Comparative study of the inductive effect of two psychomoderators--tetrabamate and complexe 1656--and of phenobarbital on liver microsomal enzymes in rats.

Tetrabamate (Atrium) and Complexe 1656, two psychosedative drugs, are complexes formed by phenobarbital with its N-substituted derivatives. We have studied the induction effect of all these compounds on the microsomal liver enzymes in male rats. Tetrabamate and Complexe 1656 were found to be equally effective in increasing the amount of microsomal proteins, cytochrome P-450 and the in vitro aniline p-hydroxylation and amino-pyrine N-demethylation activity. These effects were similar to those obtained after a treatment with an equivalent dose of phenobarbital. Febarbamate and difebarbamate had

Hyperamylasemia in heroin addicts. Characterization by isoamylase analysis.

Hyperamylasemia was noted in 17 (19%) of a group of 91 hospitalized heroin addicts. Thirteen of the 17 were in acute respiratory distress (12 with so-called "heroin lung" syndrome and one with status asthmaticus). Isoamylase analysis in the hyperamylasemic patients demonstrated S-type isoamylase dominance in 15, P-type isoamylase dominance in one and essentially equivalent P- and S-type isoamylase elevations in one. It would appear from these data that the hyperamylasemia after heroin in most persons addicted to the use of this drug arises from the sources other than the pancreas. Changes in t

Cytologic changes following the use of oral contraceptives.

Abnormal cytologic findings in gynecologic smears were more common among patients using oral contraceptives than among control patients. Cytologic examination should be routinely performed before oral contraceptives are prescribed. The exfoliation of endometrial cells was essentially the same between control and oral contraceptive patients. No evidence of excessive stimulation of epithelial element in endometrium accompanied the continued use of hormonal contraceptives. In general, the cytohormonal effect associated with oral contraceptives showed a depressed superficial cell count in the vagi

Structural and mechanistic insights into OCTN1-mediated transport and cancer cell proliferation.

The Novel Organic Cation Transporter 1 (OCTN1) facilitates the uptake of endogenous metabolites and therapeutic drugs across diverse tissues. Recognized as a key transporter for the antioxidant ergothioneine (ET), OCTN1 also mediates the cellular entry and release of various cationic compounds. Dysregulation of OCTN1 has been implicated in cancer progression and drug interaction. In non-neuronal systems, OCTN1-mediated acetylcholine (ACh) transport has been implicated in promoting cancer progression by enhancing tumor cell proliferation and invasion. Despite its physiological and clinical rele

Bilateral Lower Extremity Edema During Donepezil Treatment: A Case Report.

Introduction/ Background: Acute bilateral lower-extremity edema is uncommon and often results from medication side effects, acute heart failure, nephrotic syndrome, or bilateral deep vein thrombosis. Drugs such as dihydropyridine calcium channel blockers, corticosteroids, thiazolidinediones, and nonsteroidal anti-inflammatory agents are known to cause edema. Donepezil, a widely used acetylcholinesterase inhibitor for Alzheimer's disease, has not been previously linked to leg edema. Recognizing rare adverse reactions is essential for patient safety, as edema may indicate serious conditions. Cas

Systematic standardization of organ-on-a-chip under controlled flow conditions: Use case with Caki-1 and A549 cell lines.

New in vitro models are urgently needed to improve the preclinical development of new drugs. Current standardised cellular models are mainly based on 2D cell culture, which lacks physiological flow conditions and often involves limited or simplified co-culture settings. • Organ-on-a-chip (OoC) systems use microchips incorporating extracellular matrices, surface coatings and cell cultures to mimic physiological conditions, enabling the formation of 3D structures, co-culture systems and dynamic flow. However, to perform reproducible and controlled experiments using OoC systems, it is necessary t

Harnessing pyroptosis to restrain melanoma through aripiprazole.

Melanoma exhibits significant resistance to conventional apoptosis-based therapies, underscoring the need for alternative strategies to induce cancer cell death. Pyroptosis is a pro-inflammatory form of programmed cell death characterized by caspase-1 activation, gasdermin D cleavage, and interleukin-1β (IL-1β) release. In this study, transcriptomic analysis revealed that aripiprazole induces robust inflammatory signaling in melanoma cells. The antipsychotic drug aripiprazole was found to trigger pyroptosis in BRAF-mutant melanoma cells, leading to caspase-1 activation, gasdermin D cleavage, a

The ESKAPE Challenge: Understanding Resistance and Exploring Alternative Treatments.

Antimicrobial resistance (AMR) constitutes a critical and escalating global public health challenge, severely limiting the potential of existing antimicrobial drugs and escalating infection-associated morbidity and mortality rates. This analysis focuses on the ESKAPE pathogens (Enterococcus faecium, Staphylococcus aureus, Klebsiella pneumoniae, Acinetobacter baumannii, Pseudomonas aeruginosa, and Enterobacter species), which are prioritized by the World Health Organization (WHO) and represent a significant cause of nosocomial infections due to their extensive drug resistance. Flow cytometry evaluation of cytotoxicity exerted by effector immune cells against tumor cells.

Adoptive cell therapies based on immune effector cells - Natural Killer (NK) cells, Cytotoxic T Lymphocytes (CTL) - represent a promising approach for the treatment of resistant tumors, especially for the management of hematological malignancies. The rapid growth of this field emphasises the need for reliable in vitro methods to evaluate the cytotoxic potential of the cell products developed against the desired target cells, many of which are cells in suspension and consequently pose additional technical challenges. Here, we present a flow-cytometry-based protocol for the quantification of tum

Cell type-selective targeting by heterobifunctional protein binders via in-cell enrichment.

