A machine learning approach to predicting high-risk irritability trajectories across the transition to adolescence.

Youth irritability, characterized as proneness to anger/frustration, is associated with negative mental health outcomes. This study identified irritability trajectories across the transition to early adolescence and applied machine learning to identify preadolescent predictors of irritability trajectories. Data are from the Adolescent Brain Cognitive Development (ABCD) study, a large longitudinal study that tracks youth development from preadolescence (n = 11,862, ages 8.92-11.08 years, M = 9.91, SD = 0.62). The baseline sample was 47.83% female (n = 5,680), 63.35% White, 15.74% Black, 12.07%

Dynamic relationship and pleiotropic loci of attention deficit hyperactivity disorder with sleep traits.

Attention deficit hyperactivity disorder (ADHD) is a common neurodevelopmental disorder modulated by genetic and environmental factors. Approximately 20-55% of individuals with ADHD experience concurrent sleep disturbances, yet the dynamic relationship between the two and their shared genetic mechanisms remain unclear. This study analyzed 11,288 participants (52.1% male) from the Adolescent Brain Cognitive Development Study using the Sleep Disturbance Scale for Children. We found a robust positive correlation between ADHD and sleep traits, though ADHD medication weakened this relationship. Lat

A neuroanatomical basis for co-occurring low attention and low reading ability.

Attention deficit hyperactivity disorder (ADHD) and dyslexia are two common disorders that can have devastating impacts on child development and occur together more often than one would expect by chance. Identifying imaging markers characteristic of a combined ADHD and dyslexia phenotype could improve our ability to diagnose these entities early and inform our understanding of the underlying disease processes. In this study, we set out to determine whether there are unique cortical, white matter, or subcortical characteristics in adolescents with combined low attention (LA) and low reading abi

Amygdala subregion volumes and apportionment in preadolescents - Associations with age, sex, and body mass index.

The amygdala, a key limbic structure, is critical to emotional, social, and appetitive behaviors that develop throughout adolescence. Composed of a heterogeneous group of nuclei, questions remain about potential differences in the maturation of its subregions during development. In 3953 9- and 10-year-olds from the Adolescent Brain Cognitive Development℠ Study, the CIT168 Atlas was used to segment nine amygdala subregions. Linear mixed-effects models were used to examine the effects of age, sex, pubertal stage, and body mass index z-score (BMIz) on subregion volumes and their relative apportio

DNA methylation as a possible mechanism linking childhood adversity and health: results from a 2-sample mendelian randomization study.

Childhood adversity is an important risk factor for adverse health across the life course. Epigenetic modifications, such as DNA methylation (DNAm), are a hypothesized mechanism linking adversity to disease susceptibility. Yet, few studies have determined whether adversity-related DNAm alterations are causally related to future health outcomes or if their developmental timing plays a role in these relationships. Here, we used 2-sample mendelian randomization to obtain stronger causal inferences about the association between adversity-associated DNAm loci across development (ie, birth, childhoo

The Monitoring Illicit Substance Use Consortium: A Study Protocol.

OBJECTIVE: The global impact of substance use, including cannabis, amphetamine, cocaine, ecstasy, hallucinogens, and opioids, is increasing, although the overall prevalence is low. Australia and New Zealand are among the few regions of the world in which use (typically illicit) of these classes of substances remains within the top 10 causes of disease burden. The period of adolescence and young adulthood, during which substance use behaviors accelerate in prevalence, is associated with a particular risk for harm. However, the ability to study each substance class has been limited by their low

Effects of different types of induced neonatal inflammation on development and behavior of C57BL/6 and BTBR mice.

Neuroinflammation in the early postnatal period can disturb trajectories of the completion of normal brain development and can lead to mental illnesses, such as depression, anxiety disorders, and personality disorders later in life. In our study, we focused on evaluating short- and long-term effects of neonatal inflammation induced by lipopolysaccharide, poly(I:C), or their combination in female and male C57BL/6 and BTBR mice. We chose the BTBR strain as potentially more susceptible to neonatal inflammation because these mice have behavioral, neuroanatomical, and physiological features of auti

Cognitive processing speed and accuracy are intrinsically different in genetic architecture and brain phenotypes.

