In vivo brain imaging of tangle burden in humans.

Cerebral neurofibrillary tangles (NFTs) accumulate in a predictable sequence decades before the clinical symptoms of Alzheimer's disease emerge, and the degree of tangle degeneration correlates with the severity of cognitive impairment. A valid in vivo marker of tangle burden, therefore, would be useful for presymptomatic and symptomatic disease detection and treatment monitoring. Recent advances using positron emission tomography (PET) indicate the feasibility of in vivo imaging that provides a combined signal of both neurofibrillary tangles and senile plaques. Such results are encouraging th

The rate of the reaction between CN and C2H2 at interstellar temperatures.

The rate coefficient for the important interstellar reaction between CN and C2H2 has been calculated as a function of temperature between 10 and 300 K. The potential surface for this reaction has been determined through ab initio quantum chemical techniques; the potential exhibits no barrier in the entrance channel but does show a small exit channel barrier, which lies below the energy of reactants. Phase-space calculations for the reaction dynamics, which take the exit channel barrier into account, show the same unusual temperature dependence as determined by experiment, in which the rate coe

Network-based Transcriptomic Profiling of Fetal Astrocyte Differentiation Reveals Therapeutic Targets for Neurodegenerative Disease.

INTRODUCTION: Neurodegenerative disorders, including Alzheimer's Disease (AD), Parkinson's Disease (PD), and Age-related Macular Degeneration (AMD), are marked by the progressive loss of specific neuronal populations. Astrocytes, the glial cells surrounding neurons, play a critical role in maintaining neuronal health by providing neurotrophic support, producing antioxidants, and clearing waste. Dysfunctional astrocytes contribute to disease progression, yet their developmental trajectory and molecular regulation remain incompletely understood. METHOD: This study aims to computationally charac

Evolution, structural diversity and functional plasticity of plant derived multifunctional proteases and α-amylases inhibitors.

Plant derived multifunctional inhibitors (MFIs) are an important class of plant defense proteins that disrupt insect digestion by simultaneously impairing protein and carbohydrate catabolism. With the increasing knowledge of inhibitors of plant enzymes, MFIs have become a separate functional group. In this review we propose an extended definition that encompasses the classical inhibitors acting on structurally unrelated enzymes and also inhibitors acting on enzymes of the same substrate pathway, e.g. α-amylase/α-glucosidase and trypsin/chymotrypsin systems. Most of the MFIs consist of single p

Tau protein in Parkinson's disease: advances in phosphoproteomics and molecular dynamics simulations.

Tau dysregulation, particularly abnormal phosphorylation, is increasingly implicated in Parkinson's disease (PD) and in the pathogenic interaction between Tau and α-synuclein. This review proposes an integrated PD-focused workflow connecting phosphoproteomic discovery, structural modeling, computational drug discovery, and experimental validation. Phosphoproteomic analyses of post-mortem brain tissue, biofluids, and patient-derived dopaminergic neuronal models can identify candidate Tau phosphosignatures associated with vulnerable brain regions, α-synuclein pathology, genetic background, or cl

Targeting epigenetic regulators: In-silico discovery of natural inhibitors against histone demethylase KDM4C.

Cancer is a multifaceted disease driven by genetic mutations and epigenetic dysregulation. Among epigenetic modifiers, histone demethylases like KDM4C (lysine demethylase 4C) play a pivotal role in tumor progression by removing repressive methylation mark at Histone H3K9/H3K36 and altering chromatin structure and gene expression. Overexpression of KDM4C has been implicated in various malignancies, including breast, prostate, colorectal, and hepatocellular carcinomas, hence it is promising drug target. This study employs a structure-based drug discovery strategy to identify natural polyphenolic

Mechanistic insights into pH-dependent ofloxacin adsorption on nanoporous carbons.

