Microencapsulation of lactase by W/O/W emulsion followed by complex coacervation: Effects of enzyme source, addition of potassium and core to shell ratio on encapsulation efficiency, stability and kinetics of release.

This study evaluated the technological viability of the formation of lactase microcapsules by coacervation (gelatin/gum arabic) containing potassium ions (cofactor). The impacts of the encapsulation and the cofactor on the enzyme properties obtained from Aspergillus oryzae and Kluyveromyces lactis were evaluated as a function of different pH values, temperatures, and storage times. The best microcapsules formed by coacervation showed good functional properties, such as low water activity (≤ 0.4) and particle size (≤ 93.52 μm), as well as high encapsulation efficiency (≥ 98.67%). The potassium

Cryptopain-1, a cysteine protease of Cryptosporidium parvum, does not require the pro-domain for folding.

Cryptosporidium parvum is an intracellular protozoan parasite that causes cryptosporidiosis in mammals including humans. In the current study, the gene encoding the cysteine protease of C. parvum (cryptopain-1) was identified and the biochemical properties of the recombinant enzyme were characterized. Cryptopain-1 shared common structural properties with cathepsin L-like papain family enzymes, but lacked a typical signal peptide sequence and contained a possible transmembrane domain near the amino terminus and a unique insert in the front of the mature domain. The recombinant cryptopain-1 expr

High-level expression and bulk crystallization of recombinant L-methionine gamma-lyase, an anticancer agent.

L-Methionine gamma-lyase is a pyridoxal 5'-phosphate-dependent enzyme which has tumor selective anticancer activity. An efficient production process for the recombinant enzyme was constructed by using the overexpression plasmid in Escherichia coli, large-scale cultivation, and practical crystallization on an industrial scale. The plasmid was optimized with a promoter and the region of the ribosome-binding site. Plasmid pMGLTrc03, which has a trc promoter and a spacing of 12 nucleotides between the Shine-Dalgarno sequence and the ATG translation initiation codon, was selected as the most suitab

[New aspects on prostaglandin D synthases].

Prostaglandin (PG) D2 is a major prostanoid produced in the central nervous system and mast cells, acting as a neuromodulator and an allergic and inflammatory mediator. PGD2 is readily dehydrated to produce PGs of the J series, such as PGJ2, delta 12-PGJ2, and 15-deoxy-delta 12, 14-PGJ2. We identified two distinct types of PGD synthase: one is glutathione independent, the lipocalin-type enzyme; and the other is glutathione-dependent, the hematopoietic enzyme. Lipocalin-type PGD synthase is localized in the central nervous system and genital organs, dominantly produced in the leptomeninges of t

Flavoprotein structure and mechanism. 5. Trypanothione reductase and lipoamide dehydrogenase as targets for a structure-based drug design.

Trypanothione reductase (TR) is a flavoenzyme that has been found only in parasitic protozoa of the order Kinetoplastida. The enzyme catalyzes the NADPH-dependent reduction of glutathionylspermidine conjugates and is a key enzyme of the parasite's thiol metabolism. Consequently, TR is an attractive target molecule for a structure-based drug development against Chagas' disease, African sleeping sickness, and other diseases caused by trypanosomes and leishmanias. The three-dimensional structures of TR and of three enzyme substrate complexes have been solved. Several classes of compounds are disc

Activity of mushroom tyrosinase on catechol and on a catechol estrogen in an organic solvent.

A suspension of tyrosinase-coated glass beads in butanol effectively oxidizes catechol substrate. The enzyme is not soluble in the organic solvent and activity can be stopped by removal of the solid state enzyme after low-speed centrifugation or decantation. The product was assayed by HPLC and by its reactivity towards Besthorn's reagent, which gave a reaction typical for o-quinones. Addition of water to the extent of 0.5 to 4% raised the rate of substrate utilization but the accumulation of quinone first increased and then began to decrease. It is suggested that the product in dry butanol is

How different immobilization techniques affect enzyme performance: a biosensor case study.