Non-catalytic heterobifunctional protein binders promise to expand the range of therapeutic options by establishing complexes between key target proteins and accessory presenter proteins equipped with additional properties. Here, we systematically investigate the rational design of such molecules, explore the biochemical basis of complex formation and determine how they achieve cellular efficacy using the endogenously expressed immunophilin FKBP12 as presenter protein and the transcriptional regulator BRD4 as target protein. We present classes of bifunctional molecules that enable selective, F

Inhibitors Targeting SARS-CoV-2 Papain-Like Protease: Screening, Design, Synthesis, and Biological Evaluation.

Despite the availability of vaccines and antiviral drugs, SARS-CoV-2 still presents a global health threat, emphasizing the need for new therapeutic targets beyond the main protease and RNA-dependent RNA polymerase. The papain-like protease (PLpro) is a highly conserved and essential viral enzyme that drives polyprotein processing and suppresses host innate immune responses through its deubiquitinating and deISGylating activities. This study employed two approaches to identify new PLpro inhibitors: (i) the design and synthesis of a range of non-covalent PLpro inhibitors based on a lead compoun

Usnic Acid Derivatives as Inhibitors of Mycobacterium tuberculosis Uracil-DNA Glycosylase.

Tuberculosis (TB) remains a global health issue exacerbated by spreading drug resistance and lengthy treatment regimens. Targeting bacterial DNA-repair pathways, particularly those counteracting host-generated genotoxic stress, represents a promising strategy to sensitize Mycobacterium tuberculosis to existing antibiotics. Through structure-based virtual screening of a compound library, we identified novel small-molecule inhibitors of M. tuberculosis uracil-DNA glycosylase (MtbUng), an enzyme essential for the repair of DNA damage inflicted by macrophage-produced reactive nitrogen species. Neoagarohexaose Attenuates Inflammatory and Oxidative Joint Injury in MIA/CIOA Mouse Models of Osteoarthritis.

Osteoarthritis (OA) is a prevalent chronic joint disease lacking disease-modifying drugs. Animal models with distinct pathogenic mechanisms-monosodium iodoacetate (MIA) for metabolic toxicity and collagenase-induced osteoarthritis (CIOA) for matrix degradation-are essential for therapeutic evaluation. In this study, topical application of neoagarohexaose (NA6) at 5 and 10 mg/kg twice daily was assessed in MIA- and CIOA-induced mouse OA models. NA6 at both doses reduced paw swelling, improved serum oxidative stress markers (catalase (CAT), malondialdehyde (MDA), myeloperoxidase (MPO)), ameliora

Candida glabrata and the host: a neglected affair.

Candida glabrata (Nakaseomyces glabratus), a prevalent opportunistic pathogenic yeast of humans, has been classified as a fungal pathogen of high priority by the World Health Organization. C. glabrata is the leading cause of invasive Candida infections in hospitalized and immunocompromised patients and displays co-resistance to azole and echinocandin drugs. C. glabrata is known for its distinct virulence attributes that facilitate infections of the host in the absence of classical virulence factors, viz. filament formation and secreted toxin. The inability of C. glabrata to induce a strong inf

Therapies for bronchiectasis: mechanisms and potential effects.

Bronchiectasis is a complex and heterogeneous disease, and our current understanding of its pathophysiological mechanisms remains in its infancy. Currently, few specifically approved treatments for non-cystic fibrosis bronchiectasis are applied globally, and most guideline-recommended therapies have limited evidence of efficacy. Consequently, the development of new treatment methods is particularly urgent. To gain insight into new drugs under clinical development for adults with bronchiectasis, we conducted a review of the scientific literature and clinical trial registries. In this review, we

[Interpretation of the American Dental Association practice guidelines for the management of acute dental pain with pharmacotherapy (2023-2024) and its clinical implications for China].

Acute toothache is a common dental emergency. In 2023 and 2024, the American Dental Association successively issued evidence‑based clinical practice guidelines for the pharmacologic management of acute dental pain in children, adolescents, adults, and older adults, establishing a new system that prioritizes nonsteroidal anti‑inflammatory drugs, strictly limits opioids, and implements age‑specific precise management. This article systematically interprets the evidence‑based foundation and age‑specific medication regimens of the guidelines. By integrating relevant Chinese diagnostic and treatmen

Inhibition of PfCLK3 interferes with malaria parasite RNA splicing and explains the mechanism of action of a new class of antimalarial candidates.

Emerging resistance to front-line antimalarials means there is an urgent need to discover new drugs with novel mechanisms of action that remain effective across multiple stages of the parasite life cycle. Previously, we reported a malaria parasite protein kinase, PfCLK3, as a target offering a cure, prophylaxis, and transmission blocking. The homology between PfCLK3 and human kinases suggested that the mechanism of parasiticidal activity of PfCLK3 inhibitors is disruption of RNA processing. Here, we use RNA sequencing to reveal that selective PfCLK3 inhibition extensively affects RNA splicing

Exploring the Scope and Status of Biological Interventions in Malaria Control.

Malaria is a major health concern worldwide, with an estimated 282 million infections and 610,000 deaths in 2024. Plasmodium parasites, spread by female Anopheles mosquitoes, are the cause. Several interventions, such as indoor residual spraying, insecticide-treated bed nets, and antimalarial drugs, have reduced transmission; however, challenges such as insecticide resistance, environmental concerns, and climate change have led to the exploration of alternative approaches. To address these limitations, biological control, which involves the use of natural organisms and biological agents to tar
