Tremor and Craniofacial Dystonia Preceding Ataxia in SCA43: Expanding the Phenotypic Spectrum of MME-related Ataxia.

Spinocerebellar ataxia type 43 (SCA43) is a rare autosomal dominant ataxia caused by pathogenic variants in the MME gene and typically presents with late-onset gait ataxia and sensory neuropathy. Hyperkinetic movement disorders are uncommon, and prominent craniofacial orobuccolingual dystonia-dominant presentations have not been documented. We report a 63-year-old woman who developed progressive orobuccolingual dystonia and stereotypical choreiform movements of the right hand several years before the onset of gait ataxia. These involuntary movements significantly interfered with feeding and sp

The Bustelo Bolus: Caffeine Withdrawal Mimicking Status Migrainosus in a Patient With Systemic Lupus Erythematosus and Systemic Sclerosis Overlap.

Caffeine is the most widely consumed psychoactive substance in the world, and abrupt cessation in habitual users produces a stereotyped withdrawal syndrome whose dominant feature is headache. Although caffeine withdrawal is a familiar diagnosis in ambulatory and perioperative practice, it is easily overlooked in hospitalized patients on a complex medical service, where the differential for new headache is dominated by autoimmune, infectious, and vascular etiologies. We report a 33-year-old woman with overlap connective tissue disease (systemic lupus erythematosus, mixed connective tissue disea

Implicit Biases and Empathy for Rape Victims: Neural and Behavioural Responses to Facial Cues.

Society's empathic response to a rape victim plays a crucial role in her physical and psychological recovery. However, these responses are often hindered by biases shaped by sociocultural norms and gender stereotypes. Such biases may arise from judgements about a woman's clothing, appearance, place of residence or occupation. This study examines a subtle yet impactful form of discriminatory bias formed from facial images of presumed rape victims. The visual characteristics of the female faces were systematically manipulated to represent (a) cultural or traditional prototypical signature ("Bind

From Awareness to Action: Advancing Counselor Education Policy to Improve Early Detection of Eating Disorders Through Embodied Reflection.

Eating disorders are associated with substantial morbidity and mortality yet remain underidentified in general mental health practice, delaying intervention and worsening outcomes. Counselor preparation often provides limited eating disorder content within broader psychopathology coursework, leaving trainees underprepared to recognize subtle, culturally diverse, and nonstereotypical presentations. This conceptual paper draws on scholarship in counselor education, body image, embodied self-awareness, and eating disorder training to propose a four-domain framework intended to strengthen counselo

Non-allometric expansion and enhanced compartmentalization of Purkinje cell dendrites in the human cerebellum.

Purkinje cell (PC) dendrites are optimized to integrate the vast cerebellar input array and drive the sole cortical output. PCs are classically seen as stereotypical computational units, yet mouse PCs are morphologically diverse and those with multi-branched structure can receive non-canonical climbing fiber (CF) multi-innervation that confers independent compartment-specific signaling. While otherwise uncharacterized, human PCs are universally multi-branched. Do they exceed allometry to achieve enhanced integrative capacities relative to mouse PCs? To answer this, we used several comparative

Purkinje Cell Spatial Correlation Dynamics Are Key to Cerebellar Cortical Contributions to Behavior.

A major challenge in cerebellar physiology is determining how the stereotypic, conserved circuitry of the cerebellar cortex, with its dominant parasagittal and transverse architectures, underlies its fundamental computations and contributions to behavior. Recent advances have allowed for the resolution of Purkinje cell dendritic activity at large scales, but the full roles of these Purkinje cell dynamics during behavior remain undetermined. To interrogate Purkinje cell dynamics at the population level during behavior, we implemented a novel approach for awake, chronic, wide-field Ca2+ imaging

Using personalised brain stimulation to modulate social cognition in adults with autism-spectrum-disorder: protocol for a randomised single-blind rTMS study.

BACKGROUND: Autism spectrum disorder (ASD) is a neurodevelopmental disorder characterized by impairments of social interaction and communication as well as repetitive, stereotyped behaviour. Previous research indicates that ASD without intellectual impairment is associated with underactivity and reduced functional connectivity of the brain's mentalizing pathway, to which the right temporo-parietal junction (rTPJ) serves as an important entry point and hub. In this study, we aim to utilize functional magnetic resonance imaging (fMRI) to localize activation maxima in the rTPJ and other regions i

Celebrating the unseen benefits of surgical careers for mothers in cardiothoracic surgery.