The immobilization of enzymes onto support materials is a widely adopted strategy in biosensor development, yet its impact on enzyme activity, stability, and overall performance remains system-dependent. Comparative studies evaluating physical adsorption and covalent binding under identical conditions are scarce. In this work, we systematically investigate the immobilization of glucose oxidase (GOx) and horseradish peroxidase (HRP) onto a hydrophilic polyurethane matrix for the fabrication of a conductometric glucose biosensor. Both immobilization techniques were characterized using thermograv

Proteomic analysis of Viscozyme L and its major enzyme components for pectic substrate degradation.

Enzymatic degradation of plant biomass requires the coordinated action of various enzymes. In this study, the production of reducing sugars from pectic substrates and sugar beet pulp (SBP) was investigated and compared using commercial enzyme preparations, including M2, pectinase (E1), Viscozyme L (V-L) and L-40. V-L, a cellulolytic enzyme mix produced by Aspergillus sp. was further evaluated as the most robust enzyme cocktail with the strongest SBP degradation ability in terms of the release of monosaccharides, methanol, and acetate from SBP. Mass-spectrometry-based proteomics analysis of V-L

Irreversible inactivation of snake venom l-amino acid oxidase by covalent modification during catalysis of l-propargylglycine.

Snake venom l-amino acid oxidase (SV-LAAO, a flavor-enzyme) has attracted considerable attention due to its multifunctional nature, which is manifest in diverse clinical and biological effects such as inhibition of platelet aggregation, induction of cell apoptosis and cytotoxicity against various cells. The majority of these effects are mediated by H2O2 generated during the catalytic conversion of l-amino acids. The substrate analog l-propargylglycine (LPG) irreversibly inhibited the enzyme from Crotalus adamanteus and Crotalus atrox in a dose- and time-dependent manner. Inactivation was irrev

Enzyme replacement therapy in Fabry disease: clinical implications.

PURPOSE OF REVIEW: Fabry disease is an X-linked lysosomal storage disorder caused by a deficiency of the enzyme alpha-galactosidase A. The lack of enzyme activity results in an intracellular accumulation of glycosphingolipids, mainly globotriaosylceramide, in various tissues. Significant morbidity is caused by progressive effects on the vascular endothelium, heart, brain and kidney leading to end-stage renal disease. In this review we would like to give a current overview on recent advances in therapy and an outlook on future aspects in the management of Fabry disease. RECENT FINDINGS: Beside

Purification and properties of a new testosterone 17beta-dehydrogenase (NADP+) from guinea-pig liver.

As a result of studies of guinea-pig live testosterone 17beta-dehydrogenase (NADP+) (EC 1.1.1.64), a new testosterone 17beta-dehydrogenase was discovered. The new enzyme was purified to a single homogeneous protein from the 105 000 g-supernatant fraction of guinea-pig liver by (NH4)2SO4 fractional precipitation and two gel-filtration stages, DEAE-cellulose column chromatography and hydroxyapatite column chromatography. It was characterized by many properties. The enzyme has almost the same properties as the classical testosterone 17beta-dehydrogenase (NADP+) (EC 1.1.1.64), with respect to cofa

Cyclic nucleotide phosphodiesterase from bone: characterization of the enzyme and studies of inhibition by thyroid hormones.

Studies were carried out to characterize the cyclic nucleotide phosphodiesterase from rat calvaria. 25-40% of the total enzyme activity was membrane-bound. pH, magnesium, and temperature requirements conformed closely to those established for phosphodiesterase from other tissues. Kinetic evidence was found for dual enzyme activities with different substrate affinities for both the particulate and soluble enzyme. Apparent Kms for the soluble enzyme (3.5 times 10-6 and 2.5 times 10-5M) approximated those for the particulate enzyme (5.7 times 10-6 and 2.5 times 10-5M). L-Thyroxine, 10-5M, inhibit

[Dehydrogenase activity in mononuclear cells of peripheral blood in infectious mononucleosis in children].