The adsorption of large organic molecules such as ofloxacin onto disordered nanoporous carbons presents significant challenges due to the complex interplay of adsorbate-adsorbent charge interactions, molecular dipole moments, pH-dependent speciation, and high electronegativity effects. Understanding these mechanisms is critical for the design of efficient water remediation systems targeting pharmaceutical micropollutants. This study employs a multi-scale modeling approach combining density functional theory (DFT) calculations, Hybrid Reverse Monte Carlo (HRMC) simulations, Grand Canonical Mont

From gene to function: GABA receptor-associated protein (GABARAP) interacts with and potentiates the GABA receptor (RDL) in Megalurothrips usitatus (Bagnall) (Thysanoptera: Thripidae).

The GABA receptor-associated protein (GABARAP), a member of the autophagy-related protein 8 (ATG8) family, regulates autophagy and GABAA receptor trafficking in mammals, yet its functional role in insects remains completely unexplored. The bean flower thrips, Megalurothrips usitatus, is a globally invasive pest whose control is increasingly challenged by widespread insecticide resistance. Understanding novel regulatory mechanisms in its nervous system, particularly those involving the insecticidal target GABA receptor (RDL), is therefore crucial. Spatiotemporal expression profiling revealed th

Integrative transcriptomics and electrophysiological profiling of hiPSC-derived neurons identifies novel druggable pathways in Koolen-de Vries Syndrome.

Koolen-de Vries Syndrome (KdVS) is a neurodevelopmental disorder (NDD) caused by KANSL1 haploinsufficiency with no treatment options. To investigate neuronal network activity in KdVS, human induced pluripotent stem cell (hiPSC)-derived neurons from KdVS patients and controls were cultured on microelectrode arrays (MEAs). KdVS networks exhibited reduced burst rates and increased variability in burst rhythmicity. To bridge molecular and functional aspects of the syndrome, we applied MEA-seq, integrating electrophysiological recordings with high-throughput transcriptome profiling. This analysis r

Simulation-guided design of peptide-metal coordination interfaces for next-generation metallo-immunotherapy.

Metal ions are crucial regulators of immune signaling, metabolism, and redox homeostasis, but their therapeutic deployment in cancer immunotherapy is limited by systemic toxicity and inadequate spatiotemporal control. Peptide–metal coordination interfaces offer a programmable solution by combining sequence-encoded recognition with tunable coordination chemistry, enabling controlled metal speciation, bioavailability, and stimulus-responsive functions in complex biological environments. This review summarizes how biologically relevant ions, including Mn2+, Zn2+, Cu2+, and Fe2+/Fe3+, modulate inn

Biophysical and computational insights into the activation mechanism of α-glucosidase by aromatic organophosphate esters.

Organophosphate esters (OPEs) are emerging environmental pollutants with potential estrogenic activity. However, long-term OPE exposure has also been linked to an increased risk of type 2 diabetes mellitus (T2DM), though the molecular mechanisms remain unclear. α-glucosidase plays a critical role in carbohydrate digestion, making it a key target for T2DM. In this study, two typical aromatic organophosphate esters (AOPEs), triphenyl phosphate (TPHP) and tricresyl phosphate (TCP), were selected to investigate how they affect the structure and function of α-glucosidase. Enzyme activity assays rev

Pulmonary Cytology in Transformation.

BACKGROUND: Traditionally, pulmonary cytology has been used to diagnose and screen for a variety of respiratory diseases, including lung cancer. Owing to its rapid turnaround time compared with histological diagnosis, therapeutic decisions may occasionally need to be made based on cytological findings alone. In the era of precision medicine, however, biomarker assessment and molecular testing have become increasingly important, particularly in the management of lung cancer. In addition, the digitalization of pathological slides has facilitated the development of artificial intelligence (AI)-ba

Streamlined design and production of bioactive variants of recombinant human keratinocyte growth factor to improve acidic pH stability; promising therapeutic candidates for wound treatment.

Keratinocyte Growth Factor (KGF) plays a crucial role in promoting epithelial cell proliferation, making it a valuable candidate for wound healing therapies. However, its instability in acidic environments limits its effectiveness in early-stage wound care. This study addresses the instability of recombinant human Keratinocyte Growth Factor (rhKGF) in acidic environments. Using pH molecular dynamics (pH-MD) simulations, we previously investigated the influence of pH on rhKGF stability and identified positively charged residues as contributors to its instability. Building on these findings, we