BACKGROUND: While careers in cardiothoracic (CT) surgery can enrich home life, emphasis is often placed on potential negative impacts on surgeons' families, particularly when surgeons are mothers. We aimed to elucidate the beneficial impacts on family dynamics for female cardiothoracic surgeons. METHODS: A survey was designed and disseminated electronically to partners/spouses, coparents, and adult children of female cardiothoracic surgeons. Participants answered questions regarding the impacts of their career on their lives and relationships. Results were analyzed descriptively. RESULTS: A

A transdiagnostic examination of sex- and race and ethnicity-based mental health treatment disparities among publicly insured youth.

BACKGROUND: Low-income, publicly insured youth face numerous barriers to adequate mental health care, which may be compounded for those with multiple marginalized identities. However, no research has examined how identity and diagnosis may interact to shape the treatment experiences of under-resourced youth with psychiatric conditions. Applying an intersectional lens to treatment disparities is essential for developing targeted interventions to promote equitable care. METHODS: Analyses included youth ages 7-18 with eating disorders (EDs; n = 3,311), mood/anxiety disorders (n = 3,219), or psyc

Input/Output Relationships for the Primary Hippocampal Circuit.

The hippocampus is the most studied brain region, but little is known about signal throughput-the simplest, yet most essential of circuit operations-across its multiple stages from perforant path input to CA1 output. Using hippocampal slices derived from male mice, we have found that single-pulse lateral perforant path (LPP) stimulation produces a two-part CA1 response generated by LPP projections to CA3 ("direct path") and the dentate gyrus ("indirect path"). The latter, indirect path was far more potent in driving CA1 but did so only after a lengthy delay. Rather than operating as expected f

L-tyrosine alleviates autism-like behavior in mice by remodeling the gut microbiota.

Autism spectrum disorder (ASD) is characterized by impaired social interaction and repetitive stereotyped behavior, and effective interventions for the core autistic symptoms are currently limited. This study examines the protective role of L-tyrosine in alleviating ASD-like behavioral disorders in a valproic acid (VPA)-induced ASD mouse model and explores the underlying mechanisms via integrated multi-omics. We first investigated the potential of dietary L-tyrosine in mitigating autistic behavior. Subsequently, 16S rRNA sequencing, hippocampal transcriptomics, and neurotransmitter metabolome

Developmental, regenerative, and behavioral dynamics in acoel reproduction.

Acoel worms belong to an enigmatic and understudied animal lineage in the phylum Xenacoelomorpha. Sparse taxonomic and histological work suggests that these worms exhibit a diversity of reproductive anatomies and likely a corresponding diversity in reproductive behavior. Here, we study the reproductive life history of the three-banded panther worm Hofstenia miamia, an acoel that is emerging as a lab-tractable model system. Using confocal microscopy and histology, we describe H. miamia's reproductive organs, identifying structures previously unknown in acoels. Following a cohort of worms from z

Quercetin exerts neuroprotective effects through activation of PI3K/AKT/CREB/BDNF signaling pathway in double blow model-induced schizophrenia-like mice.

OBJECTIVE: Cognitive impairment represents a core clinical feature of schizophrenia, yet effective therapeutic interventions remain lacking. Given quercetin's demonstrated anti-inflammatory and neuroprotective properties, this study investigates its potential to ameliorate cognitive deficits in schizophrenia while elucidating the underlying mechanisms. METHODS: A schizophrenia-like model was established through perinatal injection of MK-801 combined with social isolation. Behavioral assessments evaluated quercetin's antipsychotic potential and its effects on cognitive deficits in this model.

Sleep Mediates the Association Between Emotion Dysregulation and Repetitive Behaviors in Autistic Children.

PURPOSE: Emotion dysregulation (ED) is recognized as a transdiagnostic risk factor for psychopathology, influencing both autistic and non-autistic populations. Among autistic children, ED has been linked to the occurrence of restricted and repetitive behaviors (RRBs), a core feature of autism spectrum disorder. However, the mechanisms underlying this relationship remain poorly understood. Given that sleep problems are common in autistic children and play a critical role in shaping emotional and behavioral functioning, this study investigates whether sleep difficulties mediate the link between

Neurodegenerative Disease Movement Disorders and Dorsal Striatum-Mediated Imbalance of Habitual Motor Sequences.

Goal-directed regulation of locomotor behavior represents an important physiological process essential for the survival of humans and other animals. The striatum constitutes a critical component of the basal ganglia and is primarily responsible for processing external inputs. In rodents, the dorsal striatum is primarily divided into two subregions: the dorsolateral striatum (DLS) and the dorsomedial striatum (DMS), both of which are hypothesized to play distinct roles in regulating goal-directed versus habitual behaviors. Recent studies have demonstrated that different subtypes of medium spiny
