet to test gene-environment interplay comprehensively. We examined how early adversity and genetic predispositions for educational attainment, internalizing, and externalizing behaviours uniquely and jointly predict parents and friends support at age 11 and explored associations for different timing of adversity. METHODS: Participants were drawn from TRAI

Wolbachia-induced cytoplasmic incompatibility produces heritable chromatin modifications that suppress position-effect variegation.

Maternally transmitted Wolbachia often cause cytoplasmic incompatibility (CI), a sperm modification that kills host embryos lacking the endosymbiont. CI produces defects in paternal chromosome replication, condensation, and segregation during the first zygotic cell cycle, but a significant fraction of embryos progress normally through this and subsequent cycles and only exhibit defects at later developmental stages. These results, together with documented CI-induced epigenetic chromatin modifications, suggest heritable chromatin modifications are responsible for the developmentally delayed def