The study of the activity of dehydrogenases in blood cells could be an additional method at diagnostics of etiological variants of infectious mononucleosis (IM) of the viral and bacterial types. The activity of succinatedehydrogenasa (SDG) and of alpha-glycerophosphatedehydrogenase (alpha-GPDG) was increasing in lymphocytes and monocyte of the peripheral blood as compared to the same activity in healthy children. Such changes were preserved in the reconvalescense phase. The activity of enzymes in the lymphoid and monocytic cells was found at viral IM to meet the normal value. The atypical mono

[Use of enzyme test in chemotherapy of patients with cancer of the breast].

The activity of thymidine kinase, thymidine phosphorylase, adenosine deaminase, AMP 5'-nucleotidase was assessed in the serum of healthy females, patients with mastopathia cystica and those with stage IIIB breast cancer. The females age ranged from 23 to 70 years. The activity of the enzymes had significant differences in cancer patients. Minimal thymidine phosphorylase activity was found to suggest fibrous cancer. Changes in the enzymes levels in cancer patients on combined treatment may serve a biochemical test indicating the efficacy of the chemotherapy conducted.

Changes in the acetylcholinesterase activity of the nematode Nippostrongylus brasiliensis following treatment with benzimidazoles in vivo.

Acetylcholinesterase (acetylcholine hydrolase, EC 3.1.1.7) activities of Nippostrongylus brasiliensis were measured following treatment of the hosts with oxfendazole and mebendazole. Drug treatment of N. Brasiliensis caused large, sustained increases in enzyme activity in male and female worms which coincided with the expulsion of the nematodes from the small intestine of the host. It is suggested that the increases in acetylcholinesterase activity may be due to an inhibition by the benzimidazole carbamates of the secretion of enzyme to the exterior of the worms which, in turn, leads to expuls

Studies of beta-glucuronidase in first-trimester placenta, term placenta and hydatidiform mole.

The specific activity and isoenzyme pattern of beta-glucuronidase (beta-D-glucuronide glucurononosohydrolase, EC 3.2.1.31) from first-trimester placenta, term placenta and hydatidiform mole were compared. Enzyme activity is highest in term placenta and lowest in hydatidiform mole, reflecting the steroid conjugation requirements of the tissues. The difference in enzyme activity is due to variation in the level of one isoenzyme form (isoenzyme B) of beta-glucuronidase, which is absent in molar tissue. The possible cause for the changing isoenzyme pattern is discussed.

[Influence of dietary lactose on the development of lactase activity in the digestive tract of the pig].

Lactase activity has been measured in the pancreas, the small and large intestine of 79 pigs varying in age from 6-7 months. The endogenous enzyme level fluctuated along the small intestine but the activity did not vary according to the level of lactose fed in the diet either just after weaning or from 25 to 100 kg of live weight. Measurements performed in the content of the intestine showed that lactose could be digested by the enzymes of the microflora mostly active in the ileum, the caecum and the large intestine.

Lactate dehydrogenase and transaminase activities in the cerebrospinal fluid of protein-energy malnourished children.

The present study is aiming to assess whether there are variations in the activities of the enzymes glutamic-oxalacetic transaminase (GOT) and lactate dehydrogenase (LDH) in the cerebrospinal fluid (CSF) of children suffering from protein-energy malnutrition (PEM). In this respect, serum and CSF activities of GOT and LDH were assayed in thirteen cases suffering from kwashiorkor and ten normal cases serving as controls. Increased activities of both enzymes in sera and CSF of PEM children compared with normals were observed. The significance of these variations was discussed.

Vitamin D -- induced changes in the renal membrane ATPase system.

Ca-dependent and Mg-dependent ATPase activity was found to be depressed significantly in renal membrane preparations from vitamin D-deficient as compared to normal rats. Administratiin of 2000 i.u. vitamin D2 to deficient rats 30 hours before sacrifice restored the membrane enzyme activity to normal. ATP binding by enzymes was also reduced in vitamin D deficiency and raised by repletion. Vitamin D may therefore be required for normal Ca-and Mg-dependent ATPase activity.