Since the birth of cognitive science, researchers have used reaction time and accuracy to measure cognitive ability. Although recognition of these two measures is often based on empirical observations, the underlying consensus is that most cognitive behaviors may be along two fundamental dimensions: cognitive processing speed (CPS) and cognitive processing accuracy (CPA). In this study, we used genomic-wide association studies (GWAS) data from 14 cognitive traits to show the presence of those two factors and revealed the specific neurobiological basis underlying them. We identified that CPS an

Prolonged and specific spatial training during adolescence reverses adult hippocampal network impairments in a mouse model of fragile X syndrome.

The fragile X syndrome (FXS) is the leading monogenetic cause of cognitive impairment and autism. A hallmark of FXS in patients and the FXS mouse model (Fmr1 KO) is an overabundance of immature appearing dendritic spines in the cortex and hippocampus which is associated with behavioral deficits. Spine analysis in the different hippocampal subregions and at different developmental stages revealed that in adult mice, hippocampal spine pathology occurs specifically in the CA3 subregion, which plays a pivotal role in pattern completion processes important for efficient memory recall from parts of

Ambiguous genitalia: clinical management of adult female with male assigned gender: a case report.

BACKGROUND: Disorders of sex development are anomalies in which the development of urogenital ridge is undifferentiated for the male and female child. Imaging plays a vital role in investigating the gross anatomy and associated anomalies. Ultrasonography, such as genitography and magnetic resonance, is the primary modality for demonstrating internal gonads and genitalia. Early multidisciplinary approach in the management of ambiguous genitalia including early surgical intervention is the predominant practice, with few current considerations on deferral of genital reconstruction until adolescen

Characterization of a novel Lbx1 mouse loss of function strain.

Adolescent Idiopathic Scoliosis (AIS) is the most common type of spine deformity affecting 2-3% of the population worldwide. The etiology of this disease is still poorly understood. Several GWAS studies have identified single nucleotide polymorphisms (SNPs) located near the gene LBX1 that is significantly correlated with AIS risk. LBX1 is a transcription factor with roles in myocyte precursor migration, cardiac neural crest specification, and neuronal fate determination in the neural tube. Here, we further investigated the role of LBX1 in the developing spinal cord of mouse embryos using a CRI

A Maternal High-Fat Diet during Early Development Provokes Molecular Changes Related to Autism Spectrum Disorder in the Rat Offspring Brain.

Autism spectrum disorder (ASD) is a disruptive neurodevelopmental disorder manifested by abnormal social interactions, communication, emotional circuits, and repetitive behaviors and is more often diagnosed in boys than in girls. It is postulated that ASD is caused by a complex interaction between genetic and environmental factors. Epigenetics provides a mechanistic link between exposure to an unbalanced maternal diet and persistent modifications in gene expression levels that can lead to phenotype changes in the offspring. To better understand the impact of the early development environment o

Sex differences in the neurochemistry of frontal cortex: Impact of early life stress.

Traumatic events during early life have been linked with later life psychopathology. To understand this risk factor, researchers have studied the effects of prenatal and postnatal early life stress on neurochemical changes. Here we review the rodent literature on sex differences and sex-specific impact of early life stress on frontal cortex neurochemistry. This region is implicated in regulating motivation and emotion, which are often disrupted in psychological disorders. The prefrontal cortex (PFC) in particular is one of the last brain regions to develop, and there are sex differences in the

Gpr126 (Adgrg6) is expressed in cell types known to be exposed to mechanical stimuli.

GPR126 (ADGRG6) is an adhesion G protein-coupled receptor that plays an important role in a variety of tissues/organs, such as heart, sciatic nerve, cartilage, and ear. Moreover, GPR126 (ADGRG6) mutations are associated with human diseases, like adolescent idiopathic scoliosis, lung disease, bladder cancer, and intellectual disability. Despite its clinical importance, it remains elusive how GPR126 is activated and mediates signal transduction and what cellular processes depend on GPR126 signaling. Here, we generated a lacZ reporter mouse line to determine endogenous Gpr126 (Adgrg6) expression

HPV vaccines to prevent cervical cancer and genital warts: an update.

Cervical cancer is an important public health problem worldwide, and especially in developing countries. The link between cervical cancer and oncogenic human papillomavirus (HPV) infection has been clearly established. Furthermore, non-oncogenic HPV are responsible for the majority of genital warts. Two prophylactic HPV vaccines are available, which have the potential of considerably reducing HPV-related morbidity and mortality. Both vaccines are based on virus-like particles of the L1 capsid protein, and are highly efficacious and immunogenic if given before exposure to HPV, i.e. to adolescen
